Purpose: To evaluate the effect of the essential oil of Origanum vulgare Linnaeus (Lamiaceae) on the growth of Sporothrix schenckii and Sporothrix brasiliensis. Methods: The chemical composition of the essential oil was investigated by gas chromatography/flame ionization detector (GC-FID). The minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) were determined by broth micro-dilution method. Scanning electron microscopy (SEM) was also performed to reveal morphological alterations in Sporothrix spp. cells. Results: The major components of the essential oil were γ-terpinene (30.5%), carvacrol (15.7 %) and 4-terpineol (13.0 %). γ-Terpinene showed potential antifungal activity with MIC ranging from 62.5 to 500.0 μg mL-1 for S. schenckii, and 125.0 to 250.0 μg mL-1 for S. brasiliensis. SEM micrographs revealed morphological alterations in hyphae and reduction of the adhered conidia numbers. Conclusion: Origanum vulgare Linnaeus essential oil possesses potential antifungal activity, and can, therefore, can be developed as an alternative agent for the treatment of sporotrichosis. Keywords: Antifungal Activity, Essential Oil, Gas Chromatography, Origanum vulgare, Sporotrichosi

This is the final version. Available on open access from Inter-Research via the DOI in this recordKnowledge of life history parameters is essential for the effective management of species of conservation concern. For migratory marine vertebrates such as hawksbill sea turtles Eretmochelys imbricata, feeding aggregations are important developmental habitats, allowing the study of population dynamics. Here, we used data from a 31 yr mark-recapture study of juvenile hawksbill sea turtles in the Fernando de Noronha Archipelago, Brazil, to estimate key demographic parameters. Turtles recruit to the neritic habitat at similar sizes to those of other Atlantic hawksbill aggregations. The curved carapace length (CCL) at the first capture ranged from 28 to 84 cm (mean ± SD: 44.6 ± 9.8 cm). Median minimum residence time of turtles captured at least twice was 3.2 yr, whilst long-term minimum residence of up to 14 yr was also observed, with turtles showing site fidelity within the archipelago. The mean size of turtles captured was constant throughout time. Turtles grew on average 3.4 ± 2.2 cm yr-1, with a monotonic expected growth rate function generally decreasing with increasing size. At these rates, hawksbill turtles in Fernando de Noronha would need to spend ca. 14-18 yr to reach minimum adult breeding size (~74 cm CCL). This mark-recapture study has been essential to understanding the ecology and demographic parameters of this regional hawksbill turtle neritic foraging ground.National Council for Scientific and Technological Development (CNPq), Brazi

Background: The aim of this study was to report the overall survival and baseline factors associated with OS for breast, cervical and ovarian cancer in Florianópolis, Southern Brazil, a region with quality-of-life indicators comparable to high-income countries. Methods: Cohort study was performed from probabilistic record linkage of the Mortality Information System and the Population-based cancer registry of Florianópolis. It was included breasts, cervical and ovarian cancer diagnosis during the period of 2008–2012 with a follow up of 60 months. Cox regression and Kaplan-Meier method were used for associations with overall survival and risk factors. Findings: 1857 cases of the three malignancies were included in the analysis. We identified 202 deaths in breast cancer subjects, 53 for cervical cancer and 51 for ovarian cancer. Metastatic disease at diagnosis was present in 31%, 9.6%, and 55% of the cases, respectively. Overall survival was statistically correlated with age, educational level and stage for breast cancer; age and stage for cervical cancer; age and stage for ovarian cancer. Interpretation: Metastatic disease and age are the main prognostic factors for the malignancies studied, as they were associated with both overall survival and risk of death. Better screening and preventive tests for early diagnosis are needed. Cytochemical techniques and energy-filtering transmission electron microscopy applied to the study of parasitic protozoa

    Get PDF
The study of parasitic protozoa plays a major role in cell biology, biochemistry and molecular biology. Numerous cytochemical techniques have been developed in order to unequivocally identify the nature of subcellular compartments. Enzyme and immuno-cytochemistry allow the detection of, respectively, enzymatic activity products and antigens in particular sites within the cell. Energy-filtering transmission electron microscopy permits the detection of specific elements within such compartments. These approaches are particularly useful for studies employing antimicrobial agents where cellular compartments may be destroyed or remarkably altered and thus hardly identified by standard methods of observation. In this regard cytochemical and spectroscopic techniques provide valuable data allowing the determination of the mechanisms of action of such compounds

Extension of Yeast Chronological Lifespan by Methylamine

Background: Chronological aging of yeast cells is commonly used as a model for aging of human post-mitotic cells. The yeast Saccharomyces cerevisiae grown on glucose in the presence of ammonium sulphate is mainly used in yeast aging research. We have analyzed chronological aging of the yeast Hansenula polymorpha grown at conditions that require primary peroxisome metabolism for growth. Methodology/Principal Findings: The chronological lifespan of H. polymorpha is strongly enhanced when cells are grown on methanol or ethanol, metabolized by peroxisome enzymes, relative to growth on glucose that does not require peroxisomes. The short lifespan of H. polymorpha on glucose is mainly due to medium acidification, whereas most likely ROS do not play an important role. Growth of cells on methanol/methylamine instead of methanol/ammonium sulphate resulted in further lifespan enhancement. This was unrelated to medium acidification. We show that oxidation of methylamine by peroxisomal amine oxidase at carbon starvation conditions is responsible for lifespan extension. The methylamine oxidation product formaldehyde is further oxidized resulting in NADH generation, which contributes to increased ATP generation and reduction of ROS levels in the stationary phase. Conclusion/Significance: We conclude that primary peroxisome metabolism enhanced chronological lifespan of H. polymorpha. Moreover, the possibility to generate NADH at carbon starvation conditions by an organic nitrogen source supports further extension of the lifespan of the cell. Consequently, the interpretation of CLS analyses in yeast should include possible effects on the energy status of the cell.

    Magnetism in Gd-W films

Vapor condensation techniques are useful to prepare magnetic alloys whose components have low or even negligible equilibrium mutual solubility. In this work, one of these techniques-sputtering-was used to obtain Gd(x)W(1-x) alloys whose magnetic properties were investigated as a function of the Gd atomic concentration x. Gadolinium and various Gd-based alloys are promising materials for magnetic refrigeration and this was one of the motivations for this study. The Gd(x)-W(1-x) films were sputter deposited from Gd and W targets with x ranging from 0 to 1 as determined by x-ray energy-dispersive spectroscopic analyses. X-ray diffraction patterns indicate that crystalline structures were formed at low and high Gd concentrations, while at intermediate concentrations, the films were amorphous. Magnetization measurements, performed as a function of temperature and with static and alternating applied fields, reveal a spin glasslike behavior in all the W-containing samples for temperatures below the freezing temperature T(f). For low and intermediate Gd concentrations, and for T>T(f), the films were paramagnetic, while a ferromagnetic phase was observed in the Gd-W alloy of the highest Gd content. The magnetocaloric effect was investigated from the magnetization isotherms M versus H, from which the isothermal magnetic entropy variation Delta S(M) as a function of T, for the removal of an applied field of 50 kOe, was determined. It was observed that the maximum value of Delta S(M) for each Delta S(M) versus T curve and the temperature at which these maxima occur, are strongly dependent on x. Evidence for distinct coastal and offshore communities of bottlenose dolphins in the north east Atlantic.

Bottlenose dolphin stock structure in the northeast Atlantic remains poorly understood. However, fine scale photo-id data have shown that populations can comprise multiple overlapping social communities. These social communities form structural elements of bottlenose dolphin (Tursiops truncatus) [corrected] populations, reflecting specific ecological and behavioural adaptations to local habitats. We investigated the social structure of bottlenose dolphins in the waters of northwest Ireland and present evidence for distinct inshore and offshore social communities. Individuals of the inshore community had a coastal distribution restricted to waters within 3 km from shore. These animals exhibited a cohesive, fission-fusion social organisation, with repeated resightings within the research area, within a larger coastal home range. The offshore community comprised one or more distinct groups, found significantly further offshore (>4 km) than the inshore animals. In addition, dorsal fin scarring patterns differed significantly between inshore and offshore communities with individuals of the offshore community having more distinctly marked dorsal fins. Specifically, almost half of the individuals in the offshore community (48%) had characteristic stereotyped damage to the tip of the dorsal fin, rarely recorded in the inshore community (7%). We propose that this characteristic is likely due to interactions with pelagic fisheries. Social segregation and scarring differences found here indicate that the distinct communities are likely to be spatially and behaviourally segregated. Together with recent genetic evidence of distinct offshore and coastal population structures, this provides evidence for bottlenose dolphin inshore/offshore community differentiation in the northeast Atlantic. We recommend that social communities should be considered as fundamental units for the management and conservation of bottlenose dolphins and their habitat specialisations
