    GWAS of random glucose in 476,326 individuals provide insights into diabetes pathophysiology, complications and treatment stratification

    Conventional measurements of fasting and postprandial blood glucose levels investigated in genome-wide association studies (GWAS) cannot capture the effects of DNA variability on ‘around the clock’ glucoregulatory processes. Here we show that GWAS meta-analysis of glucose measurements under nonstandardized conditions (random glucose (RG)) in 476,326 individuals of diverse ancestries and without diabetes enables locus discovery and innovative pathophysiological observations. We discovered 120 RG loci represented by 150 distinct signals, including 13 with sex-dimorphic effects, two cross-ancestry and seven rare frequency signals. Of these, 44 loci are new for glycemic traits. Regulatory, glycosylation and metagenomic annotations highlight ileum and colon tissues, indicating an underappreciated role of the gastrointestinal tract in controlling blood glucose. Functional follow-up and molecular dynamics simulations of lower frequency coding variants in glucagon-like peptide-1 receptor (GLP1R), a type 2 diabetes treatment target, reveal that optimal selection of GLP-1R agonist therapy will benefit from tailored genetic stratification. We also provide evidence from Mendelian randomization that lung function is modulated by blood glucose and that pulmonary dysfunction is a diabetes complication. Our investigation yields new insights into the biology of glucose regulation, diabetes complications and pathways for treatment stratification.</p

    Las instrucciones de relevancia en combinación con la interrogación elaborativa facilitan la lectura estratégica: evidencias desde los movimientos oculares

    The aim of the present study was to examine effects of relevance instructions and elaborative interrogation on the processing of and memory for expository texts. Eye movements of 132 undergraduate students were tracked while they read expository texts. After reading each text, they produced an oral summary. Participants were divided into four experimental conditions that differed by the presence or absence of the why question and the specific or general relevance instruction they received. Results showed that readers who received the why question embedded in the texts and also received the specific instruction of answering the question demonstrated more strategic reading, as reflected in their first-pass and look-back reading times and also in their better recall of question-relevant information. These results can be readily applied to real-life learning contexts, as they suggest that employing specific relevance instructions in combination with elaborative interrogation may elicit more efficient and strategic reading.El objetivo del presente estudio fue examinar el efecto de las Instrucciones de relevancia y de Interrogación elaborativa en el procesamiento y el posterior recuerdo de textos expositivos. A tal fin se registraron los movimientos oculares de 132 estudiantes universitarios mientras leían textos expositivos. Después de leer cada texto, realizaron un resumen oral del mismo. Se asignó a los participantes a cuatro condiciones experimentales que se diferenciaban por la presencia o ausencia de una pregunta de tipo “por qué” y la instrucción específica o general de relevancia que recibieron. Los resultados mostraron que los lectores que recibieron la pregunta de tipo “por qué” insertada en los textos y que también recibieron la instrucción específica de contestar a la pregunta mostraron unos patrones de lectura más estratégicos, como quedó reflejado en sus tiempos de lectura inicial y de refijaciones y también en su mejor recuerdo de la información relacionada con la pregunta.     Effects of Gamified Augmented Reality in Public Spaces

    Advancements in smartphone technology have resulted in the proliferation of Augmented Reality (AR) applications and games. Researchers have acknowledged the great potential of AR applications to enhance entertainment and improve learning experiences. In this study, we examined the potential effects of gamified AR in public places. We developed ARQuiz, an AR-based quiz game, for a public exhibition space and conducted a user study with respondents via survey (N = 176; 55.68% female, mean age 35.94 and SD = 11.89) and face-To-face interview (N = 28; 57.14% female, mean age 31.07 and SD = 7.42). We analyzed the relationship between perceived application usefulness, perceived application enjoyment, perceived exhibition enjoyment, and perceived quiz enjoyment. In addition, we examined perceived sociability before and after the quiz, quiz score, and user behavior in the exhibition space. The results indicate that visitors who enjoyed playing the ARQuiz game enjoyed the exhibition more, obtained better quiz results and felt more social after visiting the exhibition. Furthermore, the ARQuiz was regarded as a possible platform for improving visitors learning and overall experiences in public exhibitions. Although some players expressed concerns about the privacy and intrusiveness of AR, our results indicate that a well-designed AR game may boost the overall satisfaction of an exhibition visit and increase players sociabilitys. © 2013 IEEE.Peer reviewe

    Eye tracking as an MT evaluation technique

    Eye tracking has been used successfully as a technique for measuring cognitive load in reading, psycholinguistics, writing, language acquisition etc. for some time now. Its application as a technique for measuring the reading ease of MT output has not yet, to our knowledge, been tested. We report here on a preliminary study testing the use and validity of an eye tracking methodology as a means of semi-automatically evaluating machine translation output. 50 French machine translated sentences, 25 rated as excellent and 25 rated as poor in an earlier human evaluation, were selected. Ten native speakers of French were instructed to read the MT sentences for comprehensibility. Their eye gaze data were recorded non-invasively using a Tobii 1750 eye tracker. The average gaze time and fixation count were found to be higher for the “bad” sentences, while average fixation duration and pupil dilations were not found to be substantially different for output rated as good and output rated as bad. Comparisons between HTER scores and eye gaze data were also found to correlate well with gaze time and fixation count, but not with pupil dilation and fixation duration. We conclude that the eye tracking data, in particular gaze time and fixation count, correlate reasonably well with human evaluation of MT output but fixation duration and pupil dilation may be less reliable indicators of reading difficulty for MT output. We also conclude that eye tracking has promise as a semi-automatic MT evaluation technique, which does not require bi-lingual knowledge, and which can potentially tap into the end users’ experience of machine translation output

After reading, participants produced an oral summary of the text. Based on the quality of the summaries, participants were divided into three groups: High, Medium and Low Quality Summaries. The results revealed that readers who produced High Quality Summaries made significantly more and longer fixations and regressions in the question-relevant parts of texts when compared to the other two summary groups. These results suggest that the summary task performance could be a good predictor of the reading strategies utilized during reading

    Bifidobacterium Is Enriched in Gut Microbiome of Kashmiri Women with Polycystic Ovary Syndrome

    Polycystic ovary syndrome (PCOS) is a very common endocrine condition in women in India. Gut microbiome alterations were shown to be involved in PCOS, yet it is remarkably understudied in Indian women who have a higher incidence of PCOS as compared to other ethnic populations. During the regional PCOS screening program among young women, we recruited 19 drug naive women with PCOS and 20 control women at the Sher-i-Kashmir Institute of Medical Sciences, Kashmir, North India. We profiled the gut microbiome in faecal samples by 16S rRNA sequencing and included 40/58 operational taxonomic units (OTUs) detected in at least 1/3 of the subjects with relative abundance (RA) ≥ 0.1%. We compared the RAs at a family/genus level in PCOS/non-PCOS groups and their correlation with 33 metabolic and hormonal factors, and corrected for multiple testing, while taking the variation in day of menstrual cycle at sample collection, age and BMI into account. Five genera were significantly enriched in PCOS cases: Sarcina, Megasphaera, and previously reported for PCOS Bifidobacterium, Collinsella and Paraprevotella confirmed by different statistical models. At the family level, the relative abundance of Bifidobacteriaceae was enriched, whereas Peptococcaceae was decreased among cases. We observed increased relative abundance of Collinsella and Paraprevotella with higher fasting blood glucose levels, and Paraprevotella and Alkalibacterium with larger hip, waist circumference, weight, and Peptococcaceae with lower prolactin levels. We also detected a novel association between Eubacterium and follicle-stimulating hormone levels and between Bifidobacterium and alkaline phosphatase, independently of the BMI of the participants. Our report supports that there is a relationship between gut microbiome composition and PCOS with links to specific reproductive health metabolic and hormonal predictors in Indian women

    Aeolian dust supply from the Yellow River floodplain to the Pleistocene loess deposits of the Mangshan Plateau, central China : Evidence from zircon U-Pb age spectra

    The thick loess-palaeosol sequences in the Mangshan Loess Plateau (MLP; central China) along the south bank of the lower reach of the Yellow River provide high-resolution records of Quaternary climate change. In addition, substantial increases in grain-size and accumulation rate have been inferred in the upper part of the loess sequence, above palaeosol layer S2. This study investigates the sources of the long-term dust supply to the MLP and explores the mechanism behind the sudden increase in sediment delivery and coarsening of the loess deposits since S2 (similar to 240 ka) by using end member modelling of the loess grain-size dataset and single-grain zircon U-Pb dating. Our results indicate that the lower Yellow River floodplain, directly north of the MLP, served as a major dust supply for the plateau at least since the deposition of loess unit L9 and indirectly suggest that the integration of the Yellow River and the disappearance of the Sanmen palaeolake took place before L9 (similar to 900 ka). The sudden change in sedi-mentology of the Mangshan sequence above palaeosol unit S2 may result from an increased fluvial sediment flux being transported to the lower reaches of the Yellow River because of tectonic movements (initiated) in the Weihe Basin around 240 ka. Furthermore, sediment coarsening can be explained by the gradual southward migration of the lower Yellow River floodplain towards the MLP since the deposition of palaeosol S2. The migration is evidenced by the formation of an impressive scarp, and is likely caused by tectonic tilting of the floodplain area. (C) 2018 Elsevier Ltd. All rights reserved.Peer reviewe

    Intra-channel Interference Avoidance with the OGFDM Boosts Channel Capacity of Future Wireless Mobile Communication

    The Orthogonal Generalized Frequency Division Multiplexing (OGFDM) with Intra-Channel Interference Avoidance (ICIA) approach is, for the first time, proposed, explored and evaluated. Since the interference manipulation currently represents a hot topic for wireless mobile communication, the conventional approach of mitigating the interference is no longer acceptable. As a result, a novel method for addressing the interference between adjacent filtered sub-carriers (in-phase/out-phase) is comprehensively investigated herein. The proposed approach utilises the oversampling factor to effectively avoid interference and improve the quality of service of affected filters under bad transmission states. Thus, this supportive method which is essentially aware of propagation conditions is employed for removing the roll-off ( α ) impact yet improving the level of Bandwidth (BW) efficiency for applied filters of the OGFDM waveform. Besides, in terms of the system performance, the trade-off relation between the channel capacity and the key Hilbert filter parameter is theoretically and practically discussed. This requires investigation of the influence of α factor on the maximum achieved bit-rate at the acceptable limit of the Bit Error Rate (BER).     Site fertility drives temporal turnover of vegetation at high latitudes

    Experimental evidence shows that site fertility is a key modulator underlying plant community changes under climate change. Communities on fertile sites, with species having fast dynamics, have been found to react more strongly to climate change than communities on infertile sites with slow dynamics. However, it is still unclear whether this generally applies to high-latitude plant communities in natural environments at broad spatial scales. We tested a hypothesis that vegetation of fertile sites experiences greater changes over several decades and thus would be more responsive under contemporary climate change compared to infertile sites that are expected to show more resistance. We resurveyed understorey communities (vascular plants, bryophytes, and lichens) of four infertile and four fertile forest sites along a latitudinal bioclimatic gradient. Sites had remained outside direct human disturbance. We analyzed the magnitude of temporal community turnover, changes in the abundances of plant morphological groups and strategy classes, and changes in species diversity. In agreement with our hypothesis, temporal turnover of communities was consistently greater on fertile sites compared to infertile sites. However, our results suggest that the larger turnover of fertile communities is not primarily related to the direct effects of climatic warming. Furthermore, community changes in both fertile and infertile sites showed remarkable variation in terms of shares of plant functional groups and strategy classes and measures of species diversity. This further emphasizes the essential role of baseline environmental conditions and nonclimatic drivers underlying vegetation changes. Our results show that site fertility is a key determinant of the overall rate of high-latitude vegetation changes but the composition of plant communities in different ecological contexts is variously impacted by nonclimatic drivers over time.Peer reviewe
