    The influence of COMT Val158Met genotype on the character dimension cooperativeness in healthy females

    Objectives: Although the Val(158)Met catechol-O-methyltransferase (COMT) gene has been linked with the temperament dimension Novelty Seeking (NS), new insights in this polymorphism might point to a major role for character features as well. Given that individual life experiences may influence Val(158) and Met(158) allele carriers differently it has been suggested that the character trait cooperativeness could be implicated. Case report: A homogeneous group of eighty right-handed Caucasian healthy female university students were assessed with the TCI and genotyped for the COMT Val(158)Met polymorphism (rs4680). Gene determination showed that eighteen were Val(158) homozygotes, forty-four Val/Met(158) heterozygotes, and eighteen were Met(158) homozygotes. All were within the same age range and never documented to have suffered from any neuropsychiatric illness. Bonferroni corrected non-parametric analyses showed that only for the character scale cooperativeness Val(158) homozygotes displayed significant higher scores when compared to Met(158) homozygotes. No significant differences on cooperativeness scores were found between Val(158) and Val/Met(158) carriers or between Met(158) and Val/Met(158) carriers. No differences were observed for the COMT Val(158) Met polymorphism and the other temperament and character scales. Conclusions: Our findings support the assumption that the Val(158)Met single nucleotide polymorphism (SNP) influences character traits and not only temperament. Our results add to the notion that Val(158) homozygotes are considered to be helpful and empathic and it suggest that these cooperativeness character traits are related to the dopaminergic system

    Quantum thermodynamics of a charged magneto-oscillator coupled to a heat bath

    Explicit results for various quantum thermodynamic function (QTF) of a charged magneto-oscillator coupled to a heat bath at arbitrary temperature are demonstrated in this paper. Discernible expressions for different QTF in the two limits of very low and very high temperatures are presented for three popular heat bath models : Ohmic, single relaxation time and blackbody radiation. The central result is that the effect of magnetic field turns out to be important at low temperatures yet crucial at high temperatures. It is observed that the dissipation parameter, Îł\gamma, and the cyclotron frequency, ωc\omega_c, affect the decaying or rising behaviour of various QTF in just the opposite way to each other at low temperatures.     The effects of PTSD treatment during pregnancy: systematic review and case study

    Background: PTSD in pregnant women is associated with adverse outcomes for mothers and their children. It is unknown whether pregnant women with PTSD, or symptoms of PTSD, can receive targeted treatment that is safe and effective. Objective: The purpose of the present paper was to assess the effectiveness and safety of treatment for (symptoms of) PTSD in pregnant women. Method: A systematic review was conducted in accordance with the PRISMA guidelines in Pubmed, Embase, PsychINFO, and Cochrane. In addition, a case is presented of a pregnant woman with PTSD who received eye-movement desensitization and reprocessing (EMDR) therapy aimed at processing the memories of a previous distressing childbirth. Results: In total, 13 studies were included, involving eight types of interventions (i.e. trauma-focused cognitive behavioural therapy, exposure therapy, EMDR therapy, interpersonal psychotherapy, explorative therapy, self-hypnosis and relaxation, Survivor Moms Companion, and Seeking Safety Intervention). In three studies, the traumatic event pertained to a previous childbirth. Five studies reported obstetrical outcomes. After requesting additional information, authors of five studies indicated an absence of serious adverse events. PTSD symptoms improved in 10 studies. However, most studies carried a high risk of bias. In our case study, a pregnant woman with a PTSD diagnosis based on DSM-5 no longer fulfilled the criteria of PTSD after three sessions of EMDR therapy. She had an uncomplicated pregnancy and delivery. Conclusion: Despite the fact that case studies as the one presented here report no adverse events, and treatment is likely safe, due to the poor methodological quality of most studies it is impossible to allow inferences on the effects of any particular treatment of PTSD (symptoms) during pregnancy. Yet, given the elevated maternal stress and cortisol levels in pregnant women with PTSD, and the fact that so far no adverse effects on the unborn child have been reported associated with the application of trauma-focused therapy, treatment of PTSD during pregnancy is most likely safe

    Ten steps or climbing a mountain: A study of Australian health professionals' perceptions of implementing the baby friendly health initiative to protect, promote and support breastfeeding

    Background: The Baby Friendly Hospital (Health) Initiative (BFHI) is a global initiative aimed at protecting, promoting and supporting breastfeeding and is based on the ten steps to successful breastfeeding. Worldwide, over 20,000 health facilities have attained BFHI accreditation but only 77 Australian hospitals (approximately 23%) have received accreditation. Few studies have investigated the factors that facilitate or hinder implementation of BFHI but it is acknowledged this is a major undertaking requiring strategic planning and change management throughout an institution. This paper examines the perceptions of BFHI held by midwives and nurses working in one Area Health Service in NSW, Australia. Methods: The study used an interpretive, qualitative approach. A total of 132 health professionals, working across four maternity units, two neonatal intensive care units and related community services, participated in 10 focus groups. Data were analysed using thematic analysis. Results: Three main themes were identified: ‘Belief and Commitment’; ‘Interpreting BFHI’ and ‘Climbing a Mountain’. Participants considered the BFHI implementation a high priority; an essential set of practices that would have positive benefits for babies and mothers both locally and globally as well as for health professionals. It was considered achievable but would take commitment and hard work to overcome the numerous challenges including a number of organisational constraints. There were, however, differing interpretations of what was required to attain BFHI accreditation with the potential that misinterpretation could hinder implementation. A model described by Greenhalgh and colleagues on adoption of innovation is drawn on to interpret the findings. Conclusion: Despite strong support for BFHI, the principles of this global strategy are interpreted differently by health professionals and further education and accurate information is required. It may be that the current processes used to disseminate and implement BFHI need to be reviewed. The findings suggest that there is a contradiction between the broad philosophical stance and best practice approach of this global strategy and the tendency for health professionals to focus on the ten steps as a set of tasks or a checklist to be accomplished. The perceived procedural approach to implementation may be contributing to lower rates of breastfeeding continuation

    Epigenetic regulation in neonatal ECFCs following intrauterine exposure to gestational diabetes

    poster abstractGestational diabetes (GDM) complicates up to 10% of pregnancies. In addition to acute risks, the children of diabetic mothers have an increased risk of obesity, diabetes, and hypertension, starting in childhood. While the causes of this increased risk are unknown, previous studies in our lab have identified functional deficits in endothelial colony forming cells (ECFCs) isolated from the cord blood of GDM pregnancies. This study focused on identifying genes that have altered epigenetic modifications that result in abnormal mRNA and protein expression in ECFCs from the cord blood GDM pregnancies. The objective of this study was to identify mRNA expression and DNA methylation alterations in ECFCs that may help identify the causes of ECFC dysfunction following intrauterine exposure to GDM. ECFCs were obtained from control and GDM pregnancies. DNA, RNA, and protein samples were isolated in parallel from ECFCs. RNA microarray analysis using the Affymetrix Human 1.0 Gene Array was used to identify gene expression alterations in GDM ECFCs compared to control ECFCs. Genome-wide DNA methylation was assessed using an Infinium 450K Methylation Array for DNA samples at >450,000 CpG sites. Correlation analysis was performed to identify possible sites that have altered CpG methylation and RNA expression. RNA expression results were validated using qRT-PCR and western blotting. Bisulfite sequencing of genomic DNA from the ECFCs was performed to identify additional sites with altered methylation for regions not included in the DNA methylation array. Of the 28,000 genetic loci tested, 596 mRNAs were altered between control and GDM ECFCs (p<0.01). More stringent criteria identified 38 genes for further investigation by limiting analysis to genes that exhibited increased or decreased expression by at least 50%, with a p<0.01. PLAC8 was identified as being increased 5-fold by microarray analysis, a result which was confirmed in two cohorts by qRT-PCR and western blotting. Analysis of the methylation array and bisulfite sequencing results revealed 3 regions surrounding the transcriptional start site of PLAC8 gene whose CpG methylation negatively correlate with RNA expression in samples from control and GDM ECFCs. In contrast, a CpG island is fully unmethylated in both control and GDM ECFCs. The discovery of CpG sites whose methylation correlates with PLAC8 mRNA expression in ECFCs is consistent with the hypothesis that intrauterine exposure to GDM results in epigenetic changes. Analysis of methylation at this site could be used as a biomarker for children of mothers with GDM who may be at risk for disease later in life. Using bisulfite pyrosequencing, we are currently developing assays to quickly determine if methylation of the PLAC8 putative promoter region is altered in cord blood mononuclear cells obtained from GDM or healthy control pregnancies. We are also investigating the role of methylation in regulating PLAC8 RNA expression, determining if there is altered histone modifications and transcription factor binding in these regions, and examining other genes that may comprise a molecular signature of ECFC dysfunction

    The magnetic storms of 3_4 August 2010 and 5_6 August 2011: 1. Ground- and space-based observations

    We have used total electron content (TEC) values from low, middle, and high latitudes recorded over the American continent and density and ion temperature measured in situ by the DMSP-F15 and F17 satellites during the geomagnetic storms of 3_4 August 2010 and 5_6 August 2011 to study the formation and dynamics of plasma density enhancements that developed during these two storms. Common to both storms are the timing of the main phase that extends between 20 and 24 UT and their seasonality with both storms occurring near the end of the Northern Hemisphere summer solstice. During both storms, TEC data show incipient equatorial anomalies lacking a poleward expansion beyond 20Á magnetic latitude. Two large-scale TEC enhancements were observed at middle latitudes showing a complicated pattern of structuring and merging. The first TEC enhancement corresponds to a storm-enhanced density (SED) seen between 21 and 01 UT on the following day. The second TEC enhancement was observed over Central America, located equatorward of the SED and apparently moving northward. However, careful analysis of the TEC values indicates that this second TEC enhancement is not transported from lower latitudes through a superfountain effect. Instead, the enhanced plasma has a local origin and is driven by a southward directed meridional wind that moves plasma up the tilted magnetic field lines. DMSP flights passing over the second TEC enhancement show a reduction of the ion temperature, confirming an adiabatic expansion of the plasma as it moves up the field lines. It is concluded that the midlatitude TEC enhancements do not arise from a low-latitude ionospheric fountain effect. ©2017. American Geophysical Union. All Rights Reserved

    Iron-chalcogenide FeSe0.5_{0.5}Te0.5_{0.5} coated superconducting tapes for high field applications

    The high upper critical field characteristic of the recently discovered iron-based superconducting chalcogenides opens the possibility of developing a new type of non-oxide high-field superconducting wires. In this work, we utilize a buffered metal template on which we grow a textured FeSe0.5_{0.5}Te0.5_{0.5} layer, an approach developed originally for high temperature superconducting coated conductors. These tapes carry high critical current densities (>1×104\times10^{4}A/cm2^{2}) at about 4.2K under magnetic field as high as 25 T, which are nearly isotropic to the field direction. This demonstrates a very promising future for iron chalcogenides for high field applications at liquid helium temperatures. Flux pinning force analysis indicates a point defect pinning mechanism, creating prospects for a straightforward approach to conductor optimization.Comment: Accepted for publication in Appl. Phys.     Temporal reproduction and its neuroanatomical correlates in adults with attention deficit hyperactivity disorder and their unaffected first-degree relatives

    Background: Little is known about time perception, its putative role as cognitive endophenotype, and its neuroanatomical underpinnings in adults with attention deficit hyperactivity disorder (ADHD). Method: Twenty adults with ADHD, 20 unaffected first-degree relatives and 20 typically developing controls matched for age and gender undertook structural magnetic resonance imaging scans. Voxel-based morphometry with DARTEL was performed to obtain regional grey-matter volumes. Temporal processing was investigated as a putative cognitive endophenotype using a temporal reproduction paradigm. General linear modelling was employed to examine the relationship between temporal reproduction performances and grey-matter volumes. Results: ADHD participants were impaired in temporal reproduction and unaffected first-degree relatives performed in between their ADHD probands and typically developing controls. Increased grey-matter volume in the cerebellum was associated with poorer temporal reproduction performance. Conclusions: Adults with ADHD are impaired in time reproduction. Performances of the unaffected first-degree relatives are in between ADHD relatives and controls, suggesting that time reproduction might be a cognitive endophenotype for adult ADHD. The cerebellum is involved in time reproduction and might play a role in driving time performances

    Habitat Use by Middle Mississippi River Pallid Sturgeon

    Little is known about the habitat preferences and needs of pallid sturgeon Scaphirhynchus albus, which was federally listed as endangered in 1990. To learn more about habitat use and selection by pallid sturgeon, sonic transmitters were surgically implanted in 27 individuals from the middle Mississippi River. Study fish were located 184 times (1–23 times/individual) from November 1995 to December 1999. Of the seven macrohabitats identified, pallid sturgeon were found most often in main-channel habitats (39% of all relocations) and main-channel border habitats (26%); the between-wing-dam habitats were used less often (14%). Strauss’s linear selectivity index (Li) values indicated that study fish exhibited positive selection for the main-channel border, downstream island tips, between-wing-dam, and wing-dam-tip habitats; they showed negative selection for main-channel, downstream of wing dams, and upstream of wing dam habitats. Comparison of Li values for four temperature ranges and three daily mean discharge ranges revealed little change in habitat selection due to temperature or discharge. Habitat use patterns also were similar across seasons and discharge regimes, except during spring months when between-wingdam habitats saw greater use and main-channel and main-channel border habitat use declined. These changes may have been a response to high river stages associated with spring flooding, which may create favorable feeding areas in the between-wing-dam habitats. Enhancement and restoration of habitat diversity, particularly downstream island tip and between-wing-dam habitats, may be necessary for the recovery of pallid sturgeon in the middle Mississippi River
