    Endothelial Connexin37 and Connexin40 participate in basal but not agonist-induced NO release.

    BACKGROUND: Connexin37 (Cx37) and Cx40 are crucial for endothelial cell-cell communication and homeostasis. Both connexins interact with endothelial nitric oxide synthase (eNOS). The exact contribution of these interactions to the regulation of vascular tone is unknown. RESULTS: Cx37 and Cx40 were expressed in close proximity to eNOS at cell-cell interfaces of mouse aortic endothelial cells. Absence of Cx37 did not affect expression of Cx40 and a 50 % reduction of Cx40 in Cx40(+/-) aortas did not affect the expression of Cx37. However, absence of Cx40 was associated with reduced expression of Cx37. Basal NO release and the sensitivity for ACh were decreased in Cx37(-/-) and Cx40(-/-) aortas but not in Cx40(+/-) aortas. Moreover, ACh-induced release of constricting cyclooxygenase products was present in WT, Cx40(-/-) and Cx40(+/-) aortas but not in Cx37(-/-) aortas. Finally, agonist-induced NO-dependent relaxations and the sensitivity for exogenous NO were not affected by genotype. CONCLUSIONS: Cx37 is more markedly involved in basal NO release, release of cyclooxygenase products and the regulation of the sensitivity for ACh as compared to Cx40

    Design Space Exploration for Partially Reconfigurable Architectures in Real-Time Systems

    International audienceIn this paper, we introduce FoRTReSS (Flow for Reconfigurable archiTectures in Real-time SystemS), a methodology for the generation of partially reconfigurable architectures with real-time constraints, enabling Design Space Exploration (DSE) at the early stages of the development. FoRTReSS can be completely integrated into existing partial reconfiguration flows to generate physical constraints describing the architecture in terms of reconfigurable regions that are used to floorplan the design, with key metrics such as partially reconfigurable area, real-time or external fragmentation. The flow is based upon our SystemC simulator for real-time systems that helps develop and validate scheduling algorithms with respect to application timing constraints and partial reconfiguration physical behaviour. We tested our approach with a video stream encryption/decryption application together with Error Correcting Code and showed that partial reconfiguration may lead to an area improvement up to 38% on some resources without compromising application performance, in a very small amount of time: less than 30 s

    A sulfur-rich pi-electron acceptor derived from 5,5 '-bithiazolidinylidene: charge-transfer complex vs. charge-transfer salt

    International audienceNovel pi-electron acceptors are still highly desirable for the formation of conducting salts or as n-dopable semiconductors. We describe here two synthetic approaches to substitute a dicyanovinylidene group, C=C(CN)(2) to a thioketone (C=S) in the recently described DEBTTT acceptor where DEBTTT stands for (E)-3,3'-diethyl-5,5'-bithiazolidinylidene-2,4,2',4'-tetrathione. These electron withdrawing groups enhance the electron accepting ability as demonstrated through electrochemical investigations, without hindering the formation of short intra-and intermolecular S center dot center dot center dot S contacts in the solid state. Association of this acceptor 1 with tetramethyltetrathiafulvalene (TMTTF) and decamethylferrocene (Fe(Cp*)(2)) afforded 1 : 1 adducts which were analyzed by single crystal X-ray diffraction. Combined with vibrational and magnetic properties, it appears that [TMTTF][1] behaves as a neutral charge-transfer complex while [Fe(Cp*)(2)][1] is an ionic salt. The concentration of the spin density on the exocyclic sulfur atoms in 1(-center dot) favors the setting of direct anti-ferromagnetic interactions in [Fe(Cp*)(2)][1

    Endothelial connexin40 (Cx40) contributes to regulate the structure and function of vessels. We have examined whether the protein also modulates the altered growth of vessels in tumor models established in control mice (WT), mice lacking Cx40 (Cx40-/-), and mice expressing the protein solely in endothelial cells (Tie2-Cx40). Tumoral angiogenesis and growth were reduced, whereas vessel perfusion, smooth muscle cell (SMC) coverage and animal survival were increased in Cx40-/- but not Tie2-Cx40 mice, revealing a critical involvement of endothelial Cx40 in transformed tissues independently of the hypertensive status of Cx40-/- mice. As a result, Cx40-/- mice bearing tumors survived significantly longer than corresponding controls, including after a cytotoxic administration. Comparable observations were made in WT mice injected with a peptide targeting Cx40, supporting the Cx40 involvement. This involvement was further confirmed in the absence of Cx40 or by peptide-inhibition of this connexin in aorta-sprouting, matrigel plug and SMC migration assays, and associated with a decreased expression of the phosphorylated form of endothelial nitric oxide synthase. The data identify Cx40 as a potential novel target in cancer treatment

    Smooth muscle tone is controlled by Ca(2+) signaling in the endothelial layer. Mouse endothelial cells are interconnected by gap junctions made of Connexin40 (Cx40) and Cx37, which allow the exchange of signaling molecules to coordinate their activity. Here, we investigated the role of Cx40 in the endothelial Ca(2+) signaling of the mouse aorta. Ca(2+) imaging was performed on intact aortic endothelium from both wild type (Cx40+/+) and Connexin40-deficient (Cx40 -/-) mice. Acetylcholine (ACh) induced early fast and high amplitude Ca(2+) transients in a fraction of endothelial cells expressing the M3 muscarinic receptors. Inhibition of intercellular communication using carbenoxolone or octanol fully blocked the propagation of ACh-induced Ca(2+) transients toward adjacent cells in WT and Cx40-/- mice. As compared to WT, Cx40-/- mice displayed a reduced propagation of ACh-induced Ca(2+) waves, indicating that Cx40 contributes to the spreading of Ca(2+) signals. The propagation of those Ca(2+) responses was not blocked by suramin, a blocker of purinergic ATP receptors, indicating that there is no paracrine effect of ATP release on the Ca(2+) waves. Altogether our data show that Cx40 and Cx37 contribute to the propagation and amplification of the Ca(2+) signaling triggered by ACh in endothelial cells expressing the M3 muscarinic receptors

    Zebra : Building Efficient Network Message Parsers for Embedded Systems

To overcome this issue, a promising approach is to build parsers directly in hardware. Unfortunately, developing such parsers is a daunting task for most developers as it is at the crossroads of several areas of expertise, such as low-level network programming, or hardware design. In this paper, we propose Zebra, a generative approach to drastically ease the development of hardware parsers and their use in network applications. To validate our approach, we have used Zebra to generate hardware parsers for widely used protocols, namely HTTP, SMTP, SIP, and RTSP. Our experiments show that Zebra-based parsers are up to 11 times faster than software-based parsers

    PESSTO monitoring of SN 2012hn: further heterogeneity among faint type I supernovae

    We present optical and infrared monitoring data of SN 2012hn collected by the Public ESO Spectroscopic Survey for Transient Objects (PESSTO). We show that SN 2012hn has a faint peak magnitude (MR ~ -15.7) and shows no hydrogen and no clear evidence for helium in its spectral evolution. Instead, we detect prominent Ca II lines at all epochs, which relates this transient to previously described 'Ca-rich' or 'gap' transients. However, the photospheric spectra (from -3 to +32 d with respect to peak) of SN 2012hn show a series of absorption lines which are unique, and a red continuum that is likely intrinsic rather than due to extinction. Lines of Ti II and Cr II are visible. This may be a temperature effect, which could also explain the red photospheric colour. A nebular spectrum at +150d shows prominent CaII, OI, CI and possibly MgI lines which appear similar in strength to those displayed by core-collapse SNe. To add to the puzzle, SN 2012hn is located at a projected distance of 6 kpc from an E/S0 host and is not close to any obvious starforming region. Overall SN 2012hn resembles a group of faint H-poor SNe that have been discovered recently and for which a convincing and consistent physical explanation is still missing. They all appear to explode preferentially in remote locations offset from a massive host galaxy with deep limits on any dwarf host galaxies, favouring old progenitor systems. SN 2012hn adds heterogeneity to this sample of objects. We discuss potential explosion channels including He-shell detonations and double detonations of white dwarfs as well as peculiar core-collapse SNe.Comment: 14 pages, 14 figures, accepted to MNRAS on 14/10/201

    Subharmonic bifurcation cascade of pattern oscillations caused by winding number increasing entrainment

    Convection structures in binary fluid mixtures are investigated for positive Soret coupling in the driving regime where solutal and thermal contributions to the buoyancy forces compete. Bifurcation properties of stable and unstable stationary square, roll, and crossroll (CR) structures and the oscillatory competition between rolls and squares are determined numerically as a function of fluid parameters. A novel type of subharmonic bifurcation cascade (SC) where the oscillation period grows in integer steps as n(2π)/(ω)n (2\pi)/(\omega) is found and elucidated to be an entrainment process.Comment: 7 pages, 4 figure

    The supernova CSS121015:004244+132827: a clue for understanding super-luminous supernovae

    We present optical photometry and spectra of the super luminous type II/IIn supernova CSS121015:004244+132827 (z=0.2868) spanning epochs from -30 days (rest frame) to more than 200 days after maximum. CSS121015 is one of the more luminous supernova ever found and one of the best observed. The photometric evolution is characterized by a relatively fast rise to maximum (~40 days in the SN rest frame), and by a linear post-maximum decline. The light curve shows no sign of a break to an exponential tail. A broad Halpha is first detected at ~ +40 days (rest-frame). Narrow, barely-resolved Balmer and [O III] 5007 A lines, with decreasing strength, are visible along the entire spectral evolution. The spectra are very similar to other super luminous supernovae (SLSNe) with hydrogen in their spectrum, and also to SN 2005gj, sometimes considered a type Ia interacting with H-rich CSM. The spectra are also similar to a subsample of H-deficient SLSNe. We propose that the properties of CSS121015 are consistent with the interaction of the ejecta with a massive, extended, opaque shell, lost by the progenitor decades before the final explosion, although a magnetar powered model cannot be excluded. Based on the similarity of CSS121015 with other SLSNe (with and without H), we suggest that the shocked-shell scenario should be seriously considered as a plausible model for both types of SLSN.Comment: 17 pages, 10 figures and 5 tables.     An Unbiased ALMA Spectral Survey of the LkCa 15 and MWC 480 Protoplanetary Disks

    The volatile contents of protoplanetary disks both set the potential for planetary chemistry and provide valuable probes of defining disk system characteristics such as stellar mass, gas mass, ionization, and temperature structure. Current disk molecular inventories are fragmented, however, giving an incomplete picture: unbiased spectral line surveys are needed to assess the volatile content. We present here an overview of such a survey of the protoplanetary disks around the Herbig Ae star MWC 480 and the T Tauri star LkCa 15 in ALMA Band 7, spanning ~36 GHz from 275 to 317 GHz and representing an order of magnitude increase in sensitivity over previous single-dish surveys. We detect 14 molecular species (including isotopologues), with five species (C34S, 13CS, H2CS, DNC, and C2D) detected for the first time in protoplanetary disks. Significant differences are observed in the molecular inventories of MWC 480 and LkCa 15, and we discuss how these results may be interpreted in light of the different physical conditions of these two disk systems
