Citrus psorosis virus movement protein contains an aspartic protease required for autocleavage and the formation of tubule-like structures at plasmodesmata

Plant virus cell-to-cell movement is an essential step in viral infections. This process is facilitated by specific virus-encoded movement proteins (MPs), which manipulate the cell wall channels between neighboring cells known as plasmodesmata (PD). Citrus psorosis virus (CPsV) infection in sweet orange involves the formation of tubule-like structures within PD, suggesting that CPsV belongs to "tubuleforming" viruses that encode MPs able to assemble a hollow tubule extending between cells to allow virus movement. Consistent with this hypothesis, we show that the MP of CPsV (MPCPsV) indeed forms tubule-like structures at PD upon transient expression in Nicotiana benthamiana leaves. Tubule formation by MPCPsV depends on its cleavage capacity, mediated by a specific aspartic protease motif present in its primary sequence. A single amino acid mutation in this motif abolishes MPCPsV cleavage, alters the subcellular localization of the protein, and negatively affects its activity in facilitating virus movement. The amino-terminal 34-kDa cleavage product (34KCPsV), but not the 20-kDa fragment (20KCPsV), supports virus movement. Moreover, similar to tubule-forming MPs of other viruses, MPCPsV (and also the 34KCPsV cleavage product) can homooligomerize, interact with PD-located protein 1 (PDLP1), and assemble tubule-like structures at PD by a mechanism dependent on the secretory pathway. 20KCPsV retains the protease activity and is able to cleave a cleavage-deficient MPCPsV in trans. Altogether, these results demonstrate that CPsV movement depends on the autolytic cleavage of MPCPsV by an aspartic protease activity, which removes the 20KCPsV protease and thereby releases the 34KCPsV protein for PDLP1-dependent tubule formation at PD. IMPORTANCE Infection by citrus psorosis virus (CPsV) involves a self-cleaving aspartic protease activity within the viral movement protein (MP), which results in the production of two peptides, termed 34KCPsV and 20KCPsV, that carry the MP and viral protease activities, respectively. The underlying protease motif within the MP is also found in the MPs of other members of the Aspiviridae family, suggesting that protease-mediated protein processing represents a conserved mechanism of protein expression in this virus family. The results also demonstrate that CPsV and potentially other ophioviruses move by a tubule-guided mechanism. Although several viruses from different genera were shown to use this mechanism for cell-to-cell movement, our results also demonstrate that this mechanism is controlled by posttranslational protein cleavage. Moreover, given that tubule formation and virus movement could be inhibited by a mutation in the protease motif, targeting the protease activity for inactivation could represent an important approach for ophiovirus control.Fil: Robles Luna, Gabriel. Narratives of Loss: Exploring Grief through Photography

Grief is not an exclusively private and intrapsychic phenomenon but a dynamic process whereby the bereaved negotiates the meaning of the loss in a way that may challenge his/her personal self-narrative. Drawing on a social constructionist model of grief, this paper features a case study where we analyse narratives of mourning elicited through a personal photographic project. The visual-based narrative methodology used in our study (photo-production) not only allows for multi-modal forms of expression and communication in the study of grief, but it also serves as an aid for individuals to scaffold further meaning-making processes which cannot be conveyed through narrative alone. The paper concludes with an invitation to reflect on the use of photography as both a possible methodological tool to explore narratives of grief and a therapeutic tool for the construction of meanings and continuing bonds with the deceased

Optimizar la mano de obra administrada en Ternium Siderar desde la génesis

Este proyecto se elaboró en el marco de la experiencia realizada en la Práctica Estudiantil de Verano que otorga la Organización Techint en sus distintas empresas. En este caso el trabajo se llevó a cabo en Ternium Siderar, estando en el sector de logística de Mano de Obra Administrada en la Gerencia de Mantenimiento. Es un trabajo de aplicación directa, ya que las soluciones se plantearon y quedaron en funcionamiento; para dar con el resultado, los problemas que se tenían en el sector fueron propuestos por el tutor de la Práctica, y las investigaciones se realizaron para dar con la solución a dichos inconvenientes. En este caso los objetivos a alcanzar fueron tres particularmente; generar perfiles para los usuarios que componen el grupo de trabajo de programación de mano de obra en lo que respecta a las actividades de mantenimiento en las plantas de Ternium; realizar un único informe de fin de reparaciones programadas para todos los sectores de mantenimiento que se abocan a cada área productiva; y por último actualizar, unificar y crear especificaciones técnicas de compras que debían ser enviadas al sector de compras para poder licitar determinados servicios de mantenimiento. Más allá de que se llegó a lo propuesto, en este proyecto final se pueden observar diferentes herramientas adquiridas a lo largo de la carrera, utilizadas para abordar a las soluciones necesarias.Fil: Luna, María Belén. Graphical survey, diagnosis and proposals for intervention of "Guardia y Custodia" of "El Polvorín"

This project, carried out for the subject of the DAC of Rehabilitation, is based on one of the two military buildings that are in the foothills of Montjuic, in the district of “El Polvorín”, now, part of the “Marina del Port”. In this building resided the guard that guarded the warehouse where the gunpowder was stored to supply the arms of the Castle of Montjuic. Through it a tour of history, environment and the whole is made to understand the meaning and importance of the building. In this study a description is made as detailed as possible to understand how it was built and its deterioration over the years. And it is helped by means of photographs, graphs and drawings to be able to visualize it. The process carried out for the graphic survey is detailed, explaining which information sources have been consulted, which tools have been used for the measurement and finally the plans derived from all this work have been included. The project will consist of different sections. In them we will describe the building in its current state, locating all those injuries as well as detecting all those points for which the current legal regulations are not being met. In the second section, the necessary proposals will be made to solve the localized injuries as well as for the building to comply with the stipulations of the Technical Building Code. Likewise, the realization of some functional and / or aesthetic improvements will be studied to make the building more useful. With the improvements in communication, and the inauguration this September, of two new subway stations in this neighborhood, access to the building, almost unknown in Barcelona, has been facilitated in this way the search for information and data collection It has been easier. El femicidio: dogmática y aplicación judicial

El Ecuador incorporó el femicidio como delito en la normativa, con la aprobación del Código Orgánico Integral Penal; reconociendo la muerte violenta de mujeres causadas por el hecho de serlo o por su condición de género, como un problema social y público, asociado a las relaciones asimétricas de poder entre hombres y mujeres. La construcción del femicidio en el contexto social obedece a lucha de las mujeres por sus derechos; y, ha sido reconocida en el ámbito normativo, plano vinculado con la dogmática, que incorpora el análisis de las categorías del delito: 1) tipicidad, 2) antijuridicidad; y, 3) culpabilidad. Elementos materia de discusión en los procesos penales. El objetivo de la presente investigación, es analizar dogmáticamente con perspectiva de género la aplicación del tipo penal femicidio en un contexto íntimo, de tal manera que permita al lector entender este fenómeno desde su naturaleza más compleja, partiendo por un recorrido de su construcción social, la necesidad de su inclusión en el Ecuador, las discusiones relevantes vinculadas con el tipo penal, el análisis de las categorías dogmáticas del delito, los problemas que se pueden generar en cada elemento del delito; todo con la finalidad de responder a la interrogante ¿Cómo realizar un análisis dogmático con enfoque de género del tipo penal de femicidio

Perfil del turista de salud que visita los baños de Barro termomedicinal de hervideros, zorritos – Tumbes 2018

La presente tesis tiene por objetivo determinar el Perfil del Turista de Salud que visita los Baños de Barro Termomedicinal de Hervideros, Zorritos – Tumbes 2018. Dentro de ello incluye identificar el perfil demográfico y perfil psicográfico del turista que llega ha dicho lugar; por ende, también desarrollar la creación de un perfil del turista de salud de acuerdo a los resultados establecidos por la encuesta. Se aplicó un estudio descriptivo de la variable de estudio para explicar su comportamiento. La pregunta de esta investigación es ¿Cuál es el Perfil del Turista que visita los baños de barro termomedicinal de Hervideros en el distrito de Zorritos 2018? Los resultados se encontraron utilizando la técnica de la encuesta con el instrumento cuestionario para los turistas que visitan el lugar de Hervideros, con nuestra de 200 visitantes. Se realizó la investigación no experimental, es decir no hay ninguna manipulación en la variable; teniendo así una investigación aplicada donde los problemas se convertirán de inmediato en conclusiones para poder dar soluciones en un tiempo determinado. Es así que este trabajo de acuerdo a los resultados se concluyó con un perfil del turista de Salud en los Baños de Barro Termomedicinal de Hervideros en el distrito de Zorritos, provincia de Contralmirante Villar Zorritos. Se obtuvo características demográficas y psicográficas de los turistas para poder así mejorar los servicios que cumplen las expectativas de los visitantes

La educación bimodal como puerta de acceso a la Universidad

Este trabajo presentará el relato de una experiencia de educación bimodal llevada a cabo en el primer y segundo año de la carrera de Psicopedagogía de la Escuela de Humanidades de la Universidad Nacional de San Martin (EH-UNSAM). Dicha experiencia plantea el propósito de ampliar la cobertura educativa y democratizar el acceso a los conocimientos y a la formación universitaria, propone otra línea de enriquecimiento que aprovecha las propiedades de lo virtual para potenciar el intercambio entre docentes y estudiantes. La estrategia se basa en reconocer la riqueza que nos ofrecen las tecnologías para potenciar el acceso y gestión de la información, la comunicación entre actores y la apropiación del conocimiento, en los procesos de enseñanza, aprendizaje y gestión. Ofrecer una propuesta bimodal implica comenzar un largo y variado camino hacia la innovación, redistribución de recursos, estrategias y habilidades en el aula, aprovechando la potencialidad de las tecnologías de la información y reflexionando sobre nuestras prácticas de enseñanza.Secretaría de Asuntos Académico
