Pharmacokinetic profiles of the active metamizole metabolites in healthy horses
Metamizole (MT) is an analgesic and antipyretic drug labelled for use in humans, horses, cattle, swine and dogs. MT is rapidly hydrolysed to the active primary metabolite 4-methylaminoantipyrine (MAA). MAA is formed in much larger amounts compared with other minor metabolites. Among other secondary metabolites, 4-aminoantipyrine (AA) is also relatively active. The aim of this research was to evaluate the pharmacokinetic profiles of MAA and AA after dose of 25 mg/kg MT by intravenous (i.v.) and intramuscular (i.m.) routes in healthy horses. Six horses were randomly allocated to two equally sized treatment groups according to a 2 9 2 crossover study design. Blood was collected at predetermined times within 24 h, and plasma was analysed by a validated HPLC-UV method. No behavioural changes or alterations in health parameters were observed in the i.v. or i.m. groups of animals during or after (up to 7 days) drug administration. Plasma concentrations of MAA after i.v. and i.m. administrations of MT were detectable from 5 min to 10 h in all the horses. Plasma concentrations of AA were detectable in the same range of time, but in smaller amounts. Maximum concentration (Cmax), time to maximum concentration (Tmax) and AUMC0-last of MAA were statistically different between the i.v. and i.m. groups. The AUCIM/AUCIV ratio of MAA was 1.06. In contrast, AUC0-last of AA was statistically different between the groups (P < 0.05) with an AUCIM/AUCIV ratio of 0.54.
This study suggested that the differences in the MAA and AA plasma concentrations found after i.m. and i.v. administrations of MT might have minor consequences on the pharmacodynamics of the drug
Efeitos do etil-cianoacrilato ou do octil-cianoacrilato sobre lesões corneais experimentais em coelhos
The use of ethyl-cyanoacrylate and octyl-cyanoacrylate were clinically and histopathologically compared on the corneas of 36 rabbits after lamellar keratectomy (standardized diameter and depth). The animals were distributed into two groups, one for each type of adhesive. From each group, six subgroups were histopathologically evaluated on the 3rd, 7th, 14th, 21st, 30th, and 60th day post-operative. General (daily) and ophthalmic (days 0, 1, 3, 5, 7, 14, 21, 30, 44, and 60) evaluations clinically indicated that there were significant differences for the variables water intake, attitude, blepharitis, corneal edema, and fluorescein test. The adhesive permanence time for octyl-cyanoacrylate (17.22 days) was greater than that for ethyl-cyanoacrylate (7.66 days). With respect to the histopathological evaluation, corneal epithelization and collagen organization occurred without severe complications. However, treatment with ethyl-cyanoacrylate led to a moderate inflammatory reaction in the initial phases. With octyl-cyanoacrylate, re-epithelization and collagen organization proceeded more slowly with a discrete inflammatory reaction in the initial phases. From clinical and histopathologic points of view, octyl-cyanoacrylate showed advantages over ethyl-cyanoacrylate, whereas wound healing was achieved in both groups without major complications.Comparou-se o uso do etil-cianoacrilato e do octil-cianoacrilato em córneas de 36 coelhos após ceratectomia lamelar (diâmetro e profundidade padronizados). Os animais foram distribuídos em dois grupos, segundo o tipo de adesivo, e redistribuídos em seis subgrupos com três animais cada, para as avaliações histológicas aos 3, 7, 14, 21, 30 e 60 dias de pós-operatório. As avaliações clínicas gerais (diárias) e as oftálmicas (dias 0, 1, 3, 5, 7, 14, 21, 30, 44 e 60), indicaram diferença entre os dois grupos, quanto ao consumo de água, atitude, blefarite, edema da córnea e teste da fluoresceína. O Tempo de permanência, sobre o leito corneal, do adesivo octil-cianoacrilato (17,22 dias), foi maior que o do etil-cianoacrulato (7,66 dias). A histopatologia, para ambos os grupos, mostrou que a re-epitelização e a organização do colágeno ocorreram sem graves intercorrências. O grupo tratado com o etil-cianoacrilato apresentou, nas fases iniciais, reação inflamatória mais evidente que o tratado com octil-cianoacrilato. Neste, a re-epitelização e a organização do colágeno ocorreram mais lentamente e com reação inflamatória discreta. Sob os pontos de vista clínico e de avaliação histológica simples, os resultados mostraram vantagens do octil-cianoacrilato, entretanto, a cicatrização da córnea ocorreu em ambos os grupos

Interpenetrating Hydrogel Networks Enhance Mechanical Stability, Rheological Properties, Release Behavior and Adhesiveness of Platelet-Rich Plasma
Platelet‐rich plasma (PRP) has attracted much attention for the treatment of articular cartilage defects or wounds due to its intrinsic content of growth factors relevant for tissue repair. However, the short residence time of PRP in vivo, due to the action of lytic enzymes, its weak mechanical properties and the consequent short‐term release of bioactive factors has restricted its application and efficacy. The present work aimed at designing new formulation strategies for PRP, based on the use of platelet concentrate (PC)‐loaded hydrogels or interpenetrating polymer networks, directed at improving mechanical stability and sustaining the release of bioactive growth factors over a prolonged time‐span. The interpenetrating hydrogels comprised two polymer networks interlaced on a molecular scale: (a) a first covalent network of thermosensitive and biodegradable vinyl sulfone bearing p(hydroxypropyl methacrylamide‐lacate)‐polyethylene glycol triblock copolymers, tandem cross‐linked by thermal gelation and Michael addition when combined with thiolated hyaluronic acid, and (b) a second network composed of cross‐linked fibrin. The PC‐loaded hydrogels, instead, was formed only by network (a). All the designed and successfully synthesized formulations greatly increased the stability of PRP in vitro, leading to significant increase in degradation time and storage modulus of PRP gel. The resulting viscoelastic networks showed the ability to controllably release platelet derived growth factor and transforming growth factr β1, and to improve the tissue adhesiveness of PRP. The newly developed hydrogels show great potential for application in the field of wound healing, cartilage repair and beyond
Cell instructive Liquid Crystalline Networks for myotube formation
Development of biological tissues in vitro is not a trivial task and requires the correct maturation of the selected cell line. To this aim, many attempts were done mainly by mimicking the biological environment using micro/nanopatterned or stimulated scaffolds. However, the obtainment of functional tissues in vitro is still far from being achieved. In contrast with the standard methods, we here present an easy approach for the maturation of myotubes toward the reproduction of muscular tissue. By using liquid crystalline networks with different stiffness and molecular alignment, we demonstrate how the material itself can give favorable interactions with myoblasts helping a correct differentiation. Electrophysiological studies demonstrate that myotubes obtained on these polymers have more adult-like morphology and better functional features with respect to those cultured on standard supports. The study opens to a platform for the differentiation of other cell lines in a simple and scalable way

Antioxidant capacity of durum wheat large flour particles may be evaluated by QUENCHER<sub>ABTS</sub> assay by adopting a proper calculation mode
Assessment of Antioxidant Capacity (AC) of foods is useful to consider cumulative/ synergistic action of all dietary antioxidants, thus providing a more integrated information than the simple sum of measurable antioxidants. Among the different AC assays, the QUENCHERABTS (QUick, Easy, New, CHEap and Reproducible) procedure is based on the direct reaction of ABTS•+ reagent with fine solid food particles without extraction of antioxidants. This assay is able to measure both soluble and insoluble antioxidants, that simultaneously come into contact with ABTS•+ molecules by either liquid–liquid or solid–liquid interactions, respectively. These interactions may change depending on the particle diameter. Usually, particles having 0.1–0.3 mm size are used. Here, AC was evaluated on whole flour (WF), derived from a mix of grains of ten durum wheat varieties, characterized by three different particle sizes: a smaller one, ≤0.2 mm (control, WF0.2), and two larger ones, ≤0.5 mm and ≤1 mm (WF0.5 and WF1, respectively). Moreover, a novel AC calculation procedure based on the slope value of the regression line of ABTS•+ response vs flour amount is presented in detail. The classical QUENCHERABTS procedure provided for WF0.2 an AC value of 42.0±2.7 μmol eq. Trolox/g d.w. A similar result was obtained for WF0.5 (38.3±0.9 μmol eq. Trolox/g d.w.), thus indicating that these large particles may be analyzed by the QUENCHERABTS assay provided that the “slope” calculation procedure is used. On the contrary, WF1 showed about half AC (20.3±0.2 μmol eq. Trolox/g d.w.), thus showing that very large particles cannot be used even adopting the “slope” calculation
Hydroxy double salts intercalated with Mn(II) complexes as potential contrast agents
A series of Mn(II) aminophosphonate complexes were successfully synthesized and intercalated into the hydroxy double salt [Zn5(OH)8]Cl2·yH2O. Complex incorporation led to an increase in the interlayer spacing from 7.8 to 10–12 Å. Infrared spectroscopy showed the presence of the characteristic vibration peaks of the Mn(II) complexes in the intercalates' spectra, indicating successful incorporation. The complex-loaded composites had somewhat lower proton relaxivities than the pure complexes. Nevertheless, these intercalates may have use as MRI contrast agents for patients with poor kidney function, where traditional Gd(III)-based contrast agents cause severe renal failure
