Dynamic approach to solve the daily drayage problem with travel time uncertainty
The intermodal transport chain can become more e cient by means of a good organization of
drayage movements. Drayage in intermodal container terminals involves the pick up and delivery
of containers at customer locations, and the main objective is normally the assignment
of transportation tasks to the di erent vehicles, often with the presence of time windows. This
scheduling has traditionally been done once a day and, under these conditions, any unexpected
event could cause timetable delays. We propose to use the real-time knowledge about vehicle
position to solve this problem, which permanently allows the planner to reassign tasks in case
the problem conditions change. This exact knowledge of the position of the vehicles is possible
using a geographic positioning system by satellite (GPS, Galileo, Glonass), and the results show
that this additional data can be used to dynamically improve the solution

a identificação de compostos orgânicos voláteis (VOC’s) emitidos por materiais
celulósicos, nomeadamente livros. O objectivo principal foi o estabelecimento de
perfis quÃmicos do material de origem, celulose, de modo a tornar possÃvel a
identificação de marcadores do tipo de celulose – celulose de algodão ou madeira,
bem como do seu estado de conservação/degradação.
Utilizou-se a micro-extracção em fase sólida associada à cromatografia gasosa e
espectrometria de massa (HS-SPME-GC-MS). Para validação do método, foram
estudados parâmetros experimentais: tipo de fibra, tempo de exposição, tempo de
dessorção, temperaturas do injector e detector, programa cromatográfico, tendo sido
igualmente estabelecidos os melhores parâmetros espectrométricos.
Como resultado do estudo dos VOCs emitidos pelos livros foram identificados 22
compostos. Pela primeira vez, foram identificados marcadores de um dos tipos de
celulose utilizada na manufactura de livros pertencentes ao património cultural. O
estudo da quiralidade dos compostos foi pela primeira vez utilizado em materiais de
interesse histórico, tendo-se constatado ser uma área importante, uma vez que apenas um dos enantiómeros de um dos compostos alvo está presente nos livros
Which criteria matter most in the evaluation of venture capital investments?
Structured Abstract
Purpose - This study identifies the importance assigned to the various criteria used by the Portuguese
Venture Capitalists (VCs) to evaluate and select early stage venture capital projects.
Design/methodology/approach - The data was collected through a questionnaire answered by 20
Portuguese VCs. We use descriptive statistics techniques and non-parametric tests to identify the most
valued criteria and test differences in the importance assigned to the criteria of several types of VCs and
investments.
Findings - The study reveals that personality and experience of the entrepreneur and of the management
team are the most valued groups of criteria. VCs with a majority of private share capital value more the
personality of the entrepreneur and management team than the companies with a majority of public share
capital. Additionally, the VCs that did not yet internationalize consider the personality of the entrepreneur
and management team and the financial aspects, to be more important than the VCs that have already
expanded abroad.
Originality/value – It provides evidence on the VCs behaviour in a small venture capital market. Since
most of the existing literature on this area refer to large VC markets, the present study is important to
investigate whether the conclusions reached by the previous studies can be extended to a small VC
market. Also, this study is a contribution to the literature on the internationalization of VCs and it is the
first study that explores the impact of the VCs being internationalized on the value given to the various
selection criteria of early stage venture capital projects
The effect of pH on bromelain partition from Ananas comosus by PEG4000/phosphate ATPS
In this work, the thermodynamic equilibrium and applying of PEG4000/Phosphate ATPS on the purification of bromelain extracted from pineapple was studied. A rigorous study of the equilibrium curves and tie-line length from PEG4000/phosphate ATPS were done for the pH 6-11 at 25ºC. Results showed that there was augment in the PEG and salt contents with the high pH value from PEG4000/Phosphate ATPS and two-phase formation needed only increasing the PEG content. Two tie-line length at pH 11 from PEG4000/Phosphate ATPS were optimal condition for bromelain purification, one on composition of 14% PEG and 13% salt and other at 12.6% PEG and 12.2% salt, while a 25-62 folds of enzyme was found. SDS-PAGE electrophoreses had one band only, which showed that bromelain was purified. The impact of concentration among venture capitalists:revisiting the determinants of venture capital
This article analyzes the impact of the level of concentration among Venture Capitalists (VCs) on the supply of venture capital (VC), through the reduced form model for the equilibrium amount of VC (using a simultaneous equation model on aggregated data from 15 European countries). It is shown that the level of concentration among VCs has a positive effect on VC supply, so creating condi-tions to increase the level of concentration can stimulate VC supply. The findings reveal the importance of unemployment and personal income rate on VC demand and the positive impact of stock market capitalization on VC supply.info:eu-repo/semantics/publishedVersio
Caracterização molecular e funcional de peptÃdeos de anuros e seus análogos : efeitos sobre patógenos microbianos e células tumorais
Tese (doutorado) — Universidade de BrasÃlia, Instituto de Ciências Biológicas, Departamento de Biologia Celular, Programa de Pós-Graduação em Biologia Molecular, 2022.A resistência antimicrobiana representa um dos maiores desafios da medicina moderna e implica que esforços devem ser empenhados na pesquisa e desenvolvimento de novas drogas sob o risco de retornarmos à era pré-antibiótica. Dentre as alternativas, os peptÃdeos antimicrobianos representam uma fonte interessante de moléculas que atuam principalmente na membrana plasmática das bactérias, um sÃtio de ação que é notadamente menos propenso ao desenvolvimento de resistência. Até o momento, mais de três mil peptÃdeos antimicrobianos já foram descritos, sendo a secreção cutânea de anfÃbios a principal fonte dessas moléculas com quase um terço dos peptÃdeos descritos. Além da atividade antimicrobiana, muitos destes peptÃdeos também apresentam atividade sobre células tumorais. Na primeira parte desta tese, investigamos os efeitos de dois peptÃdeos isolados da secreção cutânea do anuro Boana raniceps sobre bactérias, fungos e formas epimastigotas de Trypanosoma cruzi, além seus efeitos sobre células eucarióticas e eritrócitos. Na segunda parte, desenvolvemos dez análogos tendo como base o peptÃdeo citotóxico pentadactilina, isolada inicialmente da rã Leptodactylus labyrinthicus, com o objetivo de potencializar os efeitos biológicos desejados e, ao mesmo tempo, reduzir os efeitos deletérios. De maneira geral, observamos que alterações estruturais nos análogos foram associadas com uma atividade biológica mais potente. Por outro lado, observamos que algumas alterações nos peptÃdeos favoreceu substancialmente a atividade sobre a bactéria Gram-positiva Staphylococcus aureus, enquanto que para a bactéria Gram-negativa Escherichia coli houve diminuição na atividade. Por outro lado, outras mudanças estruturais culminou na perda total de atividade sobre bactérias e também sobre as células eucarióticas testadas. Para avaliarmos os efeitos dos análogos sobre células eucarióticas usamos técnicas de citometria de fluxo e proteômica. Usando a citometria de fluxo, observamos que os análogos 6 e 8 (AN6 e AN8), induziram várias alterações nas linhagens de células B16F10 e MCF-7, tais como mudanças no tamanho e granulosidade e marcação positiva para anexina V/FITC e iodeto de propÃdio, sugerindo a ocorrência de necrose. Também observamos que houve uma marcação expressiva para caspases quando testamos as células B16F10 tratadas com os peptÃdeos. O tratamento de células B16F10 e MCF-7 com os peptÃdeos AN6 e AN8 induziu a despolarização da membrana mitocondrial, já a permeabilidade lisossômica não foi afetada nas células B16F10, enquanto que houve uma permeabilidade lisossômica nas células MCF-7 tratadas com ambos os peptÃdeos com significância estatÃstica. Observou-se também uma significante fragmentação de DNA nas duas linhagens tratadas tanto com o AN6 quanto ao AN8. Por outro lado, as alterações no ciclo celular foram observadas majoritariamente nas células B16F10. Na análise proteômica, usamos a linhagem B16F10 tratada com o peptÃdeo AN6. Após análise estatÃstica, encontramos 242 proteÃnas diferencialmente expressas das quais 103 estavam com o padrão de abundância aumentada e 139 proteÃnas com abundância diminuÃda em relação ao controle. Diferentemente do que observamos nos ensaios de citometria, não encontramos proteÃnas relacionadas com processos de morte celular com a expressão aumentada.Coordenação de Aperfeiçoamento de Pessoal de NÃvel Superior (CAPES); Conselho Nacional de Desenvolvimento CientÃfico e Tecnológico (CNPq) e Fundação de Apoio à Pesquisa do Distrito Federal (FAP/DF).Antimicrobial resistance represents one of the greatest challenges of modern medicine and implies that efforts must be devoted to the research and development of new drugs at the risk of returning to the pre-antibiotic era. Among the alternatives, antimicrobial peptides represent an interesting source of molecules that act mainly on the plasma membrane of bacteria, a site of action that is notably less prone to the development of resistance. To date, more than three thousand antimicrobial peptides were described, with amphibian skin secretions as the main source of these molecules, with almost a third of the peptides described. In addition to antimicrobial activity, many of these peptides also have activity on tumor cells. In the first part of this thesis, we investigated the effects of two peptides isolated from the cutaneous secretion of the anuran Boana raniceps on bacteria, fungi and epimastigotes forms of Trypanosoma cruzi, in addition to their effects on eukaryotic cells and erythrocytes. In the second part, we developed ten analogs based on the cytotoxic peptide pentadactylin, initially isolated from the frog Leptodactylus labyrinthicus, with the aim of potentiating the desired biological effects and, at the same time, reducing the deleterious effects. Overall, we observed that structural changes in the analogs were associated with more potent biological activity, for example, substantially increased the activity on the Gram-positive bacteria Staphylococcus aureus while the activity on the Gram-negative bacterium Escherichia coli decreased. On the other hand, we observed that some structural changes culminate in the total loss of activity on bacteria and on the eukaryotic cells tested. To evaluate the effects of the analogs on eukaryotic cells we used flow cytometry and proteomics methods. Using flow cytometry, we observed that analogs 6 and 8 (AN6 and AN8) induced several changes in the B16F10 and MCF-7 cell lines such as changes in size and granularity and positive staining for annexin V/FITC and propidium iodide, suggesting occurrence of necrosis. We also observed expressive labeling for caspases when B16F10 cells were treated with peptides. The treatment of B16F10 and MCF-7 cells with AN6 and AN8 peptides induced mitochondrial membrane depolarization, whereas lysosomal permeability was not affected in B16F10 cells, while there was a significant lysosomal permeability in MCF-7 cells treated with both peptides. A significant DNA fragmentation in the two strains treated with both AN6 and AN8 was detected. On the other hand, changes in the cell cycle were observed mostly in B16F10 cells. In the proteomic analysis, we used the B16F10 strain treated with the AN6 peptide. After statistical analysis, we found 242 differentially expressed proteins of which 103 were with the pattern of increased abundance and 139 proteins with decreased abundance compared to the control. Unlike what we observed in the cytometry assays, we did not find proteins related to cell death processes with increased expression
Acurácia da ressonância magnética para identificar lesões traumáticas intra-articulares do joelho
PURPOSE: To evaluate the diagnostic accuracy of magnetic resonance imaging of the knee in identifying traumatic intraarticular knee lesions. METHOD: 300 patients with a clinical diagnosis of traumatic intraarticular knee lesions underwent prearthoscopic magnetic resonance imaging. The sensitivity, specificity, positive predictive value, negative predictive value, likelihood ratio for a positive test, likelihood ratio for a negative test, and accuracy of magnetic resonance imaging were calculated relative to the findings during arthroscopy in the studied structures of the knee (medial meniscus, lateral meniscus, anterior cruciate ligament, posterior cruciate ligament, and articular cartilage). RESULTS: Magnetic resonance imaging produced the following results regarding detection of lesions: medial meniscus: sensitivity 97.5%, specificity 92.9%, positive predictive value 93.9%, positive negative value 97%, likelihood positive ratio 13.7, likelihood negative ratio 0.02, and accuracy 95.3%; lateral meniscus: sensitivity 91.9%, specificity 93.6%, positive predictive value 92.7%, positive negative value 92.9%, likelihood positive ratio 14.3, likelihood negative ratio 0.08, and accuracy 93.6%; anterior cruciate ligament: sensitivity 99.0%, specificity 95.9%, positive predictive value 91.9%, positive negative value 99.5%, likelihood positive ratio 21.5, likelihood negative ratio 0.01, and accuracy 96.6%; posterior cruciate ligament: sensitivity 100%, specificity 99%, positive predictive value 80.0%, positive negative value 100%, likelihood positive ratio 100, likelihood negative ratio 0.01, and accuracy 99.6%; articular cartilage: sensitivity 76.1%, specificity 94.9%, positive predictive value 94.7%, positive negative value 76.9%, likelihood positive ratio 14.9, likelihood negative ratio 0.25, and accuracy 84.6%. CONCLUSION: Magnetic resonance imaging is a satisfactory diagnostic tool for evaluating meniscal and ligamentous lesions of the knee, but it is unable to clearly identify articular cartilage lesions.OBJETIVO: Avaliar a validade da ressonância magnética do joelho no diagnóstico das lesões intra-articulares traumáticas do joelho. MÉTODO: População de 300 pacientes, com quadro clÃnico sugestivo de lesões intra-articulares traumáticas do joelho, que tiveram seus laudos de ressonância magnética comparados com os resultados obtidos nas artroscopias realizadas posteriormente. Foram calculados a sensibilidade, especificidade, valor preditivo positivo, valor preditivo negativo, razão de verossimilhança positiva, razão de verossimilhança negativa e acurácia da ressonância magnética do joelho para o diagnóstico de lesões em cada estrutura intra-articular estudada do joelho (menisco medial, menisco lateral, ligamento cruzado anterior, ligamento cruzado posterior e cartilagem articular). RESULTADOS: Em relação à s lesões do menisco medial, a sensibilidade da ressonância magnética foi de 97.5%, a especificidade de 92.9% o valor preditivo positivo de 93.9%, o valor preditivo negativo de 97%, a razão de verossimilhança positiva de 13.7, a razão de verossimilhança negativa de 0.02 e a acurácia de 95.3%. Para o menisco lateral, a sensibilidade da ressonância magnética foi de 91.9%, a especificidade de 93.6%, o valor preditivo positivo de 92.7%, o valor preditivo negativo de 92.9%, a razão de verossimilhança positiva de 14.3, a razão de verossimilhança negativa de 0.08 e a acurácia de 93.6%. Para o ligamento cruzado anterior, a sensibilidade da ressonância magnética foi de 99.0%, a especificidade de 95.4%, o valor preditivo positivo de 91.9%, o valor preditivo negativo de 99.5%, a razão de verossimilhança positiva de 21.5, a razão de verossimilhança negativa de 0.01 e a acurácia de 96.6%. Para o ligamento cruzado posterior, a sensibilidade da ressonância magnética foi de 100%, a especificidade de 99%, o valor preditivo positivo de 80%, o valor preditivo negativo de 100%, a razão de verossimilhança positiva de 100, a razão de verossimilhança negativa de 0.01 e a acurácia de 99.6%. Para as lesões condrais a sensibilidade da ressonância magnética foi de 76.1%, a especificidade de 94.9%, o valor preditivo positivo de 94.7%, o valor preditivo negativo de 76.9%, a razão de verossimilhança positiva de 14.9, a razão de verossimilhança negativa de 0.25 e a acurácia de 84.6%. CONCLUSÃO: A ressonância magnética apresenta alta acurácia para identificar as lesões meniscais e ligamentares do joelho, mas é insatisfatória para diagnosticar as lesões da cartilagem articular
