The pharmacological potential of the macromycetes (Cordyceps sinensis and Pleurotus ostreatus) cultived by submerged fermentation

Resumo: Este trabalho objetivou a avaliação das potencialidades farmacológicas dos macromicetos Cordyceps sinensis e Pleurotus ostreatus, obtidos por fermentação submersa, quanto à ação hipolipidêmica, antiaterosclerótica, e protetor da disfunção puberal em modelos hiperlipídicos de ratos machos Wistar, bem como a ação antitumoral do extrato aquoso de Cordyceps sinensis contra células do neuroblastoma humano. Para a instauração do modelo hiperlipídico, uma dieta comercial basal foi aditivada com gordura vegetal hidrogenada (6% p/p) e gordura de porco (14% p/p). Para as determinações bioquímicas, foram realizadas as dosagens do colesterol plasmático, lipoproteína de baixa densidade (LDL), atividade da aspartato transaminase, uréia e testosterona. A atividade antiaterosclerótica foi inferida pela determinação da ativação macrofágica: capacidade fagocítica, volume lisossomal e produção de íons (ânion superóxido, peróxido de hidrogênio e óxido nítrico). Os resultados indicaram que o Cordyceps sinensis diminuiu significativamente os parâmetros bioquímicos colesterol plasmático (37%), triglicerídeos (35%), LDL (40%), bem como normalizou os níveis de testosterona e restaurou a função hepática. Similarmente, o Pleurotus ostreatus também melhorou o perfil lipídico, bem como diminuiu a ativação macrofágica em aproximadamente 70%. Em adição, a ação antitumoral do Cordyceps sinensis contra células do neuroblastoma humano foi caracterizada por uma taxa de inibição de 20%. Sendo assim, estes cogumelos possivelmente não substituirão os tradicionais medicamentos já existentes para o tratamento destas patologias, mas poderão de forma valiosa complementá-los

Xilanases: principais metodologias e parâmetros cinéticos

Endo-1,4-β-xylanases are extracellular enzymes that make the hydrolysis of substrates β 1,4. Xylan is the main representative among the substrates that are susceptible to hydrolytic attack of xylanases. Xylanases have a wide application in different sectors of the economy. The purpose of this study was to review the published literature on the main methodologies used to study these enzymes, as well as gather some kinetic parameters. The reviewed methodologies are centered mainly in the purification process, since that is an essential step for the optimization of its enzymatic activity and therefore industrial application. The highlights are the techniques that use the saline precipitation and refined methodologies as chromatography and its variations, as well as factorial design studies and enzyme immobilization. These methodologies aim respectively, among other things, optimal conditions for enzyme production and recycling.As endo β 1,4 xilanases são enzimas extracelulares que fazem a hidrólise de substratos que possuem ligações do tipo β 1,4. A xilana é o principal representante dentre os substratos susceptíveis ao ataque hidrolítico das xilanases. Encontra vasta aplicação em diferentes setores da economia. O objetivo do presente trabalho foi revisar a literatura publicada sobre as principais metodologias utilizadas para se trabalhar com estas enzimas, bem como reunir alguns parâmetros cinéticos. As metodologias revisadas estão centralizadas principalmente no processo de purificação, haja vista que é uma etapa essencial para otimização da sua atividade enzimática e conseguinte aplicação industrial. Destacam-se as técnicas que utilizam a precipitação por sais e metodologias refinadas como a cromatografia e suas variações. Tem ganhado destaque, em pesquisas acadêmicas, os estudos de planejamentos fatoriais e imobilização enzimática. Tais metodologias buscam respectivamente, dentre outras coisas, condições otimizadas de produção e reciclagem enzimática

A Percepção dos Gestores sobre os Objetivos do Esporte nos Municípios

The purpose of this inventory was to characterize the profile of sports managers from cities belonging to an intermunicipal consortium in the state of São Paulo and to know their view on the objectives of the sport in the city. A qualitative research was carried out, with the participation of 12 sports managers belonging to the Union of the municipalities of Media Sorocabana, in São Paulo. It was possible to verify that the profile of the municipal sports managers is similar to that identified in other studies. Also it is verified the existence of a culture that disregards the professional skills and abilities for the sport management, inherent to the sport organizations. In their interviews, managers claim that sport in their cities aims to contribute to social inclusion, improve health and quality of life, identify new talent for sports and democratize access to sports practices. We realize that some of the proposed objectives cannot be achieved only with the practice of sports, but that they depend on the elaboration of a public policy that integrates different municipal secretariats.El presente estudio tuvo como objetivos caracterizar el perfil de los gestores del deporte, en ciudades que constituyen  un consorcio intermunicipal de São Paulo y conocer su mirada sobre los objetivos del deporte en la ciudad. Se realizó una investigación cualitativa, con la participación de 12 gestores de deporte pertenecientes a la Unión de los municipios de la Media Sorocabana del Estado de São Paulo (UMMES). Fue posible verificar que el perfil de los gestores deportivos municipales se asemeja al identificado en otros estudios realizados. También se constata la existencia de una cultura que desconsidera las competencias y habilidades necesarias para el manejo deportivo inherente a las organizaciones deportivas. En sus entrevistas, los gestores declaran que el deporte tiene como objetivos contribuir a la inclusión social, mejorar la salud y la calidad de vida de los practicantes, identificar nuevos talentos para el deporte y democratizar el acceso a las prácticas deportivas. Se percibe que parte de los objetivos propuestos no pueden ser alcanzados sólo con la práctica del deporte, sino que dependen de la elaboración de una política pública que integre diferentes secretarías municipales.O presente estudo teve como objetivos caracterizar o perfil dos gestores municipais de esporte de cidades pertencentes a um consórcio intermunicipal no estado de São Paulo e conhecer seu olhar sobre os objetivos do esporte no município. Foi possível verificar que o perfil dos gestores esportivos municipais se assemelha ao identificado em outros estudos realizados. Também se constata a existência de uma cultura que desconsidera as competências e habilidades necessárias para o gerenciamento esportivo inerente às organizações esportivas. Em suas entrevistas, os gestores declaram que o esporte em seus municípios tem como objetivos contribuir para a inclusão social, melhorar a saúde e a qualidade de vida dos praticantes, identificar novos talentos para o esporte e democratizar o acesso às práticas esportivas. Eudragit ® FS 30 D polymeric films containing chondroitin sulfate as candidates for use in coating seeking modified delivery of drugs

Polymeric films associating different concentrations of Eudragit(r) FS 30 D (EFS) and chondroitin sulfate (CS) were produced by casting for the development of a new target-specific site material. Formed films kept a final polymer mass of 4% (w/v) in the following proportions: EFS 100:00 CS (control), EFS 95:05 CS, EFS 90:10 CS and EFS 80:20 CS. They were analyzed for physical and chemical characteristics using Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM) and Raman spectroscopy. Furthermore, they were characterized by their water vapor permeability and degree of hydration at different conditions simulating the gastrointestinal tract. No chemical interactions were observed between CS and EFS, suggesting only a physical interaction between them in the different combinations tested. The results suggest that EFS and CS, when combined, may form films that are candidates for coating processes seeking a modified drug delivery, especially due to the synergism between pH dependency and specific biodegradability properties by the colonic microbiota. EFS 90:10 CS proved to be the most suitable for this purpose considering hydration and permeability characteristics of different associations analyzed

Technological and Biological Properties of Buttermilk: A Minireview

Buttermilk is a by-product obtained from the batting insertion in the process of obtaining cream and butter and it is constituted by fat globules which are surrounded by milk fat globule membranes (MFGMs). During the stirring process, the membrane is ruptured and the various components present therein are released. Because it has a high nutritional content and low cost, buttermilk has drawn attention in the prospect of new forms of application. In addition, its disposal is expensive and not biologically viable. The objective of this work is to present a compilation of the technological and biological activities of buttermilk. Among the technological properties, it is worth mentioning its application as in the production of functional foods, a conduit for the incorporation of probiotics, inhibition of bacterial adherence on industrial surfaces, as well as the encapsulation of easily degraded activities and fermentative processes. Among the biological properties, its antioxidant, hypocholesterolemic, antimicrobial, and anticancer activities stand out. In conclusion, the reuse of buttermilk is economically and sustainably viable and encourages increasing research related to its use

El concepto de congruencia y el respaldo de las celebridades deportivas

O segmento dos esportes, bem como o endosso de celebridades e o marketing esportivo vem apresentando crescimento significativo no cenário nacional e global, o que acarreta estudos e planejamentos cada vez mais precisos com o objetivo de atingir o público alvo de maneira mais assertiva. Este trabalho propõe investigar os antecedentes da congruência do endosso de celebridades, bem como o nível de congruência entre celebridades esportivas com alto reconhecimento e diferentes categorias de produtos. Como metodologia, foi desenvolvido um estudo de natureza quantitativa e os dados foram analisados basicamente por meio de duas técnicas \ud estatísticas: regressão linear e análise de variância. Os resultados apontam que duas das três antecedentes testadas são, de fato, significantes para o entencimento da congruência, assim como evidenciam diferenças significativas na percepção do público em relação à participação de atletas no endosso das três categorias de produtos testadas, de acordo com o nível de congruência percebido, indicando, desta forma, a necessidade de estudos prévios por parte das empresas que desejam ter suas marcas e produtos promovidos por celebridades esportivas.The segment of sports, as well as celebrity endorsement and sports marketing has shown significant growth in the national and global marketing, which leads to the development of more precise studies and plans focused on reaching the target audience in the most assertive way. The purpose of this study is to investigate the antecedents of celebrity endorsement congruence, as well its evaluation by consumer, when considering high performance sports celebrities and different product categories. A quantitative research was conducted and data were analysed basicaly by two statistic \ud techniques: linear regression and analisys of variance. Results points out that two of the three antecedents tested are, indeed, significant to better understand congruence. Significant differences were found between sports celebrity congruence with the three different product categories, indicating the need of previous market research by enterprises which aim on using sport celebrities to endorse their brands and productsEl segmento de los deportes, así como el respaldo de celebridades y marketing deportivo ha mostrado un crecimiento significativo en el escenario nacional y mundial, lo que implica el estudio y una planificación más precisa con el fin de llegar al público objetivo de una manera más asertiva. En este trabajo se propone investigar los antecedentes de la coherencia de la celebridad, así como el nivel de congruencia entre celebridades del deporte de alto reconocimiento y diferentes categorías de productos. Como métodos, un estudio cuantitativo fue desarrollado y se analizaron los datos sobre todo por dos técnicas estadísticas: regresión y análisis de varianza lineal. Los resultados \ud indican que dos de los tres antecedentes probados son realmente significativo para entendimiento de congruencia, así como aspectos más destacados diferencias significativas en la percepción del público con respecto a la participación de los atletas en la aprobación de las tres categorías de productos probados de conformidad con el nivel percepción de congruencia, lo que indica la necesidad de estudios previos de las empresas que deseen que sus marcas y productos promovidos por celebridades del deporte

The Effect of Aluminium Surface Treatments on the Bonding Properties of Silica-Modified Epoxy Adhesive Joints: A Statistical Approach

A full factorial design is carried out to investigate the effects of different surface treatments, the inclusion of silica microparticles and the use of wash primer on the apparent shear strength and adherent strength of single-lap aluminium joints. Scanning electron microscopy, surface energy and roughness measurements are performed to characterise the aluminium surface. The results show that the use of wash primer decreases the apparent shear strength of the joints significantly. The cohesive failure of the primer is the main cause of the reduction in strength. On the other hand, the inclusion of 10 wt.% of silica microparticles in the adhesive layers increases the shear strength by 26%. Surfaces treated with NaOH for one minute, without using a wash primer, result in the most resistant joint. In contrast to the apparent shear strength, adherent strength is most effective when only degreasing is considered

Erythrina velutina Willd. alkaloids : piecing biosynthesis together from transcriptome analysis and metabolite profiling of seeds and leaves

Introduction: Natural products of pharmaceutical interest often do not reach the drug market due to the associated low yields and difficult extraction. Knowledge of biosynthetic pathways is a key element in the development of biotechnological strategies for plant specialized metabolite production. Erythrina species are mainly used as central nervous system depressants in folk medicine and are important sources of bioactive tetracyclic benzylisoquinoline alkaloids (BIAs), which can act on several pathology-related biological targets. Objectives: In this sense, in an unprecedented approach used with a non-model Fabaceae species grown in its unique arid natural habitat, a combined transcriptome and metabolome analyses (seeds and leaves) is presented. Methods: The Next Generation Sequencing-based transcriptome (de novo RNA sequencing) was carried out in a NextSeq 500 platform. Regarding metabolite profiling, the High-resolution Liquid Chromatography was coupled to DAD and a micrOTOF-QII mass spectrometer by using electrospray ionization (ESI) and Time of Flight (TOF) analyzer. The tandem MS/MS data were processed and analyzed through Molecular Networking approach. Results: This detailed macro and micromolecular approach applied to seeds and leaves of E. velutina revealed 42 alkaloids, several of them unique. Based on the combined evidence, 24 gene candidates were put together in a putative pathway leading to the singular alkaloid diversity of this species. Conclusion: Overall, these results could contribute by indicating potential biotechnological targets formodulation of erythrina alkaloids biosynthesis as well as improve molecular databases with omic data from a non-model medicinal plant, and reveal an interesting chemical diversity of Erythrina BIA harvested in Caatinga
