    Estimación del índice de área foliar en un huerto de manzanos (Malus domestica) cv. Cripp’s Pink usando el sensor LAI-2000

    43 P.Se realizó un estudio para evaluar el equipo LAI-2000 (LI-COR, Lincoln, Nebraska, USA) en mediciones del índice de área foliar (IAF) en manzanos cv. Cripp’s Pink. Estas mediciones fueron comparadas con datos obtenidos a través de deshoje parcial. El ensayo se llevó a cabo en la comuna de Pelarco, Región del Maule, Chile (35° 25’ 16’’ L.S 71° 23’ 58’’ L.O, 198 m.s.n.m). Para la medición con LAI-2000 se utilizaron dos protocolos de medición (vista de 90° y 180°). Para evaluar la efectividad del equipo a diferentes niveles de radiación, se registró, el área foliar durante un día completo desde las 10:00 am hasta las 20:00 pm con el objetivo de establecer el horario adecuado para la medición de IAF. Con estos datos se logró establecer que la cantidad de radiación, orientación de las hileras, y los movimientos del sol durante el día, influencias fuertemente las mediciones del sensor LAI-2000. Las mediciones realizadas con LAI-2000, presentaron una buena correlación con los datos obtenidos del deshoje. Los mejores resultados fueron obtenidos con el protocolo 180° (sin el anillo 1) con un coeficiente de determinación (R2) de 0,65 y una raíz del cuadrado medio del error (RCME) de 0,054. Los resultados anteriores, permiten establecer que LAI-2000 es un equipo dinámico y fácil de utilizar, que entrega mediciones en tiempo real. Sin embargo, LAI-2000 es altamente dependiente de las condiciones de radiación y características del dosel, por lo que su uso debe ser comparado con métodos directos para establecer los ajustes y protocolos adecuados para su uso en forma práctica./ABSTRACT: A field experiment was carried out to evaluate the LAI-2000 sensor (LI-COR, Lincoln, Nebraska, USA) for measuring leaf area index (LAI) in apple trees cv. Cripps's Pink. LAI-2000 measurements were compared with data obtained by partial defoliation of the trees. The field experiment was located in Pelarco, Region del Maule, Chile (35 ° 25 '16'' L.S 71 ° 23' 58'' L.W, 198 m.a.s.l). Two protocols (view 90 ° and 180 °) were used for LAI-2000 measurements. To evaluate the performance of the sensor at different radiation levels, LAI was recorded from 10:00 am to 20:00 pm with the objective of establishing the appropriate time for measuring LAI. With these data was possible to establish that radiation, row orientation, and sun movements during the day,has a strong influences in the LAI-2000 measurements. LAI values obtained with LAI-2000, showed a good correlation with data obtained from defoliation. The best results were obtained with the protocol 180° when the ring 1 was eliminated, with determination coefficient (R2) of 0.65 and root mean square error (RMSE) of 0.054 m2 m-2. These results show that LAI-2000 allow to obtain real-time measurements with low errors. However, LAI-200 is highly dependent of the irradiation conditions and characteristics of the canopy, so its use should be compared with direct methods to adjust and setting the suitable protocols for its practical use

    Objetivo. A Lei de Acesso à Informação (LAI) n. 12.527, de 18 de novembro de 2011, regula o acesso a informações públicas em todos os âmbitos do Poder Público brasileiro. Este estudo objetiva discutir aspectos da LAI e sua relação com a gestão documental, além da necessidade da atuação do profissional arquivista nos arquivos públicos e privados. Método. A metodologia caracteriza-se por pesquisa bibliográfica e documental. Resultados. Fundamenta-se a revisão de literatura com abordagem sobre a LAI, a gestão documental arquivística e os ciclos de vida dos documentos. Conclusões. Conclui-se que a ausência da gestão documental nas diferentes fases arquivísticas, dificulta a aplicação da LAI, assim como a ausência do profissional arquivista para organizar a massa documental e disponibilizá-la aos usuários.Objetivo. La Ley de Acceso a la Información (LAI) n. 12.527, de 18 de noviembre de 2011, regula el acceso a la información pública en todos los ámbitos del Poder Público brasileño. Este estudio tiene como propósito discutir aspectos de la LAI y su relación con la gestión documental, además de la necesidad de la atuación del profesional archivista en los archivos públicos y privados. Método. La metodología se caracteriza por la investigación bibliográfica y documental. Resultados. Se fundamenta la revisión de literatura con abordaje sobre a LAI, la gestión documental archivística y los ciclos de vida de los documentos. Conclusiones. Se concluye que la ausencia de la gestión documental en las diferentes fases archivísticas, dificulta la aplicación de la LAI, así como la ausencia del profesional archivista para organizar la masa documental y disponibilizarla a los usuarios.Objective. It concludes that the absence of archival document management in different phases, hinders the implementation of LAI, as well as the absence of professional archivist to organize the mass of documents and make it available to users

    Proteomic Techniques in the Physiological Proteomics Core Facility

    poster abstractA new software package, IdentiQuantXL, has been developed in the Physiological Proteomics Core Facility to provide large-scale protein identification and label-free quantification using either low or high resolution LC-MS/MS data. Though many software packages have been developed to perform label-free quantification of proteins in complex biological samples using peptide intensities generated by liquid chromatography - tandem mass spectrometry (LC-MS/MS), two important issues hinder the use of peptide intensity measurements: (i) It is difficult to accurately determine the retention time of each peptide peak, especially for low resolution data, and (ii) many peptides cannot be used for protein quantification. To address these two key issues, we have developed a new method to enable accurate peptide peak retention time determination and multiple filters to eliminate unqualified peptides for protein quantification. Repeatability and linearity have been tested using ion trap-derived low resolution data from six very different samples, i.e., standard peptides, kidney tissue lysates, HT29-MTX cell lysates, depleted human serum, human serum albumin-bound proteins, and standard proteins spiked in kidney tissue lysates. In all these unique experiments, at least 90.8% of proteins (up to 1,390) had CVs ≤ 3 0% across 10 technique replicates, and at least 92.1% of proteins (up to 2,013) had R2 ≥ 0.9500 across 7 concentrations. The performance of our strategy was verified using identical amounts of standard protein (lysozyme) spiked in complex biological samples (cell culture media containing secreted proteins) with a CV of 8.6% across eight injections. The excellent performance was further confirmed by comparing label-free mass spectrometry to Western blot detection of prolactin, which was decreased 17.1fold in dwarfed mice compared to wild-type using the label-free quantification strategy and very low or undetectable using Western blot. The results indicate that our new platform, named IdentiQuantXL, accurately quantifies thousands of peptides and proteins in complex samples. It has been applied in the aqueous humor proteome in patients with Fuchs endothelial corneal dystrophy. While many software packages focus only on high resolution data, our strategy is designed for both high and low resolution data. Consequently, it is very useful for data generated by low resolution mass spectrometers such as the LTQ, especially when the dynamic exclusion of ions in data acquisition is enabled to obtain more MS/MS fragments of low-abundance peptides to maximally identify proteins in a complex biological sample.     Skrining Fitokimia Dan Uji Aktivitas Antioksidan Bunga Lai (Durio kutejensis)

    ABSTRAKPenelitian ini bertujuan untuk mengidentifikasi/ skrining fitokimia dan uji aktivitas antioksidan Bunga Lai (Durio kutejensis). Bunga Lai memiliki potensi untuk dapat dikembangkan menjadi salah satu produk sedian farmasi, namun Bunga Lai masih kurang dikenal jika dibandingkan dengan jenis tanaman Durio zibethinus. Dalam penelitian ini, peneliti menggunakan menggunakan metode kualitatif dan kuantitatif yang bersifat eksperimental. Pada pengujian skrining fitokomia menggunakan metode kualitatif dan uji aktivitas antioksidan menggunakan metode kuantitatif. Metode skrining fitokimia dilakukan dengan melihat reaksi pengujian warna dengan menggunakan suatu pereaksi warna dan uji aktivitas antioksidan  menggunakan metode DPPH yaitu dengan cara mengukur penangkap radikal sintetik dalam pelarut polar. Metode DPPH (Difenil pikrilhidrazil) untuk mengukur daya peredaman sampel (ekstrak) terhadap radikal bebas DPPH yang akan bereaksi dengan atom hidrogen dari senyawa peredaman radikal bebas membentuk DPPH yang lebih stabil. Pada pengujian antioksidan bunga lai (Durio kutejensis) dengan menggunkan ekstrsk kental dengan kosentrasi berbeda beda yaitu : 10 mg/L, 20 mg/L, 40 mg/L, 80 mg/L, 100 mg/L, 150 mg/L. Didapatkan hasil % inhibisi Ekstrak Bunga Lai  berturut-turut 68,16% untuk formula I, 56,25% untuk formula II, dan 41,21% untuk formula III, sedangkan nilai IC50 sebesar 2,38 ppm. Berdasarkan hasil penelitian, aktivitas antioksidan ekstrak bunga Lai memiliki nilai IC50 yang lebih rendah dibandingkan vitamin C. Hal ini menunjukkan bahwa aktivitas antioksidan pada bunga Lai lebih tinggi dibandingkan dengan Vitamin C. Dapat disimpulkan, dilihat dari aktivitas antioksidannya ekstrak bunga Lai sangat berpotensi sebagai alternatif bahan antioksidan alami. Kata kunci : Durio; Kutejensis; Lai; Bunga Durian. ABSTRACTThis study aims to identify/screen phytochemicals and test the antioxidant activity of Flower Lai (Durio kutejensis). Flower Lai has the potential to be developed into a pharmaceutical product, but Flower Lai is still less well known when compared to Durio zibethinus. In this study, researchers used qualitative and quantitative methods that were experimental. The phytochemical screening test used qualitative methods and the antioxidant activity test used quantitative methods. The phytochemical screening method was carried out by observing the color test reaction using a color reagent and the antioxidant activity test using the DPPH method, namely by measuring synthetic radical scavengers in polar solvents. The DPPH (Diphenyl picrylhydrazil) method is used to measure the reducing power of the sample (extract) against DPPH free radicals which will react with hydrogen atoms of free radical scavenging compounds to form more stable DPPH. Free radical scavenging compounds that react with DPPH will become new radicals that are more stable or non-radical compounds. In testing the antioxidants of other flowers (Durio kutejensis) using a thick extract with different concentrations, namely: 10 mg/L, 20 mg/L, 40 mg/L, 80 mg/L, 100 mg/L, 150 mg/L. The results of the % inhibition of Flower Lai Extract were 68.16% for formula I, 56.25% for formula II, and 41.21% for formula III, while the IC50 value was 2.38 ppm. Based on the results of the study, the antioxidant activity of Lai flower extract has a lower IC50 value than vitamin C. This indicates that the antioxidant activity of Lai flower is higher than that of Vitamin C. Judging from its antioxidant activity, Lai flower extract has the potential as an alternative natural antioxidant. Keywords : Durio; Kutejensis; Lai; Flower Lai

    Uso de fotos hemiesféricas para la elaboración de modelos de altura de la regeneración en Nothofagus pumilio

UNPA-INTA-CONICET, Río Gallegos, Argentina.El crecimiento en altura de la regeneración es la principal variable empleada para caracterizar la continuidad de bosques primarios y manejados. Entre los condicionantes bióticos y abióticos, la cobertura del dosel determina la cantidad de luz disponible. Se desarrollaron modelos de predicción de altura de regeneración de Nothofagus pumilio en relación con parámetros de cobertura y radiación obtenidos mediante fotos hemiesféricas, para bosques primarios y manejados. Se emplearon 3700 pares de datos (edad-altura) obtenidos durante 2002-2007 en parcelas permanentes de muestreo (n=16), caracterizadas con fotos hemiesféricas empleando un lente Sigma 8mm y analizadas usando el programa Gap Light Analyzer v.2.0. Se ajustaron modelos (Altura=a.Edadb.(c+d.X+e.X²)) por medio de técnicas de regresión no-linear que utilizaron como variables independientes a la altura y la edad de las plántulas, y como variables dependientes a la cobertura de copas (CC), el índice de área foliar efectivo (IAF), la radiación global (RG) y el porcentaje de radiación global incidente (PRG). Los modelos ajustados tuvieron un ajuste aceptable (r²=61%-63%) generando curvas polimórficas donde H aumentó hasta un máximo (20-25 cm en plántulas de 6 en CC 55%, LAI 1.0, GR 12.0 W.m² o PGR 50%) para luego decrecer en todas las edades analizadas con mayores o menores valores de dichas variables. Los bosques primarios presentaron CC=89.5%, LAI=2.6, GR=3.5 W.m² y PGR=14.3%, mientras que la corta de protección presentó CC=56.5%, LAI=0.8, GR=14.1 W.m² and PGR= 57.2%.
