    The traditional production of machinery and tools at ŽĎAS, a. s., and the increasing requirements placed on the final product, particularly those related to the internal quality of forgings, requires new technological measures for the ingot’s casting and solidification. The development and use of the new mould type 8K9.2 for tool-steel ingot casting has reduced the share of forgings with unacceptable quality. A substantial improvement has been achieved, especially in terms of the occurrence of defects, such as cavities and cracks in the ingot body.Tradicionalna proizvodnja strojev in orodij v podjetju ŽĎAS, a. s., in naraščajoče zahteve pri lastnostih končnega proizvoda, ki so povezane z notranjo kakovostjo izkovkov, so zahtevale nove tehnološke ukrepe pri litju in strjevanju ingotov.     Elexacaftor-tezacaftor-ivacaftor in patients with cystic fibrosis ineligible for clinical trials: a 24-week observational study

    Introduction: Seminal clinical trials with the triple combination of elexacaftor-tezacaftor-ivacaftor (ETI) demonstrated clinical efficacy in people with cystic fibrosis (pwCF) who carry at least one F508del mutation. However, due to exclusion criteria of these clinical trials, the effect of ETI was not studied in a substantial number of pwCF. Thus, we ran a single center trial to evaluate a clinical efficacy of ETI treatment in adult pwCF who were ineligible for enrollment in registration studies.Methods: PwCF on ETI with prior lumacaftor-ivacaftor therapy, severe airway obstruction, well-preserved lung function, or with airway infection with pathogens at risk of more rapid decline in lung function formed the study group, while all the others on ETI formed the control group. Lung function, nutritional status and sweat chloride concentration were assessed before and after initialization of ETI therapy over a 6-month period.Results: Approximately a half of the ETI-treated pwCF at the adult Prague CF center (49 of 96) were assigned to the study group. Their mean changes in body mass index ( + 1.04 kg/m2) and in sweat chloride concentration (−48.4 mmol/L) were similar to the control group ( + 1.02 kg/m2; −49.7 mmol/L), while the mean change in percent predicted forced expiratory volume in 1 s (ppFEV1; + 10.3 points) was significantly lower than in the control group ( + 15.8 points) (p = 0.0015). In the subgroup analysis, pwCF with severe airway obstruction (ppFEV1 <40) and pwCF with well-preserved lung function (ppFEV1 >90) showed a less potential for improvement in lung function during the ETI treatment than controls (median change in ppFEV1 + 4.9 points and + 9.5 points, respectively).Conclusion: PwCF not eligible for inclusion in clinical trials demonstrated improvement in lung function and nutritional status following the initiation of treatment with the ETI combination. Moderate increase in ppFEV1 was observed in those with severe airway obstruction or well-preserved lung function

    Zeolites are stable microporous aluminosilicates with numerous applications in chemical technology such as separation of species and catalytic transformations. Our study is focused on a weakly explored zeolite SSZ-16 with pore constrictions defined by 8-membered oxygen rings. Key results are the preparation of Et6-diquat-5 dication used as a structure directing agent (SDA) and finding the optimum synthesis conditions with respect to zeolite phase purity. Stability of SDA was examined in conditions similar to those of autoclave synthesis (concentration, pH, temperature). Moreover, the content and location of SDA species in zeolite phase and conditions of SDA decomposition were investigated

    The importance of determination of oxidative stress markers in exhaled breath condensate for the assessment of lung disease progression in patients with cystic fibrosis

    The aim of this study was to evaluate the relationship of oxidative stress (OS) marker s in exhaled breath condensate (EBC) in adult patients with cystic fibrosis (CF) to the severity of lung disease, nutritional status and systemic antioxidants and inflammatory markers, as well as to short - and medium - term development of pulmonary function and nutritional status, and finally to assess the response to treatment with inhaled corticosteroids (ICS). Methods: CF patients were examined in a stable phase of the disease during routine outpatient controls. EBC was collected using E C oScreen device (J aeger) in CF patients and in control group members. Nitrites and nitrates and 8 - isoprostane were examined using liquid chromatography and competitive enzyme immunoanalysis, respectively, in EBC as OS markers. Demographic data including the dominant pathogen of airway colonization and ICS treatment were recorded in CF patients. Lung function tests, chest X-ray s, nutritional statuses and systemic antioxidants and inflammatory markers were also examined using standard methods. The values of OS markers in EBC in patients with CF were compared with the control group and correlated to clinical parameters.     The production of heavy steel forgings of micro-alloyed steels gives the possibility to obtain advantages associated with the benefit of the application of micro-alloying elements and thermomechanical treatments for improving the mechanical properties of forgings to level by sheets, strips and tubes. The paper presents the influence of quenching temperature on the mechanical properties and microstructure of F60 steel according to ASTM A694. The verification of the influence of quenching temperature contributes to an optimization of the method of micro-alloyed steel heat treatment. The steel’s microstructure and mechanical properties after quenching constitute the initial and basic criteria to achieve the required mechanical properties after a properly chosen tempering temperature.Web of Science45662261
