Numerical evolutions of a black hole-neutron star system in full General Relativity: I. Head-on collision
We present the first simulations in full General Relativity of the head-on
collision between a neutron star and a black hole of comparable mass. These
simulations are performed through the solution of the Einstein equations
combined with an accurate solution of the relativistic hydrodynamics equations
via high-resolution shock-capturing techniques. The initial data is obtained by
following the York-Lichnerowicz conformal decomposition with the assumption of
time symmetry. Unlike other relativistic studies of such systems, no limitation
is set for the mass ratio between the black hole and the neutron star, nor on
the position of the black hole, whose apparent horizon is entirely contained
within the computational domain. The latter extends over ~400M and is covered
with six levels of fixed mesh refinement. Concentrating on a prototypical
binary system with mass ratio ~6, we find that although a tidal deformation is
evident the neutron star is accreted promptly and entirely into the black hole.
While the collision is completed before ~300M, the evolution is carried over up
to ~1700M, thus providing time for the extraction of the gravitational-wave
signal produced and allowing for a first estimate of the radiative efficiency

Summary of the First Workshop on Sustainable Software for Science: Practice and Experiences (WSSSPE1)
Challenges related to development, deployment, and maintenance of reusable software for science are becoming a growing concern. Many scientists’ research increasingly depends on the quality and availability of software upon which their works are built. To highlight some of these issues and share experiences, the First Workshop on Sustainable Software for Science: Practice and Experiences (WSSSPE1) was held in November 2013 in conjunction with the SC13 Conference. The workshop featured keynote presentations and a large number (54) of solicited extended abstracts that were grouped into three themes and presented via panels. A set of collaborative notes of the presentations and discussion was taken during the workshop.
Unique perspectives were captured about issues such as comprehensive documentation, development and deployment practices, software licenses and career paths for developers. Attribution systems that account for evidence of software contribution and impact were also discussed. These include mechanisms such as Digital Object Identifiers, publication of “software papers”, and the use of online systems, for example source code repositories like GitHub. This paper summarizes the issues and shared experiences that were discussed, including cross-cutting issues and use cases. It joins a nascent literature seeking to understand what drives software work in science, and how it is impacted by the reward systems of science. These incentives can determine the extent to which developers are motivated to build software for the long-term, for the use of others, and whether to work collaboratively or separately. Structure-transport correlation reveals anisotropic charge transport in coupled PbS nanocrystal superlattices
Semiconductive nanocrystals (NCs) can be self-assembled into ordered
superlattices (SLs) to create artificial solids with emerging collective
properties. Computational studies have predicted that properties such as
electronic coupling or charge transport are determined not only by the
individual NCs but also by the degree of their organization and structure.
However, experimental proof for a correlation between structure and charge
transport in NC SLs is still pending. Here, we perform X-ray nano-diffraction
and apply Angular X-ray Cross-Correlation Analysis (AXCCA) to characterize the
structures of coupled PbS NC SLs, which are directly correlated with the
electronic properties of the same SL microdomains. We find strong evidence for
the effect of SL crystallinity on charge transport and reveal anisotropic
charge transport in highly ordered monocrystalline hexagonal close-packed PbS
NC SLs, caused by the dominant effect of shortest interparticle distance. This
implies that transport anisotropy should be a general feature of weakly coupled

Use of IFNγ/IL10 Ratio for Stratification of Hydrocortisone Therapy in Patients With Septic Shock
Large clinical trials testing hydrocortisone therapy in septic shock have produced conflicting results. Subgroups may benefit of hydrocortisone treatment depending on their individual immune response. We performed an exploratory analysis of the database from the international randomized controlled clinical trial Corticosteroid Therapy of Septic Shock (CORTICUS) employing machine learning to a panel of 137 variables collected from the Berlin subcohort comprising 83 patients including demographic and clinical measures, organ failure scores, leukocyte counts and levels of circulating cytokines. The identified theranostic marker was validated against data from a cohort of the Hellenic Sepsis Study Group (HSSG) (n = 246), patients enrolled in the clinical trial of Sodium Selenite and Procalcitonin Guided Antimicrobial Therapy in Severe Sepsis (SISPCT, n = 118), and another, smaller clinical trial (Crossover study, n = 20). In addition, in vitro blood culture experiments and in vivo experiments in mouse models were performed to assess biological plausibility. A low serum IFNγ/IL10 ratio predicted increased survival in the hydrocortisone group whereas a high ratio predicted better survival in the placebo group. Using this marker for a decision rule, we applied it to three validation sets and observed the same trend. Experimental studies in vitro revealed that IFNγ/IL10 was negatively associated with the load of (heat inactivated) pathogens in spiked human blood and in septic mouse models. Accordingly, an in silico analysis of published IFNγ and IL10 values in bacteremic and non-bacteremic patients with the Systemic Inflammatory Response Syndrome supported this association between the ratio and pathogen burden. We propose IFNγ/IL10 as a molecular marker supporting the decision to administer hydrocortisone to patients in septic shock. Prospective clinical studies are necessary and standard operating procedures need to be implemented, particularly to define a generic threshold. If confirmed, IFNγ/IL10 may become a suitable theranostic marker for an urging clinical need
Ligelizumab for Chronic Spontaneous Urticaria
Background: In the majority of patients with chronic spontaneous urticaria, most currently available therapies do not result in complete symptom control. Ligelizumab is a next-generation high-affinity humanized monoclonal anti-IgE antibody. Data are limited regarding the dose–response relationship of ligelizumab and the efficacy and safety of ligelizumab as compared with omalizumab and placebo in patients who have moderate-to-severe chronic spontaneous urticaria that is inadequately controlled with H1-antihistamines at approved or increased doses, alone or in combination with H2-antihistamines or leukotriene-receptor antagonists.
Methods: In a phase 2b dose-finding trial, we randomly assigned patients to receive ligelizumab at a dose of 24 mg, 72 mg, or 240 mg, omalizumab at a dose of 300 mg, or placebo, administered subcutaneously every 4 weeks for a period of 20 weeks, or a single 120-mg dose of ligelizumab. Disease symptoms of hives, itch, and angioedema were monitored by means of weekly activity scores. The main objective was to determine a dose–response relationship for the complete control of hives (indicated by a weekly hives-severity score of 0, on a scale from 0 to 21, with higher scores indicating greater severity); the primary end point of this response was assessed at week 12. Complete symptom control was indicated by a weekly urticaria activity score of 0 (on a scale from 0 to 42, with higher scores indicating greater severity). Safety was analyzed throughout the trial.
Results: A total of 382 patients underwent randomization. At week 12, a total of 30%, 51%, and 42% of the patients treated with 24 mg, 72 mg, and 240 mg, respectively, of ligelizumab had complete control of hives, as compared with 26% of the patients in the omalizumab group and no patients in the placebo group. A dose–response relationship was established. At week 12, a total of 30%, 44%, and 40% of the patients treated with 24 mg, 72 mg, and 240 mg, respectively, of ligelizumab had complete control of symptoms, as compared with 26% of the patients in the omalizumab group and no patients in the placebo group. In this small and short trial, no safety concerns regarding ligelizumab or omalizumab emerged.
Conclusions: A higher percentage of patients had complete control of symptoms of chronic spontaneous urticaria with ligelizumab therapy of 72 mg or 240 mg than with omalizumab or placebo. (Funded by Novartis Pharma; ClinicalTrials.gov number, NCT02477332. opens in new tab.

Reabilitação de tartaruga-cabeçuda (Caretta caretta Linnaeus, 1758) após traumatismo craniano
Traumas cranianos em tartarugas marinhas geralmente estão relacionados à ação antrópica por colisão com embarcações motorizadas. Em sua maioria, estes eventos são fatais, e quando não, várias sequelas podem permanecer. Desta forma, objetivou-se relatar um caso de uma tartaruga-cabeçuda (Caretta caretta) com politraumatismos craniano. O espécime foi resgatado encalhado no dia 17 de novembro de 2018, na praia de Retirinho, município de Aracati, Ceará, Brasil. O animal foi encontrado ativo e responsivo, com bom escore corpóreo e elevado grau de desidratação. Na inspeção, verificou-se uma lesão traumática na cabeça, que atingiu as placas frontoparietal, parietais e temporais esquerda e supraocular esquerda, com exposição de tecido mole adjacente. No ramo mandibular esquerdo, presença de lesão perfuro-incisa, e na cavidade oral, anzol de pesca aderido à musculatura da sínfise mandibular direita. No exame neurológico, observou-se excitação e hipersensibilidade ao toque, resposta ao teste de ameaça no olho direito, mas apresentou reflexo reduzido no olho esquerdo. Os quatro membros responderam satisfatoriamente ao teste de retirada, não manifestando movimentos incoordenados. O animal recebeu suporte terapêutico com administração de antibióticos, analgésicos, anti-inflamatórios, polivitamínicos, fluidoterapia e tratamento tópico. Após 291 dias de iniciado o processo de reabilitação, o animal foi reintroduzido em ambiente natural. Conclui-se que o protocolo terapêutico utilizado na reabilitação deste espécime de tartaruga-cabeçuda foi eficiente, principalmente no que diz respeito à terapia tópica. Além disso, a partir dos exames radiográficos foi possível visualizar a evolução do processo cicatricial das fraturas ósseas, auxiliando assim na tomada de decisão em liberar o animal à natureza

Retrospective evaluation of whole exome and genome mutation calls in 746 cancer samples
Funder: NCI U24CA211006Abstract: The Cancer Genome Atlas (TCGA) and International Cancer Genome Consortium (ICGC) curated consensus somatic mutation calls using whole exome sequencing (WES) and whole genome sequencing (WGS), respectively. Here, as part of the ICGC/TCGA Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium, which aggregated whole genome sequencing data from 2,658 cancers across 38 tumour types, we compare WES and WGS side-by-side from 746 TCGA samples, finding that ~80% of mutations overlap in covered exonic regions. We estimate that low variant allele fraction (VAF < 15%) and clonal heterogeneity contribute up to 68% of private WGS mutations and 71% of private WES mutations. We observe that ~30% of private WGS mutations trace to mutations identified by a single variant caller in WES consensus efforts. WGS captures both ~50% more variation in exonic regions and un-observed mutations in loci with variable GC-content. Together, our analysis highlights technological divergences between two reproducible somatic variant detection efforts
Fabrication of nanocrystal superlattice microchannels by soft-lithography for electronic measurements of single-crystalline domains
We report a high-throughput and easy-to-implement approach to fabricate
microchannels of nanocrystal superlattices with dimensions of ~4 micrometer^2,
thus approaching the size of typical single-crystalline domains. By means of
microcontact printing, highly ordered superlattices with microscale dimensions
are transferred onto photolithographically prepatterned microelectrodes,
obtaining well-defined superlattice microchannels. We present step-by-step
guidelines for microfabrication, nanocrystal self-assembly and patterning to
archive large quantities of up to 330 microchannels per device for
statistically meaningful investigations of charge transport in
single-crystalline superlattice domains. As proof-of-concept, we perform
conductivity and field-effect transistor measurements on microchannels of PbS
nanocrystal superlattices. We find that the electric transport within
microchannel superlattices is orders of magnitude more efficient than within
conventional large-scale channels, highlighting the advantage of the near
single-crystalline microchannels presented in this paper.Comment: 22 pages, 12 Figure
