    Cloning and DNA sequence of the 5'-exonuclease gene of bacteriophage T5

    AbstractThe nucleotide sequence of the BalI-PstI fragment of T5 DNA, 1347 bp in length, coding for 5'-exonuclease (D15 gene), has been determined. A coding region of the gene contains 873 bp and is preceded by a typical Shine-Dalgarno sequence. The D15 gene belongs to a cluster, consisting of at least 3 genes, in which a termination codon of a preceding gene overlaps an initiation codon of the following one. The sequence contains an open reading frame for 291 amino acid residues. The molecular mass of the 5'-exonuclease calculated from the predicted amino acid sequence is 33400 Da

    The existence of a new phenomena - giant electron-gamma bursts is established. The bursts are generated in thunderclouds as a result of the combined action of runaway breakdown and extensive atmosphere showers (RB-EAS). The experiments were fulfilled at the Tien Shan Mountain Scientific Station using EAS-Radio installation.     Pharmacogenetics of Bleeding and Thromboembolic Events in Direct Oral Anticoagulant Users

This was a retrospective real-world study linking genotype data of three Finnish biobanks with national register data on drug dispensations and healthcare encounters. We investigated several single-nucleotide variants (SNVs) in the ABCG2, ABCB1, CES1, and CYP3A5 genes potentially associated with bleeding or thromboembolic events in direct oral anticoagulant (DOAC) users based on earlier research. We used Cox regression models to compare the incidence of clinical outcomes between carriers and noncarriers of the SNVs or haplotypes. In total, 1,806 patients on apixaban, dabigatran, or rivaroxaban were studied. The ABCB1 c.3435C>T (p.Ile1145=, rs1045642) SNV (hazard ratio (HR) 0.42, 95% confidence interval (CI), 0.18-0.98, P = 0.044) and 1236T-2677T-3435T (rs1128503-rs2032582-rs1045642) haplotype (HR 0.44, 95% CI, 0.20-0.95, P = 0.036) were associated with a reduced risk for thromboembolic outcomes, and the 1236C-2677G-3435C (HR 2.55, 95% CI, 1.03-6.36, P = 0.044) and 1236T-2677G-3435C (HR 5.88, 95% CI, 2.35-14.72, P A (rs4148738) SNV associated with a lower risk for bleeding events (HR 0.37, 95% CI, 0.16-0.89, P = 0.025) in apixaban users. ABCB1 variants are potential factors affecting thromboembolic events in rivaroxaban users and bleeding events in apixaban users. Studies with larger numbers of patients are warranted for comprehensive assessment of the pharmacogenetic associations of DOACs and their relevance for clinical practice.Peer reviewe

    Serotonin limits generation of chromaffin cells during adrenal organ development

    Adrenal glands are the major organs releasing catecholamines and regulating our stress response. The mechanisms balancing generation of adrenergic chromaffin cells and protecting against neuroblastoma tumors are still enigmatic. Here we revealed that serotonin (5HT) controls the numbers of chromaffin cells by acting upon their immediate progenitor "bridge" cells via 5-hydroxytryptamine receptor 3A (HTR3A), and the aggressive HTR3A(high) human neuroblastoma cell lines reduce proliferation in response to HTR3A-specific agonists. In embryos (in vivo), the physiological increase of 5HT caused a prolongation of the cell cycle in "bridge" progenitors leading to a smaller chromaffin population and changing the balance of hormones and behavioral patterns in adulthood. These behavioral effects and smaller adrenals were mirrored in the progeny of pregnant female mice subjected to experimental stress, suggesting a maternal-fetal link that controls developmental adaptations. Finally, these results corresponded to a size-distribution of adrenals found in wild rodents with different coping strategies

    Power quality in high voltage mains supplying mainline railroads’ traction substations

    The increase in the trains weight and their operation speeds leads to considerable growth of abrupt-variable, non-linear and one-phase traction loads. This, in its own turn, leads to significant deviations, unsymmetry and voltage harmonic distortions in 110-220 kV mains adjacent to traction substations of AC railroads. Significant deterioration in power quality factors is observed in Siberian and Far East regions where 110-220 kV main power transmission lines are immediately adjacent to Transsib traction substations; in this case, in points with lower levels of short circuit power, power quality factors exceed by far the permissible levels. The articles provides the results of studies aimed at solution of power quality enhancement is-sues for mains adjacent to traction substations of Trans-Siberian Railway. To enhance power quality in mains supplying traction substations, smart grid technologies can be used together with multiagent control systems. Technical solutions, based on the results obtained, are proposed that allow to bring the power quality factors to the regulatory values. Due to a big investments amount, an implementation of tar-get program is needed to resolve the large-scale issue of enhancing power quality in mains adjacent to traction substations of AC railroads
