    Analysis of four DLX homeobox genes in autistic probands

    BACKGROUND: Linkage studies in autism have identified susceptibility loci on chromosomes 2q and 7q, regions containing the DLX1/2 and DLX5/6 bigene clusters. The DLX genes encode homeodomain transcription factors that control craniofacial patterning and differentiation and survival of forebrain inhibitory neurons. We investigated the role that sequence variants in DLX genes play in autism by in-depth resequencing of these genes in 161 autism probands from the AGRE collection. RESULTS: Sequencing of exons, exon/intron boundaries and known enhancers of DLX1, 2, 5 and 6 identified several nonsynonymous variants in DLX2 and DLX5 and a variant in a DLX5/6intragenic enhancer. The nonsynonymous variants were detected in 4 of 95 families from which samples were sequenced. Two of these four SNPs were not observed in 378 undiagnosed samples from North American populations, while the remaining 2 were seen in one sample each. CONCLUSION: Segregation of these variants in pedigrees did not generally support a contribution to autism susceptibility by these genes, although functional analyses may provide insight into the biological understanding of these important proteins

    Effects of the whole seed and a protein isolate of faba bean (Vicia faba) on the cholesterol metabolism of hypercholesterolaemic rats

    The aim of the present work was to analyse the hypocholesterolaemic efficiency of a Vicia fabaprotein isolate in relation to the intact legume. In addition, the mechanisms underlying the effects of this isolate were investigated. Hypercholesterolaemic rats were divided into three groups n10 3 and fed high-fat diets rich in cholesterol-containing casein, whole seeds of Vicia faba or the protein isolate of faba beans as protein source, for 2 weeks ad libitum. The protein isolate was prepared by isoelectric precipitation and spray dried. Analyses of serum, liver and faeces, as well as of the activity of hepatic 3-hydroxy-3-methylglutaryl (HMG)-CoA reductase, were assessed by enzymatic methods. The rats fed on Vicia faba diets showed significantly lower body weights and energy intakes than rats fed on casein diets. The wholeseed diet induced a significant reduction in plasma triacylglycerol. Feeding rats on diets containing faba bean seeds, or the protein isolate, induced a significant decrease in plasma (LDL+VLDL)-cholesterol but not in HDL-cholesterol. Hepatic cholesterol and triacylglycerol were also reduced. The hypocholesterolaemic effects of Vicia faba were not the result of a reduction in cholesterol synthesis as assessed from HMG-CoA reductase activity, but the result of an increase in steroid faecal excretion. The faba bean-protein isolate obtained under our experimental conditions was useful in improving the metabolic alterations induced by feeding with a hypercholesterolaemic diet compared with casein.     A genome-wide association study for blood lipid phenotypes in the Framingham Heart Study

    <p>Abstract</p> <p>Background</p> <p>Blood lipid levels including low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), and triglycerides (TG) are highly heritable. Genome-wide association is a promising approach to map genetic loci related to these heritable phenotypes.</p> <p>Methods</p> <p>In 1087 Framingham Heart Study Offspring cohort participants (mean age 47 years, 52% women), we conducted genome-wide analyses (Affymetrix 100K GeneChip) for fasting blood lipid traits. Total cholesterol, HDL-C, and TG were measured by standard enzymatic methods and LDL-C was calculated using the Friedewald formula. The long-term averages of up to seven measurements of LDL-C, HDL-C, and TG over a ~30 year span were the primary phenotypes. We used generalized estimating equations (GEE), family-based association tests (FBAT) and variance components linkage to investigate the relationships between SNPs (on autosomes, with minor allele frequency ≥10%, genotypic call rate ≥80%, and Hardy-Weinberg equilibrium p ≥ 0.001) and multivariable-adjusted residuals. We pursued a three-stage replication strategy of the GEE association results with 287 SNPs (P < 0.001 in Stage I) tested in Stage II (n ~1450 individuals) and 40 SNPs (P < 0.001 in joint analysis of Stages I and II) tested in Stage III (n~6650 individuals).</p> <p>Results</p> <p>Long-term averages of LDL-C, HDL-C, and TG were highly heritable (h<sup>2 </sup>= 0.66, 0.69, 0.58, respectively; each P < 0.0001). Of 70,987 tests for each of the phenotypes, two SNPs had p < 10<sup>-5 </sup>in GEE results for LDL-C, four for HDL-C, and one for TG. For each multivariable-adjusted phenotype, the number of SNPs with association p < 10<sup>-4 </sup>ranged from 13 to 18 and with p < 10<sup>-3</sup>, from 94 to 149. Some results confirmed previously reported associations with candidate genes including variation in the lipoprotein lipase gene (<it>LPL</it>) and HDL-C and TG (rs7007797; P = 0.0005 for HDL-C and 0.002 for TG). The full set of GEE, FBAT and linkage results are posted at the <b>d</b>ata<b>b</b>ase of <b>G</b>enotype <b>a</b>nd <b>P</b>henotype (dbGaP). After three stages of replication, there was no convincing statistical evidence for association (i.e., combined P < 10<sup>-5 </sup>across all three stages) between any of the tested SNPs and lipid phenotypes.</p> <p>Conclusion</p> <p>Using a 100K genome-wide scan, we have generated a set of putative associations for common sequence variants and lipid phenotypes. Validation of selected hypotheses in additional samples did not identify any new loci underlying variability in blood lipids. Lack of replication may be due to inadequate statistical power to detect modest quantitative trait locus effects (i.e., <1% of trait variance explained) or reduced genomic coverage of the 100K array. GWAS in FHS using a denser genome-wide genotyping platform and a better-powered replication strategy may identify novel loci underlying blood lipids.</p

    The metallicity gradient as a tracer of history and structure : the Magellanic Clouds and M33 galaxies

The stellar metallicity and its gradient place constraints on the formation and evolution of galaxies. Aims. This is a study of the metallicity gradient of the LMC, SMC and M33 galaxies derived from their asymptotic giant branch (AGB) stars. Methods. The [Fe/H] abundance was derived from the ratio between C- and M-type AGB stars and its variation analysed as a function of galactocentric distance. Galaxy structure parameters were adopted from the literature. Results. The metallicity of the LMC decreases linearly as −0.047±0.003 dex kpc−1 out to ∼8 kpc from the centre. In the SMC, [Fe/H] has a constant value of ∼−1.25 ± 0.01 dex up to ∼12 kpc. The gradient of the M33 disc, until ∼9 kpc, is −0.078 ± 0.003 dex kpc−1 while the outer disc/halo, out to ∼25 kpc, has [Fe/H] ∼ −1.7 dex. Conclusions. The metallicity of the LMC, as traced by different populations, bears the signature of two major star forming episodes: the first one constituting a thick disc/halo population and the second one a thin disc and bar due to a close encounter with the Milky Way and SMC. The [Fe/H] of the recent episode supports an LMC origin for the Stream. The metallicity of the SMC supports star formation, ∼3 Gyr ago, as triggered by LMC interaction and sustained by the bar in the outer region of the galaxy. The SMC [Fe/H] agrees with the present-day abundance in the Bridge and shows no significant gradient. The metallicity of M33 supports an “insideout” disc formation via accretion of metal poor gas from the interstellar medium.Peer reviewe

    In-situ trace metal (Cd, Pb, Cu) speciation along the Po River plume (Northern Adriatic Sea) using submersible systems

    Highlights • In-situ trace metal speciation in the Adriatic Sea by submersible voltammetric sensors • Metal species distribution mainly affected by Po River outflow. • Metal dynamic concentrations below legal limits • Cu dynamic concentrations toxic to sensitive phytoplankton Abstract Information on the distribution and speciation of trace metals is of critical importance for our ability to interpret the links between the bioavailability and uptake of an element, and its biogeochemical cycle in coastal environments. Within the framework of the European Project “In-situ automated Monitoring of Trace metal speciation in Estuaries and Coastal zones in relation with the biogeochemical processes (IMTEC)”, the chemical speciation of Cd, Pb and Cu was carried out along the Po River plume in the period 27 October – 2 November 2002. During the cruise, five Voltammetric In-situ Profiling systems and one Multi Physical Chemical Profiler, as well as conventional voltammetric instruments, were successfully applied in order to evaluate the distribution of Cd, Pb and Cu between different fractions (free ion, dynamic, colloidal, dissolved and particulate fractions) and to assess the evolution of these fractions during estuarine mixing and in the water column. Dynamic concentrations were 0.05–0.2 nmol L−1 Cd, 0.02–0.2 nmol L−1 Pb, and 0.15–4.0 nmol L−1Cu. Cd was mainly present as dynamic fraction (40–100% of the dissolved Cd). High proportions of Pb (~70%) and Cu (~80%) were present as colloids probably of biogenic origin. Principal components analysis reveals a strong influence of the Po River discharge on the spatial and vertical distributions of metal species. Almost all the metal fractions globally decreased following the salinity gradient. Metal concentrations are far below (at least one order of magnitude lower) the Environmental Quality Standard established by the Italian law. However, the Cu dynamic fraction showed concentrations likely to be toxic to sensitive phytoplankton community and to have negative effects on larva development of coastal macroinvertebrate species (toxicity data extracted from literature)

    Integrated water vapor obtained by satellite-borne instruments: evaluation with GPS measurements in the Iberian Peninsula

Los instrumentos satelitales son: Global Ozone Monitoring Instrument (GOME-2), Moderate-Resolution Imaging Spectroradiometer (MODIS), Ozone Monitoring Instrument (OMI), Spining Enhanced Visible and InfraRed Imager (SEVIRI), Atmospheric Infrared Sounder (AIRS), y Scanning Imaging Absorption Spectrometer for Atmospheric Chartography (SCIAMACHY). Los productos de estos satélites muestran una buena correlación con respecto al producto de GPS (R2 ~ 0.7). Todos los satélites muestran cierta tendencia a sobrestimar los valores bajos de vapor de agua y a subestimar los altos. Además, la precisión, medida mediante el rango intercuartílico (IQR) también disminuye rápidamente al aumentar el IWV. Por otro lado, otro factor importante es el ángulo solar zenital (SZA) que influye en el rendimiento de los instrumentos satelitales, especialmente aquellos que dependen de la radiación solar. Al aumentar el SZA acercándose a 90°, los instrumentos pierden rendimiento aumentando la sobrestimación y el IQR. Sin embargo, a valores de SZA mayores de 90° (esto es, la noche) los índices calculados no tienen grandes dependencias con el SZA.[EN]This work focuses in the comparison of several integrated water vapor (IWV) from several satellites with respect to a dataset from ground-based GPS IWV, in nine stations at the Iberian Peninsula. The satellite instruments are Global Ozone Monitoring Instrument (GOME-2), Moderate-Resolution Imaging Spectroradiometer (MODIS), Ozone Monitoring Instrument (OMI), Spinning Enhanced Visible and InfraRed Imager (SEVIRI), Atmospheric Infrared Sounder (AIRS), and Scanning Imaging Absorption Spectrometer for Atmospheric Chartography (SCIAMACHY). The products of this instruments show a fair correlation with respect to GPS product (R2 ~ 0.7). All satellites show a certain tendency to overestimation of low IWV values, while underestimating large IWV values. Moreover, the precision is studied using the inter-quartile range (IQR), which also decreases quickly when IWV increases. Another important factor is the solar zenith angle (SZA), which affects the performance of satellite instruments, especially those that are dependent on solar radiation. When SZA increases, going closer to 90°, the instruments had worse performance, increasing overestimation and IQR. Nevertheless, for SZA larger than 90° (that is to say, the night), the indexes do not show large dependencies on SZA
