Ornithine Decarboxylase Antizyme Induces Hypomethylation of Genome DNA and Histone H3 Lysine 9 Dimethylation (H3K9me2) in Human Oral Cancer Cell Line
Background: Methylation of CpG islands of genome DNA and lysine residues of histone H3 and H4 tails regulates gene transcription. Inhibition of polyamine synthesis by ornithine decarboxylase antizyme-1 (OAZ) in human oral cancer cell line resulted in accumulation of decarboxylated S-adenosylmethionine (dcSAM), which acts as a competitive inhibitor of methylation reactions. We anticipated that accumulation of dcSAM impaired methylation reactions and resulted in hypomethylation of genome DNA and histone tails. Methodology/Principal Findings: Global methylation state of genome DNA and lysine residues of histone H3 and H4 tails were assayed by Methylation by Isoschizomers (MIAMI) method and western blotting, respectively, in the presence or absence of OAZ expression. Ectopic expression of OAZ mediated hypomethylation of CpG islands of genome DNA and histone H3 lysine 9 dimethylation (H3K9me2). Protein level of DNA methyltransferase 3B (DNMT3B) and histone H3K9me specific methyltransferase G9a were down-regulated in OAZ transfectant. Conclusions/Significance: OAZ induced hypomethylation of CpG islands of global genome DNA and H3K9me2 by down-regulating DNMT3B and G9a protein level. Hypomethylation of CpG islands of genome DNA and histone H3K9me2 is a potent mechanism of induction of the genes related to tumor suppression and DNA double strand break repair

Psychosocial twin cohort studies in Japan: the Keio Twin Research Center (KoTReC)
The Keio Twin Research Center (KoTReC) was established in 2009 at Keio University to combine two longitudinal cohort projects — the Keio Twin Study (KTS) for adolescence and adulthood and the Tokyo Twin Cohort Project (ToTCoP) for infancy and childhood. KoTReC also conducted a two-time panel study of self-control and psychopathology in twin adolescence in 2012 and 2013 and three independent anonymous cross-sectional twin surveys (ToTcross) before 2012 — the ToTCross, the Junior and Senior High School Survey and the High School Survey. This article introduces the recent research designs of KoTReC and its publications
3-Layer Public Key Cryptosystem with Short Tandem Repeat DNA
While the digital technology spreads through the society, reliable personal authentication is becoming an urgent issue.
As shown in digital taxation (e-Tax) and blockchain, etc., high reliable link between the private key of a public key and the owner who has it in card or smartphone etc. is required.
This paper proposes 3 layer public key cryptosystem in which Individual Number (a.k.a. My Number ) and STR (Short Tandem Repeat)
as personal identification data installed.
Individual Number is a national identification number issued by government, like social security number in USA.
STR is a kind of DNA data which does not contain any subtle personal information such as inherited character and has very accurate personal identification.
The proposed system satisfies requirements of integrity, soundness and zero knowledge characteristics which analog biometrics such as face authentications cannot provide
Type I Gaucher disease with exophthalmos and pulmonary arteriovenous malformation
BACKGROUND: Gaucher disease type I, the non-neuropathic type, usually presents in adulthood with hepatosplenomegaly. We report here an adult with type I Gaucher disease presented with unusual and severe clinical manifestations. CASE PRESENTATION: Hepatosplenomegaly, bone crisis and fractures occurred at early childhood, and splenectomy was performed at the age of 5. Exophthalmos with increase in retrobulbar space was noted when the patient was 30. Cerezyme infusion started at the age of 32; but unfortunately, pulmonary arteriovenous malformation with dyspnea and hypoxemia was found two years later. Gene analysis revealed V375L/L444P mutations in the β-glucocerebrosidase gene. CONCLUSION: Although both eye and lung diseases have been associated with Gaucher disease, this is the first reported demonstration of exophthalmos and pulmonary arteriovenous malformation in the same patient. This case may therefore present an extremely severe and unusual form of type I Gaucher disease
Conditional deletion of Npt2b in phosphate transport
Background
Hyperphosphatemia is common in chronic kidney disease and is associated with morbidity and mortality. The intestinal Na+-dependent phosphate transporter Npt2b is thought to be an important molecular target for the prevention of hyperphosphatemia. The role of Npt2b in the net absorption of inorganic phosphate (Pi), however, is controversial.
Methods
In the present study, we made tamoxifen-inducible Npt2b conditional knockout (CKO) mice to analyze systemic Pi metabolism, including intestinal Pi absorption.
Results
Although the Na+-dependent Pi transport in brush-border membrane vesicle uptake levels were significantly decreased in the distal intestine of Npt2b CKO mice compared with control mice, plasma Pi and fecal Pi excretion levels were not significantly different. Data obtained using the intestinal loop technique showed that Pi uptake in Npt2b CKO mice was not affected at a Pi concentration of 4 mM, which is considered the typical luminal Pi concentration after meals in mice. Claudin, which may be involved in paracellular pathways, as well as claudin-2, 12, and 15 protein levels were significantly decreased in the Npt2b CKO mice. Thus, Npt2b deficiency did not affect Pi absorption within the range of Pi concentrations that normally occurs after meals.
Conclusion
These findings indicate that abnormal Pi metabolism may also be involved in tight junction molecules such as Cldns that are affected by Npt2b deficiency
The GRANDMA network in preparation for the fourth gravitational-wave observing run
GRANDMA is a world-wide collaboration with the primary scientific goal ofstudying gravitational-wave sources, discovering their electromagneticcounterparts and characterizing their emission. GRANDMA involves astronomers, astrophysicists, gravitational-wave physicists, and theorists. GRANDMA is now atruly global network of telescopes, with (so far) 30 telescopes in bothhemispheres. It incorporates a citizen science programme (Kilonova-Catcher)which constitutes an opportunity to spread the interest in time-domainastronomy. The telescope network is an heterogeneous set of already-existingobserving facilities that operate coordinated as a single observatory. Withinthe network there are wide-field imagers that can observe large areas of thesky to search for optical counterparts, narrow-field instruments that dotargeted searches within a predefined list of host-galaxy candidates, andlarger telescopes that are devoted to characterization and follow-up of theidentified counterparts. Here we present an overview of GRANDMA after the thirdobserving run of the LIGO/VIRGO gravitational-wave observatories in and its ongoing preparation for the forthcoming fourth observational campaign(O4). Additionally, we review the potential of GRANDMA for the discovery andfollow-up of other types of astronomical transients.<br
The Belle II Physics Book
We present the physics program of the Belle II experiment, located on the
intensity frontier SuperKEKB collider. Belle II collected its first
collisions in 2018, and is expected to operate for the next decade. It is
anticipated to collect 50/ab of collision data over its lifetime. This book is
the outcome of a joint effort of Belle II collaborators and theorists through
the Belle II theory interface platform (B2TiP), an effort that commenced in
2014. The aim of B2TiP was to elucidate the potential impacts of the Belle II
program, which includes a wide scope of physics topics: B physics, charm, tau,
quarkonium, electroweak precision measurements and dark sector searches. It is
composed of nine working groups (WGs), which are coordinated by teams of
theorist and experimentalists conveners: Semileptonic and leptonic B decays,
Radiative and Electroweak penguins, phi_1 and phi_2 (time-dependent CP
violation) measurements, phi_3 measurements, Charmless hadronic B decay, Charm,
Quarkonium(like), tau and low-multiplicity processes, new physics and global
fit analyses. This book highlights "golden- and silver-channels", i.e. those
that would have the highest potential impact in the field. Theorists
scrutinised the role of those measurements and estimated the respective
theoretical uncertainties, achievable now as well as prospects for the future.
Experimentalists investigated the expected improvements with the large dataset
expected from Belle II, taking into account improved performance from the
upgraded detector.Comment: 689 page
