A signal sequence is not required for protein export in prlA mutants of Escherichia coli.
The prA/secY gene, which codes for an integral membrane
protein component of the Escherichia coli protein export
machinery, is the locus of the strongest suppressors of
signal sequence mutations. We demonstrate that two
exported proteins of E.coli, maltose-binding protein and
alkaline phosphatase, each lacking its entire signal
sequence, are exported to the periplasm in several priA
mutants. The export efficiency can be substantial; in a
strain carrying the prUA4 allele, 30% of signal-sequenceless alkaline phosphatase is exported to the periplasm.
Other components of the E.coli export machinery,
including SecA, are required for this export. SecB is
required for the export of signal-sequenceless alkaline
phosphatase even though the normal export of alkaline
phosphatase does not require this chaperonin. Our
findings indicate that signal sequences confer speed and
efficiency upon the export process, but that they are not
always essential for export. Entry into the export pathway
may involve components that so overlap in function that
the absence of a signal sequence can be compensated for,
or there may exist one or more means of entry that do
not require signal sequences at all
A Public, K-Selected, Optical-to-Near-Infrared Catalog of the Extended Chandra Deep Field South (ECDFS) from the MUltiwavelength Survey by Yale-Chile (MUSYC)
We present a new K-selected, optical-to-near-infrared photometric catalog of
the Extended Chandra Deep Field South (ECDFS), making it publicly available to
the astronomical community. The dataset is founded on publicly available
imaging, supplemented by original zJK imaging data obtained as part of the
MUltiwavelength Survey by Yale-Chile (MUSYC). The final photometric catalog
consists of photometry derived from nine band U-K imaging covering the full
0.5x0.5 sq. deg. of the ECDFS, plus H band data for approximately 80% of the
field. The 5sigma flux limit for point-sources is K = 22.0 (AB). This is also
the nominal completeness and reliability limit of the catalog: the empirical
completeness for 21.75 < K < 22.00 is 85+%. We have verified the quality of the
catalog through both internal consistency checks, and comparisons to other
existing and publicly available catalogs. As well as the photometric catalog,
we also present catalogs of photometric redshifts and restframe photometry
derived from the ten band photometry. We have collected robust spectroscopic
redshift determinations from published sources for 1966 galaxies in the
catalog. Based on these sources, we have achieved a (1sigma) photometric
redshift accuracy of Dz/(1+z) = 0.036, with an outlier fraction of 7.8%. Most
of these outliers are X-ray sources. Finally, we describe and release a utility
for interpolating restframe photometry from observed SEDs, dubbed InterRest.
Particularly in concert with the wealth of already publicly available data in
the ECDFS, this new MUSYC catalog provides an excellent resource for studying

Hubble Space Telescope WFC3 Early Release Science: Emission-Line Galaxies from Infrared Grism Observations
We present grism spectra of emission-line galaxies (ELGs) from 0.6-1.6
microns from the Wide Field Camera 3 on the Hubble Space Telescope. These new
infrared grism data augment previous optical Advanced Camera for Surveys G800L
0.6-0.95 micron grism data in GOODS-South from the PEARS program, extending the
wavelength covereage well past the G800L red cutoff. The ERS grism field was
observed at a depth of 2 orbits per grism, yielding spectra of hundreds of
faint objects, a subset of which are presented here. ELGs are studied via the
Ha, [OIII], and [OII] emission lines detected in the redshift ranges 0.2<z<1.4,
1.2<z<2.2 and 2.0<z<3.3 respectively in the G102 (0.8-1.1 microns; R~210) and
G141 (1.1-1.6 microns; R~130) grisms. The higher spectral resolution afforded
by the WFC3 grisms also reveals emission lines not detectable with the G800L
grism (e.g., [SII] and [SIII] lines). From these relatively shallow
observations, line luminosities, star-formation rates, and grism spectroscopic
redshifts are determined for a total of 48 ELGs to m(AB)~25 mag. Seventeen
GOODS-South galaxies that previously only had photometric redshifts now have
new grism-spectroscopic redshifts, in some cases with large corrections to the
photometric redshifts (Delta(z)~0.3-0.5). Additionally, one galaxy had no
previously-measured redshift but now has a secure grism-spectroscopic redshift,
for a total of 18 new GOODS-South spectroscopic redshifts. The faintest source
in our sample has a magnitude m(AB)=26.9 mag. The ERS grism data also reflect
the expected trend of lower specific star formation rates for the highest mass
galaxies in the sample as a function of redshift, consistent with downsizing
and discovered previously from large surveys. These results demonstrate the
remarkable efficiency and capability of the WFC3 NIR grisms for measuring
galaxy properties to faint magnitudes and redshifts to z>2.Comment: Accepted for publication in AJ. Updated to include referee comments.

The Hubble Space Telescope Wide Field Camera 3 Early Release Science data: Panchromatic Faint Object Counts for 0.2-2 microns wavelength
We describe the Hubble Space Telescope (HST) Wide Field Camera 3 (WFC3) Early
Release Science (ERS) observations in the Great Observatories Origins Deep
Survey (GOODS) South field. The new WFC3 ERS data provide calibrated, drizzled
mosaics in the UV filters F225W, F275W, and F336W, as well as in the near-IR
filters F098M (Ys), F125W (J), and F160W (H) with 1-2 HST orbits per filter.
Together with the existing HST Advanced Camera for Surveys (ACS) GOODS-South
mosaics in the BViz filters, these panchromatic 10-band ERS data cover 40-50
square arcmin at 0.2-1.7 {\mu}m in wavelength at 0.07-0.15" FWHM resolution and
0.090" Multidrizzled pixels to depths of AB\simeq 26.0-27.0 mag (5-{\sigma})
for point sources, and AB\simeq 25.5-26.5 mag for compact galaxies.
In this paper, we describe: a) the scientific rationale, and the data taking
plus reduction procedures of the panchromatic 10-band ERS mosaics; b) the
procedure of generating object catalogs across the 10 different ERS filters,
and the specific star-galaxy separation techniques used; and c) the reliability
and completeness of the object catalogs from the WFC3 ERS mosaics. The
excellent 0.07-0.15" FWHM resolution of HST/WFC3 and ACS makes star- galaxy
separation straightforward over a factor of 10 in wavelength to AB\simeq 25-26

Precis of Vaulting Ambition: Sociobiology and the Quest for Human Nature
The debate about the credentials of sociobiology has persisted because scholars have failed to distinguish the varieties of sociobiology and because too little attention has been paid to the details of the arguments that are supposed to support the provocative claims about human social behavior. I seek to remedy both dcfieieneies. After analysis of the relationships among different kinds of sociobiology and contemporary evolutionary theory, I attempt to show how some of the studies of the behavior of nonhuman animals meet the methodological standards appropriate to evolutionary research. I contend that the efforts of E. O. Wilson, Richard Alexander, Charles Lumsden, and others to generate conclusions about human nature are flawed, both because they apply evolutionary ideas in an unrigorous fashion and because they use dubious assumptions to connect their evolutionary analyses with their conclusions. This contention rests on analyses of many of the major sociobiological proposals about human social behavior, including: differences in sex roles, racial hostility, homosexuality, conflict between parents and adolescent offspring, incest avoidance, the avunculate, alliances in combat, female infanticide, and gene-culture coevolution. Vaulting Ambition thus seeks to identify what is good in sociobiology, to expose the errors of premature speculations about human nature, and to prepare the way for serious study of the evolution of human social behavior
Family Firms and Firm Performance: Evidence from Japan

By applying a calibrated force to a substrate disulfide, the chemical mechanisms of Trx catalysis can be examined in detail at the single-molecule level. Here we use single-molecule force-clamp spectroscopy to explore the chemical evolution of Trx catalysis by probing the chemistry of eight different Trx enzymes. All Trxs show a characteristic Michaelis-Menten mechanism that is detected when the disulfide bond is stretched at low forces, but at high forces, two different chemical behaviors distinguish bacterial-origin from eukaryotic-origin Trxs. Eukaryotic-origin Trxs reduce disulfide bonds through a single-electron transfer reaction (SET), whereas bacterial-origin Trxs show both nucleophilic substitution (SN2) and SET reactions. A computational analysis of Trx structures identifies the evolution of the binding groove as an important factor controlling the chemistry of Trx catalysis
Risk factors for Coronavirus disease 2019 (Covid-19) death in a population cohort study from the Western Cape province, South Africa
Risk factors for coronavirus disease 2019 (COVID-19) death in sub-Saharan Africa and the effects of human immunodeficiency virus (HIV) and tuberculosis on COVID-19 outcomes are unknown. We conducted a population cohort study using linked data from adults attending public-sector health facilities in the
Western Cape, South Africa. We used Cox proportional hazards models, adjusted for age, sex, location, and comorbidities, to examine the associations between HIV, tuberculosis, and COVID-19 death from 1 March to 9 June 2020 among (1) public-sector “active patients” (≥1 visit in the 3 years before March 2020); (2) laboratory-diagnosed COVID-19 cases; and (3) hospitalized COVID-19
cases. We calculated the standardized mortality ratio (SMR) for COVID-19, comparing adults living with and without HIV using
modeled population estimates.Among 3 460 932 patients (16% living with HIV), 22 308 were diagnosed with COVID-19, of whom 625 died. COVID19 death was associated with male sex, increasing age, diabetes, hypertension, and chronic kidney disease. HIV was associated with
COVID-19 mortality (adjusted hazard ratio [aHR], 2.14; 95% confidence interval [CI], 1.70–2.70), with similar risks across strata of
viral loads and immunosuppression. Current and previous diagnoses of tuberculosis were associated with COVID-19 death (aHR,
2.70 [95% CI, 1.81–4.04] and 1.51 [95% CI, 1.18–1.93], respectively). The SMR for COVID-19 death associated with HIV was 2.39
(95% CI, 1.96–2.86); population attributable fraction 8.5% (95% CI, 6.1–11.1)
Investigation of hospital discharge cases and SARS-CoV-2 introduction into Lothian care homes
Background
The first epidemic wave of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) in Scotland resulted in high case numbers and mortality in care homes. In Lothian, over one-third of care homes reported an outbreak, while there was limited testing of hospital patients discharged to care homes.
Aim
To investigate patients discharged from hospitals as a source of SARS-CoV-2 introduction into care homes during the first epidemic wave.
Methods
A clinical review was performed for all patients discharges from hospitals to care homes from 1st March 2020 to 31st May 2020. Episodes were ruled out based on coronavirus disease 2019 (COVID-19) test history, clinical assessment at discharge, whole-genome sequencing (WGS) data and an infectious period of 14 days. Clinical samples were processed for WGS, and consensus genomes generated were used for analysis using Cluster Investigation and Virus Epidemiological Tool software. Patient timelines were obtained using electronic hospital records.
Findings
In total, 787 patients discharged from hospitals to care homes were identified. Of these, 776 (99%) were ruled out for subsequent introduction of SARS-CoV-2 into care homes. However, for 10 episodes, the results were inconclusive as there was low genomic diversity in consensus genomes or no sequencing data were available. Only one discharge episode had a genomic, time and location link to positive cases during hospital admission, leading to 10 positive cases in their care home.
Conclusion
The majority of patients discharged from hospitals were ruled out for introduction of SARS-CoV-2 into care homes, highlighting the importance of screening all new admissions when faced with a novel emerging virus and no available vaccine
