A process of ârewritingâ rural territories is currently underway, based on the importance of the ever-present connection between landscape and agriculture, which enhance the social value of both categories. In recent decades, in fact, agriculture has begun to express a new, different and more âconsciousâ meaning: its value does not consist just in its capacity to provide goods and services, but also in its possibility to create new territorial identities, new values, a more effective ârelationalityâ between producers and consumers, new business and local development models, and new opportunities to rebalance the relationship between town and countryside. Similarly, the rural landscape has been interpreted as a continuous process of social construction. The new reciprocity between landscape and agriculture was evaluated starting from the analysis of the family farming model. To get a better idea of the performance of this kind of enterprises it was conducted an assessment at local-scale using as case study the agritourist farms in the Valle di Comino
Radiolabeled native low-density lipoprotein injected into patients with carotid stenosis accumulates in macrophages of atherosclerotic plaque
BACKGROUND:
Accumulation of LDL within the arterial wall appears to play a crucial role in the initiation and progression of . The dynamic sequence of this event has not been fully elucidated in humans.
METHODS AND RESULTS:
In 7 with previous transient ischemic attack or stroke and critical (>70%) , autologous [(125)I]-labeled LDL or [(125)I]-labeled human serum albumin were 24 to 72 hours before endarterectomy. specimens obtained at endarterectomy were analyzed by autoradiography and immunohistochemistry. Autoradiographic study showed that LDL was localized prevalently in the foam cells of plaques, whereas the accumulation in the lipid core was negligible. Immunohistochemistry revealed that foam cells that had accumulated LDL were mostly CD68 positive, whereas a small number were alpha-actin positive. No accumulation of the radiotracer was detected in plaques after injection of human serum albumin. In 3 treated for 4 weeks with vitamin E (900 mg/d), an almost complete suppression of LDL uptake by was observed.
CONCLUSIONS:
This study shows that circulating LDL rapidly in human . The prevalent accumulation of LDL by provides strong support to the hypothesis that these cells play a crucial role in the pathogenesis of atherosclerosis
Synchronized onset of nuclear and cell surface modifications in U937 cells during apoptosis
In this study we investigated the relationship between nuclear and cell surface modifications (i.e. blebbing, phosphatidylserine [PS] and sugar residues exposure) in a monocytic cell line, U937, during apoptosis induced by oxidative stress (1mM H2O2) or inhibition of protein synthesis (10 mg/ml puromycin). Dying cells were simultaneously observed for nuclear modifications, presence of superficial blebs and plasma membrane alterations. Morphological analysis performed by conventional fluorescence microscopy, or by transmission and scanning electron microscopy showed that the courses of nuclear and membrane alterations occured concomitantly, but the phenotype was dependent on the stage of the apoptotic process and the type of apoptogenic inducer used. The progression of apoptosis in U937 cells beyond early stages resulted in the extensive formation of blebs which concomitantly lost some typical markers of apoptosis, such as PS and sugar residues. Therefore, the modality by which the nucleus condenses, or the amount and the pattern of distribution of PS on the cell surface were, for each cell line, strictly related to the apoptogenic inducer. The morphological data reported in the present paper should lead to a more precise quantification of apoptosis by improving the detection of apoptotic cells in vivo (i.e. in tissue, organs), which is a crucial point in the evaluation of efficiency of antiproliferative drugs, such as antiblastic or immunosuppressive compounds
The expression and the nuclear activity of the caretaker gene Ku86 are modulated by somatostatin
Somatostatin is a peptide hormone that exerts antisecretory and antiproliferative activities on some human tumors. The Ku70/86 heterodimer acts as regulatory subunit of the DNA dependent protein kinase and its DNA binding activity mediates DNA double strands breaks repair that is crucial to maintain the genetic integrity of the genome. The activation of the heterodimer regulates cell cycle progression and the activity of nuclear transcription factors involved in DNA replication and cell proliferation. Moreover Ku86 behaves as a receptor for the growth inhibitory tetradecapeptide, somatostatin. Herein we report that somatostatin treatment to a colon carcinoma cell line (Caco-2) inhibits cell growth and, at same time, strongly modulates the activation of Ku70/86 heterodimer and the levels of Ku86 in the nucleus by increasing its specific mRNA level. Our findings are consistent with the hypothesis that somatostatin controls cell cycle progression and DNA repair through a new signalling pathway that involves the regulation of Ku86 level and modulates the Ku70/86 activity in the nucleus
Low Expression of Programmed Death 1 (PD-1), PD-1 Ligand 1 (PD-L1), and Low CD8+ T Lymphocyte Infiltration Identify a Subgroup of Patients With Gastric and Esophageal Adenocarcinoma With Severe Prognosis
Prognosis of gastric and esophageal cancer is poor and treatment improvements are needed. Programmed cell death 1 receptor (PD-1) interaction with its ligand PD-L1 in tumor micro-environment promotes immune tolerance and blocking monoclonal antibodies have entered clinical practice. However, clinical significance of PD-1 and PD-L1 expression in gastric and esophageal adenocarcinomas, particularly in non-Asian patients, is still unclear. Three tissue microarrays including 190 clinically annotated esophageal (n = 31) and gastric (n = 159) adenocarcinomas and 58 paired mucosa specimens, were stained with PD-1, PD-L1, and CD8-specific reagents in indirect immunohistochemistry assays. PD-L1 expression was detectable in 23.2% of cancer specimens. High PD-1 expression was detectable in 37.3% of cases and high CD8+ infiltration in 76%. PD-L1 and high PD1 expression significantly correlated with each other (rs = 0.404, P < 0.0001) and both significantly correlated with CD8+ infiltration (rs = 0.435, P = 0.0003, and rs = 0.444; P = 0.0004, respectively). CD8+ lymphocyte infiltration correlated with improved survival in univariate (P = 0.009), but not multivariate analysis. Most interestingly, multivariate analysis and Kaplan-Meier curves indicate that combined low PD-1/PD-L1 expression and low CD8+ lymphocyte infiltration significantly correlate with poor prognosis. Our data document the clinical significance of a microenvironmental signature including PD-1/PD-L1 expression and CD8+ lymphocyte infiltration in gastric and esophageal adenocarcinomas and contribute to identify a patients' subset requiring more aggressive peri-operative treatments
High myeloperoxidase positive cell infiltration in colorectal cancer is an independent favorable prognostic factor
BACKGROUND
Colorectal cancer (CRC) infiltration by adaptive immune system cells correlates with favorable prognosis. The role of the innate immune system is still debated. Here we addressed the prognostic impact of CRC infiltration by neutrophil granulocytes (NG).
METHODS
A TMA including healthy mucosa and clinically annotated CRC specimens (nâ=â1491) was stained with MPO and CD15 specific antibodies. MPO+ and CD15+ positive immune cells were counted by three independent observers. Phenotypic profiles of CRC infiltrating MPO+ and CD15+ cells were validated by flow cytometry on cell suspensions derived from enzymatically digested surgical specimens. Survival analysis was performed by splitting randomized data in training and validation subsets.
RESULTS
MPO+ and CD15+ cell infiltration were significantly correlated (p<0.0001; râ=â0.76). However, only high density of MPO+ cell infiltration was associated with significantly improved survival in training (Pâ=â0.038) and validation (Pâ=â0.002) sets. In multivariate analysis including T and N stage, vascular invasion, tumor border configuration and microsatellite instability status, MPO+ cell infiltration proved an independent prognostic marker overall (Pâ=â0.004; HRâ=â0.65; CI:±0.15) and in both training (Pâ=â0.048) and validation (Pâ=â0.036) sets. Flow-cytometry analysis of CRC cell suspensions derived from clinical specimens showed that while MPO+ cells were largely CD15+/CD66b+, sizeable percentages of CD15+ and CD66b+ cells were MPO-.
CONCLUSIONS
High density MPO+ cell infiltration is a novel independent favorable prognostic factor in CRC
Hyperbaric exposure and oxidative Stress in occupational activities (HEOxS): the study protocol
Background: Hyperbaric exposure (HE) is proven to be a stressor to several mechanisms in living cells.
Even if after homeostasis restoration, harmful effects are expected, in particular a presence of free
radicals. These latter are the stimulus to negative phenomenon as inflammation or cancer. In Italy,
with 7500 km of sea shores, a large quantity of workers is exposed to HE during occupational
activities. A deep knowledge of HE and bodily effects is not well defined; hence a multidisciplinary
assessment of risk is needed. To detect one or more indicators of HE a research group is organised,
under the INAIL sponsorship. The research project focused on the oxidative stress (OxS) and this
paper details on the possible protocol to estimate, with a large amount of techniques on several
human liquids, the relationship between OxS and HE. Specific attention will be paid to identify
confounding factors and their influence.
Methods: Blood and urine will be sampled. Several lab techniques will be performed on samples, both
targeted, to measure the level of well-known biomarkers, and untargeted. Regard the formers:
products of oxidation of DNA and RNA in urine; inflammation and temperature cytokines and
protein carbonyles in blood. Untargeted evaluation will be performed for a metabolomics analysis in
urine. Confounding factors: temperature, body fat, fitness, allergies and dietary habits. These factors
will be assessed, directly or indirectly, prior and after HE. The final scope of the project is to determine
one or more indicators that relates to HE in hits twofold nature: depth and duration.
Conclusion: The relationship between OxS and HE is not deeply investigated and literature proposes
diverging results. The project aims to define the time dependence of biomarkers related to OxS, to
rise knowledge in risk assessment in workers exposed to HE

Spectrum of mutations in Italian patients with familial hypercholesterolemia: New results from the LIPIGEN study
Background Familial hypercholesterolemia (FH) is an autosomal dominant disease characterized by elevated plasma levels of LDL-cholesterol that confers an increased risk of premature atherosclerotic cardiovascular disease. Early identification and treatment of FH patients can improve prognosis and reduce the burden of cardiovascular mortality. Aim of this study was to perform the mutational analysis of FH patients identified through a collaboration of 20 Lipid Clinics in Italy (LIPIGEN Study). Methods We recruited 1592 individuals with a clinical diagnosis of definite or probable FH according to the Dutch Lipid Clinic Network criteria. We performed a parallel sequencing of the major candidate genes for monogenic hypercholesterolemia (LDLR, APOB, PCSK9, APOE, LDLRAP1, STAP1). Results A total of 213 variants were detected in 1076 subjects. About 90% of them had a pathogenic or likely pathogenic variants. More than 94% of patients carried pathogenic variants in LDLR gene, 27 of which were novel. Pathogenic variants in APOB and PCSK9 were exceedingly rare. We found 4 true homozygotes and 5 putative compound heterozygotes for pathogenic variants in LDLR gene, as well as 5 double heterozygotes for LDLR/APOB pathogenic variants. Two patients were homozygous for pathogenic variants in LDLRAP1 gene resulting in autosomal recessive hypercholesterolemia. One patient was found to be heterozygous for the ApoE variant p.(Leu167del), known to confer an FH phenotype. Conclusions This study shows the molecular characteristics of the FH patients identified in Italy over the last two years. Full phenotypic characterization of these patients and cascade screening of family members is now in progress
