    Synergistic Antifungal Study of PEGylated Graphene Oxides and Copper Nanoparticles against Candida albicans

This article is an open access article distributed under the terms and conditions of the Creative Commons Attribution (CC BY) license (http://creativecommons.org/licenses/by/4.0/).The coupling reactions of polyethylene glycol (PEG) with two different nano-carbonaceous materials, graphene oxide (GO) and expanded graphene oxide (EGO), were achieved by amide bond formations. These reactions yielded PEGylated graphene oxides, GO-PEG and EGO-PEG. Whilst presence of the newly formed amide links (NH-CO) were confirmed by FTIR stretches observed at 1732 cm−1 and 1712 cm−1, the associated Raman D- and G-bands resonated at 1311/1318 cm−1 and 1584/1595 cm−1 had shown the carbonaceous structures in both PEGylated products remain unchanged. Whilst SEM images revealed the nano-sheet structures in all the GO derivatives (GO/EGO and GO-PEG/EGO-PEG), TEM images clearly showed the nano-structures of both GO-PEG and EGO-PEG had undergone significant morphological changes from their starting materials after the PEGylated processes. The successful PEGylations were also indicated by the change of pH values measured in the starting GO/EGO (pH 2.6–3.3) and the PEGylated GO-PEG/EGO-PEG (pH 6.6–6.9) products. Initial antifungal activities of selective metallic nanomaterials (ZnO and Cu) and the four GO derivatives were screened against Candida albicans using the in vitro cut-well method. Whilst the haemocytometer count indicated GO-PEG and copper nanoparticles (CuNPs) exhibited the best antifungal effects, the corresponding SEM images showed C. albicans had, respectively, undergone extensive shrinkage and porosity deformations. Synergistic antifungal effects all GO derivatives in various ratio of CuNPs combinations were determined by assessing C. albicans viabilities using broth dilution assays. The best synergistic effects were observed when a 30:70 ratio of GO/GO-PEG combined with CuNPs, where MIC50 185–225 μm/mL were recorded. Moreover, the decreased antifungal activities observed in EGO and EGO-PEG may be explained by their poor colloidal stability with increasing nanoparticle concentrations.Peer reviewe

    Identification of rare-earth minerals associated to K-feldspar: Capacsaya project in Peru

    A recently discovered the rare-earth-rich site in Capacsaya, located at 123 km northwest of Cusco, at the south of Peru, contains significant quantities of light and heavy rare-earth elements such as neodymium, lanthanum, cerium, europium, and yttrium. This work reports the identification of rare-earth elements and their associated minerals using scanning electron microscopy, energy-dispersive X-ray spectroscopy and X-ray diffraction analyses. Five (5) samples extracted from different locations at the Capacsaya site were characterized and identified K-feldspar as the mineral associated with the rare-earth elements in a representative sample with a high concentration of lanthanum and cerium. The results showed rare-earth elements contained within the mineral phase monazite, being cerium the dominant element in the phase (La, Ce, Nd)PO4. Finally, through the electrostatic separation process we demonstrate that it was possible to achieve an efficient separation of the K-feldspar phase in the particle size range 75–150 μ m.Fil: Ochoa, J..     Abstract Heating, cooling and ventilation accounts for 30% energy usage and for 50% of the electricity usage in the United States. Currently, most modern buildings still condition rooms assuming maximum occupancy rather than actual usage. As a result, rooms are often over-conditioned needlessly. This paper proposes an HVAC control strategy based on occupancy prediction and real time occupancy monitoring via a sensor network of cameras. This strategy shows 20.0% potential energy savings while still maintaining ASHRAE building standards

    Abstract Thermal comfort has traditionally been measured solely by temperature. While other methods such as Predicted Mean Vote (PMV) are available for measuring thermal comfort, the parameters required for an accurate value are overly complicated to obtain and require a great deal of sensory input. This paper proposes to bypass overly cumbersome or simplistic measures thermal comfort by bringing humans in the loop. By using humans as sensors, we can accurately adjust temperatures to improve occupant comfort. We show that occupants are more comfortable with a system that continually adjusts to thermal preference than a system that attempts to predict user comfort based on environmental factors. In addition, we also show that such a system is able to save 10.1% energy while improving the quality of service

A partir de estas caracterizaciones se desarrolla una estrategia de cálculo para la confección de la carga metalúrgica, que permite obtener, simultáneamente, al ser procesada por reducción carbotérmica en un homo de arco eléctrico, una ferroaleación multicomponente y una escoria útil para la confección de consumibles de soldadura por arco eléctrico. El polvo de la escoria obtenida se aglomera con vidrio líquido. Los pelets resultantes y su comportamiento en la soldadura automática por arco sumergido (SAW), presentan propiedades metalúrgicas y tecnológicas que satisfacen los requisitos de una matriz de un fundente aglomerado. La composición química de la ferroaleación multicomponente está constituida por elementos metálicos: vanadio, cromo, molibdeno, titanio y niobio (V, Cr, Mo, Ti y Nb) de alto valor metalúrgico y aleante, apropiada para la formulación de cargas aleantes de consumibles tanto para la soldadura manual (SMAW) como para la SAW.[EN] In this paper, chemical and mineralogical characterizations of the black sands of the Mejias placer of Sagua de Tánamo (the most important beach littoral placer of the northwest of oriental Cuba) are exposed. Starting from these characterizations a calculation strategy is developed for the making of the metallurgical load that allows to obtain simultaneously, when processed by carbothermic reduction in an electrical arc furnace, a multicomponent ferroalloy and a useful slag for the making of electric arch welding consumables. The powder of the obtained slag is agglomerated with liquid glass. The resulting pellets, due to their behavior on the submerged arc welding (SAW) present technological and metallurgical properties that correspond with the requirements of an agglomerated flux matrix. The chemical composition of the multicomponent ferroalloy is constituted by metallic elements of high metallurgical and alloyed values (V, Cr, Mo, Ti, Nb).     The insertion/deletion in the DNA-binding region allows the discrimination and subsequent identification of the glucocorticoid receptor 1 (gr1) and gr2 nucleotide sequences in gilthead sea bream (Sparus aurata): Standardizing the gr nomenclature for a better understanding of the stress response in teleost fish species

    Cortisol carries out its physiological mechanism of action through the recognition by the mineralocorticoid receptor (MR) and the glucocorticoid receptor (GR) 1 (GR1) and GR2. Previous studies reported that the main difference between gr1 and gr2 nucleotide sequences resides in a 27-nucleotide insertion/deletion in the DNA-binding region, respectively. However, in gilthead sea bream (Sparus aurata) the annotation for gr1 and gr2 seems contradictory. The gr2 sequence possesses the characteristic 27-nucleotide insertion that, in fact, is associated with the gr1 nucleotide sequence. Thus, this study aimed to elucidate the nucleotide sequences for the gr1 and gr2 in gilthead sea bream. The Clustal Omega alignment for different fish species corroborated the presence of such 27-nucleotide insertion/deletion in the DNA-binding region for gr1 and gr2, respectively. Then, we design specific primers set for the amplification of the gilthead sea bream gr1 by polymerase chain reaction (PCR). Importantly, the gr1 nucleotide partial sequence has a high similarity with other gr1 sequences already published for other fish species, being present in all of them the 27-nucleotide insertion in the DNA-binding region. We also detected that in European sea bass the gr1 and gr2 sequences had not been named according to the 27-nucleotide insertion/deletion criteria in the DNA-binding region.     Patient-Reported Outcomes After Complex Adult Spinal Deformity Surgery: 5-Year Results of the Scoli-Risk-1 Study

Objective: To prospectively evaluate PROs up to 5-years after complex ASD surgery. Methods: The Scoli-RISK-1 study enrolled 272 ASD patients undergoing surgery from 15 centers. Inclusion criteria was Cobb angle of >80°, corrective osteotomy for congenital or revision deformity, and/or 3-column osteotomy. The following PROs were measured prospectively at intervals up to 5-years postoperative: ODI, SF36-PCS/MCS, SRS-22, NRS back/leg. Among patients with 5-year follow-up, comparisons were made from both baseline and 2-years postoperative to 5-years postoperative. PROs were analyzed using mixed models for repeated measures. Results: Seventy-seven patients (28.3%) had 5-year follow-up data. Comparing baseline to 5-year data among these 77 patients, significant improvement was seen in all PROs: ODI (45.2 vs. 29.3, P 0.05) and proportion achieving MCID did not differ significantly in patients with major surgery-related complications compared to those without. Conclusions: After complex ASD surgery, significant improvement in PROs were seen at 5-years postoperative in ODI, SF36-PCS/MCS, SRS-22r, and NRS-back/leg pain. No significant changes in PROs occurred during the 2 to 5-year postoperative period.     The gene expression profile of the glucocorticoid receptor 1 (gr1) but not gr2 is modulated in mucosal tissues of gilthead sea bream (Sparus aurata) exposed to acute air-exposure stress

    The perception of an acute stressor (short-duration; high-intensity) induces a physiological response that activates the hypothalamic-pituitary-interrenal (HPI) axis and the subsequent release of cortisol. Cortisol carries out its effect at the molecular level through its recognition by the mineralocorticoid receptor (MR) and the glucocorticoid receptor (GR). Recently, we unveiled the nucleotide sequence of the glucocorticoid receptor 1 (gr1) and gr2 in gilthead sea bream (Sparus aurata). Importantly, GR1 and GR2 respond to different levels of cortisol concentration in fish and, consequently, play a differential role in the stress response. To date, and despite their relevance, no data describes the modulation of these receptors in response to an acute stressor in gilthead sea bream (S. aurata). In this study, we evaluated the kinetics of modulation of cortisol receptors expression (gr1, gr2, mr), and its similarity with the expression pattern of selected genes associated with stress (hsp70; enolase) and immune response (lysozyme; c3; il-1 beta; tnf-alpha; il-10; tgf-beta 1) in gilthead sea bream mucosal tissues (skin; gills; anterior gut). To do it, fish were acutely stressed by three-minute air exposure, and the expression profile was evaluated at zero, 1 h, 6 h, and 24 h post-stress (hps). The cortisol level in plasma and skin mucus peaked at 1 hps. All the mucosal tissues showed a time-dependent and tissue-specific upregulation of gr1 and mr. The immune-related genes showed the upregulation of il-1 beta at 6 hps (gills; anterior gut), and tnf-alpha and c3 at 24 hps (anterior gut). Taking together, our study concludes that fish subjected to three-minute air exposure modulated the expression of gr1 but not gr2 in mucosal tissues (skin; gills; anterior gut). Furthermore, our data reinforce the idea of a stimulatory effect induced in genes associated with the innate immune response after acute stress but focused at the mucosal level and in a time- and tissue-dependent manner

    In-Network Outlier Detection in Wireless Sensor Networks

    To address the problem of unsupervised outlier detection in wireless sensor networks, we develop an approach that (1) is flexible with respect to the outlier definition, (2) computes the result in-network to reduce both bandwidth and energy usage,(3) only uses single hop communication thus permitting very simple node failure detection and message reliability assurance mechanisms (e.g., carrier-sense), and (4) seamlessly accommodates dynamic updates to data. We examine performance using simulation with real sensor data streams. Our results demonstrate that our approach is accurate and imposes a reasonable communication load and level of power consumption.Comment: Extended version of a paper appearing in the Int'l Conference on Distributed Computing Systems 200

    Antimicrobial functionalized genetically engineered spider silk

    Genetically engineered fusion proteins offer potential as multifunctional biomaterials for medical use. Fusion or chimeric proteins can be formed using recombinant DNA technology by combining nucleotide sequences encoding different peptides or proteins that are otherwise not found together in nature. In the present study, three new fusion proteins were designed, cloned and expressed and assessed for function, by combining the consensus sequence of dragline spider silk with three different antimicrobial peptides. The human antimicrobial peptides human neutrophil defensin 2 (HNP-2), human neutrophil defensins 4 (HNP-4) and hepcidin were fused to spider silk through bioengineering. The spider silk domain maintained its self-assembly features, a key aspect of these new polymeric protein biomaterials, allowing the formation of b-sheets to lock in structures via physical interactions without the need for chemical crosslinking. These new functional silk proteins were assessed for antimicrobial activity against Gram e Escherichia coli and Gram þ Staphylococcus aureus and microbicidal activity was demonstrated. Dynamic light scattering was used to assess protein aggregation to clarify the antimicrobial patterns observed. Attenuated-total reflectance Fourier transform infrared spectroscopy (ATR-FTIR) and circular dichroism (CD) were used to assess the secondary structure of the new recombinant proteins.
