    Aplicación de PCR-RFLP para subtipificar Campylobacter jejuni PCR-RFLP for Campylobacter jejuni subtyping

    Diez cepas de Campylobacter jejuni aisladas de fetos porcinos abortados fueron identificadas por pruebas bioquímicas: 8 como C. jejuni biotipo II de Lior, y 2 como C. jejuni biotipo I. Para poder subtipificarlas se utilizó la técnica de reacción en cadena de la polimerasa (PCR) para amplificar el gen flaA y al producto obtenido se lo digirió con la enzima de restricción DdeI (RFLP). Se pudieron obtener 6 subtipos a partir de C. jejuni biotipo II, mientras que los dos aislamientos de biotipo I correspondieron a un mismo subtipo. Aunque existe una amplia variedad de técnicas de biología molecular que son aplicadas con fines epidemiológicos para Campylobacter, PCR-RFLP, demostró ser una técnica simple y accesible, capaz de subtipificar a C. jejuni.<br>Ten Campylobacter jejuni isolates, 8 identified as C. jejuni biotype II of Lior and 2 as C. jejuni biotipe I, were recovered from aborted pig fetuses. In order to discriminate among strains, restriction fragment length polymorphism (RFLP) using DdeI of polymerase chain reaction (PCR) products of flaA gen was used. C. jejuni biotype II strains could be diferenciated in 6 by PCR-RFLP, and one subtype was obtained from C. jejuni biotype I. Although there is great variability of molecular techniques applied to the Campylobacter epidemiological studies, PCR-RFLP demonstrated to be a simple and accessible technique to discriminate Campylobacter jejuni isolates

    Ecología de comunidades de malezas de arroz (Oryza sativa L.) como aporte hacia una agricultura sustentable

    En este trabajo se analizan desde un enfoque ecológico comunidades de malezas y plantas indicadoras en sitios bajo cultivo de arroz a una escala regional en la provincia de Corrientes. Los sitios se sometieron a un procedimiento de clasificación jerárquica en función de la presencia-ausencia y la abundancia-dominancia de las especies. Se halló una correspondencia clara entre la tipología a priori de hábitats de los sitios y la tipología de la clasificación. La estructura de formas de vida de las comunidades demostró el predominio, en términos de frecuencia de especies, de plantas terrestres sobre palustres y acuáticas, y de perennes sobre anuales. Desde el punto de vista de las plantas indicadoras, especies como Digitaria cilirias, Echinochloa colona y Stapfochloa elata tuvieron una distribución preferente por hábitats de posiciones altas en el paisaje y de hidromorfismo moderado. Por el contrario, especies como Juncus pallescens y Panicum dichotomiflorum mostraron una mayor amplitud ecológica, con afinidad por posiciones topográficas de un rango más amplio, y tolerancia a condiciones de hidromorfismo acentuado y de alto contenido de aluminio en el suelo.Los resultados de este estudio pueden proporcionar pautas para el desarrollo de estrategias de manejo ecológico de malezas, dirigidas a modificar los nichos ocupados por las malezas y someterlas a diferentes condiciones de estrés ambiental. También ponen de relieve la contribución de los humedales arroceros a la preservación de la diversidad vegetal regional

    Remote sensing and the future of landscape ecology

    Landscape ecology focuses on the analysis of spatial pattern and its relationship to ecological processes. As a scientifi c discipline, landscape ecology has grown rapidly in recent years, supported by developments in GIS and spatial analysis techniques. Although remote sensing data are widely employed in landscape ecology research, their current and potential roles have not been evaluated critically. To provide an overview of current practice, 438 research papers published in the journal Landscape Ecology for the years 2004–2008 were examined for information about use of remote sensing. Results indicated that only 36% of studies explicitly mentioned remote sensing. Of those that did so, aerial photographs and Landsat satellite sensor images were most commonly used, accounting for 46% and 42% of studies, respectively. The predominant application of remote sensing data across these studies was for thematic mapping purposes. This suggests that landscape ecologists have been relatively slow to recognize the potential value of recent developments in remote sensing technologies and methods. The review also provided evidence of a frequent lack of key detail in studies recently published in Landscape Ecology, with 75% failing to provide any assessment of uncertainty or error relating to image classifi cation and mapping. It is suggested that the role of remote sensing in landscape ecology might be strengthened by closer collaboration between researchers in the two disciplines, by greater integration of diverse remote sensing data with ecological data, and by increased recognition of the value of remote sensing beyond land-cover mapping and pattern description. This is illustrated by case studies drawn from Latin America (focusing on forest loss and fragmentation) and the UK (focusing on habitat quality for woodland birds). Such approaches might improve the analytical and theoretical rigour of landscape ecology, and be applied usefully to issues of outstanding societal interest, such as the impacts of environmental change on biodiversity and ecosystem services

    A western blotting (WB) procedure has been developed for detecting antibodies to bovine leukosis virus (BLV) in cattle sera. Two hundred and thirty three serum samples from naturally infected cattle with BLV virus and serial bleedings from experimentally BLV infected cows were used. An agar gel immunodiffusion test (AGID) was used for comparing with the results obtained by WB. The AGID positive sera showed a different degree of reactivity by WB test against the two most important viral antigens (gp51 and p24), or against one of them. Other proteins (gp30, p15, p12 and p10) were not detected with any AGID positive sera, being observed occasionally three bands corresponding to the p24 protein. Using sera obtained by BLV experimental inoculation, the antibodies directed to p24 appeared early (between the 2nd and 4th week post inoculation) and thereafter antibodies to gp51were detected in some animals. The analysis of field serum samples by AGID as compared to WB showed an agreement of 90.9%. Only 1.7% of sera were negative by AGID and positive by WB and 7.2% that were not conclusive by AGID and were defined by WB (4.2% as positive and 3.0% as negative).<br>Um sistema de western blotting (WB) foi desenvolvido para detecção de anticorpos contra o vírus da leucose em soros de bovinos. Foram utilizadas amostras de soros de 233 animais naturalmente infectados e soros de vacas experimentalmente infectadas. O teste de imunodifusão em ágar (AGID) foi usado para comparação dos resultados. Graus diferentes de reatividade foram observados em soros positivos ao AGID, quando testados em WB frente a um ou aos dois antígenos mais importantes (gp51 e p24). Outras proteínas (gp30, p15, p12 e p10) não foram detectadas por nenhum soro positivo ao AGID, sendo que três bandas correspondentes à proteína p24 foram observadas ocasionalmente. Em soros obtidos por inoculação experimental, anticorpos contra a proteína p24 foram detectados entre a segunda e a quarta semanas após a inoculação e, em alguns animais, detectaram-se anticorpos anti-gp51 mais tardiamente. O estudo de soros de campo com AGID e WB mostrou concordância de 90,9% sendo que apenas 1,7% dos soros negativos pelo AGID foram positivos ao WB e 7,2% dos resultados não conclusivos por AGID foram definidos por WB (4,2% como positivos e 3% como negativos)

    Repercussion of anthropogenic landscape changes on pedodiversity and preservation of the pedological heritage

    Over a period of time people have lived in and with their surrounding landscapes and for several thousand years transformed the soilscapes and the vegetation into cultural landscape types important for their economy and to meet their needs (Richter 2007, Ellis 2011, Hjelle 2012). The sustainable provision of goods and services depends critically on managing soils without damaging the natural soilscapes and the related natural resources. To support the transition towards sustainable development, science needs to understand how land-use change affects the environment and how this, in turn, feeds back into human livelihood strategies or infl uences the vulnerability of the environment (Rounsevell et al. 2012a). Interactions between decision-making, governance structures, production and consumption, technology, ecosystem services and global environmental change infl uence human activities at the local and regional scale, and are infl uenced by and feed back to the global scale, thereby shaping trajectories of human–environment interaction in land systems (Lambin and Meyfroidt 2011)

    Effects of serelaxin in patients with acute heart failure

    BACKGROUND Serelaxin is a recombinant form of human relaxin-2, a vasodilator hormone that contributes to cardiovascular and renal adaptations during pregnancy. Previous studies have suggested that treatment with serelaxin may result in relief of symptoms and in better outcomes in patients with acute heart failure. METHODS In this multicenter, double-blind, placebo-controlled, event-driven trial, we enrolled patients who were hospitalized for acute heart failure and had dyspnea, vascular congestion on chest radiography, increased plasma concentrations of natriuretic peptides, mild-to-moderate renal insufficiency, and a systolic blood pressure of at least 125 mm Hg, and we randomly assigned them within 16 hours after presentation to receive either a 48-hour intravenous infusion of serelaxin (30 μg per kilogram of body weight per day) or placebo, in addition to standard care. The two primary end points were death from cardiovascular causes at 180 days and worsening heart failure at 5 days. RESULTS A total of 6545 patients were included in the intention-to-treat analysis. At day 180, death from cardiovascular causes had occurred in 285 of the 3274 patients (8.7%) in the serelaxin group and in 290 of the 3271 patients (8.9%) in the placebo group (hazard ratio, 0.98; 95% confidence interval [CI], 0.83 to 1.15; P=0.77). At day 5, worsening heart failure had occurred in 227 patients (6.9%) in the serelaxin group and in 252 (7.7%) in the placebo group (hazard ratio, 0.89; 95% CI, 0.75 to 1.07; P=0.19). There were no significant differences between the groups in the incidence of death from any cause at 180 days, the incidence of death from cardiovascular causes or rehospitalization for heart failure or renal failure at 180 days, or the length of the index hospital stay. The incidence of adverse events was similar in the two groups. CONCLUSIONS In this trial involving patients who were hospitalized for acute heart failure, an infusion of serelaxin did not result in a lower incidence of death from cardiovascular causes at 180 days or worsening heart failure at 5 days than placebo.     Predictors for anastomotic leak, postoperative complications, and mortality after right colectomy for cancer: Results from an international snapshot audit

    Background: A right hemicolectomy is among the most commonly performed operations for colon cancer, but modern high-quality, multination data addressing the morbidity and mortality rates are lacking. Objective: This study reports the morbidity and mortality rates for right-sided colon cancer and identifies predictors for unfavorable short-term outcome after right hemicolectomy. Design: This was a snapshot observational prospective study. Setting: The study was conducted as a multicenter international study. Patients: The 2015 European Society of Coloproctology snapshot study was a prospective multicenter international series that included all patients undergoing elective or emergency right hemicolectomy or ileocecal resection over a 2-month period in early 2015. This is a subanalysis of the colon cancer cohort of patients. Main Outcome Measures: Predictors for anastomotic leak and 30-day postoperative morbidity and mortality were assessed using multivariable mixed-effect logistic regression models after variables selection with the Lasso method. Results: Of the 2515 included patients, an anastomosis was performed in 97.2% (n = 2444), handsewn in 38.5% (n = 940) and stapled in 61.5% (n = 1504) cases. The overall anastomotic leak rate was 7.4% (180/2444), 30-day morbidity was 38.0% (n = 956), and mortality was 2.6% (n = 66). Patients with anastomotic leak had a significantly increased mortality rate (10.6% vs 1.6% no-leak patients; p 65 0.001). At multivariable analysis the following variables were associated with anastomotic leak: longer duration of surgery (OR = 1.007 per min; p = 0.0037), open approach (OR = 1.9; p = 0.0037), and stapled anastomosis (OR = 1.5; p = 0.041). Limitations: This is an observational study, and therefore selection bias could be present. For this reason, a multivariable logistic regression model was performed, trying to correct possible confounding factors. Conclusions: Anastomotic leak after oncologic right hemicolectomy is a frequent complication, and it is associated with increased mortality.
