    The lectotype for the Asian elephant, Elephas maximus Linnaeus, 1758 (Mammalia, Proboscidea) and comments on ‘primary, secondary and tertiary syntypes’ and ‘virtual lectotype designation’

    Last November a group of colleagues and ourselves designated a lectotype for the Asian elephant, Elephas maximus Linnaeus, 1758, having used morphology and genetic and proteomic sequencing to confirm that Linnaeus’s syntypes included both Asian and African elephants. The article was published (Cappellini et al., 2013) online in the Zoological Journal of the Linnean Society, together with eight items of Supplementary Information, and appeared on paper in the ZJLS in January 2014. The paper and SI items are available online at DOI:10.1111/zoj.12084. The lectotype is a very nearly complete mounted skeleton on display in the Natural History Museum of the University of Florence. John Ray described the specimen in 1673 and 1693 and Linnaeus cited Ray’s 1693 publication. The lectotype designation is available and valid. Dubois, Nemésio & Bour, however, have criticised our choice of selected specimen (published in Bionomina, June 2014; a preview is available online at http://mapress.com/bionomina/content.htm). We are concerned because they have demonstrated misunderstanding or ignorance of a number of aspects of the International Code of Zoological Nomenclatur

    Learning styles of 3rd year genetics students and their affection and perception of effectiveness of a range of instructional modes

    Different students come into our classes with different past learning experiences with diverse backgrounds. They have acquired their preferred way to assimilating information and have settled into a mode of learning that is most effective for them. Teaching them in using a single dimensional approach inevitably will only facilitate effective learning to a small population of the student cohort. A questionnaire was administered to ascertain the range of preferred learning styles of a cohort of third year genetics students in an effort to understand the diversity of their learning styles and to fine tune instructional modes for the coming year. The questionnaire assessed the preferred sensory modes in which the students choose in the way they perceive, process and acquire information. Through a series of lectures, use of multi-media and guided inquiry case studies, group discussion and group presentation, students were also able to provide additional feedback as to the mode of instructional activity which they perceived that they enjoy and is most effective in learning human molecular genetics. This paper analyses the correlation of gender and the different learning styles of these students, the preference and perceived effectiveness of a range of instructional activities. Based of these findings a number of recommendations for improvements to instructional are also discussed

    Resolution of the type material of the Asian elephant, Elephas maximus Linnaeus, 1758 (Proboscidea, Elephantidae)

    The understanding of Earth’s biodiversity depends critically on the accurate identification and nomenclature of species. Many species were described centuries ago, and in a surprising number of cases their nomenclature or type material remain unclear or inconsistent. A prime example is provided by Elephas maximus, one of the most iconic and well-known mammalian species, described and named by Linnaeus (1758) and today designating the Asian elephant. We used morphological, ancient DNA (aDNA), and high-throughput ancient proteomic analyses to demonstrate that a widely discussed syntype specimen of E. maximus, a complete foetus preserved in ethanol, is actually an African elephant, genus Loxodonta. We further discovered that an additional E. maximus syntype, mentioned in a description by John Ray (1693) cited by Linnaeus, has been preserved as an almost complete skeleton at the Natural History Museum of the University of Florence. Having confirmed its identity as an Asian elephant through both morphological and ancient DNA analyses, we designate this specimen as the lectotype of E. maximus

    Diagnostic applications of next generation sequencing: working towards quality standards

    Over the past 6 years, next generation sequencing (NGS) has been established as a valuable high-throughput method for research in molecular genetics and has successfully been employed in the identification of rare and common genetic variations. All major NGS technology companies providing commercially available instruments (Roche 454, Illumina, Life Technologies) have recently marketed bench top sequencing instruments with lower throughput and shorter run times, thereby broadening the applications of NGS and opening the technology to the potential use for clinical diagnostics. Although the high expectations regarding the discovery of new diagnostic targets and an overall reduction of cost have been achieved, technological challenges in instrument handling, robustness of the chemistry and data analysis need to be overcome. To facilitate the implementation of NGS as a routine method in molecular diagnostics, consistent quality standards need to be developed. Here the authors give an overview of the current standards in protocols and workflows and discuss possible approaches to define quality criteria for NGS in molecular genetic diagnostics

    A new perspective on turbulent Galactic magnetic fields through comparison of linear polarisation decomposition techniques

    We compare two rotationally invariant decomposition techniques on linear polarization data: the spin-2 spherical harmonic decomposition in two opposite parities, the E- and B-mode, and the multiscale analysis of the gradient of linear polarization, |∇ P|. We demonstrate that both decompositions have similar properties in the image domain and the spatial frequency domain. They can be used as complementary tools for turbulence analysis of interstellar magnetic fields in order to develop a better understanding of the origin of energy sources for the turbulence, the origin of peculiar magnetic field structures and their underlying physics. We also introduce a new quantity |∇EB| based on the E- and B-modes and we show that in the intermediate- and small-scale limit |∇EB| |∇ P|. Analysis of the 2.3 GHz S-band Polarization All Sky Survey shows many extended coherent filament-like features appearing as ‘double jumps’ in the |∇ P| map that are correlated with negative and positive filaments of B-type polarization. These local asymmetries between the two polarization types, E and B, of the non-thermal Galactic synchrotron emission have an influence on the E- and B-mode power spectra analyses. The wavelet-based formalism of the polarization gradient analysis allows us to locate the position of E- or B-mode features responsible for the local asymmetries between the two polarization types. In analysed subregions, the perturbations of the magnetic field are trigged by star clusters associated with H II regions, the Orion–Eridanus superbubble and the North Polar Spur at low Galactic latitude

    High expression of the MADS-box gene VRT2 increases the number of rudimentary basal spikelets in wheat

    Spikelets are the fundamental building blocks of Poaceae inflorescences, and their development and branching patterns determine the various inflorescence architectures and grain yield of grasses. In wheat (Triticum aestivum), the central spikelets produce the most and largest grains, while spikelet size gradually decreases acropetally and basipetally, giving rise to the characteristic lanceolate shape of wheat spikes. The acropetal gradient corresponds with the developmental age of spikelets; however, the basal spikelets are developed first, and the cause of their small size and rudimentary development is unclear. Here, we adapted G&T-seq, a low-input transcriptomics approach, to characterize gene expression profiles within spatial sections of individual spikes before and after the establishment of the lanceolate shape. We observed larger differences in gene expression profiles between the apical, central, and basal sections of a single spike than between any section belonging to consecutive developmental time points. We found that SHORT VEGETATIVE PHASE MADS-box transcription factors, including VEGETATIVE TO REPRODUCTIVE TRANSITION 2 (VRT-A2), are expressed highest in the basal section of the wheat spike and display the opposite expression gradient to flowering E-class SEPALLATA1 genes. Based on multi-year field trials and transgenic lines, we show that higher expression of VRT-A2 in the basal sections of the spike is associated with increased numbers of rudimentary basal spikelets. Our results, supported by computational modeling, suggest that the delayed transition of basal spikelets from vegetative to floral developmental programs results in the lanceolate shape of wheat spikes. This study highlights the value of spatially resolved transcriptomics to gain insights into developmental genetics pathways of grass inflorescences

    Identifying factors which influence eating disorder risk during behavioral weight management: A consensus study

    This study aimed to understand clinician, researcher and consumer views regarding factors which influence eating disorder (ED) risk during behavioral weight management, including individual risk factors, intervention strategies and delivery features. Eighty-seven participants were recruited internationally through professional and consumer organizations and social media and completed an online survey. Individual characteristics, intervention strategies (5-point scale) and delivery features (important/unimportant/unsure) were rated. Participants were mostly women (n = 81), aged 35-49 y, from Australia or United States, were clinicians and/or reported lived experience of overweight/obesity and/or ED. There was agreement (64% to 99%) that individual characteristics were relevant to ED risk, with history of ED, weight-based teasing/stigma and weight bias internalization having the highest agreement. Intervention strategies most frequently rated as likely to increase ED risk included those with a focus on weight, prescription (structured diets, exercise plans) and monitoring strategies, e.g., calorie counting. Strategies most frequently rated as likely to decrease ED risk included having a health focus, flexibility and inclusion of psychosocial support. Delivery features considered most important were who delivered the intervention (profession, qualifications) and support (frequency, duration). Findings will inform future research to quantitatively assess which of these factors predict eating disorder risk, to inform screening and monitoring protocols
