    "Music with a Message:" A Proposed Evaluation Model for the KRUNK Movement Health Initiative for Youth

    Adolescent health is a growing concern among professionals since it has been shown that the majority of deaths and disabilities that occur in this age group are largely determined by behaviors. Many adolescents engage in risky behaviors such as early sexual promiscuity, substance abuse and violence, which can have long lasting or permanent consequences. In order to significantly reduce the incidence of health disparities in these areas, public health interventions must be innovative, specific and comprehensive in addressing the behaviors that lead to such issues. It is also imperative that existing interventions address barriers many youth face including lack of knowledge and empowerment about the negative effects of risky behavior. Although challenges remain in educating and promoting health services to an ever-changing adolescent population, interventionists should focus on identifying the causes of early initiation of risky behaviors and the rationale behind why they continue. This is of public health significance because there are a number of health issues facing at-risk youth in underserved populations such as early sexual behavior, violence and substance abuse. Interventions will be identified utilizing music as a mechanism to inform youth about healthy behaviors. Adolescents are influenced by music and other forms of media which has a direct effect on their behavior. The influences of hip-hop music and its effects among adolescent behavior will be discussed. A qualitative evaluation strategy is proposed for the KRUNK Movement health initiative for youth located in the greater Pittsburgh area

    Constructivism and distance education : meeting the needs of adult learners in distance education

    This review addresses literature regarding how the implementation of constructivist principles in designing online learning environments can meet adults\u27 needs. This literature review identifies the specific characteristics and concerns associated with distance learning environments. It also explores the usefulness of using constructivism as a means to meet the unique needs of adult learners in distance learning courses. Resources consist primarily of books and articles published from 1995 to the present, dealing with issues of constructivism, andragogy, and distance education. The topics covered include constructivism, adult learners, and constructivist-based strategies integration with particular attention being paid to the problem-based learning model, scaffolded discussions, dynamic learning communities, and WebQuests. The collaborative nature of problem-based learning, scaffolded online discussion boards, and dynamic learning communities, address many of the needs of adults. The flexibility, authenticity and autonomous nature of the WebQuest address the needs of adults as well. Changes in available technologies and responsibilities of 21st century adults give educators and researchers cause to revisit the founding precepts of andragogy and distance learning. Educators and scholars continue to develop effective and innovative methods of meeting the needs of adult learners

    Live, Attenuated Influenza A H5N1 Candidate Vaccines Provide Broad Cross-Protection in Mice and Ferrets

    BACKGROUND: Recent outbreaks of highly pathogenic influenza A H5N1 viruses in humans and avian species that began in Asia and have spread to other continents underscore an urgent need to develop vaccines that would protect the human population in the event of a pandemic. METHODS AND FINDINGS: Live, attenuated candidate vaccines possessing genes encoding a modified H5 hemagglutinin (HA) and a wild-type (wt) N1 neuraminidase from influenza A H5N1 viruses isolated in Hong Kong and Vietnam in 1997, 2003, and 2004, and remaining gene segments derived from the cold-adapted (ca) influenza A vaccine donor strain, influenza A/Ann Arbor/6/60 ca (H2N2), were generated by reverse genetics. The H5N1 ca vaccine viruses required trypsin for efficient growth in vitro, as predicted by the modification engineered in the gene encoding the HA, and possessed the temperature-sensitive and attenuation phenotypes specified by the internal protein genes of the ca vaccine donor strain. More importantly, the candidate vaccines were immunogenic in mice. Four weeks after receiving a single dose of 10(6) 50% tissue culture infectious doses of intranasally administered vaccines, mice were fully protected from lethality following challenge with homologous and antigenically distinct heterologous wt H5N1 viruses from different genetic sublineages (clades 1, 2, and 3) that were isolated in Asia between 1997 and 2005. Four weeks after receiving two doses of the vaccines, mice and ferrets were fully protected against pulmonary replication of homologous and heterologous wt H5N1 viruses. CONCLUSIONS: The promising findings in these preclinical studies of safety, immunogenicity, and efficacy of the H5N1 ca vaccines against antigenically diverse H5N1 vaccines provide support for their careful evaluation in Phase 1 clinical trials in humans

    Detecting forest response to droughts with global observations of vegetation water content

    Droughts in a warming climate have become more common and more extreme, making understanding forest responses to water stress increasingly pressing. Analysis of water stress in trees has long focused on water potential in xylem and leaves, which influences stomatal closure and water flow through the soil-plant-atmosphere continuum. At the same time, changes of vegetation water content (VWC) are linked to a range of tree responses, including fluxes of water and carbon, mortality, flammability, and more. Unlike water potential, which requires demanding in situ measurements, VWC can be retrieved from remote sensing measurements, particularly at microwave frequencies using radar and radiometry. Here, we highlight key frontiers through which VWC has the potential to significantly increase our understanding of forest responses to water stress. To validate remote sensing observations of VWC at landscape scale and to better relate them to data assimilation model parameters, we introduce an ecosystem-scale analog of the pressure-volume curve, the non-linear relationship between average leaf or branch water potential and water content commonly used in plant hydraulics. The sources of variability in these ecosystem-scale pressure-volume curves and their relationship to forest response to water stress are discussed. We further show to what extent diel, seasonal, and decadal dynamics of VWC reflect variations in different processes relating the tree response to water stress. VWC can also be used for inferring belowground conditions-which are difficult to impossible to observe directly. Lastly, we discuss how a dedicated geostationary spaceborne observational system for VWC, when combined with existing datasets, can capture diel and seasonal water dynamics to advance the science and applications of global forest vulnerability to future droughts

    Shipping blood to a central laboratory in multicenter clinical trials: effect of ambient temperature on specimen temperature, and effects of temperature on mononuclear cell yield, viability and immunologic function

    <p>Abstract</p> <p>Background</p> <p>Clinical trials of immunologic therapies provide opportunities to study the cellular and molecular effects of those therapies and may permit identification of biomarkers of response. When the trials are performed at multiple centers, transport and storage of clinical specimens become important variables that may affect lymphocyte viability and function in blood and tissue specimens. The effect of temperature during storage and shipment of peripheral blood on subsequent processing, recovery, and function of lymphocytes is understudied and represents the focus of this study.</p> <p>Methods</p> <p>Peripheral blood samples (n = 285) from patients enrolled in 2 clinical trials of a melanoma vaccine were shipped from clinical centers 250 or 1100 miles to a central laboratory at the sponsoring institution. The yield of peripheral blood mononuclear cells (PBMC) collected before and after cryostorage was correlated with temperatures encountered during shipment. Also, to simulate shipping of whole blood, heparinized blood from healthy donors was collected and stored at 15°C, 22°C, 30°C, or 40°C, for varied intervals before isolation of PBMC. Specimen integrity was assessed by measures of yield, recovery, viability, and function of isolated lymphocytes. Several packaging systems were also evaluated during simulated shipping for the ability to maintain the internal temperature in adverse temperatures over time.</p> <p>Results</p> <p>Blood specimen containers experienced temperatures during shipment ranging from -1 to 35°C. Exposure to temperatures above room temperature (22°C) resulted in greater yields of PBMC. Reduced cell recovery following cryo-preservation as well as decreased viability and immune function were observed in specimens exposed to 15°C or 40°C for greater than 8 hours when compared to storage at 22°C. There was a trend toward improved preservation of blood specimen integrity stored at 30°C prior to processing for all time points tested. Internal temperatures of blood shipping containers were maintained longer in an acceptable range when warm packs were included.</p> <p>Conclusions</p> <p>Blood packages shipped overnight by commercial carrier may encounter extreme seasonal temperatures. Therefore, considerations in the design of shipping containers should include protecting against extreme ambient temperature deviations and maintaining specimen temperature above 22°C or preferably near 30°C.</p
