Regional association of pCASL-MRI with FDG-PET and PiB-PET in people at risk for autosomal dominant Alzheimer's disease.
Autosomal dominant Alzheimer's disease (ADAD) is a small subset of Alzheimer's disease that is genetically determined with 100% penetrance. It provides a valuable window into studying the course of pathologic processes that leads to dementia. Arterial spin labeling (ASL) MRI is a potential AD imaging marker that non-invasively measures cerebral perfusion. In this study, we investigated the relationship of cerebral blood flow measured by pseudo-continuous ASL (pCASL) MRI with measures of cerebral metabolism (FDG PET) and amyloid deposition (Pittsburgh Compound B (PiB) PET). Thirty-one participants at risk for ADAD (age 39 ± 13 years, 19 females) were recruited into this study, and 21 of them received both MRI and FDG and PiB PET scans. Considerable variability was observed in regional correlations between ASL-CBF and FDG across subjects. Both regional hypo-perfusion and hypo-metabolism were associated with amyloid deposition. Cross-sectional analyses of each biomarker as a function of the estimated years to expected dementia diagnosis indicated an inverse relationship of both perfusion and glucose metabolism with amyloid deposition during AD development. These findings indicate that neurovascular dysfunction is associated with amyloid pathology, and also indicate that ASL CBF may serve as a sensitive early biomarker for AD. The direct comparison among the three biomarkers provides complementary information for understanding the pathophysiological process of AD
ATF-4 and hydrogen sulfide signalling mediate longevity from inhibition of translation or mTORC1 [preprint]
Inhibition of mTORC1 (mechanistic target of rapamycin 1) slows ageing, but mTORC1 supports fundamental processes that include protein synthesis, making it critical to elucidate how mTORC1 inhibition increases lifespan. Under stress conditions, the integrated stress response (ISR) globally suppresses protein synthesis, resulting in preferential translation of the transcription factor ATF-4. Here we show in C. elegans that the ATF-4 transcription program promotes longevity and that ATF-4 upregulation mediates lifespan extension from mTORC1 inhibition. ATF-4 activates canonical anti-ageing mechanisms but also increases expression of transsulfuration enzymes to promote hydrogen sulfide (H2S) production. ATF-4-induced H2S production mediates longevity and stress resistance from C. elegans mTORC1 suppression, and ATF4 drives H2S production in mammalian dietary restriction. This H2S boost increases protein persulfidation, a protective modification of redox-reactive cysteines. Increasing H2S levels, or enhancing mechanisms that H2S modulates through persulfidation, may represent promising strategies for mobilising therapeutic benefits of the ISR or mTORC1 inhibition

Bioengineered 3D models of human pancreatic cancer recapitulate in vivo tumour biology
Patient-derived in vivo models of human cancer have become a reality, yet their turnaround time is inadequate for clinical applications. Therefore, tailored ex vivo models that faithfully recapitulate in vivo tumour biology are urgently needed. These may especially benefit the management of pancreatic ductal adenocarcinoma (PDAC), where therapy failure has been ascribed to its high cancer stem cell (CSC) content and high density of stromal cells and extracellular matrix (ECM). To date, these features are only partially reproduced ex vivo using organoid and sphere cultures. We have now developed a more comprehensive and highly tuneable ex vivo model of PDAC based on the 3D co-assembly of peptide amphiphiles (PAs) with custom ECM components (PA-ECM). These cultures maintain patient-specific transcriptional profiles and exhibit CSC functionality, including strong in vivo tumourigenicity. User-defined modification of the system enables control over niche-dependent phenotypes such as epithelial-to-mesenchymal transition and matrix deposition. Indeed, proteomic analysis of these cultures reveals improved matrisome recapitulation compared to organoids. Most importantly, patient-specific in vivo drug responses are better reproduced in self-assembled cultures than in other models. These findings support the use of tuneable self-assembling platforms in cancer research and pave the way for future precision medicine approaches
Blood-Brain Barrier Breakdown in the Aging Human Hippocampus
The blood-brain barrier (BBB) limits entry of blood-derived products, pathogens, and cells into the brain that is essential for normal neuronal functioning and information processing. Post-mortem tissue analysis indicates BBB damage in Alzheimer’s disease (AD). The timing of BBB breakdown remains, however, elusive. Using an advanced dynamic contrast-enhanced MRI protocol with high spatial and temporal resolutions to quantify regional BBB permeability in the living human brain, we show an age-dependent BBB breakdown in the hippocampus, a region critical for learning and memory that is affected early in AD. The BBB breakdown in the hippocampus and its CA1 and dentate gyrus subdivisions worsened with mild cognitive impairment that correlated with injury to BBB-associated pericytes, as shown by the cerebrospinal fluid analysis. Our data suggest that BBB breakdown is an early event in the aging human brain that begins in the hippocampus and may contribute to cognitive impairment
Shadowing of the Nascent Jet in NGC 4261 by a Line-Emitting Supersonic Accretion Disk
NGC 4261 (3C 270) is a low-luminosity radio galaxy with two symmetric
kiloparsec-scale jets. Earlier Hubble Space Telescope observations indicated
the presence of a hundred-parsec scale disk of cool dust and gas surrounding a
central, supermassive (\sim 4.9\times 10^8\msun) black hole. The recent
detection of free-free radio absorption by a small, geometrically-thin disk,
combined with earlier studies of the disk's large scale properties, provide the
strictest constraints to date on the nature of the accretion process in this
system. We show here that a supersonic disk, illuminated by the active galactic
nucleus (AGN), can not only account for the observed radio shadowing, but can
also produce the optical broad lines emitted from this region. Beyond a
critical radius , line cooling dominates over gravitational dissipation
and the gas is effectively cooled down to temperatures below K. Within
, however, heating due to the release of gravitational energy overwhelms
line cooling and the plasma is heated very quickly to a temperature close to
its virial value as it falls toward the central engine. The disk is
optically-thin to UV and X-ray radiation within , so the ionizing
radiation from the AGN is preferentially absorbed near , affecting the
disk structure significantly. To include the ensuing photoionization effect, we
have used the algorithm Cloudy with additional heating introduced by
gravitational dissipation to calculate the temperature profile and line
emission from the disk in a self-consistent manner. The results of our model
calculation are consistent with current multiwavelength observations of the

Recent advances in experimental techniques allow one to measure and control
systems at the level of single molecules and atoms. Here gaining information
about fluctuating thermodynamic quantities is crucial for understanding
nonequilibrium thermodynamic behavior of small systems. To achieve this aim,
stochastic thermodynamics offers a theoretical framework, and nonequilibrium
equalities such as Jarzynski equality and fluctuation theorems provide key
information about the fluctuating thermodynamic quantities. We review the
recent progress in quantum fluctuation theorems, including the studies of
Maxwell's demon which plays a crucial role in connecting thermodynamics with

Retinal glycoprotein enrichment by concanavalin a enabled identification of novel membrane autoantigen synaptotagmin-1 in equine recurrent uveitis.
Complete knowledge of autoantigen spectra is crucial for understanding pathomechanisms of autoimmune diseases like equine recurrent uveitis (ERU), a spontaneous model for human autoimmune uveitis. While several ERU autoantigens were identified previously, no membrane protein was found so far. As there is a great overlap between glycoproteins and membrane proteins, the aim of this study was to test whether pre-enrichment of retinal glycoproteins by ConA affinity is an effective tool to detect autoantigen candidates among membrane proteins. In 1D Western blots, the glycoprotein preparation allowed detection of IgG reactions to low abundant proteins in sera of ERU patients. Synaptotagmin-1, a Ca2+-sensing protein in synaptic vesicles, was identified as autoantigen candidate from the pre-enriched glycoprotein fraction by mass spectrometry and was validated as a highly prevalent autoantigen by enzyme-linked immunosorbent assay. Analysis of Syt1 expression in retinas of ERU cases showed a downregulation in the majority of ERU affected retinas to 24%. Results pointed to a dysregulation of retinal neurotransmitter release in ERU. Identification of synaptotagmin-1, the first cell membrane associated autoantigen in this spontaneous autoimmune disease, demonstrated that examination of tissue fractions can lead to the discovery of previously undetected novel autoantigens. Further experiments will address its role in ERU pathology
Macroscopic superposition states of ultracold bosons in a double-well potential
We present a thorough description of the physical regimes for ultracold
bosons in double wells, with special attention paid to macroscopic
superpositions (MSs). We use a generalization of the Lipkin-Meshkov-Glick
Hamiltonian of up to eight single particle modes to study these MSs, solving
the Hamiltonian with a combination of numerical exact diagonalization and
high-order perturbation theory. The MS is between left and right potential
wells; the extreme case with all atoms simultaneously located in both wells and
in only two modes is the famous NOON state, but our approach encompasses much
more general MSs. Use of more single particle modes brings dimensionality into
the problem, allows us to set hard limits on the use of the original two-mode
LMG model commonly treated in the literature, and also introduces a new mixed
Josephson-Fock regime. Higher modes introduce angular degrees of freedom and MS

Indication for the disappearance of reactor electron antineutrinos in the Double Chooz experiment
The Double Chooz Experiment presents an indication of reactor electron
antineutrino disappearance consistent with neutrino oscillations. A ratio of
0.944 0.016 (stat) 0.040 (syst) observed to predicted events was
obtained in 101 days of running at the Chooz Nuclear Power Plant in France,
with two 4.25 GW reactors. The results were obtained from a single 10
m fiducial volume detector located 1050 m from the two reactor cores. The
reactor antineutrino flux prediction used the Bugey4 measurement as an anchor
point. The deficit can be interpreted as an indication of a non-zero value of
the still unmeasured neutrino mixing parameter \sang. Analyzing both the rate
of the prompt positrons and their energy spectrum we find \sang = 0.086
