    Sorghum [Sorghum bicolor (L.) Moench] Seed Quality as Affected by Variety, Harvesting Stage and Fertilizer Application in Bomet Country of Kenya

    A set of laboratory experiments were conducted to evaluate the physiological quality of Sorghum bicolor (L.) Moench seeds subjected to different field cultural management practices. The main aim of this study was to contribute towards improved production of Sorghum bicolor (L.) Moench through better understanding of the crop’s agronomy. The experimental design was a three factorial split-split block design in the rain fed Sorghum bicolor (L.) Moench producing areas of Bomet County of Kenya. Treatments within the block were randomized. There were three factors which included variety at two levels (improved and local varieties); fertilizer application at two levels (with and without fertilizer); and seed selection at three levels (continuous, from booting stage and at harvest). Percentages of crop emergence and days to 50% physiological maturity were determined. The following seed quality tests were carried out: 1000-seed weight, standard germination, mean germination time and electrical conductivity. The improved variety had a field emergence of 100% while that of the local variety ranged from 60 - 97%. In regard to field emergence and maturity, there were significance differences (P≤ 0.001) between plots with fertilizer and those without regardless of the variety. Those plots with fertilizer outperformed those without. Germination and vigour tests indicated that seed selection time did not influence seed vigour, viability and yield (P ≤ 0.05). However, there were significant differences (P ≤ 0.001) between the improved variety (E1291) and local (Ochuti) variety. In relation to this, variety E1291 showed better seed vigour, viability and yield as compared to Ochuti. Similarly, there were significant differences (P ≤ 0.001) between plots to which fertilizer was applied and those without. Fertilizer application led to an increase in seed yield, seed weight, seed vigour and viability. It was therefore concluded that (i) E1291 was more adapted to the study area as compared to Ochuti and hence should be adopted by farmers; (ii) farmers should apply optimal phosphate and nitrogenous fertilizers to their soils so as to increase the quality and yield of their seed; and (iii) farmers could continue selecting their seed using their indigenous knowledge.Key words: Seed, Sorghum bicolor, viability, vigour, yiel

    A survey of farmers’ perceptions and management strategies of the sweet potato weevil in Homa bay county, Kenya

    Sweet potato (Ipomoea batatas L.) is one of the most widely grown root cropsworldwide. In Africa, it is grown in small plots by poorer farmers. Production of thecrop is extremely low in Kenya as compared to other African countries due to theexistence of common insect pests. Sweet potato weevil (Cylas spp.) is known as thebiggest pit fall for production and productivity of the crop in the country. This studysought to determine the opinion of sweet potato farmers concerning sweet potatoresistance to Cylas spp. and determine control strategies employed by sweet potatofarmers in managing the pest. The study also sought to determine the sweet potatoproduction constraints faced by the farmers in Homa Bay County, Kenya. The studywas conducted using a Participatory Rural Appraisal approach in which 269 farmers inthe County were interviewed on the sweet potato varieties with field resistance to Cylasspp., the crops’ production constraints (with emphasis on damage by Cylas spp.) andfarmers’ control strategies in regard to the weevil. Data were also collected from thefarmers whose sample size was determined using the table on sample size selection andstandardization equation. Data were analysed using descriptive statistical techniquesthat were frequencies, percentages and standard errors. The study established thatmajority of the farmers from Rachuonyo (89.7%) and Ndhiwa (91.9%) were not awareof any variety that had field resistance to Cylas spp. However, 10.5% of the farmers inRachuonyo and 8.1% of the farmers in Ndhiwa identified nine varieties which haveshown relative field resistance to root damage by Cylas spp. The varieties that wereidentified to be resistant to Cylas spp. by farmers in Rachuonyo were Kalamb Nyerere,Tombra, Sinia, Odinga, Kemb 10, Wera and Zapallo. However, the varieties that wereidentified to be resistant to Cylas spp. by farmers in Ndhiwa were Amina, Mugandeand Ndege Oyiejo. Further, the findings revealed that Cylas spp. was the mostproblematic pest by 90.3% and 96.8% of households in Rachuonyo and Ndhiwa,respectively. Majority (64.5%) of the farmers in Ndhiwa did not use any methods tomanage Cylas spp. However, farmers in Rachuonyo (26.2%) and Ndhiwa (15.3%)preferred re-ridging during weeding as a management strategy in regard to Cylas spp.These findings reiterate the importance of the sweet potato weevil in rural sweet potatofarming systems and thus innovative management strategies are necessary.Key words: Sweet potato, control strategies, Cylas spp., resistance, pest, variety,constraint

    Analysis of the genetic diversity of selected East African sweet potato (Ipomea batatas [L.] Lam.) accessions using microsatellite markers

    Sweet potato (Ipomea batatas [L.] Lam.) is an economically important crop in East Africa chiefly grown by small holder farmers. Sharing of vines for planting is a very common occurrence among these farmers and eventually varieties are given local names, making it hard to trace the original pedigree. It is therefore important to characterise the sweet potato germplasm for purposes of breeding and germplasm conservation. In this study, 68 sweet potato accessions were evaluated for diversity using 12 microsatellite markers. The genetic relationship of the germplasm was evaluated using the Jaccard’s coefficient for dissimilarity analysis, unweighted pair group method with arithmetic means (UPGMA) tree and principal component analysis (PCoA) on DARwin software, while summary statistics was done using PowerMarker and Popgene softwares. The polymorphic information content of the markers ranged from 0.1046 for markers J67b and J67 to 0.3671 for marker J1809a, with a mean value of 0.2723. The total number of alleles amplified was 21. The major allele frequency ranged from 0.5882 for marker JB1809a to 0.9412 for markers J67b and J67c. Cluster analysis divided the accessions into four major clusters. Principle component analysis divided the accession into four groups which were different from those by cluster analysis. This study was able to identify several distinct accessions as well as a few possible duplicate accessions that overlapped on the cluster analysis.Key words: Sweet potato, cluster analysis, genetic diversity, principal component analysis, Simple SequenceRepeats

    Prognostic value of galectin-3, a novel marker of fibrosis, in patients with chronic heart failure: data from the DEAL-HF study

    Biomarkers are increasingly being used in the management of patients with chronic heart failure (HF). Galectin-3 is a recently developed biomarker associated with fibrosis and inflammation, and it may play a role in cardiac remodeling in HF. We determined its prognostic value in patients with chronic HF. Patients with chronic HF (New York Heart Association functional class III or IV) who participated in the Deventer-Alkmaar heart failure study were studied. Galectin-3 levels were determined at baseline using a novel optimized enzyme-linked immunosorbent assay. Univariate and multivariate analyses were used to determine the prognostic value of this biomarker. We studied 232 patients; their mean age was 71 +/- A 10 years, 72% were male, and 96% were in NYHA class III. During a follow-up period of 6.5 years, 98 patients died. Galectin-3 was a significant predictor of mortality risk after adjustment for age and sex, and severity of HF and renal dysfunction, as assessed by NT-proBNP and estimated glomerular filtration rate, respectively (hazard ratio per standard deviation 1.24, 95% CI 1.03-1.50, P = 0.026). Plasma galectin-3 is a novel prognostic marker in patients with chronic HF. Its prognostic value is independent of severity of HF, as assessed by NT-proBNP levels, and it may potentially be used in the management of such patients

    Latitudinal Patterns in European Seagrass Carbon Reserves: Influence of Seasonal Fluctuations versus Short-Term Stress and Disturbance Events

    Seagrass meadows form highly productive and valuable ecosystems in the marine environment. Throughout the year, seagrass meadows are exposed to abiotic and biotic variations linked to (i) seasonal fluctuations, (ii) short-term stress events such as, e.g., local nutrient enrichment, and (iii) small-scale disturbances such as, e.g., biomass removal by grazing. We hypothesized that short-term stress events and smallscale disturbances may affect seagrass chance for survival in temperate latitudes. To test this hypothesis we focused on seagrass carbon reserves in the form of starch stored seasonally in rhizomes, as these have been defined as a good indicator for winter survival. Twelve Zostera noltei meadows were monitored along a latitudinal gradient in Western Europe to firstly assess the seasonal change of their rhizomal starch content. Secondly, we tested the effects of nutrient enrichment and/or biomass removal on the corresponding starch content by using a short-term manipulative field experiment at a single latitude in the Netherlands. At the end of the growing season, we observed a weak but significant linear increase of starch content along the latitudinal gradient from south to north. This agrees with the contention that such reserves are essential for regrowth after winter, which is more severe in the north. In addition, we also observed a weak but significant positive relationship between starch content at the beginning of the growing season and past winter temperatures. This implies a lower regrowth potential after severe winters, due to diminished starch content at the beginning of the growing season. Short-term stress and disturbances may intensify these patterns, because our manipulative experiments show that when nutrient enrichment and biomass loss co-occurred at the end of the growing season, Z. noltei starch content declined. In temperate zones, the capacity of seagrasses to accumulate carbon reserves is expected to determine carbon-based regrowth after winter. Therefore, processes affecting those reserves might affect seagrass resilience. With increasing human pressure on coastal systems, short- and small-scale stress events are expected to become more frequent, threatening the resilience of seagrass ecosystems, particularly at higher latitudes, where populations tend to have an annual cycle highly dependent on their storage capacity

    The Role of Galectin-1 and Galectin-3 in the Mucosal Immune Response to Citrobacter rodentium Infection

    Despite their abundance at gastrointestinal sites, little is known about the role of galectins in gut immune responses. We have therefore investigated the Citrobacter rodentium model of colonic infection and inflammation in Galectin-1 or Galectin-3 null mice. Gal-3 null mice showed a slight delay in colonisation after inoculation with C. rodentium and a slight delay in resolution of infection, associated with delayed T cell, macrophage and dendritic cell infiltration into the gut mucosa. However, Gal-1 null mice also demonstrated reduced T cell and macrophage responses to infection. Despite the reduced T cell and macrophage response in Gal-1 null mice, there was no effect on C. rodentium infection kinetics and pathology. Overall, Gal-1 and Gal-3 play only a minor role in immunity to a gut bacterial pathogen

    Cancer of the breast: 5-year survival in a tertiary hospital in Uganda

    The objective was to investigate survival of breast cancer patients at Mulago Hospital. A retrospective study of the medical records of 297 breast cancer patients referred to the combined breast clinic housed in the radiotherapy department between 1996 and 2000 was done. The female/male ratio was 24 : 1. The age range was 22–85 years, with a median of 45 years and peak age group of 30–39 years. Twenty-three percent had early disease (stages 0–IIb) and 26% had metastatic disease. Poorly differentiated was the most common pathological grade (58%) followed by moderately differentiated (33%) and well-differentiated (9%) tumours. The commonest pathological type encountered was ‘not otherwise specified' (76%). Of all patients, 75% had surgery, 76% had radiotherapy, 60% had hormonotherapy and 29% had chemotherapy. Thirty-six (12%) patients received all the four treatment modalities. The 5-year survival probabilities (Kaplan–Meier) for early disease were 74 and 39% for advanced disease (P=0.001). The overall 5-year survival was 56%, which is lower than the rates in the South African blacks (64%) and North American whites (82–88%)

    Molecular biology of breast cancer metastasis: Genetic regulation of human breast carcinoma metastasis

    The present is an overview of recent data that describes the genetic underpinnings of the suppression of cancer metastasis. Despite the explosion of new information about the genetics of cancer, only six human genes have thus far been shown to suppress metastasis functionally. Not all have been shown to be functional in breast carcinoma. Several additional genes inhibit various steps of the metastatic cascade, but do not necessarily block metastasis when tested using in vivo assays. The implications of this are discussed. Two recently discovered metastasis suppressor genes block proliferation of tumor cells at a secondary site, offering a new target for therapeutic intervention

    Mortality from gastrointestinal congenital anomalies at 264 hospitals in 74 low-income, middle-income, and high-income countries: a multicentre, international, prospective cohort study

    Background: Congenital anomalies are the fifth leading cause of mortality in children younger than 5 years globally. Many gastrointestinal congenital anomalies are fatal without timely access to neonatal surgical care, but few studies have been done on these conditions in low-income and middle-income countries (LMICs). We compared outcomes of the seven most common gastrointestinal congenital anomalies in low-income, middle-income, and high-income countries globally, and identified factors associated with mortality. // Methods: We did a multicentre, international prospective cohort study of patients younger than 16 years, presenting to hospital for the first time with oesophageal atresia, congenital diaphragmatic hernia, intestinal atresia, gastroschisis, exomphalos, anorectal malformation, and Hirschsprung's disease. Recruitment was of consecutive patients for a minimum of 1 month between October, 2018, and April, 2019. We collected data on patient demographics, clinical status, interventions, and outcomes using the REDCap platform. Patients were followed up for 30 days after primary intervention, or 30 days after admission if they did not receive an intervention. The primary outcome was all-cause, in-hospital mortality for all conditions combined and each condition individually, stratified by country income status. We did a complete case analysis. // Findings: We included 3849 patients with 3975 study conditions (560 with oesophageal atresia, 448 with congenital diaphragmatic hernia, 681 with intestinal atresia, 453 with gastroschisis, 325 with exomphalos, 991 with anorectal malformation, and 517 with Hirschsprung's disease) from 264 hospitals (89 in high-income countries, 166 in middle-income countries, and nine in low-income countries) in 74 countries. Of the 3849 patients, 2231 (58·0%) were male. Median gestational age at birth was 38 weeks (IQR 36–39) and median bodyweight at presentation was 2·8 kg (2·3–3·3). Mortality among all patients was 37 (39·8%) of 93 in low-income countries, 583 (20·4%) of 2860 in middle-income countries, and 50 (5·6%) of 896 in high-income countries (p<0·0001 between all country income groups). Gastroschisis had the greatest difference in mortality between country income strata (nine [90·0%] of ten in low-income countries, 97 [31·9%] of 304 in middle-income countries, and two [1·4%] of 139 in high-income countries; p≤0·0001 between all country income groups). Factors significantly associated with higher mortality for all patients combined included country income status (low-income vs high-income countries, risk ratio 2·78 [95% CI 1·88–4·11], p<0·0001; middle-income vs high-income countries, 2·11 [1·59–2·79], p<0·0001), sepsis at presentation (1·20 [1·04–1·40], p=0·016), higher American Society of Anesthesiologists (ASA) score at primary intervention (ASA 4–5 vs ASA 1–2, 1·82 [1·40–2·35], p<0·0001; ASA 3 vs ASA 1–2, 1·58, [1·30–1·92], p<0·0001]), surgical safety checklist not used (1·39 [1·02–1·90], p=0·035), and ventilation or parenteral nutrition unavailable when needed (ventilation 1·96, [1·41–2·71], p=0·0001; parenteral nutrition 1·35, [1·05–1·74], p=0·018). Administration of parenteral nutrition (0·61, [0·47–0·79], p=0·0002) and use of a peripherally inserted central catheter (0·65 [0·50–0·86], p=0·0024) or percutaneous central line (0·69 [0·48–1·00], p=0·049) were associated with lower mortality. // Interpretation: Unacceptable differences in mortality exist for gastrointestinal congenital anomalies between low-income, middle-income, and high-income countries. Improving access to quality neonatal surgical care in LMICs will be vital to achieve Sustainable Development Goal 3.2 of ending preventable deaths in neonates and children younger than 5 years by 2030
