Reducing Wind Sensitivity for Blower Door Testing
The fan pressurization method is a common practice in many countries for measuring the air leakage of houses. The test results are sensitive to uncertainties in the measured pressures and airflows. In particular, changing wind conditions during a test result in some pressure stations having more or less uncertainty than others.
Usually, it is necessary to fit the measured data to the power-law equation. Using the ordinary least square (OLS) fitting method, the pressure exponent and flow coefficient can be determined, and the reported data at high pressures can be extrapolated to small pressures where natural infiltration occurs. However, this fitting method neglects the existing of the uncertainty of these measurements, which may lead to errors in the prediction of flows at low pressures and therefore to unreliable input data for energy simulations. The weighted line of organic correlation (WLOC) takes the uncertainty at each pressure station into account and minimizes the fitting residuals for both pressure and flow.
This paper shows the results of a statistical analysis of an extensive data set of over 7.400 fan pressurization test of six houses in 109 different leakage configurations. It was found that in over 90 % of the analyzed cases, WLOC enables a more reliable prediction of pressure exponent and flow coefficient at low pressure compared to OLS and appears to be a better fitting technique

Comparison of Airflow and Acoustic Measurements for Evaluation of Building Air Leakage Paths in a Laboratory Test Apparatus
Unintended Infiltration in buildings is responsible for a significant portion of the global housing stock energy demand. Today, the fan pressurization method, also known as blower-door test, is the most frequently used measurement method to evaluate the airtightness of buildings and determining the total air change rate of a building or a building element. However, the localization and quantification of single leaks in the building envelope remain difficult and time-consuming.
In this paper, an acoustic method is introduced to estimate the leakage size of single leaks in buildings. Sound transmission measurements and measurements of airflow have been conducted in a laboratory test apparatus. The objective of this investigation is to compare acoustic measurements with airflow measurements of leaks under the same boundary conditions. The test apparatus consists of two chambers, which are separated by a test wall. This test wall represents a single characteristic air leakage path in the building envelope. Various types of wall structures with different slit geometries, wall thicknesses and insulation materials have been investigated. The acoustic measurements have been performed with a sound source placed in one chamber and ultrasonic microphones located in both chambers. The results of the acoustic measurements were compared to airflows through the test wall measured using a flow nozzle to provide estimates of the uncertainty in the acoustic approach

24S‐hydroxycholesterol: cellular effects and variations in brain diseases
The adult brain exhibit a characteristic cholesterol homeostasis, with low synthesis rate and active catabolism. Brain cholesterol turnover is possible thanks to the action of the enzyme Cytochrome P450 46A1 (CYP46A1) or 24-cholesterol hydroxylase, that transforms cholesterol into 24S-hydroxycholesterol (24S-HC). But before crossing the blood-brain barrier (BBB), this oxysterol that is the most abundant in the brain can act locally, affecting the functioning of neurons, astrocytes, oligodendrocytes, and vascular cells. The first part of this review addresses different aspects of 24S-HC production and elimination from the brain. The second part concentrates in the effects of 24S-HC at the cellular level, describing how this oxysterol affects cell viability, amyloid beta production, neurotransmission, and transcriptional activity. Finally, the role of 24S-HC in Alzheimer, Huntington and Parkinson diseases, multiple sclerosis and amyotrophic lateral sclerosis, as well as the possibility of using this oxysterol as predictive and/or evolution biomarker in different brain disorders is discussed.Fil: Sodero, Alejandro Omar. Diagnostic biomarkers for adult haemophagocytic lymphohistiocytosis in critically ill patients (HEMICU): a prospective observational study protocol
INTRODUCTION: Haemophagocytic lymphohistiocytosis (HLH) in adults is characterised by toxic immune activation and a sepsis-like syndrome, leading to high numbers of undiagnosed cases and mortality rates of up to 68%. Early diagnosis and specific immune suppressive treatment are mandatory to avoid fatal outcome, but the diagnostic criteria (HLH-2004) are adopted from paediatric HLH and have not been validated in adults. Experimental studies suggest biomarkers to sufficiently diagnose HLH. However, biomarkers for the diagnosis of adult HLH have not yet been investigated.
METHODS AND ANALYSIS: The HEMICU (Diagnostic biomarkers for adult haemophagocytic lymphohistiocytosis in critically ill patients) study aims to estimate the incidence rate of adult HLH among suspected adult patients in intensive care units (ICUs). Screening for HLH will be performed in 16 ICUs of Charité - Universitätsmedizin Berlin. The inclusion criteria are bicytopaenia, hyperferritinaemia (≥500 µg/L), fever or when HLH is suspected by the clinician. Over a period of 2 years, we expect inclusion of about 100 patients with suspected HLH. HLH will be diagnosed if at least five of the HLH-2004 criteria are fulfilled, together with an expert review; all other included patients will serve as controls. Second, a panel of potential biomarker candidates will be explored. DNA, plasma and serum will be stored in a biobank. The primary endpoint of the study is the incidence rate of adult HLH among suspected adult patients during ICU stay. Out of a variety of measured biomarkers, this study furthermore aims to find highly potential biomarkers for the diagnosis of adult HLH in ICU. The results of this study will contribute to improved recognition and patient outcome of adult HLH in clinical routine
Replication by the Epistasis Project of the interaction between the genes for IL-6 and IL-10 in the risk of Alzheimer's disease
Background: Chronic inflammation is a characteristic of Alzheimer's disease (AD). An interaction associated with the risk of AD has been reported between polymorphisms in the regulatory regions of the genes for the pro-inflammatory cytokine, interleukin-6 (IL-6, gene: IL6), and the anti-inflammatory cytokine, interleukin-10 (IL-10, gene: IL10).Methods: We examined this interaction in the Epistasis Project, a collaboration of 7 AD research groups, contributing DNA samples from 1,757 cases of AD and 6,295 controls. Results: We replicated the interaction. For IL6 rs2069837 AA x IL10 rs1800871 CC, the synergy factor (SF) was 1.63 (95% confidence interval: 1.10-2.41, p = 0.01), controlling for centre, age, gender and apolipoprotein E epsilon 4 (APOE epsilon 4) genotype. Our results are consistent between North Europe (SF = 1.7, p = 0.03) and North Spain (SF = 2.0, p = 0.09). Further replication may require a meta-analysis. However, association due to linkage disequilibrium with other polymorphisms in the regulatory regions of these genes cannot be excluded. Conclusion: We suggest that dysregulation of both IL-6 and IL-10 in some elderly people, due in part to genetic variations in the two genes, contributes to the development of AD. Thus, inflammation facilitates the onset of sporadic AD
