    Medical rescuers’ occupational health during COVID-19 : contribution of coping and emotion regulation on burnout, trauma and post-traumatic growth

    The COVID-19 pandemic places unique challenges to medical rescuers’ occupational health. Thus, it is crucial to assess its direct and indirect impacts on key psychological outcomes and adaptation strategies. This study aims to analyse the impact of this pandemic on medical rescuers’ coping and emotion regulation strategies, and their levels of work-related psychological outcomes, such as burnout, trauma and post-traumatic growth. Additionally, it aims to analyse the contribution of coping and emotion regulation strategies, employed to manage the COVID-19 pandemic, on burnout, trauma and post-traumatic growth. A sample of 111 medical rescuers answered the Brief Cope, Emotion Regulation Questionnaire, Oldenburg Burnout Inventory, Impact of Event Scale-Revised and Post-Traumatic Growth Inventory. Medical rescuers resorted moderately to coping and emotion regulation strategies, since the beginning of COVID-19. They presented moderate burnout and post-traumatic growth and low trauma. Coping presented a higher weight on burnout, trauma and post-traumatic growth, than emotion regulation. Expressive suppression and dysfunctional coping predicted burnout and trauma, and problem and emotion-focused coping predicted post-traumatic growth. Dysfunctional coping mediated and, thus, exacerbated the effect of expressive suppression on burnout and on trauma. Practitioners should pay closer attention to professionals with higher burnout and trauma. Occupational practices should focus on reducing dysfunctional coping and expressive suppression and promoting problem-focused coping.Resumo: A pandemia COVID-19 coloca desafios únicos à saúde ocupacional dos profissionais do pré-hospitalar. Assim, é fundamental avaliar o seu impacto psicológico, para sintomatologia direta ou indiretamente relacionada com a COVID-19. Pretende-se analisar o impacto desta pandemia nos outcomes psicológicos laborais, como burnout, trauma e crescimento pós-traumático, e nas estratégias de coping e regulação emocional. Pretende-se também analisar o contributo do coping e regulação emocional para estes outcomes. Uma amostra de 111 profissionais de emergência médica pré-hospitalar responderam ao Brief Cope, Emotion Regulation Questionnaire, Oldenburg Burnout Inventory, Impact of Event Scale-Revised e Post-Traumatic Growth Inventory. Os profissionais recorreram moderadamente a estratégias de coping e regulação emocional, desde o início da pandemia. Reportaram moderado burnout e crescimento pós-traumático e baixo trauma. O coping apresentou maior peso para os outcomes. A supressão emocional e o coping disfuncional contribuíram para o burnout e trauma, e coping problema e emoção contribuíram para o crescimento pós-traumático. O coping disfuncional mediou e exarcebou o efeito da supressão emocional no burnout e trauma. Assim, poderá ser dada especial atenção a profissionais com elevado burnout e trauma e as práticas poderão ser focadas na redução do coping disfuncional e supressão emocional, bem como na promoção do coping problema.info:eu-repo/semantics/publishedVersio

    Robust perpendicular exchange coupling in an ultrathin CoO/PtFe double layer: strain and spin orientation

The cobalt oxide growth by reactive molecular beam epitaxy on a Pt-terminated PtFe/Pt(001) surface gives rise to a hexagonal surface and a monoclinic distorted CoO 3 nm film at room temperature. This distorted ultrathin CoO layer couples with the PtFe(001) layer establishing a robust perpendicular exchange bias shift. Soft x-ray absorption spectroscopy provides a full description of the spin orientations in the CoO/PtFe double layer. The exchange bias shift is preserved up to the Néel antiferromagnetic ordering temperature of TN = 293 K.     Cloning approach and functional analysis of anti-intimin single-chain variable fragment (scFv)

    <p>Abstract</p> <p>Background</p> <p>Intimin is an important virulence factor involved in the pathogenesis of enteropathogenic <it>Escherichia coli </it>(EPEC) and enterohemorrhagic <it>Escherichia coli </it>(EHEC). Both pathogens are still important causes of diarrhea in children and adults in many developing and industrialized countries. Considering the fact that antibodies are important tools in the detection of various pathogens, an anti-intimin IgG2b monoclonal antibody was previously raised in immunized mice with the conserved sequence of the intimin molecule (int<sub>388-667</sub>). In immunoblotting assays, this monoclonal antibody showed excellent specificity. Despite good performance, the monoclonal antibody failed to detect some EPEC and EHEC isolates harboring variant amino acids within the 338-667 regions of intimin molecules. Consequently, motivated by its use for diagnosis purposes, in this study we aimed to the cloning and expression of the single-chain variable fragment from this monoclonal antibody (scFv).</p> <p>Findings</p> <p>Anti-intimin hybridoma mRNA was extracted and reversely transcripted to cDNA, and the light and heavy chains of the variable fragment of the antibody were amplified using commercial primers. The amplified chains were cloned into <it>pGEM-T Easy </it>vector. Specific primers were designed and used in an amplification and chain linkage strategy, obtaining the scFv, which in turn was cloned into pAE vector. <it>E. coli </it>BL21(DE3)pLys strain was transformed with pAE scFv-intimin plasmid and subjected to induction of protein expression. Anti-intimin scFv, expressed as inclusion bodies (insoluble fraction), was denatured, purified and submitted to refolding. The protein yield was 1 mg protein per 100 mL of bacterial culture. To test the functionality of the scFv, ELISA and immunofluorescence assays were performed, showing that 275 ng of scFv reacted with 2 mg of purified intimin, resulting in an absorbance of 0.75 at 492 nm. The immunofluorescence assay showed a strong reactivity with EPEC E2348/69.</p> <p>Conclusion</p> <p>This study demonstrated that the recombinant anti-intimin antibody obtained is able to recognize the conserved region of intimin (Int<sub>388-667</sub>) in purified form and the EPEC isolate.</p

    Latex peptidases of Calotropis procera for dehairing of leather as an alternative to environmentally toxic sodium sulfide treatment

    Dehairing of crude leather is a critical stage performed at the beginning of its processing to obtain industrially useful pieces. Tanneries traditionally apply a chemical process based on sodium sulfide. Since this chemical reactive is environmentally toxic and inefficiently recycled, innovative protocols for reducing or eliminating its use in leather depilation are welcomed. Therefore, latex peptidases from Calotropis procera (CpLP) and Cryptostegia grandiflora (CgLP) were assayed for this purpose. Enzyme activity on substrates representative of skin such as hide powder azure (UHPA), elastin (UE), azocollagen (UAZOCOL), keratin (UK), and epidermis (UEP) was determined, while depilation activity was assayed on cow hide. Only CpLP was active against keratin (13.4 UK) and only CgLP was active against elastin (0.12 UE). CpLP (93.0 UHPA, 403.6 UAZOCOL, 36.3 UEP) showed higher activity against the other substrates than CgLP (47.6 UHPA, 261.5 UAZOCOL, 8.5 UEP). In pilot assays, CpLP (0.05% w/v with sodium sulfite 0.6% w/v as activator) released hairs from cow hide pieces. Macroscopic and microscopic analyses of the hide revealed that the dehairing process was complete and the leather structure was preserved.     Actividades corporales de aventura en la escuela: la carrera de orientación como propuesta en el desarrollo de habilidades socioemocionales

    Os esportes de aventura quando tematizados nas aulas de Educação Física têm o potencial de despertar fortes emoções, principalmente pelas experiências de risco e aventura, no contexto escolar, a Base Nacional Comum Curricular-BNCC, existe há necessidade de se trabalhar a educação emocional. O objetivo deste estudo foi analisar o comportamento socioemocional dos alunos diante de uma intervenção de corrida de orientação. 39 escolares do ensino médio participaram do estudo. Foi aplicado o questionário Big Five Inventory (BFI) após uma intervenção de corrida de orientação para a avaliação do perfil socioemocional. Foi constado que as maiores médias entre os perfis foram: Abertura (34,59), amabilidade (30,44), conscienciosidade (28,23) (tabela 1). A corrida de orientação, se mostra eficaz no desempenho emocional dos escolares pois os perfis de maior as maiores medias foram para os perfis de: Abertura, amabilidade e conscienciosidade. Está intervenção também atende aos requisitos presentes no BNCC sobre o desenvolvimento emocional como também sobre a necessidade de práticas de aventura para a Educação Física.Adventure sports when thematized in Physical Education classes have the potential to arouse strong emotions, mainly through risk and adventure experiences, in the school context, the National Common Curricular Base-BNCC, there is a need to work on emotional education. The objective of this study was to analyze the socio-emotional behavior of students in the face of an orienteering race intervention. 39 high school students participated in the study. The Big Five Inventory (BFI) questionnaire was applied after an orienteering race intervention to assess the socio-emotional profile. It was found that the highest averages among the profiles were: Openness (34.59), kindness (30.44), conscientiousness (28.23) (Table 1). The orienteering race proves to be effective in the emotional performance of the students because the profiles of the highest the highest averages were for the profiles of: Openness, kindness and conscientiousness. This intervention also meets the requirements present in the BNCC on emotional development as well as on the need for adventure practices for Physical Education.Los deportes de aventura cuando son tematizados en las clases de Educación Física tienen el potencial de despertar emociones fuertes, principalmente a través de experiencias de riesgo y aventura, en el contexto escolar, Base Curricular Común NacionalBNCC, existe la necesidad de trabajar la educación emocional. El objetivo de este estudio fue analizar el comportamiento socioemocional de los estudiantes ante una intervención de carrera de orientación. 39 estudiantes de secundaria participaron en el estudio. Se aplicó el cuestionario Big Five Inventory (BFI) después de una intervención de carrera de orientación para evaluar el perfil socioemocional. Se encontró que los promedios más altos entre los perfiles fueron: Apertura (34,59), amabilidad (30,44), escrupulosidad (28,23) (Tabla 1). La carrera de orientación demuestra ser efectiva en el desempeño emocional de los estudiantes debido a que los perfiles de los más altos los promedios más altos fueron para los perfiles de: Apertura, amabilidad y escrupulosidad. Esta intervención también cumple con los requisitos presentes en el BNCC sobre el desarrollo emocional, así como sobre la necesidad de prácticas de aventura para la Educación Física.Facultad de Humanidades y Ciencias de la Educació
