In the past the market economy was dominated by the local and regional design, characterized for strong links to the customs, traditions and natural resources from eachregion. As the consequence of the technological advances and the growing needs of consumption,at present, products are manufactured based on economical, functional and marketingcriterions, which means that their design is very offset from the regional and local signs.Between the challenges that characterize the actual economy, one highlights the existenceof more market niches, product variations and shorter life cycles. To survive in this economy, marked by the competition and the globalisation, one search to imply in product developingprocess, the design, engineering, marketing and production department, that composes aknown industrial methodology called concurrent or simultaneous engineering. In this article, one intends to exhibit the applications and advantages of the newtechnologies, like Rapid Prototyping (RP), that are valuable tools of the concurrentengineering and to analyse the specific implications in the design history and teaching.RP is a family of modern technologies that generate three-dimensional solid objects undercomputer control. Besides its designation be reported to the first major application that isindustrial prototyping, the RP equipments also allow the rapid tooling and the directmanufacture of small series or single products.Nowadays, the commercialisation of RP equipments, termed concept modellers or 3Dprintersis in great expansion. Although these machines use a very reduced class of cheapmaterials with poor mechanical characteristics, they allow the designers to verify and quicklytest the ideas that they are developing during the creative process. The designer can realize inplenitude the iterative process of creation without the traditional economical and materialslimitations, being enough to send the CAD file directly to the RP equipment placed in aoffice, like an usual inkjet printer. So it is possible to test new ideas with better accuracybefore its concretisation, avoiding misunderstandings and delays and inciting the positivecriticism through the involvement of the technical staff from all the related departments.Particularity, faced to the challenges due to the globalisation, the traditional and regionalindustries will find in these technologies the means that they need to ensure its own futuresurviving. This presentation is divided in the following sections:- Innovation loop in the past and in the present. The iterative process.- New technologies available to the design and the designers: Rapid Prototyping andInternet.- Design Pedagogy: new challenges in the formation of future generations of designers.- Conclusions and future trends

In the current situation of a constant increase in the atmospheric CO2 concentration, there is a potential risk of decreased nutritional value and food crop quality. Therefore, selecting strong-responsive varieties to elevated CO2 (eCO2) conditions in terms of yield and nutritional quality is an important decision for improving crop productivity under future CO2 conditions. Using bean and soybean varieties of contrasting responses to eCO2 and different origins, we assessed the effects of eCO2 (800 ppm) in a controlled environment on the yield performance and the concentration of protein, fat, and mineral elements in seeds. The range of seed yield responses to eCO2 was −11.0 to 32.7% (average change of 5%) in beans and −23.8 to 39.6% (average change of 7.1%) in soybeans. There was a significant correlation between seed yield enhancement and aboveground biomass, seed number, and pod number per plant. At maturity, eCO2 increased seed protein concentration in beans, while it did not affect soybean. Lipid concentration was not affected by eCO2 in either legume species. Compared with ambient CO2 (aCO2), the concentrations of manganese (Mn), iron (Fe), and potassium (K) decreased significantly, magnesium (Mg) increased, while zinc (Zn), phosphorus (P), and calcium (Ca) were not changed under eCO2 in bean seeds. However, in soybean, Mn and K concentrations decreased significantly, Ca increased, and Zn, Fe, P, and Mg concentrations were not significantly affected by eCO2 conditions. Our results suggest that intraspecific variation in seed yield improvement and reduced sensitivity to mineral losses might be suitable parameters for breeders to begin selecting lines that maximize yield and nutrition under eCO2.info:eu-repo/semantics/publishedVersio

The APOE ε2 allele increases the risk of Earlier Age at onset in Machado-Joseph disease

Background. Machado-Joseph disease (MJD) is an autosomal dominant neurodegenerative disorder of late onset, caused by a (CAG)n expansion at the ATXN3 gene (14q32.1). Variation in age-at-onset is partially explained by the size of the (CAG)n tract in expanded alleles. The remaining variation should be the product of other factors, namely modifier genes. The genotype at the APOE locus has been described as a possible modifier in different neurological disorders, namely Parkinson (PD) and Huntington disease (HD). In the CNS, apolipoprotein E constitutes an important mediator of cholesterol transport/metabolism, which is essential for synaptic integrity and neuronal function. Objective. To investigate a modulating effect of the APOE polymorphism on age-at-onset of MJD. Design and Subjects. The APOE polymorphism was typed in a series of 192 MJD patients. Results. Cases with the ε2/ε3 genotype presented an earlier onset, when compared with those with ε3/ε3 or ε3/ε4. In this series of patients, the presence of an APOE ε2 allele implies a decrease of nearly 5 years in the age-at-onset. When combining, in a general linear model, several other predictors, namely the presence/absence of the APOE ε2 allele, with the size of the (CAG)n in expanded alleles, the model was significantly improved and the explanation of onset variance was raised from 59.8% to 66.5%. Furthermore, the presence of the ε2 allele was associated with an onset below 39 years (OR=5.00; 95% CI: 1.18-21.14). Conclusions. These findings indicate that the polymorphism at the APOE gene plays a role as a genetic modifier of MJD phenotype.Fundação para a Ciência e a Tecnologia (FCT) - SFRH/BPD/63121/2009, SFRH/BPD/38659/2007, M3.1.3/F/004/2009, “Secretaria Regional da Ciência, Tecnologia e Equipamentos”.Fundação para a Ciência e a Tecnologia (FCT) - “Transcriptional variation of the ATXN3 gene as modulator of the clinical heterogeneity in Machado-Joseph disease (MJD)” (PIC/IC/83074/2007)Institute of Biotechnology and Biomedicine (IBBA) - “High prevalence diseases in the Azores Islands” (M2.1.2/I/026/2008

Botanical gardens have long contributed to plant science and have played a leading role in ex situ conservation, namely of threatened tree species. Focusing on the three botanical gardens of Lisbon (i.e., Botanical Garden of Ajuda—JBA, Lisbon Botanical Garden—JBL, and Tropical Botanical Garden—JBT), this study aims to reveal their natural heritage and to understand the historical motivations for their creation. Our results showed that these gardens contain a total of 2551 tree specimens, corresponding to 462 taxa, within 80 plant families. Of these, 85 taxa are found in the three gardens, and more than half of the taxa are hosted in JBL (334 taxa), whereas 230 and 201 taxa were recorded in JBT and JBA, respectively. The motivations for the creation of each garden are reflected in the different geographic origins of the trees they host in their living collections. The Palearctic species are dominant in JBA and JBL, and Tropical trees prevail in JBT. With more than 250 years of history, these gardens hold an invaluable natural and historical heritage, with their living collections providing valuable sources of information for the conservation of threatened plant species, at local and global scalesinfo:eu-repo/semantics/publishedVersio

Questions require a thoughtful answer and can play diverse educational roles, such as stimulating the learning of new knowledge, as is the case in Problem-Based Learning (PBL) contexts. In this context, the student is placed at the center of the teaching and learning processes and plays an active role in the learning of new knowledge. Bearing in mind the influence that textbooks exert on teaching practices, it is worthwhile questioning to which extent questions included in school science textbooks promote a PBL oriented teaching approach. This paper describes the development and validation of a checklist for analyzing the questions included in science textbooks. After introducing the checklist, an example of its application to a science unit assigned to two junior high school science courses (8th grade) is given, approaching the two disciplines in the area

Genetically determined Amerindian ancestry correlates with increased frequency of risk alleles for systemic lupus erythematosus

Objective To assess whether genetically determined Amerindian ancestry predicts increased presence of risk alleles of known susceptibility genes for systemic lupus erythematosus (SLE). Methods Single-nucleotide polymorphisms (SNPs) within 16 confirmed genetic susceptibility loci for SLE were genotyped in a set of 804 Mestizo lupus patients and 667 Mestizo healthy controls. In addition, 347 admixture informative markers were genotyped. Individual ancestry proportions were determined using STRUCTURE. Association analysis was performed using PLINK, and correlation between ancestry and the presence of risk alleles was analyzed using linear regression. Results A meta-analysis of the genetic association of the 16 SNPs across populations showed that TNFSF4 , STAT4 , ITGAM , and IRF5 were associated with lupus in a Hispanic Mestizo cohort enriched for European and Amerindian ancestry. In addition, 2 SNPs within the major histocompatibility complex region, previously shown to be associated in a genome-wide association study in Europeans, were also associated in Mestizos. Using linear regression, we predicted an average increase of 2.34 risk alleles when comparing an SLE patient with 100% Amerindian ancestry versus an SLE patient with 0% Amerindian ancestry ( P < 0.0001). SLE patients with 43% more Amerindian ancestry were predicted to carry 1 additional risk allele. Conclusion Our results demonstrate that Amerindian ancestry is associated with an increased number of risk alleles for SLE.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/78480/1/27753_ftp.pd

Sequence analysis of 5' regulatory regions of the Machado-Joseph Disease gene (ATXN3)

    Get PDF
Machado–Joseph disease (MJD) is a late-onset autosomal dominant neurodegenerative disorder, which is caused by a coding (CAG)n expansion in the ATXN3 gene (14q32.1). The number of CAG repeats in the expanded alleles accounts only for 50 to 75 % of onset variance, the remaining variation being dependent on other factors. Differential allelic expression of ATXN3 could contribute to the explanation of different ages at onset in patients displaying similar CAG repeat sizes. Variation in 5′ regulatory regions of the ATXN3 gene may have the potential to influence expression levels and, ultimately, modulate the MJD phenotype. The main goal of this work was to analyze the extent of sequence variation upstream of the ATXN3 start codon. A fragment containing the core promoter and the 5′ untranslated region (UTR) was sequenced and analyzed in 186 patients and 59 controls (490 chromosomes). In the core promoter, no polymorphisms were observed. In the 5′ UTR, only one SNP (rs3814834) was found, but no improvements on the explanation of onset variance were observed, when adding its allelic state in a linear model. Accordingly, in silico analysis predicted that this SNP lays in a nonconserved position for CMYB binding. Therefore, no functional effect could be predicted for this variant.Institute of Biotechnology and Biomedicine of the Azores - “High Prevalence Diseases in the Azores Islands” M2.1.2/I/026/2008,Fundação para a Ciência e a Tecnologia (FCT) - “Transcriptional variation of the ATXN3 gene as modulator of the clinical heterogeneity in Machado–Joseph disease (MJD)Secretaria Regional da Ciência, Tecnologia e Equipamentos - M3.1.3/F/004/2009CNP
