    Growth and yield responses to amending the sugarcane monoculture: interactions between break history and nitrogen fertiliser

    Experiments were established in the Burdekin Irrigation Area in North Queensland, Australia, to measure whether yield improvements from breaking the sugarcane monoculture or fumigating the soil could be modified by the application of different rates of nitrogen (N) fertiliser. Experiments were conducted in consecutive crop cycles (phase 1, planted 1998; phase 2, planted 2001) using the variety Q117, with the interaction between N applications and rotation histories discussed for the two plant crops. Histories consisted of alternate crop, bare fallow or mixed grass-legume pastures for periods of 42-66 months, compared with continuous cane as plough-out replant without (PORP) or with (PORP-F) soil fumigation. The N strategies involved combinations of N rates (0-180kgN/ha) and application times (at planting, 90 days after planting (DAP) or split between these times) in phase 1 and N rates (0-300kgN/ha) in phase 2. Histories had differing effects on N available to the cane crop and hence on response to N fertiliser. Some combinations of history and N rate were N-limited and strong linear relationships between biomass production or cane yield and crop N content could be developed. Critical N contents for biomass production (R2≤0.93) and fresh-weight cane yield (R2≤0.88) were 1.42 and 0.57kgN/t, respectively. Application of N fertiliser was shown to have significant impacts on both tiller addition and the retention of tillers to produce harvestable stalks. However, the application of fertiliser N had limited (phase 1) or no (phase 2) capacity to provide the quantum of yield response in soil health benefits associated with breaking the sugarcane monoculture. Increasing N application rates above that required to optimise crop yield resulted in significant decreases in sugar content of cane and thus lower sugar yields. Yield increases solely from improved soil health (i.e. exclusive of N response) constituted advantages averaging 15% (phase 1) to 20% (phase 2) compared with PORP. These effects were manifest early in the establishment of primary shoots in the plant crops, although the longevity of these benefits was limited. Replanting cane after a 3-year crop cycle (plant, 1st and 2nd ratoon) on land that had been under pasture, crop, bare fallow or PORP-F histories (phase 2, cycle 2) showed carryover effects of histories on N availability and fertiliser N responsiveness, but limited yield impacts attributable to residual soil health benefits. These results reinforce the importance of crop rotation during breaks between sugarcane cycles to maintain soil health and improve crop productivity

    Inhibition of autophagy impairs tumor cell invasion in an organotypic model

    Autophagy is a membrane-trafficking process that delivers cytoplasmic constituents to lysosomes for degradation. It contributes to energy and organelle homeostasis and the preservation of proteome and genome integrity. Although a role in cancer is unquestionable, there are conflicting reports that autophagy can be both oncogenic and tumor suppressive, perhaps indicating that autophagy has different roles at different stages of tumor development. In this report, we address the role of autophagy in a critical stage of cancer progression—tumor cell invasion. Using a glioma cell line containing an inducible shRNA that targets the essential autophagy gene Atg12, we show that autophagy inhibition does not affect cell viability, proliferation or migration but significantly reduces cellular invasion in a 3D organotypic model. These data indicate that autophagy may play a critical role in the benign to malignant transition that is also central to the initiation of metastasis

    Advanced chronic liver disease in the last year of life: a mixed methods study to understand how care in a specialist liver unit could be improved

    Objective: To identify the limitations in palliative care provision in the last year of life for people with liver cirrhosis and potential barriers to and enablers of palliative care. / Design: Mixed methods, including a retrospective case note review, qualitative focus groups and individual interviews. / Setting: A tertiary referral liver centre in the south of England (UK). / Participants: Purposively selected case notes of 30 people with cirrhosis who attended the tertiary referral liver centre and died during an 18-month period; a purposive sample of 22 liver health professionals who participated in either focus groups or individual interviews. / Primary and secondary outcomes: Data collected from case notes included hospital admissions, documented discussions of prognosis and palliative care provision. Qualitative methods explored management of people with cirrhosis, and barriers to and enablers of palliative care. / Results: Participants had high rates of hospital admissions and symptom burden. Clinicians rarely discussed prognosis or future care preferences; they lacked the skills and confidence to initiate discussions. Palliative care provision occurred late because clinicians were reluctant to refer due to their perception that reduced liver function is reversible, poor understanding of the potential of a palliative approach; palliative care was perceived negatively by patients and families. / Conclusions: People dying with cirrhosis have unpredictable trajectories, but share a common pathway of frequent admissions and worsening symptoms as death approaches. The use of clinical tools to identify the point of irreversible deterioration and joint working between liver services and palliative care may improve care for people with cirrhosis

    Palliative care for cirrhosis: a UK survey of health professionals' perceptions, current practice and future needs

    Objective: To determine the knowledge and practice patterns of a UK cohort of relevant healthcare professionals (HCPs) about delivering palliative care in cirrhosis, and to inform priorities for future research. / Design: An on-line questionnaire survey with closed and open responses. / Setting: HCPs identified from the mailing list of special interest groups in hepatology and gastroenterology (liver), general practice and specialist palliative care (SPC) across the UK. / Results: Of the 6181 potential contacts identified, 517 HCPs responded. Most believed a role exists for SPC in caring for people with cirrhosis, but many SPC HCPs felt ill prepared to provide good care to those facing death. Further training was needed in managing liver-related symptoms, symptom control and end of life issues. All HCP groups wished to increase community provision of palliative care support, but many general practitioners felt unable to manage advanced cirrhosis in the community. There were differences in the optimal trigger for SPC referral with liver HCPs less likely to refer at symptom deterioration. Prognostication, symptom management and service configuration were key areas identified for future research. / Conclusions: All who responded acknowledged the role of SPC in caring for those dying with cirrhosis and need for further training to improve confidence and enable joint working between SPC, general practice and liver teams. Low response rates make it difficult to generalise these findings, which require further validation

    Chalk cliff retreat in East Sussex and Kent 1870s to 2001

    The retreat of chalk cliffs fringing the eastern English Channel contributes shingle to the beaches which helps to protect the cliffs and slow down erosion. Conversely, cliff retreat endangers settlements and infrastructure on the clifftop. Rates of retreat have been calculated by a variety of methods over the past century, but no attempt has been made to provide a complete coverage that allows for a true comparison of retreat rates over the entire coastline. Using historic maps and recent orthophotos, cliff retreat rates have been calculated for consecutive 50 m sections of chalk cliff along the English side of the entire eastern English Channel for a period of 125 years. The chalk cliffs of East Sussex erode at an average rate of 0.25 - 0.3 m y−1 while those in Kent at a rate of 0.1 m y−1

    The Red Rectangle: Its Shaping Mechanism and its Source of Ultraviolet Photons

    The proto-planetary Red Rectangle nebula is powered by HD 44179, a spectroscopic binary (P = 318 d), in which a luminous post-AGB component is the primary source of both luminosity and current mass loss. Here, we present the results of a seven-year, eight-orbit spectroscopic monitoring program of HD 44179, designed to uncover new information about the source of the Lyman/far-ultraviolet continuum in the system as well as the driving mechanism for the bipolar outflow producing the current nebula. Our observations of the H-alpha line profile around the orbital phase of superior conjunction reveal the secondary component to be the origin of the fast (max. v~560kms km s^{-1})bipolaroutflowintheRedRectangle.ThevariationoftotalHalphafluxfromthecentralHIIregionwithorbitalphasealsoidentifiesthesecondaryoritssurroundingsasthesourceofthefarultravioletionizingradiationinthesystem.Theestimatedmassofthesecondary( 0.94M) bipolar outflow in the Red Rectangle. The variation of total H-alpha flux from the central H II region with orbital phase also identifies the secondary or its surroundings as the source of the far-ultraviolet ionizing radiation in the system. The estimated mass of the secondary (~0.94 M\sun)andthespeedoftheoutflowsuggestthatthiscomponentisamainsequencestarandnotawhitedwarf,aspreviouslysuggested.WeidentifythesourceoftheLyman/farultravioletcontinuuminthesystemasthehot,innerregion(T) and the speed of the outflow suggest that this component is a main sequence star and not a white dwarf, as previously suggested. We identify the source of the Lyman/far-ultraviolet continuum in the system as the hot, inner region (T_{max} \ge 17,000K)ofanaccretiondisksurroundingthesecondary,fedbyRochelobeoverflowfromthepostAGBprimaryatarateofabout K) of an accretion disk surrounding the secondary, fed by Roche lobe overflow from the post-AGB primary at a rate of about 2 - 5\times10^{-5}M M\sunyr yr^{-1}.Thetotalluminosityoftheaccretiondiskaroundthesecondaryisestimatedtobeatleast300L. The total luminosity of the accretion disk around the secondary is estimated to be at least 300 L\sun$, about 5% of the luminosity of the entire system.     A southern hemisphere survey of the 5780 and 6284 {\AA} diffuse interstellar bands: correlation with the extinction

    We present a new database of 5780.5 and 6283.8 {\AA} DIB measurements and also study their correlation with the reddening. The database is based on high-resolution, high-quality spectra of early-type nearby stars located in the southern hemisphere at an average distance of 300 pc. Equivalent widths of the two DIBs were determined by means of a realistic continuum fitting and synthetic atmospheric transmissions. For all stars that possess a precise measurement of their color excess, we compare the DIBs and the extinction. We find average linear relationships of the DIBS and the color excess that agree well with those of a previous survey of northern hemisphere stars closer than 550 pc. This similarity shows that there is no significant spatial dependence of the average relationship in the solar neighborhood within \simeq 600 pc. A noticeably different result is our higher degree of correlation of the two DIBs with the extinction. We demonstrate that it is simply due to the lower temperature and intrinsic luminosity of our targets. Using cooler target stars reduces the number of outliers, especially for nearby stars, confirming that the radiation field of UV bright stars has a significant influence on the DIB strength.     Alternative complement pathway inhibition does not abrogate meningococcal killing by serum of vaccinated individuals

    Dysregulation of complement activation causes a number of diseases, including paroxysmal nocturnal hemoglobinuria and atypical hemolytic uremic syndrome. These conditions can be treated with monoclonal antibodies (mAbs) that bind to the complement component C5 and prevent formation of the membrane attack complex (MAC). While MAC is involved in uncontrolled lysis of erythrocytes in these patients, it is also required for serum bactericidal activity (SBA), i.e. clearance of encapsulated bacteria. Therefore, terminal complement blockage in these patients increases the risk of invasive disease by Neisseria meningitidis more than 1000-fold compared to the general population, despite obligatory vaccination. It is assumed that alternative instead of terminal pathway inhibition reduces the risk of meningococcal disease in vaccinated individuals. To address this, we investigated the SBA with alternative pathway inhibitors. Serum was collected from adults before and after vaccination with a meningococcal serogroup A, C, W, Y capsule conjugate vaccine and tested for meningococcal killing in the presence of factor B and D, C3, C5 and MASP-2 inhibitors. B meningococci were not included in this study since the immune response against protein-based vaccines is more complex. Unsurprisingly, inhibition of C5 abrogated killing of meningococci by all sera. In contrast, both factor B and D inhibitors affected meningococcal killing in sera from individuals with low, but not with high bactericidal anti-capsular titers. While the anti-MASP-2 mAb did not impair SBA, inhibition of C3 impeded meningococcal killing in most, but not in all sera. These data provide evidence that vaccination can provide protection against invasive meningococcal disease in patients treated with alternative pathway inhibitors
