    A Method Suitable for a Mobile Phone to Improve the Fluency of Stutterer's Speech

    ÄnkyttĂ€jien puheen sujuvuuden parantamiseen on puheterapian lisĂ€ksi saatavilla erilaisia laitteita ja ohjelmistoja. NĂ€mĂ€ elektroniset apuvĂ€lineet perustuvat usein Ă€nkyttĂ€jĂ€n omasta puheestaan saamien kuulohavaintojen muuntamiseen. KĂ€ytetyin nĂ€istĂ€ menetelmistĂ€ on viivĂ€stetty auditiivinen palaute, josta kĂ€ytetÀÀn lyhennettĂ€ DAF (Delayed Auditory Feedback). NĂ€mĂ€ erikoistuneet laitteet ovat yleensĂ€ kalliita ja hankalasti saatavilla. Halvemmalle, ohjelmistoon perustuvalle, mutta silti helposti mukana kannettavalle ratkaisulle on selvÀÀ tarvetta. TĂ€lle on tarjoutunut mahdollisuus nykyaikaisten matkapuhelimien ansiosta. Niiden ominaisuudet vastaavat jo tietokoneita, ja niiden kĂ€yttöjĂ€rjestelmille on mahdollista kehittÀÀ omia sovelluksia. TĂ€mĂ€n työn tavoitteena oli kehittÀÀ Symbian Series 60 -kĂ€yttöjĂ€rjestelmÀÀ kĂ€yttĂ€vissĂ€ matkapuhelimissa toimiva DAFia hyödyntĂ€vĂ€ sovellus, joka toimii sĂ€hköisten apuvĂ€lineiden tavoin. KĂ€ytettĂ€essĂ€ se siis auttaa Ă€nkyttĂ€jÀÀ parantamaan puheensa sujuvuutta. TĂ€tĂ€ sovellusta testattiin pienellĂ€ koehenkilöjoukolla ja tutkittiin, toimiiko se vastaavalla tavalla kuin samaan tarkoitukseen rakennetut laitteet. Mittauksista saatujen havaintojen perusteella sovellus parantaa koehenkilöinĂ€ olleiden Ă€nkyttĂ€jien puheen sujuvuutta. Vaikutus tuntuu riippuvan osin koehenkilöstĂ€ ja ilmenevĂ€n paremmin lukiessa tekstiĂ€ ÀÀneen kuin spontaanissa keskustelussa. Vaikka sovelluksen vaikutus oli selvĂ€, osa sen ominaisuuksista ei vastannut DAF-laitteiden ominaisuuksia. Huomattiin, ettĂ€ sovelluksessa on paljon potentiaalia jatkokehittelylle.Apart from speech therapy, there are various devices and software meant to improve the fluency of stutterers' speech. These electronic aids are often based on altering the auditory perceptions a stutterer receives of his own speech. The most used of these methods is called delayed auditory feedback or DAF. These specialized devices are typically expensive and hard to obtain. There is a clear need for a cheaper and portable software-based solution. The modern mobile phones have enabled implementing this solution. Their features already match those of computers, and as it is possible to develop one's own applications for their operating systems. The objective of this work was to develop an application using DAF and running in mobile phones using Symbian Series 60 operating system that works like the electronic aids. When used, it improves the fluency of a stutterer's speech. This application was tested on a small group of test subjects, examining whether it works similarly to the devices built for the purpose. The observations received from the tests indicate that the application improves the fluency of the stuttering test subjects. The effect seems to depend partly on the test subject and become more evident when text is being read aloud instead of speaking in a spontaneous conversation. Even though the effect was distinctive, part of the application's features didn't match those of the devices using DAF. It was discovered that the application has much potential for further development

    Overexpression of Polysialylated Neural Cell Adhesion Molecule Improves the Migration Capacity of Induced Pluripotent Stem Cell-Derived Oligodendrocyte Precursors

    Cell replacement therapy aiming at the compensation of lost oligodendrocytes and restoration of myelination in acquired or congenital demyelination disorders has gained considerable interest since the discovery of induced pluripotent stem cells (iPSCs). Patient-derived iPSCs provide an inexhaustible source for transplantable autologous oligodendrocyte precursors (OPCs). The first transplantation studies in animal models for demyelination with iPSC-derived OPCs demonstrated their survival and remyelinating capacity, but also revealed their limited migration capacity. In the present study, we induced overexpression of the polysialylating enzyme sialyltransferase X (STX) in iPSC-derived OPCs to stimulate the production of polysialic acid-neuronal cell adhesion molecules (PSA-NCAMs), known to promote and facilitate the migration of OPCs. The STX-overexpressing iPSC-derived OPCs showed a normal differentiation and maturation pattern and were able to downregulate PSA-NCAMs when they became myelin-forming oligodendrocytes. After implantation in the demyelinated corpus callosum of cuprizone-fed mice, STX-expressing iPSC-derived OPCs demonstrated a significant increase in migration along the axons. Our findings suggest that the reach and efficacy of iPSC-derived OPC transplantation can be improved by stimulating the OPC migration potential via specific gene modulation

    Identification of genes under dynamic post-transcriptional regulation from time-series epigenomic data

    Aim: Prediction of genes under dynamic post-transcriptional regulation from epigenomic data. Materials & methods: We used time-series profiles of chromatin immunoprecipitation-seq data of histone modifications from differentiation of mesenchymal progenitor cells toward adipocytes and osteoblasts to predict gene expression levels at five time points in both lineages and estimated the deviation of those predictions from the RNA-seq measured expression levels using linear regression. Results & conclusion: The genes with biggest changes in their estimated stability across the time series are enriched for noncoding RNAs and lineage-specific biological processes. Clustering mRNAs according to their stability dynamics allows identification of post-transcriptionally coregulated mRNAs and their shared regulators through sequence enrichment analysis.     Silver ions disrupt K+ homeostasis and cellular integrity in intact barley (Hordeum vulgare L.) roots

    The heavy metals silver, gold, and mercury can strongly inhibit aquaporin-mediated water flow across plant cell membranes, but critical examinations of their side effects are rare. Here, the short-lived radiotracer 42K is used to demonstrate that these metals, especially silver, profoundly change potassium homeostasis in roots of intact barley (Hordeum vulgare L.) plants, by altering unidirectional K+ fluxes. Doses as low as 5 ΌM AgNO3 rapidly reduced K+ influx to 5% that of controls, and brought about pronounced and immediate increases in K+ efflux, while higher doses of Au3+ and Hg2+ were required to produce similar responses. Reduced influx and enhanced efflux of K+ resulted in a net loss of >40% of root tissue K+ during a 15 min application of 500 ΌM AgNO3, comprising the entire cytosolic potassium pool and about a third of the vacuolar pool. Silver also brought about major losses of UV-absorbing compounds, total electrolytes, and NH4+. Co-application, with silver, of the channel blockers Cs+, TEA+, or Ca2+, did not affect the enhanced efflux, ruling out the involvement of outwardly rectifying ion channels. Taken together with an examination of propidium iodide staining under confocal microscopy, the results indicate that silver ions affect K+ homeostasis by directly inhibiting K+ influx at lower concentrations, and indirectly inhibiting K+ influx and enhancing K+ efflux, via membrane destruction, at higher concentrations. Ni2+, Cd2+, and Pb2+, three heavy metals not generally known to affect aquaporins, did not enhance K+ efflux or cause propidium iodide incorporation. The study reveals strong and previously unknown effects of major aquaporin inhibitors and recommends caution in their application

    Differential limit on the extremely-high-energy cosmic neutrino flux in the presence of astrophysical background from nine years of IceCube data

    We report a quasi-differential upper limit on the extremely-high-energy (EHE) neutrino flux above 5×1065\times 10^{6} GeV based on an analysis of nine years of IceCube data. The astrophysical neutrino flux measured by IceCube extends to PeV energies, and it is a background flux when searching for an independent signal flux at higher energies, such as the cosmogenic neutrino signal. We have developed a new method to place robust limits on the EHE neutrino flux in the presence of an astrophysical background, whose spectrum has yet to be understood with high precision at PeV energies. A distinct event with a deposited energy above 10610^{6} GeV was found in the new two-year sample, in addition to the one event previously found in the seven-year EHE neutrino search. These two events represent a neutrino flux that is incompatible with predictions for a cosmogenic neutrino flux and are considered to be an astrophysical background in the current study. The obtained limit is the most stringent to date in the energy range between 5×1065 \times 10^{6} and 5×10105 \times 10^{10} GeV. This result constrains neutrino models predicting a three-flavor neutrino flux of $E_\nu^2\phi_{\nu_e+\nu_\mu+\nu_\tau}\simeq2\times 10^{-8}\ {\rm GeV}/{\rm cm}^2\ \sec\ {\rm sr}at at 10^9\ {\rm GeV}$. A significant part of the parameter-space for EHE neutrino production scenarios assuming a proton-dominated composition of ultra-high-energy cosmic rays is excluded.Comment: The version accepted for publication in Physical Review

    Investigation of two Fermi-LAT gamma-ray blazars coincident with high-energy neutrinos detected by IceCube

    After the identification of the gamma-ray blazar TXS 0506+056 as the first compelling IceCube neutrino source candidate, we perform a systematic analysis of all high-energy neutrino events satisfying the IceCube realtime trigger criteria. We find one additional known gamma-ray source, the blazar GB6 J1040+0617, in spatial coincidence with a neutrino in this sample. The chance probability of this coincidence is 30% after trial correction. For the first time, we present a systematic study of the gamma-ray flux, spectral and optical variability, and multi-wavelength behavior of GB6 J1040+0617 and compare it to TXS 0506+056. We find that TXS 0506+056 shows strong flux variability in the Fermi-LAT gamma-ray band, being in an active state around the arrival of IceCube-170922A, but in a low state during the archival IceCube neutrino flare in 2014/15. In both cases the spectral shape is statistically compatible (≀2σ\leq 2\sigma) with the average spectrum showing no indication of a significant relative increase of a high-energy component. While the association of GB6 J1040+0617 with the neutrino is consistent with background expectations, the source appears to be a plausible neutrino source candidate based on its energetics and multi-wavelength features, namely a bright optical flare and modestly increased gamma-ray activity. Finding one or two neutrinos originating from gamma-ray blazars in the given sample of high-energy neutrinos is consistent with previously derived limits of neutrino emission from gamma-ray blazars, indicating the sources of the majority of cosmic high-energy neutrinos remain unknown.Comment: 22 pages, 11 figures, 2 Table

    LeptonInjector and LeptonWeighter: A neutrino event generator and weighter for neutrino observatories

    We present a high-energy neutrino event generator, called LeptonInjector, alongside an event weighter, called LeptonWeighter. Both are designed for large-volume Cherenkov neutrino telescopes such as IceCube. The neutrino event generator allows for quick and flexible simulation of neutrino events within and around the detector volume, and implements the leading Standard Model neutrino interaction processes relevant for neutrino observatories: neutrino-nucleon deep-inelastic scattering and neutrino-electron annihilation. In this paper, we discuss the event generation algorithm, the weighting algorithm, and the main functions of the publicly available code, with examples.Comment: 28 pages, 10 figures, 3 table
