    Biosensors Used for Quantification of Nitrates in Plants

    Nitrogen is essential for the plant because it is used for the production of chlorophyll, proteins, nucleic acids, amino acids, and other cellular compounds; nitrogen is available in two forms: ammonium and nitrate. Several tools have been used to quantify nitrates in plants such as the Kjeldahl method and Dumas combustion digestion; however, they are destructive and long time-consuming methods. To solve these disadvantages, methods such as selective electrodes, optical sensors, reflectometers, and images based sensors have been developed; nonetheless, all these techniques show interference when carrying out measurements. Currently, biosensors based on genetic constructions, based on the response of promoter gene fused to Gene Fluorescent Protein (GFP), are gaining popularity, because they improve the accuracy of measurements of nitrate by avoiding the interference of carriers ion, high salt conditions, and other factors. The present review shows the different methods to quantify the nitrogen in plants; later, a biosensors perspective is presented, mainly focused on biosensors based on organism genetically modified. The review presents a list of promoter and reporter genes that could be used to develop different kind of sensors, and a perspective of sensors to measure quantitatively the nitrogen is presented

    Hyperhomocystinemia as a Cause of Cerebral Ischemia in a Young Patient, Presentation of a Case in Northwestern Colombia

    La asociación entre hiperhomocisteinemia y accidente cerebrovascular está bien documentada en la literatura. Sin embargo, existen informes limitados que revelan eventos de accidente cerebrovascular en la población joven afectada por hiperhomocisteinemia. En este articulo presentamos un caso que muestra una hiperhomocisteinemia que provocó un accidente cerebrovascular isquémico en un paciente de 16 años. El paciente acudió a urgencias por mareos y debilidad en las extremidades del hemicuerpo derecho, no pérdida del conocimiento. El examen neurológico preliminar nomostró signos cerebelosos positivos, el examen físico y el estudio diagnóstico final confirmó un isquémico del territorio de la cerebral media izquierda secundario a  hiperhomocisteinemia ya que el protocolo diagnostico no mostro causas arrítmicas, aterotromboticas ni malformaciones vasculares, no foramen ni comunicaciones interventriculares o auriculares, se encontró una hiperhomocistinemia marcada. Discutimos la presentación del caso, el estudio diagnóstico, en este atípico caso en un paciente proveniente de zona rural de Santander.The association between hyperhomocysteinemia and stroke is well documented in the literature. However, there are limited reports revealing stroke events in the young population affected by hyperhomocysteinemia. In this article we present a case showing hyperhomocysteinemia that caused an ischemic stroke in a 16-year-old patient. The patient went to the emergency room due to dizziness and weakness in the extremities of the right hemibody, not loss of consciousness. The preliminary neurological examination did not show positive cerebellar signs, the physical examination and the final diagnostic study confirmed an ischemic condition of the left middle cerebral territory secondary to hyperhomocysteinemia since the diagnostic protocol did not show arrhythmic, atherothrombotic or vascular malformations causes, no foramen or interventricular communications or headphones, marked hyperhomocysteinemia was found. We discuss the presentation of the case, the diagnostic study, in this atypical case in a patient from rural Santander

    FPGA-based Fused Smart Sensor for Real-Time Plant-Transpiration Dynamic Estimation

    Plant transpiration is considered one of the most important physiological functions because it constitutes the plants evolving adaptation to exchange moisture with a dry atmosphere which can dehydrate or eventually kill the plant. Due to the importance of transpiration, accurate measurement methods are required; therefore, a smart sensor that fuses five primary sensors is proposed which can measure air temperature, leaf temperature, air relative humidity, plant out relative humidity and ambient light. A field programmable gate array based unit is used to perform signal processing algorithms as average decimation and infinite impulse response filters to the primary sensor readings in order to reduce the signal noise and improve its quality. Once the primary sensor readings are filtered, transpiration dynamics such as: transpiration, stomatal conductance, leaf-air-temperature-difference and vapor pressure deficit are calculated in real time by the smart sensor. This permits the user to observe different primary and calculated measurements at the same time and the relationship between these which is very useful in precision agriculture in the detection of abnormal conditions. Finally, transpiration related stress conditions can be detected in real time because of the use of online processing and embedded communications capabilities

    Activation of AMP kinase ameliorates kidney vascular dysfunction, oxidative stress and inflammation in rodent models of obesity

    Background and Purpose Obesity is a risk factor for the development of chronic kidney disease independent of diabetes, hypertension and other co-morbidities. Obesity-associated nephropathy is linked to dysregulation of the cell energy sensor AMP-activated protein kinase (AMPK). We aimed here to assess whether impairment of AMPK activity may cause renal arterial dysfunction in obesity and to evaluate the therapeutic potential of activating renal AMPK. Experimental Approach Effects of the AMPK activator A769662 were assessed on intrarenal arteries isolated from ob/ob mice and obese Zucker rats and then mounted in microvascular myographs. Superoxide and hydrogen peroxide production were measured by chemiluminescence and fluorescence, respectively, and protein expression was analysed by western blotting. Key Results Endothelium-dependent vasodilation and PI3K/Akt/eNOS pathway were impaired in preglomerular arteries from genetically obese rats and mice, along with impaired arterial AMPK activity and blunted relaxations induced by the AMPK activator A769662. Acute ex vivo exposure to A769662 restored endothelial function and enhanced activity of PI3K/Akt/eNOS pathway in obese rats, whereas in vivo treatment with A769662 improved metabolic state and ameliorated endothelial dysfunction, reduced inflammatory markers and vascular oxidative stress in renal arteries and restored redox balance in renal cortex of obese mice. Conclusion and Implications These results demonstrate that AMPK dysregulation underlies obesity-associated kidney vascular dysfunction and activation of AMPK improves metabolic state, protects renal endothelial function and exerts potent vascular antioxidant and anti-inflammatory effects. The beneficial effects of vascular AMPK activation might represent a promising therapeutic approach to the treatment of obesity-related kidney injury

    Hypothalamic AMPK-ER Stress-JNK1 Axis Mediates the Central Actions of Thyroid Hormones on Energy Balance

    Thyroid hormones (THs) act in the brain to modulate energy balance. We show that central triiodothyronine (T3) regulates de novo lipogenesis in liver and lipid oxidation in brown adipose tissue (BAT) through the parasympathetic (PSNS) and sympathetic nervous system (SNS), respectively. Central T3 promotes hepatic lipogenesis with parallel stimulation of the thermogenic program in BAT. The action of T3 depends on AMP-activated protein kinase (AMPK)-induced regulation of two signaling pathways in the ventromedial nucleus of the hypothalamus (VMH): decreased ceramide-induced endoplasmic reticulum(ER) stress, which promotes BAT thermogenesis, and increased c-Jun N-terminal kinase (JNK) activation, which controls hepatic lipid metabolism. Of note, ablation of AMPK alpha 1 in steroidogenic factor 1 (SF1) neurons of the VMH fully recapitulated the effect of central T3, pointing to this population in mediating the effect of central THs on metabolism. Overall, these findings uncover the underlying pathways through which central T3 modulates peripheral metabolism.Peer reviewe

    Energy Estimation of Cosmic Rays with the Engineering Radio Array of the Pierre Auger Observatory

    The Auger Engineering Radio Array (AERA) is part of the Pierre Auger Observatory and is used to detect the radio emission of cosmic-ray air showers. These observations are compared to the data of the surface detector stations of the Observatory, which provide well-calibrated information on the cosmic-ray energies and arrival directions. The response of the radio stations in the 30 to 80 MHz regime has been thoroughly calibrated to enable the reconstruction of the incoming electric field. For the latter, the energy deposit per area is determined from the radio pulses at each observer position and is interpolated using a two-dimensional function that takes into account signal asymmetries due to interference between the geomagnetic and charge-excess emission components. The spatial integral over the signal distribution gives a direct measurement of the energy transferred from the primary cosmic ray into radio emission in the AERA frequency range. We measure 15.8 MeV of radiation energy for a 1 EeV air shower arriving perpendicularly to the geomagnetic field. This radiation energy -- corrected for geometrical effects -- is used as a cosmic-ray energy estimator. Performing an absolute energy calibration against the surface-detector information, we observe that this radio-energy estimator scales quadratically with the cosmic-ray energy as expected for coherent emission. We find an energy resolution of the radio reconstruction of 22% for the data set and 17% for a high-quality subset containing only events with at least five radio stations with signal.Comment: Replaced with published version.
