    Subject and Aesthetic Interface - an inquiry into transformed subjectivities

    The present PhD-thesis seeks new definitions of human subjectivity in an age of technoscience and a networked, globalized, Information Society. The perspective presented relates to Philosophy of Science, which includes the Human, the Natural, the Social and the Life Sciences. The project is directed at addressing, and aims to participate in, the further development of Philosophy of Science, or rather, the philosophy of knowing, which leaves a perspective broader than that of science. Methodologically, I combine readings of technoetic artworks, which I approach from a hermeneutical-semiotic perspective, with transdisciplinary research into existing theory concerning the human subject. These readings form my case studies. I keep a particular focus on holistic biophysics (Mae Wan Ho, James Oschman, Marko Bischof). Furthermore, Søren Brier's cybersemiotic theory of communication, cognition and consciousness, which combines a cybernetic-autopoietic and a Peircean semiotic perspective, plays a central role in the project. The project has three parts. Part one contextualizes the study within philosophy of science. It discusses relevant epistemologies, and places the case studies in an art categorical context. It further discusses the philosophical problems involved in writing an academic thesis in the form of a linear, argumentative, critical style, and how it affects the process of meaning making in a way that has consequences to my research. The second part consists of four case studies, each under an overall theme, which applies to the question of human subjectivity. Here I build the concept Extended Sentience, and the concept of an Ideal User. The Ideal User functions as a conceptual frame, which allows me to gradually add more elements to a theory of an altered human subject and knower. The third part presents new ontologies under three basic themes: Time and Relativity, The Life Cycles of Metaphors, and Logos Philosophy and Virtual Grids. These ontologies strongly affect ways of interpretation made in part one and two. Part Three allows more space to my subjective thought processes, which will take precedence over the literature applied. Thus, I, as a post-objective subject observer, will become more transparent. Finally, I will seek an overall conclusion to the project, which should clarify areas where it is evident that the human subject must be reconsidered at a pre-scientific level.     A Theoretical Model for Urban Walking Among People With Disabilities

    This paper is an attempt to advance research on walking at a neighborhood level ofanalysis for people with disabilities by proposing a theoretical model that combines theknowledge of two disciplines: traffic planning and environmental psychology. The aim isto provide guidance for a discussion and a plan for future interdisciplinary investigationsby proposing a model that accounts for the dynamic interaction between environmentalcharacteristics, human processes, and walking experience among individuals with adisability. For this purpose, traffic planners, and environmental psychologists cametogether to discuss theories, concepts, and thematic relevance in a series of focus groupmeetings. These meetings led to the selection of the Human Environment Interaction(HEI) model, originally developed from the field of environmental psychology andoperationalized to describe how walking experiences result from the interplay betweenindividual abilities, emotional processes, and the physical and social characteristics ofthe environment (K\ufcller, 1991). The proposed model aims to sustain interdisciplinarydiscussion and research planning around the topic of neighborhood walking for peoplewith disabilities. By operationalizing each dimension in the model, a good fit betweengroups with disabilities and individual differences associated with walking experiences isassumed, which, in turn, will have the potential to provide a more conscious analysisof wellbeing-related outcomes, such as usability of the environment, frequency ofmobility, and quality of life. However, to improve understanding of urban walking at aneighborhood level for people with disabilities, empirical studies must be carried out totest the proposed model

    SAGE2Splice: Unmapped SAGE Tags Reveal Novel Splice Junctions

    Serial analysis of gene expression (SAGE) not only is a method for profiling the global expression of genes, but also offers the opportunity for the discovery of novel transcripts. SAGE tags are mapped to known transcripts to determine the gene of origin. Tags that map neither to a known transcript nor to the genome were hypothesized to span a splice junction, for which the exon combination or exon(s) are unknown. To test this hypothesis, we have developed an algorithm, SAGE2Splice, to efficiently map SAGE tags to potential splice junctions in a genome. The algorithm consists of three search levels. A scoring scheme was designed based on position weight matrices to assess the quality of candidates. Using optimized parameters for SAGE2Splice analysis and two sets of SAGE data, candidate junctions were discovered for 5%–6% of unmapped tags. Candidates were classified into three categories, reflecting the previous annotations of the putative splice junctions. Analysis of predicted tags extracted from EST sequences demonstrated that candidate junctions having the splice junction located closer to the center of the tags are more reliable. Nine of these 12 candidates were validated by RT-PCR and sequencing, and among these, four revealed previously uncharacterized exons. Thus, SAGE2Splice provides a new functionality for the identification of novel transcripts and exons. SAGE2Splice is available online at http://www.cisreg.ca

    A Comparison of the Accessibility of Three Neighborhoods’ Institutions and Amenities in Frederick, MD

    Frederick is a city of neighborhoods. We studied three neighborhoods in the southern part of the City: Downtown—the area of downtown Frederick, South—the area to the immediate south and west of downtown, and West—the area west of the Frederick Bypass anchored by the Golden Mile and US-40. We based our analysis on the institutions and amenities present in each neighborhood. Institutions were landmarks or structures of civic nature such as schools, banks, churches, and parks, while amenities were necessities or conveniences such as groceries, retail, or service areas. We determined the boundaries of the neighborhoods by mapping the locations of institutions and amenities within the City and observing how they were geographically organized, then using differences in the age of the buildings in those areas as well as physical barriers between them to designate borders. We then studied selected social, demographic, and economic characteristics of the neighborhoods. These were age, income, race, and household size. We found that households tended to consist of fewer people closer to downtown Frederick, the youngest and most racially diverse areas were along the Golden Mile, the oldest and least diverse were in downtown Frederick and west of downtown, and income levels throughout the study area can be seen as having a bi-centric distribution. We also administered a survey in person at three locations within the City. Survey questions explored how often respondents visited amenities and institutions, and what form of transportation they used to get to those places. Respondents indicated that convenience stores were the type of service they used the most often, most trips they took to such destinations were less than five miles, and the mode of transportation they chose depended on their destination. Large numbers of respondents indicated support for bike lanes, traffic abatement, and crosswalks. Three recommendations result from this analysis: add bike lanes wherever possible; improve walkability; implement policies to encourage the opening of a grocery store within downtown Frederick. In the future, appropriate areas of research to extend this study may include bicyclist and pedestrian counts at key locations to guide bicycle and pedestrian policy within the City, a thorough assessment of the City’s public transportation system, and a review of neighborhood zoning to determine whether it reflects a desirable and functional land use pattern.The City of Frederic

    Evidence for polyploidy in the globally important diazotroph Trichodesmium

    Polyploidy is a well-described trait in some prokaryotic organisms; however, it is unusual in marine microbes from oligotrophic environments, which typically display a tendency towards genome streamlining. The biogeochemically significant diazotrophic cyanobacterium Trichodesmium is a potential exception. With a relatively large genome and a comparatively high proportion of non-protein-coding DNA, Trichodesmium appears to allocate relatively more resources to genetic material than closely related organisms and microbes within the same environment. Through simultaneous analysis of gene abundance and direct cell counts, we show for the first time that Trichodesmium spp. can also be highly polyploid, containing as many as 100 genome copies per cell in field-collected samples and >600 copies per cell in laboratory cultures. These findings have implications for the widespread use of the abundance of the nifH gene (encoding a subunit of the N2-fixing enzyme nitrogenase) as an approach for quantifying the abundance and distribution of marine diazotrophs. Moreover, polyploidy may combine with the unusual genomic characteristics of this genus both in reflecting evolutionary dynamics and influencing phenotypic plasticity and ecological resilience

    Crystal Structure of the HSV-1 Fc Receptor Bound to Fc Reveals a Mechanism for Antibody Bipolar Bridging

    Herpes simplex virus type-1 expresses a heterodimeric Fc receptor, gE-gI, on the surfaces of virions and infected cells that binds the Fc region of host immunoglobulin G and is implicated in the cell-to-cell spread of virus. gE-gI binds immunoglobulin G at the basic pH of the cell surface and releases it at the acidic pH of lysosomes, consistent with a role in facilitating the degradation of antiviral antibodies. Here we identify the C-terminal domain of the gE ectodomain (CgE) as the minimal Fc-binding domain and present a 1.78-Å CgE structure. A 5-Å gE-gI/Fc crystal structure, which was independently verified by a theoretical prediction method, reveals that CgE binds Fc at the C (H)2-C (H)3 interface, the binding site for several mammalian and bacterial Fc-binding proteins. The structure identifies interface histidines that may confer pH-dependent binding and regions of CgE implicated in cell-to-cell spread of virus. The ternary organization of the gE-gI/Fc complex is compatible with antibody bipolar bridging, which can interfere with the antiviral immune response

    Stakeholders' Participation in Planning and Priority Setting in the Context of a Decentralised Health Care system: the case of prevention of mother to child Transmission of HIV Programme in Tanzania.

    In Tanzania, decentralisation processes and reforms in the health sector aimed at improving planning and accountability in the sector. As a result, districts were given authority to undertake local planning and set priorities as well as allocate resources fairly to promote the health of a population with varied needs. Nevertheless, priority setting in the health care service has remained a challenge. The study assessed the priority setting processes in the planning of the prevention of mother to child transmission of HIV (PMTCT) programme at the district level in Tanzania. This qualitative study was conducted in Mbarali district, south-western Tanzania. The study applied in-depth interviews and focus group discussions in the data collection. Informants included members of the Council Health Management Team, regional PMTCT managers and health facility providers. Two plans were reported where PMTCT activities could be accommodated; the Comprehensive Council Health Plan and the Regional PMTCT Plan that was donor funded. As donors had their own globally defined priorities, it proved difficult for district and regional managers to accommodate locally defined PMTCT priorities in these plans. As a result few of these were funded. Guidelines and main priority areas of the Ministry of Health and Social Welfare (MoHSW) also impacted on the ability of the districts and regions to act, undermining the effectiveness of the decentralisation policy in the health sector. The challenges in the priority setting processes revealed within the PMTCT initiative indicate substantial weaknesses in implementing the Tanzania decentralisation policy. There is an urgent need to revive the strategies and aims of the decentralisation policy at all levels of the health care system with a view to improving health service delivery

    Enclosing a pen reduced time to response to questionnaire mailings

    OBJECTIVES: To assess the effectiveness of including a pen in postal questionnaires on response rate, necessity of reminders, time to response, and completeness of response to the primary outcome question (POQ).   STUDY DESIGN AND SETTING: A two-arm randomized controlled trial (RCT) embedded within the screening of older women for prevention of fracture trial (SCOOP). Women, aged 70-75 years, were randomized to receive a pen with their questionnaire (n = 3,826) or to receive the questionnaire alone (n = 3,829). The results were combined with another embedded RCT in a meta-analysis.   RESULTS: A response rate of 92.4% was observed in the pen group compared with 91.3% in the control group (odds ratio [OR] = 1.16; 95% confidence interval [CI]: 0.98, 1.37; P = 0.08). There was a difference in reminders required (OR = 0.88; 95% CI: 0.79, 0.98; P = 0.02), time to response (hazard ratio = 1.06; 95% CI: 1.01, 1.11; P = 0.01) and some difference in the completeness of response to the POQ (OR = 1.18; 95% CI: 1.00, 1.39; P = 0.05). The pooled OR from the meta-analysis for response rate was 1.21 (95% CI: 1.05, 1.39; P = 0.01).   CONCLUSION: Inclusion of a pen with postal questionnaires potentially has a positive impact on response rates and the number of reminders required. There may be some reduction in time to response. Studies of different participant groups are needed to test the effectiveness over more diverse populations

    Development of Type 1 Diabetes in Wild Bank Voles Associated With Islet Autoantibodies and the Novel Ljungan Virus

    Wild bank voles (Clethrionomys glareolus) may develop diabetes in laboratory captivity. The aim of this study was to test whether bank voles develop type 1 diabetes in association with Ljungan virus. Two groups of bank voles were analyzed for diabetes, pancreas histology, autoantibodies to glutamic acid decarboxylase (GAD65), IA-2, and insulin by standardized radioligand-binding assays as well as antibodies to in vitro transcribed and translated Ljungan virus antigens. Group A represented 101 trapped bank voles, which were screened for diabetes when euthanized within 24 hours of capture. Group B represented 67 bank voles, which were trapped and kept in the laboratory for 1 month before being euthanized. Group A bank voles did not have diabetes. Bank voles in group B (22/67; 33%) developed diabetes due to specific lysis of pancreatic islet beta cells. Compared to nondiabetic group B bank voles, diabetic animals had increased levels of GAD65 (P < .0001), IA-2 (P < .0001), and insulin (P = .03) autoantibodies. Affected islets stained positive for Ljungan virus, a novel picorna virus isolated from bank voles. Ljungan virus inoculation of nondiabetic wild bank voles induced beta-cell lysis. Compared to group A bank voles, Ljungan virus antibodies were increased in both nondiabetic (P < .0001) and diabetic (P = .0015) group B bank voles. Levels of Ljungan virus antibodies were also increased in young age at onset of newly diagnosed type 1 diabetes in children (P < .01). These findings support the hypothesis that the development of type 1 diabetes in captured wild bank voles is associated with Ljungan virus. It is speculated that bank voles may have a possible zoonotic role as a reservoir and vector for virus that may contribute to the incidence of type 1 diabetes in humans
