    The trafficking and targeting of P2X receptors

    The functional expression of P2X receptors at the plasma membrane is dependent on their trafficking along secretory and endocytic pathways. There are seven P2X receptor subunits, and these differ in their subcellular distributions because they have very different trafficking properties. Some are retained within the endoplasmic reticulum (ER), while others are predominantly at the cell surface or within endosomes and lysosomes. Changes in recruitment of receptors to and from the plasma membrane provides a way of rapidly up- or down-regulating the cellular response to adenosine triphosphate (ATP). An additional layer of regulation is the targeting of these receptors within the membranes of each compartment, which affects their stability, function and the nature of the effector proteins with which they form signaling complexes. The trafficking and targeting of P2X receptors is regulated by their interactions with other proteins and with lipids and we can expect this to vary in a cell-type specific manner and in response to changes in the environment giving rise to differences in receptor activity and function

    The diversity and evolution of ecological and environmental citizen science

    Citizen science—the involvement of volunteers in data collection, analysis and interpretation—simultaneously supports research and public engagement with science, and its profile is rapidly rising. Citizen science represents a diverse range of approaches, but until now this diversity has not been quantitatively explored. We conducted a systematic internet search and discovered 509 environmental and ecological citizen science projects. We scored each project for 32 attributes based on publicly obtainable information and used multiple factor analysis to summarise this variation to assess citizen science approaches. We found that projects varied according to their methodological approach from ‘mass participation’ (e.g. easy participation by anyone anywhere) to ‘systematic monitoring’ (e.g. trained volunteers repeatedly sampling at specific locations). They also varied in complexity from approaches that are ‘simple’ to those that are ‘elaborate’ (e.g. provide lots of support to gather rich, detailed datasets). There was a separate cluster of entirely computer-based projects but, in general, we found that the range of citizen science projects in ecology and the environment showed continuous variation and cannot be neatly categorised into distinct types of activity. While the diversity of projects begun in each time period (pre 1990, 1990–99, 2000–09 and 2010–13) has not increased, we found that projects tended to have become increasingly different from each other as time progressed (possibly due to changing opportunities, including technological innovation). Most projects were still active so consequently we found that the overall diversity of active projects (available for participation) increased as time progressed. Overall, understanding the landscape of citizen science in ecology and the environment (and its change over time) is valuable because it informs the comparative evaluation of the ‘success’ of different citizen science approaches. Comparative evaluation provides an evidence-base to inform the future development of citizen science activities

    More complex than expected—mapping activities and youths’ experiences at BioBlitz events to the rosette model of science communication

    Deficit, dialogue, or participation—which of these three main models of science communications is the best fit to describe activities and experiences of citizen science? One might assume that participation is the best match, but the reality of citizen science events is more complex. The rosette model of science communication offers a more detailed set of subcategories, e.g., educate, entertain, or do, in addition to the three main models—deficit, dialogue and participation. To systematically describe citizen science activities and experiences, we apply data on what activities are offered and what young people (5–19 years old) experience when participating in a citizen science event format called BioBlitzes across the rosette model. The mapping results illustrate how the rosette model can help to make citizen science project designers and practitioners more aware of the various modes of science communications that they may encounter at BioBlitz events and inform their design decisions regarding how settings can shape participants’ experiences

    A detailed analysis of more than 800 electronic high-resolution spectra of gamma Cas, which were obtained during a time interval of over 6000 days (16.84 yrs) at several observatories, documents the smooth variations in the density and/or extend of its circumstellar envelope. We found a clear anticorrelation between the peak intensity and FWHM of the H alpha emission, which seems to agree with recent models of such emission lines. The main result of this study is a confirmation of the binary nature of the object, determination of a reliable linear ephemeris T_{min.RV} = HJD (2452081.9±\pm0.6) + (203.52±\pm0.08)*E, and a rather definitive set of orbital elements. We clearly demonstrated that the orbit is circular within the limits of accuracy of our measurements and has a semi-amplitude of radial-velocity curve of 4.30±\pm0.09 (km/s). No trace of the low-mass secondary was found. The time distribution of our spectra does not allow a reliable investigation of rapid spectral variations, which are undoubtedly present in the spectra. We postpone this investigation for a future study, based on series of dedicated whole-night spectral observations

    Young people in iNaturalist: a blended learning framework for biodiversity monitoring

    Participation in authentic research in the field and online through Community and Citizen Science (CCS) has shown to bring learning benefits to volunteers. In online CCS, available platforms present distinct features, ranging from scaffolding the process of data collection, to supporting data analysis and enabling volunteers to initiate their own studies. What is yet not well understood is how best to design CCS programmes that are educational, inclusive, and accessible by diverse volunteers, including young people and those with limited prior science experiences who are rather few in CCS. In this study, we interviewed 31 young people, aged 7–20 years old, who used iNaturalist, an online biodiversity monitoring platform, and identified how different forms of participation online and in the field facilitated (or inhibited) certain forms of learning, as defined by the Environmental Science Agency framework. Findings revealed that iNaturalist enabled participation of young people including those with limited science experiences and facilitated science learning such as the development of science competence and understanding. A blended learning framework for biodiversity monitoring in CCS is presented as a means to support the development of hybrid, educational, and inclusive CCS programmes for young people

    Complexity reduction and policy consensus: asylum seekers, the right to work, and the ‘pull factor’ thesis in the UK context

    Since the early 2000s, asylum policy in Western states has become increasingly dominated by the concept of the ‘pull factor’—the idea that the economic rights afforded to asylum seekers can act as a migratory pull, and will have a bearing on the numbers of asylum applications received. The pull factor thesis has been widely discredited by researchers but remains powerful among policymakers. Through an analysis of the pull factor in the UK context, and drawing on insights from Cultural Political Economy, this article argues that the hegemony of the pull factor thesis is best understood as a ‘policy imaginary’ which has become sedimented through both discursive and extra-discursive practices and processes. The article offers a means of understanding how a common sense assumption—which is challenged by a large body of evidence—has come to dominate policymaking in a key area of concern for politicians and policymakers

    Delivery, setting and outcomes of paediatric Outpatient Parenteral Antimicrobial Therapy (OPAT): A scoping review

    Background There has been little detailed systematic consideration of the delivery, setting and outcomes of paediatric Outpatient Parenteral Antimicrobial Therapy (OPAT), although individual studies report that it is a safe and effective treatment. Objective This scoping review aimed to examine what is known about the delivery, settings and outcomes of paediatric OPAT and to identify key knowledge deficits. Design A scoping review using Arksey and O'Malley's framework was undertaken. Data sources Keywords were identified and used to search MEDLINE and CINAHL. Study appraisal methods Primary research studies were included if samples comprised children and young people 21 or under, who had received OPAT at home or in a day treatment centre. The Mixed Methods Appraisal Tool was used to review the methodological quality of the studies Main findings From a preliminary pool of 157 articles, 51 papers were selected for full review. 19 studies fitted the inclusion criteria. Factors influencing delivery of OPAT were diverse and included child's condition, home environment, child-related factors, parental compliance, training and monitoring. There is little consensus as to what constitutes success of and adverse events in OPAT. Conclusions Future studies need to clearly define and use success indicators and adverse events in order to provide evidence that paediatric OPAT is safe and effective. Implications Consensus outcomes that include child and parent perspectives need to be developed to allow a clearer appreciation of a successful paediatric OPAT service

    Pneumococcal carriage following PCV13 delivered as one primary and one booster dose (1 + 1) compared to two primary doses and a booster (2 + 1) in UK infants

    In January 2020 the UK changed from a 2 + 1 schedule for 13-valent pneumococcal conjugate vaccine (PCV13) to a 1 + 1 schedule (doses at 3 and 12 months) based on a randomized immunogenicity trial comparing the two schedules. Carriage prevalence measured at the time of booster and 6 months later in 191 of the 213 study infants was 57 % (109/191) and 60 % (114/190) respectively. There were eight episodes of vaccine-type (VT) or vaccine-related 6C carriage in the 2 + 1 and six in the 1 + 1 group; ≥4-fold rises in serotype-specific IgG in 71 children with paired post-booster and follow up blood samples at 21–33 months of age were found in 20 % (7/35) of the 2 + 1 and 15 % (6/41) of the 1 + 1 group. VTs identified in carriage and inferred from serology were similar comprising 3, 19A and 19F. Dropping a priming dose from the 2 + 1 PCV 13 schedule did not increase VT carriage in the study cohort. Ongoing population level carriage studies will be important to confirm this
