    An integrated biochemical system for nitrate assimilation and nitric oxide detoxification in Bradyrhizobium japonicum

    Rhizobia are recognized to establish N(2)-fixing symbiotic interactions with legume plants. Bradyrhizobium japonicum, the symbiont of soybeans, can denitrify and grow under free-living conditions with nitrate (NO(3)(−)) or nitrite (NO(2)(−)) as sole nitrogen source. Unlike related bacteria that assimilate NO(3)(−), genes encoding the assimilatory NO(3)(−) reductase (nasC) and NO(2)(−) reductase (nirA) in B. japonicum are located at distinct chromosomal loci. The nasC gene is located with genes encoding an ABC-type NO(3)(−) transporter, a major facilitator family NO(3)(−)/NO(2)(−) transporter (NarK), flavoprotein (Flp) and single-domain haemoglobin (termed Bjgb). However, nirA clusters with genes for a NO(3)(−)/NO(2)(−)-responsive regulator (NasS-NasT). In the present study, we demonstrate NasC and NirA are both key for NO(3)(−) assimilation and that growth with NO(3)(−), but not NO(2)(−) requires flp, implying Flp may function as electron donor to NasC. In addition, bjgb and flp encode a nitric oxide (NO) detoxification system that functions to mitigate cytotoxic NO formed as a by-product of NO(3)(−) assimilation. Additional experiments reveal NasT is required for NO(3)(−)-responsive expression of the narK-bjgb-flp-nasC transcriptional unit and the nirA gene and that NasS is also involved in the regulatory control of this novel bipartite assimilatory NO(3)(−)/NO(2)(−) reductase pathway

    Efficacy of biosecurity measures in the control of microorganisms associated to endometritis in sows. Preliminary study

    Biosecurity can be defined as all the applied measurements that take as a target to minimize the sanitary risks in a stock farm, and include measurements related to the facilities and the management. The efficacy of these measurements must be reflected in a decrease of the microorganism in different productive phases. A study was carried out to evaluate if the set of applied measurements influences the microbial uterine contamination after farrowing of healthy sows. Two swine farms were been completed about biosecurity measurements was completed and a microbiological study of uterine swabs of sows after the farrowing was carried out. A total of 60 animals were studied, and 27 (45%, 95% CI [33.3%, 56.7%]) resulted positive. Significant differences between production and selection and multiplication farms were detected (OR = 3.44, IC 95%, 1.135-11.047). The colonization frequency was 65% CI [51.3%, 78.6%] and 35% CI [21%, 49%] in production and selection farm, respectively (P = 0.02). A total of 66 isolates were obtained, represented mainly by Staphylococcus spp. (33.33%) and Aerococcus spp. (27.27%), although other species included in the genus Streptococcus (9.09%), Enterococcus (6.06%) and Pseudomonas (4.55%), as well as different fungi species were also isolated. The frequency of isolation of different microorganisms was similar in both farms, with the exception of the genus Enterococcus that was not isolated in the production farm (P = 0.01). The questionnaire showed some differences in biosecurity measures in the selection and multiplication farm when it is compared to the production farm, which together with the increased uterine microbial contamination observed in the latter leads us to propose a preliminary hypothesis about the possible risk factors associated with this process, highlighting the absence of measures to avoid the presence of vectors and the establishment of strict protocols for cleaning and disinfectio

    Distribution of Foxp3+ T cells in the liver and hepatic lymph nodes of goats and sheep experimentally infected with Fasciola hepatica

    Foxp3 regulatory T cells (Tregs) are now considered to play a key role in modulation of immune responses during parasitic helminth infections. Immunomodulation is a key factor in Fasciola hepatica infection; however, the distribution and role of Foxp3+ Tregs cells have not been investigated in F. hepatica infected ruminants. The aim of this study was to evaluate the presence of Foxp3+ Tregs in the liver and hepatic lymph nodes from experimentally infected sheep and goats during acute and chronic stages of infection. Three groups of goats (n=6) and three groups of sheep (n=6) were used in this study. Goats in groups 1-2 and sheep in groups 4-5 were orally infected with metacercarie of ovine origin. Groups 1 and 4 were killed during the acute stage of the infection, at nine days post infection (dpi); groups 2 and 5 were killed during the chronic stage, at 15 and19 weeks post infection respectively (wpi). Groups 3 (goats) and 6 (sheep) were left as uninfected controls. Fluke burdens and liver damage were assessed and the avidin-biotin-complex method was used for the immunohistochemical study. At nine dpi in acute hepatic lesions, the number of both Foxp3+ and CD3+ T lymphocytes increased significantly in goats and sheep. In the chronic stages of infection (15-19wpi), the number of Foxp3+ and CD3+ T lymphocytes were also significantly increased with respect to control livers, particularly in portal spaces with severely enlarged bile ducts (response to adult flukes) while the increase was lower in granulomas, chronic tracts and smaller portal spaces (response to tissue damage). Foxp3+ Tregs were increased in the cortex of hepatic lymph nodes of sheep (chronic infection) and goats (acute and chronic infection). The estimated proportion of T cells which were Foxp3+ was significantly increased in the large bile ducts and hepatic lymph node cortex of chronically infected goats but not sheep. This first report of the expansion of Foxp3+ Tregs in acute and chronic hepatic lesions in ruminants suggests that these cells may be involved in both parasite survival and modulation of hepatic damage.     Analysis of the Influence of Terrain Orientation on the Design of PV Facilities with Single-Axis Trackers

    This paper investigates how to optimally orient the photovoltaic solar trackers of an axis parallel to the terrain, applying the sky model of Hay–Davies. This problem has been widely studied. However, the number of studies that consider the orientation (inclination and azimuth of the terrain) is very limited. This paper provides an examination of incident solar irradiance that can be extended to terrain with variable orientation and in consideration of different azimuths of the axis of rotation. Furthermore, a case study of the south of Spain is provided, considering different inclination and orientation terrain values. The results obtained in this study indicate, as a novelty, that for lands that are not south facing, the rotation axis azimuth of solar trackers should be different from zero and adjusted to the same direction as the land azimuth in order to maximize energy production. Annual energy production is sensitive to changes in the rotation axis azimuths of solar trackers (an influence of around 3% of annual energy production)
