This study presents the integrated biostratigraphic (calcareous nannofossils and foraminifera), petrographic, geochemical and ichnological analyzes of the Gramame Formation (Maastrichtian) at the CIPASA Quarry section, in the Pernambuco-Paraíba Basin, northeastern Brazil. A high resolution calcareous nannofossil biostratigraphic study has been carried out, allowing to subdivide the studied section into three subzones of Maastrichtian age: CC25A, CC25B, and CC25C. Each biostratigraphic unit was further characterized by petrographic, geochemical and ichnological parameters. During the Maastrichtian the area was characterized by a dry and warm climate with low influx of terrigenous sediments. These conditions were fundamental to the widespread development of a carbonate ramp system in an outer neritic to bathyal setting, with deposition of alternating calcareous mudstones and argillaceous mudstones of the Gramame Formation. The Gramame Formation belongs to the transgressive system tract of second-order type, characteristic of a Passive Margin Sequence. The studied section consists of part of a third order deposicional cycle, which is arranged in a transgressive systems tract and a highstand systems tract. The lowstand systems tract was not characterized

A series of 2-[(arylidene)amino]-cycloalkyl[b]thiophene-3-carbonitriles (2a-x) was synthesized by incorporation of substituted aromatic aldehydes in Gewald adducts (1a-c). The title compounds were screened for their antifungal activity against Candida krusei and Criptococcusneoformans and for their antiproliferative activity against a panel of 3 human cancer cell lines (HT29, NCI H-292 and HEP). Forantiproliferative activity, the partial least squares (PLS) methodology was applied. Some of the prepared compounds exhibited promising antifungal and proliferative properties. The most active compounds for antifungal activity were cyclohexyl[b]thiophene derivatives, and forantiproliferative activity cycloheptyl[b]thiophene derivatives, especially 2-[(1H-indol-2-yl-methylidene)amino]-5,6,7,8-tetrahydro-4H-cyclohepta[b]thiophene-3-carbonitrile (2r), which inhibited more than 97 % growth of the three cell lines. The PLS discriminant analysis (PLS-DA)applied generated good exploratory and predictive results and showed that the descriptors having shape characteristics were strongly correlated with the biological data.

The poultry red mite (PRM), Dermanyssus gallinae (De Geer, 1778), is a worldwide distributed ectoparasite and considered a major pest affecting the laying hen industry in Europe. Based on available information in other ectoparasites, the mite microbiome might participate in several biological processes and the acquisition, maintenance and transmission of pathogens. However, little is known about the role of PRM as a mechanical carrier or a biological vector in the transmission of pathogenic bacteria. Herein, we used a metaproteomics approach to characterize the alphaproteobacteria in the microbiota of PRM, and variations in its profile with ectoparasite development (nymphs vs. adults) and feeding (unfed vs. fed). The results showed that the bacterial community associated with D. gallinae was mainly composed of environmental and commensal bacteria. Putative symbiotic bacteria of the genera Wolbachia, C. Tokpelaia and Sphingomonas were identified, together with potential pathogenic bacteria of the genera Inquilinus, Neorickettsia and Roseomonas. Significant differences in the composition of alphaproteobacterial microbiota were associated with mite development and feeding, suggesting that bacteria have functional implications in metabolic pathways associated with blood feeding. These results support the use of metaproteomics for the characterization of alphaproteobacteria associated with the D. gallinae microbiota that could provide relevant information for the understanding of mite-host interactions and the development of potential control interventions.

COL1A1 and miR-29b show lower expression levels during osteoblast differentiation of bone marrow stromal cells from Osteogenesis Imperfecta patients

Abstract\ud \ud Background\ud The majority of Osteogenesis Imperfecta (OI) cases are caused by mutations in one of the two genes, COL1A1 and COL1A2 encoding for the two chains that trimerize to form the procollagen 1 molecule. However, alterations in gene expression and microRNAs (miRNAs) are responsible for the regulation of cell fate determination and may be evolved in OI phenotype.\ud \ud \ud Methods\ud In this work, we analyzed the coding region and intron/exon boundaries of COL1A1 and COL1A2 genes by sequence analysis using an ABI PRISM 3130 automated sequencer and Big Dye Terminator Sequencing protocol. COL1A1 and miR-29b expression were also evaluated during the osteoblastic differentiation of mesenchymal stem cell (MSC) by qRT-PCR using an ABI7500 Sequence Detection System.\ud \ud \ud Results\ud We have identified eight novel mutations, where of four may be responsible for OI phenotype. COL1A1 and miR-29b showed lower expression values in OI type I and type III samples. Interestingly, one type III OI sample from a patient with Bruck Syndrome showed COL1A1 and miR-29b expressions alike those from normal samples.\ud \ud \ud Conclusions\ud Results suggest that the miR-29b mechanism directed to regulate collagen protein accumulation during mineralization is dependent upon the amount of COL1A1 mRNA. Taken together, results indicate that the lower levels observed in OI samples were not sufficient for the induction of miR-29b.Support for this work was provided by the Brazilian agencies FAPESP, CNPq,\ud and Center for Cell-based Therapy. Using a data set corresponding to an integrated luminosity of 3fb13 fb^{-1}, collected by the LHCb experiment in pppp collisions at centre-of-mass energies of 7 and 8 TeV, the effective lifetime in the Bs0J/ψηB^0_s \rightarrow J/\psi \eta decay mode, τeff\tau_{\textrm{eff}}, is measured to be τeff=1.479±0.034 (stat)±0.011 (syst)\tau_{\textrm{eff}} = 1.479 \pm 0.034~\textrm{(stat)} \pm 0.011 ~\textrm{(syst)} ps. Assuming CPCP conservation, τeff\tau_{\textrm{eff}} corresponds to the lifetime of the light Bs0B_s^0 mass eigenstate. This is the first measurement of the effective lifetime in this decay mode.Comment: All figures and tables, along with any supplementary material and additional information, are available at https://lhcbproject.web.cern.ch/lhcbproject/Publications/LHCbProjectPublic/LHCb-PAPER-2016-017.htm
