    From Falling Beams to Fallen Souls : The Ethical Development of the Hard-Boiled Detective Novel

    The hard-boiled detective story ushered in a revolution in the mystery story. The older formal mystery story saw society as a benevolent, ordering force which the detective restored by catching the murderer, while the hard-boiled mystery portrayed society as corrupt, wild and flawed, and presented an implicit criticism of it. The three best writers of the hard-boiled school, Dashiell Hammett, Raymond Chandler, and Ross Macdonald, show an ethical progression, a turning outward, from the personal, inwardly focused ethics of Hammett\u27s characters to the outwardly turned moral understanding of Macdonald\u27s hero. Dashiell Hammett writes of an ironic, tough world that the detective accepts because he cannot change it. Hammett\u27s detectives have a strong private code which forms a bulwark against the chaos of society. The detective cannot depend on anything outside himself, so these personal ethics help the detective survive, but he remains isolated and apart from society. Raymond Chandler\u27s detective turns the focus of his ethics slightly outward. His code is personal, but he believes in ideals outside himself and works towards them. He has honor and truly wants to help those in need, and his code embodies the world as he thinks it should be. He cannot accept the corruption of the world, remaining isolated because of his personal stand against it. Ross Macdonald\u27s detective has partaken of the world, and has been tainted but not corrupted. Instead he has a greater understanding of people\u27s failures. He also has a personal code, but his emphasis is on helping others understand themselves and their actions. Thus Macdonald focuses the ethical spotlight outside of his detective, on the forces in life that make people go bad

    Pilot Study: Assistive Technology as a Vocational Support for Individuals with Autism Spectrum Disorder

    The purpose of this pilot study was to determine the effectiveness of video-based instruction (VBI) to support completion of vocational tasks. A mixed-method approach was utilized to explore the use of VBI on a personal digital assistant with individuals with autism spectrum disorder (ASD). Using two assembling cooking tasks, researchers investigated the level of independence with task completion through written instruction versus VBI. The results indicated a small non-significant increase in the level of independence with task completion during the VBI task independent of intelligence quotient (IQ) levels. Participant’s feedback of VBI was also noted as positive to help learn other tasks. This study presents evidence for the use of assistive technology to support task completion in the area of vocation for individuals with ASD

    A degree-optimal, ordered peer-to-peer overlay network

Two of the most important theoretical advancements in this area were the development of degree-optimal and ordered networks. Prior to this work, it was not known whether both properties could be achieved simultaneously. This thesis presents Family Trees, the first peer-to-peer overlay network that is both degree-optimal and ordered. We develop Family Trees theoretically, proving correctness and bounds on their performance. We also describe how Family Trees can be optimized to improve latency and discuss the results of an experimental study showing that Family Trees provide good performance in practice.by Kevin C.     Campaign Finance Reform: The Unfinished Agenda

    In 1974, following the Watergate scandal, Congress enacted major campaign finance reform legislation. The legislation created a revolutionary new public financing system for our presidential campaigns, but it left congressional campaigns to be financed totally by private money. The presidential public financing system has worked well. Despite some incremental problems, the system has accomplished its basic goal of allowing individuals to run for the presidency without becoming dependent on their financial backers. The system for financing congressional cam paigns, on the other hand, is out of control and in need of fundamental reform. The inappropriate role of special interest political action commit tees (PACs) in influencing congressional elections and congressional decisions is the single biggest problem facing the political process.     Female Chromosome X Mosaicism is Age-Related and Preferentially Affects the Inactivated X Chromosome

    To investigate large structural clonal mosaicism of chromosome X, we analysed the SNP microarray intensity data of 38,303 women from cancer genome-wide association studies (20,878 cases and 17,425 controls) and detected 124 mosaic X events 4 2 Mb in 97 (0.25%) women. Here we show rates for X-chromosome mosaicism are four times higher than mean autosomal rates; X mosaic events more often include the entire chromosome and participants with X events more likely harbour autosomal mosaic events. X mosaicism frequency increases with age (0.11% in 50-year olds; 0.45% in 75-year olds), as reported for Y and autosomes. Methylation array analyses of 33 women with X mosaicism indicate events preferentially involve the inactive X chromosome. Our results provide further evidence that the sex chromosomes undergo mosaic events more frequently than autosomes, which could have implications for understanding the underlying mechanisms of mosaic events and their possible contribution to risk for chronic diseases

    Integrated genomic characterization of pancreatic ductal adenocarcinoma

    We performed integrated genomic, transcriptomic, and proteomic profiling of 150 pancreatic ductal adenocarcinoma (PDAC) specimens, including samples with characteristic low neoplastic cellularity. Deep whole-exome sequencing revealed recurrent somatic mutations in KRAS, TP53, CDKN2A, SMAD4, RNF43, ARID1A, TGFβR2, GNAS, RREB1, and PBRM1. KRAS wild-type tumors harbored alterations in other oncogenic drivers, including GNAS, BRAF, CTNNB1, and additional RAS pathway genes. A subset of tumors harbored multiple KRAS mutations, with some showing evidence of biallelic mutations. Protein profiling identified a favorable prognosis subset with low epithelial-mesenchymal transition and high MTOR pathway scores. Associations of non-coding RNAs with tumor-specific mRNA subtypes were also identified. Our integrated multi-platform analysis reveals a complex molecular landscape of PDAC and provides a roadmap for precision medicine
