Ukupna taložna tvar na području Primorsko-goranske županije u razdoblju 1975.-2008.
Dustfall measurements in Primorsko-goranska county started in 1975 in the Bakar Bay (Site 2 - Bakar and Site 3 - Kraljevica). The measurements were extended to the city of Rijeka (Site 1 - Rijeka) in 1982, to the nearby islands (Site 4 - Krk and Site 5 - Cres) in 1986, and inland to Gorski Kotar (Site 6 - Delnice and Site 7 - Lividraga) in 1995. This article brings the results of dustfall measurements from 1975 to 2008. Dustfall was low in the city, the nearby islands, and Gorski Kotar. Recommended and limit values
were occasionally exceeded in the Bakar Bay due to emissions from the coke plant and harbour at Site 2 (1979-1997) and from the shipyard at Site 3. Lead content in the dustfall was below the national limits, save for two occasions at Site 2. Deposition of sulphur and nitrogen at the inland sites were below the respective critical load values.Ispitivanja ukupne taložne tvari u Primorsko-goranskoj županiji započela su 1975. godine na području Bakarskog zaljeva. Mjerenja su se 1982. god proširila na grad Rijeku, 1986. na otoke Krk i Cres, a 1995. i na područje Gorskog kotara. U radu su prikazani rezultati mjerenja ukupne taložne tvari u razdoblju 1975.-2008. na sedam odabranih postaja: u gradu Rijeci, dvije na području Bakarskog zaljeva (Bakar i Kraljevica), dvije na otocima (Krk i Cres) te dvije u Gorskom kotaru (Delnice i Lividraga). Analizirano je kretanje prosječnih godišnjih vrijednosti ukupno istaloženih tvari te u njima prisutnih količina sagorivog pepela, sulfata, nitrata, amonija i olova. Na području grada Rijeke, otoka Krka i Cresa te Gorskog kotara
količina ukupne taložne tvari je niska. Količine ukupne taložne tvari povremeno su prelazile preporučenu graničnu vrijednost u Bakru (1979.-1994.), zbog emisija iz koksare i iz luke za rasuti teret te u Kraljevici, gdje se osamdesetih godina osjećao utjecaj koksare u Bakru, a od sredine devedesetih pojačani rad u brodogradilištu. Gaining further insight into photo-Fenton treatment of phenolic compounds commonly found in food processing industry
A mixture of eight phenolic compounds, namely 2,4-dinitrophenol, tannic, ellagic, gallic, protocatechuic, vanillic, syringic and sinapic acids, have been treated by means of a photo-Fenton process under simulated and real sunlight. An experimental design methodology, based in Doehlert matrixes, was employed to check the effect of the concentration of Fe(II) and H2O2, as well as pH. Response surfaces show that photo-Fenton can be extended to pH values clearly above 2.8, probably due to complexation of iron with the phenolic substances. Experiments performed under solar irradiation at pH = 3.9 showed that complete removal of the monitored pollutants was achieved in less than 3 min; mineralisation was also efficient, although some organics remained in the solution. Toxicity was monitored according to Pseudokirchneriella subcapitata and Daphnia magna bioassays; Recombinant Yeast Assay (RYA) was employed to assess estrogenic and dioxin-like activities. 2,4-Dinitrophenol was demonstrated to be the major concern and, in general, photo-Fenton resulted in a detoxification of the solution. Finally, excitation emission matrix (EEM) fluorimetry was employed to obtain complementary information on the behaviour of organic matter. Most peaks associated with the parent pollutants disappeared after short irradiation periods and, at 12 min of irradiation chromophores were destroyed, what can be associated with the removal of complex molecules. T-cell subpopulations αβ and γδ in cord blood of very preterm infants : The influence of intrauterine infection

Immune response mechanisms in this group of patients and factors that influence cord blood mononuclear cell populations remain poorly understood and are considered insufficient. However, competent immune functions of the cord blood mononuclear cells are also described. The aim of this work was to evaluate the T-cell population (CD3+) with its subpopulations bearing T-cell receptor (TCR) αβ or TCR γδ in the cord blood of preterm infants born before 32 weeks of gestation by mothers with or without an intrauterine infection. Being a pilot study, it also aimed at feasibility check and assessment of an expected effect size. The cord blood samples of 46 infants age were subjected to direct immunofluorescent staining with monoclonal antibodies and then analyzed by flow cytometry. The percentage of CD3+ cells in neonates born by mothers with diagnosis of intrauterine infection was significantly lower than in neonates born by mothers without infection (p = 0.005; Mann-Whitney U test). The number of cells did not differ between groups. Infection present in the mother did not have an influence on the TCR αβ or TCR γδ subpopulations. Our study contributes to a better understanding of preterm infants' immune mechanisms, and sets the stage for further investigations.Peer reviewedFinal Published versio
T-lex: a program for fast and accurate assessment of transposable element presence using next-generation sequencing data
Transposable elements (TEs) are repetitive DNA sequences that are ubiquitous, extremely abundant and dynamic components of practically all genomes. Much effort has gone into annotation of TE copies in reference genomes. The sequencing cost reduction and the newly available next-generation sequencing (NGS) data from multiple strains within a species offer an unprecedented opportunity to study population genomics of TEs in a range of organisms. Here, we present a computational pipeline (T-lex) that uses NGS data to detect the presence/absence of annotated TE copies. T-lex can use data from a large number of strains and returns estimates of population frequencies of individual TE insertions in a reasonable time. We experimentally validated the accuracy of T-lex detecting presence or absence of 768 previously identified TE copies in two resequenced Drosophila melanogaster strains. Approximately 95% of the TE insertions were detected with 100% sensitivity and 97% specificity. We show that even at low levels of coverage T-lex produces accurate results for TE copies that it can identify reliably but that the rate of ‘no data’ calls increases as the coverage falls below 15×. T-lex is a broadly applicable and flexible tool that can be used in any genome provided the availability of the reference genome, individual TE copy annotation and NGS data
Analysis of expression profiles of MAGE-A antigens in oral squamous cell carcinoma cell lines
<p>Abstract</p> <p>Background</p> <p>The immunological response to solid tumours is insufficient. Therefore, tumour specific antigens have been explored to facilitate the activation of the immune system. The cancer/testis antigen class of MAGE-A antigens is a possible target for vaccination. Their differential expression profiles also modulate the course of the cancer disease and its response to antineoplastic drugs.</p> <p>Methods</p> <p>The expression profiles of MAGE-A2, -A3, -A4, -A6 and -A10 in five own oral squamous cell carcinoma cell lines were characterised by rt-PCR, qrt-PCR and immunocytochemistry with a global MAGE-A antibody (57B) and compared with those of an adult keratinocyte cell line (NHEK).</p> <p>Results</p> <p>All tumour cell lines expressed MAGE-A antigens. The antigens were expressed in groups with different preferences. The predominant antigens expressed were MAGE-A2, -A3 and -A6. MAGE-A10 was not expressed in the cell lines tested. The MAGE-A gene products detected in the adult keratinocyte cell line NHEK were used as a reference.</p> <p>Conclusion</p> <p>MAGE-A antigens are expressed in oral squamous cell carcinomas. The expression profiles measured facilitate distinct examinations in forthcoming studies on responses to antineoplastic drugs or radiation therapy. MAGE-A antigens are still an interesting aim for immunotherapy.</p
Gene Space Dynamics During the Evolution of Aegilops tauschii, Brachypodium distachyon, Oryza sativa, and Sorghum bicolor Genomes
Nine different regions totaling 9.7 Mb of the 4.02 Gb Aegilops tauschii genome were sequenced using the Sanger sequencing technology and compared with orthologous Brachypodium distachyon, Oryza sativa (rice), and Sorghum bicolor (sorghum) genomic sequences. The ancestral gene content in these regions was inferred and used to estimate gene deletion and gene duplication rates along each branch of the phylogenetic tree relating the four species. The total gene number in the extant Ae. tauschii genome was estimated to be 36,371. The gene deletion and gene duplication rates and total gene numbers in the four genomes were used to estimate the total gene number in each node of the phylogenetic tree. The common ancestor of the Brachypodieae and Triticeae lineages was estimated to have had 28,558 genes, and the common ancestor of the Panicoideae, Ehrhartoideae, and Pooideae subfamilies was estimated to have had 27,152 or 28,350 genes, depending on the ancestral gene scenario. Relative to the Brachypodieae and Triticeae common ancestor, the gene number was reduced in B. distachyon by 3,026 genes and increased in Ae. tauschii by 7,813 genes. The sum of gene deletion and gene duplication rates, which reflects the rate of gene synteny loss, was correlated with the rate of structural chromosome rearrangements and was highest in the Ae. tauschii lineage and lowest in the rice lineage. The high rate of gene space evolution in the Ae. tauschii lineage accounts for the fact that, contrary to the expectations, the level of synteny between the phylogenetically more related Ae. tauschii and B. distachyon genomes is similar to the level of synteny between the Ae. tauschii genome and the genomes of the less related rice and sorghum. The ratio of gene duplication to gene deletion rates in these four grass species closely parallels both the total number of genes in a species and the overall genome size. Because the overall genome size is to a large extent a function of the repeated sequence content in a genome, we suggest that the amount and activity of repeated sequences are important factors determining the number of genes in a genome
Membrane insertion and topology of the translocon-associated protein (TRAP) gamma subunit
Translocon-associated protein (TRAP) complex is intimately associated with the ER translocon for the insertion or translocation of newly synthesised proteins in eukaryotic cells. The TRAP complex is comprised of three single-spanning and one multiple-spanning subunits. We have investigated the membrane insertion and topology of the multiple-spanning TRAP-γ subunit by glycosylation mapping and green fluorescent protein fusions both in vitro and in cell cultures. Results demonstrate that TRAP-γ has four transmembrane (TM) segments, an Nt/Ct cytosolic orientation and that the less hydrophobic TM segment inserts efficiently into the membrane only in the cellular context of full-length protein
Hypoglycemic and hypolipidemic effects of Saururus chinensis Baill in streptozotocin-induced diabetic rats
Saururus chinensis Baill was reported to inhibit α-glucosidase in vitro and flatten postprandial increase in blood glucose in streptozotocin (STZ)-induced diabetic rats. We studied the effect of chronic consumption of S. chinensis Baill on blood glucose and lipid profile in STZ-induced diabetic male rats fed high fat diet. Male rats weighing 100-120 g were fed 30% fat diet with and without 10% freeze-dried leaves of S. chinensis Baill for 7 weeks after 1 week of adaptation. The rats were rendered diabetic by intravenous injection of STZ (60 mg/kg) after 6-week feeding of the assigned diets. At 1 week after the injection, the rats were sacrificed after an overnight fast. Plasma glucose (380.2 ± 14.4 mg/dL), total cholesterol (93.9 ± 7.9 mg/dL) and triglyceride levels (123.6 ± 7.5 mg/dL) of the S. chinensis Baill group were significantly lower than those of the control group (418.1 ± 12.0 mg/dL, 119.9 ± 9.4 mg/dL, 152.0 ± 10.3 mg/dL, respectively, p<0.05). Chronic consumption of S. chinesis Baill significantly decreased maltase activity of the small intestinal mucosa (120.1 ± 8.7 U/g protein) compared with the control group (96.8 ± 7.0 U/g protein, p<0.05). These results suggest that S. chinensis Baill have hypoglycemic and hypolipidemic effects by inhibiting α-glucosidase activity in the animal model of diabetes mellitus
