    Procesos simultáneos de producción de enzimas hidrolíticas y remoción biológica de hemicelulosa bajo el concepto de biorrefinería

    El objetivo de este trabajo contempló el diseñó y caracterización de un biorreactor de sustrato sólido (lecho percolador con recirculación) (BLPR) para la producción de enzimas hemicelulolíticas extracelulares por parte de Cellulomonas flavigena PR-22. Se construyeron dos biorreactores BLPR, un prototipo constituido de borosilicato (al cual se le denomino BLPR “RBK”) y el otro fue construido de acero inoxidable 304 (al cual se denominó “MBQ”). Dentro del cuerpo del reactor, se encuentra dispuesto un sistema de sujeción de biomasa y un aspersor de medio de cultivo. Además, el reactor cuenta con cuatro entradas de aeración y temperatura controlada en distintos puntos del lecho. El BLPR se caracterizó térmica e hidráulicamente. Los parámetros de operación fueron establecidos de acuerdo con las condiciones adecuadas de crecimiento de C. flavigena PR-22 en material lignocelulósico (32°C, 75% H, 1.5 vvm, pH = 6 - 8), como diluyente se utilizó un medio mineral mínimo formado por sales grado industrial enriquecido con extracto de levadura (0.2%). Se utilizaron dos distintas biomasas: rastrojo de maíz (RM200) y bagazo de agave (BGA), ambas pretratadas por explosión de vapor. Con la operación del sistema utilizando RM200, se obtuvo una producción 2.2 veces mayor de hemicelulasas en comparación con un tanque agitado (BTA) durante la primera semana de cultivo. Con bagazo de agave se produjeron 2200 U/mL de actividad hemicelulasa en la primera semana de cultivo. En ambos casos, el crecimiento de C. flavigena PR-22 fue ajustado a un modelo cinético exponencial y se determinó una velocidad especifica de crecimiento máxima (μmax) de 0.00375 h-1 en RM200 y de 0.08865 h-1 en BGA. La recirculación del microorganismo y de las enzimas solubles en el medio líquido permite una constante hidrólisis de la holocelulosa del sustrato, lo cual permite el crecimiento microbiano, producción de enzimas y remoción de la fracción de hemicelulosa.     Design and implementation in USRP of a preamble-based synchronizer for OFDM systems

    The Orthogonal Frequency Division Multiplexing (OFDM) is one of the most widely adopted schemes in wireless technologies such as Wi-Fi and LTE due to its high transmission rates, and the robustness against Intersymbol Interference (ISI). However, OFDM is highly sensitive to synchronism errors, which affects the orthogonality of the carriers. We analyzed several synchronization algorithms based on the correlation of the preamble symbols through the implementation in Software-Defined Radio (SDR) using the Universal Software Radio Peripheral (USRP). Such an implementation was performed in three stages: frame detection, comparing the autocorrelation output and the average power of the received signal; time synchronism, where the cross-correlation based on the short and long preamble symbols was implemented; and the frequency synchronism, where the Carrier Frequency Offset (CFO) added by the channel was detected and corrected. The synchronizer performance was verified through the USRP implementation.     The native turkey (Meleagris gallopavo): male morphophysiology

    Objective: To describe the morphophysiology and reproductive characteristics of the male turkey. Methodology: An analysis based on the practical experiences from over 10 years with the native Mexican turkey was developed and complemented with literature reports. Results: In Mexico the production management is customs based on traditions and beliefs. In their reproductive morphophysiology, the epididymis absence stands out, as well as a rapid testicular growth during the breeding season, which is controlled by the photoperiod. Limitations of the study: Native turkey farming is practiced in backyard, in marginalized and low-resource areas that have no strategies (nutritional and reproductive) to improve their production. Conclusions: The anatomical, morphological, physiological and reproductive characteristics of the native Mexican turkey were documented. The knowledge on these characteristics will allow to develop feeding and reproductive strategies to improve the productive and reproductive performance of the native turkey and preserve their genetic resources

    The Mutational Landscape of Acute Myeloid Leukaemia Predicts Responses and Outcomes in Elderly Patients from the PETHEMA-FLUGAZA Phase 3 Clinical Trial

    Mutational profiling using a custom 43-gene next-generation sequencing panel revealed that patients with mutated DNMT3A or EZH2, or an increase in TET2 VAF and lower TP53 VAF showed a higher overall response. NRAS and TP53 variants were associated with shorter overall survival (OS), whereas only mutated BCOR was associated with a shorter relapse-free survival (RFS). Subgroup analyses of OS according to biological and genomic characteristics showed that patients with low-intermediate cytogenetic risk and mutated NRAS benefited from azacytidine therapy and patients with mutated TP53 showed a better RFS in the azacytidine arm. In conclusion, differential mutational profiling might anticipate the outcomes of first-line treatment choices (AZA or FLUGA) in older patients with AML. We sought to predict treatment responses and outcomes in older patients with newly diagnosed acute myeloid leukemia (AML) from our FLUGAZA phase III clinical trial (PETHEMA group) based on mutational status, comparing azacytidine (AZA) with fludarabine plus low-dose cytarabine (FLUGA). Mutational profiling using a custom 43-gene next-generation sequencing panel revealed differences in profiles between older and younger patients, and several prognostic markers that were useful in young patients were ineffective in older patients. We examined the associations between variables and overall responses at the end of the third cycle. Patients with mutated DNMT3A or EZH2 were shown to benefit from azacytidine in the treatment-adjusted subgroup analysis. An analysis of the associations with tumor burden using variant allele frequency (VAF) quantification showed that a higher overall response was associated with an increase in TET2 VAF (odds ratio (OR), 1.014; p = 0.030) and lower TP53 VAF (OR, 0.981; p = 0.003). In the treatment-adjusted multivariate survival analyses, only the NRAS (hazard ratio (HR), 1.9, p = 0.005) and TP53 (HR, 2.6, p = 9.8 × 10 −7) variants were associated with shorter overall survival (OS), whereas only mutated BCOR (HR, 3.6, p = 0.0003) was associated with a shorter relapse-free survival (RFS). Subgroup analyses of OS according to biological and genomic characteristics showed that patients with low-intermediate cytogenetic risk (HR, 1.51, p = 0.045) and mutated NRAS (HR, 3.66, p = 0.047) benefited from azacytidine therapy. In the subgroup analyses, patients with mutated TP53 (HR, 4.71, p = 0.009) showed a better RFS in the azacytidine arm. In conclusion, differential mutational profiling might anticipate the outcomes of first-line treatment choices (AZA or FLUGA) in older patients with AML. Using the maximally selected log-rank statistics, we established an optimal cutoff of FLT3–ITD AR of 0.44 for OS, and 0.8 for RFS. We analyzed the OS and RFS according to FLT3–ITD status in all patients, and we found that the group of FLT3–ITD-positive patients with AR 0.44, allo-HSCT was superior to auto-HSCT in terms of OS and RFS.     Characteristics and outcomes of adult patients in the PETHEMA registry with relapsed or refractory FLT3-ITD mutation-positive acute myeloid leukemia

    This retrospective study investigated outcomes of 404 patients with relapsed/refractory (R/R) FMS-like tyrosine kinase 3 (FLT3)-internal tandem duplication (ITD) acute myeloid leukemia (AML) enrolled in the PETHEMA registry, pre-approval of tyrosine kinase inhibitors. Most patients (63%) had received first-line intensive therapy with 3 + 7. Subsequently, patients received salvage with intensive therapy (n = 261), non-intensive therapy (n = 63) or supportive care only (n = 80). Active salvage therapy (i.e., intensive or non-intensive therapy) resulted in a complete remission (CR) or CR without hematological recovery (CRi) rate of 42%. More patients achieved a CR/CRi with intensive (48%) compared with non-intensive (19%) salvage therapy (p < 0.001). In the overall population, median overall survival (OS) was 5.5 months; 1- and 5-year OS rates were 25% and 7%. OS was significantly (p < 0.001) prolonged with intensive or non-intensive salvage therapy compared with supportive therapy, and in those achieving CR/CRi versus no responders. Of 280 evaluable patients, 61 (22%) had an allogeneic stem-cell transplant after they had achieved CR/CRi. In conclusion, in this large cohort study, salvage treatment approaches for patients with FLT3-ITD mutated R/R AML were heterogeneous. Median OS was poor with both non-intensive and intensive salvage therapy, with best long-term outcomes obtained in patients who achieved CR/CRi and subsequently underwent allogeneic stem-cell transplant.This study was supported by Centro de Investigación Biomédica en Red Cáncer (CIBERONC), Valencia, Spain [CB16/12/00284]

    Gemtuzumab ozogamicin (GO) is a conjugate of a monoclonal antibody and calicheamicin, which has been reapproved for the treatment of acute myeloid leukemia (AML). AML patients with the CD33 rs12459419 CC genotype might benefit from the addition of GO to intensive treatment in contrast to patients with CT/TT genotypes. Nevertheless, contradictory results have been reported. We sought to shed light on the prediction of GO response in AML patients with rs12459419 polymorphism who were treated with GO in the consolidation (n = 70) or reinduction (n = 20) phase. The frequency distribution of the rs12459419 polymorphism in the complete cohort of patients was 44.4% (n = 40), 50% (n = 45), and 5.6% (n = 5) for CC, CT, and TT genotypes, respectively. Regarding the patients treated with GO for consolidation, we performed a Kaplan-Meier analysis of overall survival and relapse-free survival according to the rs12459419 polymorphism (CC vs. CT/TT patients) and genetic risk using the European Leukemia Net (ELN) 2010 risk score. We also carried out a Cox regression analysis for the prediction of overall survival, with age and ELN 2010 as covariates. We found no statistical significance in the univariate or multivariate analysis. Additionally, we performed a global Kaplan-Meier analysis for the patients treated with GO for reinduction and did not find significant differences; however, our cohort was too small to draw any conclusion from this analysis. The use of GO in consolidation treatment is included in the approval of the compound; however, evidence regarding its efficacy in this setting is lacking. Rs12459419 polymorphism could help in the selection of patients who might benefit from GO. Regrettably, in our cohort, the rs12459419 polymorphism does not seem to be an adequate tool for the selection of patients who might benefit from the addition of GO in consolidation cycles
