Mast cell tumor (MCT), or mastocytoma, the second most common skin tumor in dogs, is characterized by neoplastic transformations and abnormal proliferation of mast cells from cutaneous or visceral origins. Mutations in the proto-oncogene c-kit are present in 15% of dogs with MCT and up to 35% in high-grade tumors. In the clinical management of the disease, some characteristics, such as anatomical occurrence, histological grading, clinical staging, age and breed, are important parameters to better drive the treatment and predict the prognosis of each patient. However, it is not clear the correlations between c-kit mutation and clinical/epidemiological data, demonstrating the necessity of new studies. In this study, 78 clinical cases of canine patients with MCT were analyzed in Campinas - SP, Brazil, at Oncovida - Veterinary Oncology Center, in the period of 2017 to 2019, to determine the frequency, prevalence and correlations between molecular, clinical and epidemiological aspects. Results presented here shows that mixed breed dogs (51%) were the most affected, followed by Labrador (17%) and Boxer (16%). Most dogs were female (67%), aged 6 to 10 years (62%); 42% of the animals had tumors located in the pelvic limb and 17% had nodules in the chest. The most used method for diagnosis was histopathology, totalizing 60% of cases, followed by aspirative cytology (26%) and immunohistochemistry (8%). Regarding histological variables, 3 patients had grade I (11.8%), 47 had grade II (61%) and 15 had grade III (19%) and, according to KIUPEL et al. (2011) classification, 77 % of the cases presented low grade and 23% were high grade. The survival of these patients was also analyzed, demonstrating that animals presenting grade I and grade II (low grade) had longer survival (68%) in relation to grade III (high grade) (16%) The variables showed a significant correlation between histological grade and c-kit mutation with breed, age, localization of the tumor, diagnostic, chosen treatment and survival, indicating that they are both decisive to the outcome of the disease. Only sex was not correlated, suggesting that both the grade of the tumor and the c-kit mutation were not found to contribute to a different development of the tumor in male or female. These results demonstrate important data on the clinical behavior of MCT in the region of Campinas-SP, presenting relevant knowledge that can be used in the future in the clinical manage of the patients with this disease and demonstrating the necessity of further studies on the epidemiology of MCT in each region of Brazil

Through the analysis of all Brazilian indigenous lands registered in said application, it is sought to investigate which concepts and categories are put into play by the social actors to refer to and analyze the Brazilian Indigenous Lands as tourist attractions. It is also important to analyze the way tourists refer to indigenous peoples in their publications, as well as the construction of stereotypes and ways of thinking about indianity. It is believed that this analysis contributes important contributions to the scientific context of tourism research. Both for using relatively new means for conducting research, and for addressing the topic of relatively undeveloped literature in the area, that is, tourism in Indigenous Lands

Variable positive end-expiratory pressure can maintain oxygenation in experimental acute respiratory distress syndrome induced by oleic acid in dogs

    Get PDF
The use of positive end-expiratory pressure (PEEP) or lung recruitment maneuvers (RM) to improve oxygenation in acute respiratory distress syndrome (ARDS) is used but it may reduce cardiac output (CO). Intermittent PEEP may avoid these complications. Our objective was to determine if variable PEEP compared with constant PEEP is capable of maintaining arterial oxygenation and minimizing hemodynamic alterations with or without RM. Eighteen dogs with ARDS induced by oleic acid were randomized into three equal groups: group 1, low variable PEEP; group 2, high variable PEEP, and group 3, RM + high variable PEEP. All groups were submitted to constant PEEP, followed by variable PEEP (PEEP was increased from 5 to 10 cmH2O in group 1, and from 5 to 18 cmH2O in the other two groups). PaO2 was higher in group 3 (356.2 ± 65.4 mmHg) than in group 1 (92.7 ± 29.7 mmHg) and group 2 (228.5 ± 72.4 mmHg), P 0.05. Variable PEEP is able to maintain PaO2 when performed in combination with RM in dogs with ARDS. After RM, CO was reduced and there was no relevant difference between the variable and constant PEEP periods.Universidade Federal de São Paulo (UNIFESP) Escola Paulista de Medicina Departamento de MedicinaCentro Universitário São CamiloUniversidade de São Paulo Faculdade de Medicina Laboratório de Investigação Médica (LIM 12)UNIFESP, EPM, Depto. de MedicinaSciEL

In vitro study of the effect of Er: YAG laser irradiation on the apical sealing of different root canal sealers

    Get PDF
OBJECTIVES: The aim of this study was to determine the effect of Er:YAG laser irradiation used to clean dentinal walls on the apical sealing of root canals filled with different types of sealers. Background Data: Laser application to the dentinal walls removed debris, rendering the root canals free of smear layers and leaving the dentinal canaliculi open. METHODS: Sixty-four maxillary canines obtained from laboratory files were instrumented with K-files (Dentsply, Maillefer, Ballaigues, Switzerland) using the crown-down technique, and irrigated with a 0.5% sodium hypochlorite solution. The specimens were divided into two groups of 32 teeth each. In group I, the teeth were instrumented and irrigated with sodium hypochlorite solution, and divided into four subgroups to be sealed with the different materials (Endofill, N-Rickert, Sealapex and Sealer 26). In group II, the root canals were subjected to Er:YAG laser irradiation (200 mJ, 7 Hz and 60 J total energy), followed by root canal sealing as in group I. RESULTS: The data showed lower levels of apical microleakage in the teeth filled with N-Rickert, Sealapex and Sealer 26 cements than in those sealed with Endofill (p ; 0.05). CONCLUSIONS: The Er:YAG laser irradiation applied to the root canal walls was not able to prevent apical microleakage.OBJETIVOS: Avaliou-se a infiltração marginal apical em caninos superiores, obturados com quatro tipos diferentes de cimento, os quais foram imersos em tinta nanquim e mantidos a uma temperatura de 37º C por 96 horas, descalcificados em solução de ácido clorídrico a 5%, desidratados em série crescente de álcoois e diafanizados em salicilato de metila.

    Get PDF
Objectives: Hospitalization during pregnancy and childbirth increases the risk of Venous Thromboembolism Risk (VTE). This study applied a VTE risk score to all hospitalized pregnant women to ascertain its effectiveness in preventing maternal death from VTE until 3 months after discharge. Methods: In this interventional study, patients were classified as low- or high-risk according to the VTE risk score (Clinics Hospital risk score). High-risk patients (score ≥ 3) were scheduled for pharmacological Thromboprophylaxis (TPX). Interaction analysis of the main risk factors was performed using Odds Ratio (OR) and Poisson regression with robust variance. Results: The data of 10694 cases (7212 patients) were analyzed; 1626 (15.2%, 1000 patients) and 9068 (84.8%, 6212 patients) cases were classified as high-risk (score ≥ 3) and low-risk (score < 3), respectively. The main risk factors (Odds Ratio, 95% Confidence Interval) for VTE were age ≥ 35 and < 40 years (1.6, 1.4–1.8), parity ≥ 3 (3.5, 3.0–4.0), age ≥ 40 years (4.8, 4.1–5.6), multiple pregnancies (2.1, 1.7–2.5), BMI ≥ 40 kg/m2 (5.1, 4.3–6.0), severe infection (4.1, 3.3–5.1), and cancer (12.3, 8.8–17.2). There were 10 cases of VTE: 7/1636 (0.4%) and 3/9068 (0.03%) in the high- and low-risk groups, respectively. No patient died of VTE. The intervention reduced the VTE risk by 87%; the number needed to treat was 3. Conclusions: This VTE risk score was effective in preventing maternal deaths from VTE, with a low indication for TPX. Maternal age, multiparity, obesity, severe infections, multiple pregnancies, and cancer were the main risk factors for VTE

The novel p.Cys65Tyr mutation in NR5A1gene in three 46,XY siblings with normal testosterone levels and their mother with primary ovarian insufficiency

Disorders of sex development (DSD) is the term used for congenital conditions in which development of chromosomal, gonadal, or phenotypic sex is atypical. Nuclear receptor subfamily 5, group A, member 1 gene (NR5A1) encodes steroidogenic factor 1 (SF1), a transcription factor that is involved in gonadal development and regulates adrenal steroidogenesis. Mutations in the NR5A1 gene may lead to different 46,XX or 46,XY DSD phenotypes with or without adrenal failure. We report a Brazilian family with a novel NR5A1 mutation causing ambiguous genitalia in 46,XY affected individuals without Müllerian derivatives and apparently normal Leydig function after birth and at puberty, respectively. Their mother, who is also heterozygous for the mutation, presents evidence of primary ovarian insufficiency. Three siblings with 46,XY DSD, ambiguous genitalia and normal testosterone production were included in the study. Molecular analyses for AR, SRD5A2 genes did not reveal any mutation. However, NR5A2 sequence analysis indicated that all three siblings were heterozygous for the p.Cys65Tyr mutation which was inherited from their mother. In silico analysis was carried out to elucidate the role of the amino acid change on the protein function. After the mutation was identified, all sibs and the mother had been reevaluated. Basal hormone concentrations were normal except that ACTH levels were slightly elevated. After 1 mcg ACTH stimulation test, only the older sib showed subnormal cortisol response. The p.Cys65Tyr mutation located within the second zinc finger of DNA binding domain was considered deleterious upon analysis with predictive algorithms. The identification of heterozygous individuals with this novel mutation may bring additional knowledge on structural modifications that may influence NR5A1 DNA-binding ability, and may also contribute to genotype-phenotype correlations in DSD. The slightly elevated ACTH basal levels in all three patients with 46,XY DSD and the subnormal cortisol response after 1 mcg ACTH stimulation in the older sib indicate that a long-term follow-up for adrenal function is important for these patients. Our data reinforce that NR5A1 analysis must also be performed in 46,XY DSD patients with normal testosterone levels without AR mutations.15
