    Cheese whey: a cost-effective alternative for hyaluronic acid production by Streptococcus zooepidemicus

    This study focuses on the optimization of cheese whey formulated media for the production of hyaluronic acid HA by Streptococcus zooepidemicus. Culture media containing whey (W; 2.1 g/L) or whey hydrolysate (WH; 2.4 g/L) gave the highest HA productions. Both W and WH produced high yields on protein consumed, suggesting cheese whey is a good nitrogen source for S. zooepidemicus production of HA. Polysaccharide concentrations of 4.0 g/L and 3.2 g/L were produced in W and WH in a further scale-up to 5 L bioreactors, confirming the suitability of the low-cost nitrogen source. Cheese whey culture media provided high molecular weight (> 3000 kDa) HA products. This study revealed replacing the commercial peptone by the low-cost alternative could reduce HA production costs by up to a 70%compared to synthetic media.Isabel Rodriguez was funded by a postdoctoral contract from the Xunta de Galicia, Spain (Plan I2C, 2012).     Hydrothermal Carbonization of Biomass Wastes: Sustainability and Geochemistry

    Introduction. To reduce the stream of solid waste going to landfills, innovative means for beneficial use are essential. The diversity and volume of organic wastes pose singular problems and opportunities for recovery and circularity. Common processes for organics include conversion to biofuels and carbonization to biochar, typically done by torrefaction (dry pyrolysis). Research on biochar explores its potential as pollutant adsorbent, agricultural or polluted soil amendment, biofuel (directly or as feedstock), and for carbon sequestration (Ighalo et al., 2022; Cavali et al., 2023). Recently, other processes at lower temperatures such as hydrothermal carbonization (HTC) offer new possibilities (Seshadri et al., 2016; Madsen et al., 2017). The properties of both biochar and hydrochar are strongly dependent on the biomass feedstock type (e.g., wood vs. algae) and on the carbonization process employed. In a real world pilot study, the public company for solid waste management in Asturias, Spain (COGERSA SAU) experimented with HTC of organic waste streams, including treated wood waste (mainly bulky waste, furniture, etc.), winery bagasse, and the organic fraction from separate collection of municipal solid waste (OFMSW). An advantage of HTC is that the feedstock can be processed wet, as received. Water was added to achieve a uniform 1:4 solid to water mass ratio and the mixtures were processed in a 2 m3 reactor vessel (195 °C, 1.3 MPa, 3 h) fed by the superheated steam available at the COGERSA plant (waste-to-energy of clinical waste by rotary kiln). Then, excess water was expelled mechanically (30 MPa) and the solids were dried in air at 100 °C. The resulting hydrochars were characterized by methods organic geochemists traditionally use in fossil fuel studies: proximate and ultimate analyses, porosimetry, thermogravimetric analysis, thermodesorption-gas chromatography-mass spectrometry (TD-GC-MS, heating in He at 350 °C for 20 s), and pyrolysis of the thermodesorption residue (Py-GC-MS, 610 °C, 20 s). For comparison, a commercial torrefied biochar (Vermichar S.L.) produced from Quercus ilex wood was also analyzed by the above methods. Results. Yields of hydrochar varied from 65% (OFMSW) to 83% (wood) of the initial feedstock mass (dry, ash-free). Molar H/C and O/C ratios decreased by mean values of 10 and 24%, respectively, relative to the raw feedstocks. HTC increased the high heating values slightly, to a mean of 21 MJ kg-1, in the range of sub-bituminous coal. Thermogravimetry overall shows a slight shift towards decomposition at higher temperatures, as well as peak narrowing after HTC. OFMSW TD-GC-MS yielded predominantly C16 and C18 saturated and unsaturated fatty acids and their methyl and ethyl esters (Fig. 1A). Subsequent Py-GC-MS results were similar, but with greatly reduced yield. In contrast, the Py-GC-MS yield for the wood was greater than that of the preceding thermodesorption, with abundant lignin (methoxyphenol) and polysaccharide markers, and minor, but notable, monoaromatic tricyclic terpenoids and methoxyphenol dimers (Fig. 1B). The TD-GC-MS of the biochar yielded little, while subsequent Py-GC-MS presented a predominance of simple monoaromatic hydrocarbons and phenols, along with phenanthrenes, retene in particular (Fig. 1C). Minor naphthalenes and monoaromatic tricyclic terpenoids are also noted. Conclusions. Analytical techniques usually applied in advanced characterization of fossil fuels, such as Py-GC-MS, were shown to be useful in unraveling molecular hydrochar and biochar composition. This opens new possibilities for the study of these products in detail, including diverse applications such as tracking of the evolution of different feedstock materials after carbonization, the possible uses as biofuels (specially for the liquid fraction obtained in the HTC process), or even the identification of potential pollutants. In this preliminary study of several hydrochars and a commercial biochar, interesting general and specific features were revealed: the molecular composition of hydrochar was strongly influenced by feedstock type, the OFMSW hydrochar revealed potential for biodiesel production, and biochar presented marked differences from “woody” biomass hydrochars

    [Abstract] Beta-type titanium alloy surfaces (Tie35Nbe10Ta (TNT) and Tie30Nbe4Sn (TNS)) weremodified by laser using a power of 1000 W and speed of 6.7 mm/s (Condition A) and a powerof 1500 W with speed of 10 mm/s (Condition B).     Self-determination in teaching formative basketball players and its influence on respect to rules and opponents

    The aim of this study was to examine adaptive behaviors in teaching formative basketball players with regard to their motivations to play. Hence, 284 federative basketball players, ranging in age from 11 to 16 years old from the Region of Extremadura took part in the study. Descriptive analysis, correlation analysis and lineal regression analysis were conducted with SPSS 19.0 statistical program. Results showed that more self-determined players had greater adaptive conducts, such as respect to rules and opponents. Finally, it is noteworthy to provide intrinsic regulations in youth basketball players to create a good learning and promotes social values

    Synthesis, structure and antileishmanial evaluation of endoperoxide–pyrazole hybrids

    Leishmaniases are among the most impacting neglected tropical diseases. In attempts to repurpose antimalarial drugs or candidates, it was found that selected 1,2,4-trioxanes, 1,2,4,5-tetraoxanes, and pyrazole-containing chemotypes demonstrated activity against Leishmania parasites. This study reports the synthesis and structure of trioxolane–pyrazole (OZ1, OZ2) and tetraoxane–pyrazole (T1, T2) hybrids obtained from the reaction of 3(5)-aminopyrazole with endoperoxide-containing building blocks. Interestingly, only the endocyclic amine of 3(5)-aminopyrazole was found to act as nucleophile for amide coupling. However, the fate of the reaction was influenced by prototropic tautomerism of the pyrazole heterocycle, yielding 3- and 5-aminopyrazole containing hybrids which were characterized by different techniques, including X-ray crystallography. The compounds were evaluated for in vitro antileishmanial activity against promastigotes of L. tropica and L. infantum, and for cytotoxicity against THP-1 cells. Selected compounds were also evaluated against intramacrophage amastigote forms of L. infantum. Trioxolane–pyrazole hybrids OZ1 and OZ2 exhibited some activity against Leishmania promastigotes, while tetraoxane–pyrazole hybrids proved inactive, most likely due to solubility issues. Eight salt forms, specifically tosylate, mesylate, and hydrochloride salts, were then prepared to improve the solubility of the corresponding peroxide hybrids and were uniformly tested. Biological evaluations in promastigotes showed that the compound OZ1•HCl was the most active against both strains of Leishmania. Such finding was corroborated by the results obtained in assessments of the L. infantum amastigote susceptibility. It is noteworthy that the salt forms of the endoperoxide–pyrazole hybrids displayed a broader spectrum of action, showing activity in both strains of Leishmania.     Aquatic contamination by heavy metals is a major concern for the serious negative consequences it has for plants, animals, and humans. Among the most toxic metals, Cd(II) stands out since selective and truly efficient methodologies for its removal are not known. We report a novel multidentate chelating agent comprising the heterocycles thiadiazole and benzisothiazole. 3-((5-(trifluoromethyl)-1,3,4-thiadiazol-2-yl)amino)benzo[d]isothiazole 1,1-dioxide (AL14) was synthesized from cheap saccharin and characterized by different techniques, including single crystal X-ray crystallography. Our studies revealed the efficiency and selectivity of AL14 for the chelation of dissolved Cd(II) (as compared to Cu(II) and Fe(II)).     Molecular and crystal structure, spectroscopy, and photochemistry of a dispiro compound bearing the tetraoxane pharmacophore

    The molecular structure and photochemistry of dispiro[cyclohexane-1,3′-[1,2,4,5]tetraoxane-6′,2′′-tricyclo[3.3.1.13,7]decan]-4-one (TX), an antiparasitic 1,2,4,5-tetraoxane was investigated using matrix isolation IR and EPR spectroscopies, together with quantum chemical calculations undertaken at the DFT(B3LYP)/6-311++G(3df,3pd) level of theory, with and without Grimme's dispersion correction. Photolysis of the matrix-isolated TX, induced by in situ broadband (λ>235 nm) or narrowband (λ in the range 220–263 nm) irradiation, led to new bands in the infrared spectrum that could be ascribed to two distinct photoproducts, oxepane-2,5-dione, and 4-oxohomoadamantan-5-one. Our studies show that these photoproducts result from initial photoinduced cleavage of an O−O bond, with the formation of an oxygen-centered diradical that regioselectivity rearranges to a more stable (secondary carbon-centered)/(oxygen-centered) diradical, yielding the final products. Formation of the diradical species was confirmed by EPR measurements, upon photolysis of the compound at λ=266 nm, in acetonitrile ice (T=10–80 K). Single-crystal X-ray diffraction (XRD) studies demonstrated that the TX molecule adopts nearly the same conformation in the crystal and matrix-isolation conditions, revealing that the intermolecular interactions in the TX crystal are weak. This result is in keeping with observed similarities between the infrared spectrum of the crystalline material and that of matrix-isolated TX. D.; Vanderspek, R.; Watanabe, D.; Wittrock, J.; Ziegler, C.; Zohrabi, F.We present the discovery and characterisation of two transiting planets observed by the Transiting Exoplanet Survey Satellite (TESS) orbiting the nearby (d∗ ≈ 22 pc), bright (J ≈ 9 mag) M3.5 dwarf LTT 3780 (TOI-732). We confirm both planets and their association with LTT 3780 via ground-based photometry and determine their masses using precise radial velocities measured with the CARMENES spectrograph. Precise stellar parameters determined from CARMENES high-resolution spectra confirm that LTT 3780 is a mid-M dwarf with an effective temperature of Teff = 3360 ± 51 K, a surface gravity of log g∗ = 4.81 ± 0.04 (cgs), and an iron abundance of [Fe/H] = 0.09 ± 0.16 dex, with an inferred mass of M∗ = 0.379 ± 0.016M· and a radius of R∗ = 0.382 ± 0.012R·. The ultra-short-period planet LTT 3780 b (Pb = 0.77 d) with a radius of 1.35-0.06+0.06 R·, a mass of 2.34-0.23+0.24 M·, and a bulk density of 5.24-0.81+0.94 g cm-3 joins the population of Earth-size planets with rocky, terrestrial composition. The outer planet, LTT 3780 c, with an orbital period of 12.25 d, radius of 2.42-0.10+0.10 R·, mass of 6.29-0.61+0.63 M·, and mean density of 2.45-0.37+0.44 g cm-3 belongs to the population of dense sub-Neptunes. With the two planets located on opposite sides of the radius gap, this planetary system is anexcellent target for testing planetary formation, evolution, and atmospheric models.     The relationship between the severity and mortality of SARS-CoV-2 infection and 25-hydroxyvitamin D concentration — a metaanalysis

    Introduction: There is increasing scientific interest in the possible association between hypovitaminosis D and the risk of SARS-CoV-2 infection severity and/or mortality.  Objective: To conduct a metanalysis of the association between 25-hydroxyvitamin D (25(OH)D) concentration and SARS-CoV-2 infection severity or mortality.Material and methods: We searched PubMed, EMBASE, Google scholar and the Cochrane Database of Systematic Reviews for studies published between December 2019 and December 2020. Effect statistics were pooled using random effects models. The quality of included studies was assessed with the Newcastle–Ottawa Scale (NOS). Targeted outcomes: mortality and severity proportions in COVID-19 patients with 25(OH)D deficiency, defined as serum 25(OH)D < 50 nmol/L.  Results: In the 23 studies included (n = 2692), the mean age was 60.8 (SD ± 15.9) years and 53.8% were men. Results suggested that vitamin 25(OH)D deficiency was associated with increased risk of severe SARS-CoV-2 disease (RR 2.00; 95% CI 1.47–2.71, 17 studies) and mortality (RR 2.45; 95% CI 1.24–4.84, 13 studies). Only 7/23 studies reported C-reactive protein values, all of which were > 10 mg/L.Conclusions 25(OH)D deficiency seems associated with increased SARS-CoV-2 infection severity and mortality. However, findings do not imply causality, and randomized controlled trials are required, and new studies should be designed to determine if decreased 25(OH)D is an epiphenomenon or consequence of the inflammatory process associated with severe forms of SARS-CoV-2. Meanwhile, the concentration of 25(OH)D could be considered as a negative acute phase reactant and a poor prognosis in COVID-19 infection
