    Splenic CD8(+) T cells secrete TGF-beta 1 to exert suppression in mice with anterior chamber-associated immune deviation

    Background CD8(+) regulatory T cells (Treg) have been considered to be involved in a model of ocular-induced tolerance, known as anterior chamber-associated immune deviation (ACAID). The mechanisms of suppression by CD8(+) T cells in ACAID remain only poorly understood. TGF-beta 1 is considered as an inhibitory cytokine for immunosuppression in some models. The production of TGF-beta 1 by CD8(+) T cells in ACAID, and whether CD8+ T cells exert suppression through TGF-beta 1, is unknown. Methods The suppressive effect of CD8(+) T cells in ACAID mice was determined by a local adoptive transfer (LAT) assay. The production of TGF-beta 1 by CD8(+) T cells was measured by enzyme-linked immunosorbent assay (ELISA). Anti-TGF-beta 1 antibodies were used in the LAT assay to test if they could block the inhibitory effect of CD8(+) T cells. Results CD8(+) T cells from ACAID mice were shown to block the delayed-type hypersensitivity (DTH) response in an antigen-specific manner in a LAT assay. These CD8+ T cells secreted TGF-beta 1, and their suppression could partially be blocked by anti-TGF-beta 1 antibodies. Conclusions Our study confirms that CD8+ T cells from ACAID mice possess inhibitory properties. This population exerts part of its suppressive function via the production of TGF-beta 1

    Numerical Simulation of the Thermomechanical Behavior of a Hot Stamping Die

    For studying the temperature field of hot stamping, the effect of the distance between cooling pipes, the distance between the cooling pipe and mold surface, and the water cooling pipe diameter were evaluated with ANSYS software. The results showed that the maximum temperature was about 50°C, and the maximum temperature difference of hot stamping was less than 35°C. The maximum temperature and the uniformity of stamping temperature can be improved by controlling the size and spacing of cooling pipes.Определен эффект расстояния между трубами охлаждения, расстояния между трубой охлаждения и поверхностью формы и диаметра трубы водяного охлаждения на температурное поле горячей штамповки с использованием программного обеспечения ANSYS.     Polysaccharides and polyphenols in sea buckthorn leaf tea have synergistic impact on studied colonic strains in vitro and bacteria in vivo

    Present research on prebiotics focuses on either polysaccharides or polyphenols. This study compared the individual and combined impact of polysaccharide, quercetin, and gallic acid (GA) treatment on three human faecal strains. In vitro pure culturing and correlation analysis confirmed that the growth of both beneficial microbe B. longum subsp. longum (0.695, 0.205: R2, slope, respectively) and pathogenic C. perfringens (0.712, 0.085: R2, slope, respectively) increased due to polysaccharide treatment, and only GA treatment would inhibit C. perfringens (0.789, –0.165: R2, slope, respectively) growth. In vivo studies also revealed that genome copies of Bifidobacterium increased and C. perfringens decreased in the faeces, when a blend of the three nutrients rather than single polysaccharide or polyphenols were fed to rats. These data suggested that combined prebiotic treatment improved human faecal strain composition better than single treatment

    Numerical investigation of conjugated heat transfer in a channel with a moving depositing front

    This article presents numerical simulations of conjugated heat transfer in a fouled channel with a moving depositing front. The depositing front separating the fluid and the deposit layer is captured using the level-set method. Fluid flow is modeled by the incompressible Navier–Stokes equations. Numerical solution is performed on a fixed mesh using the finite volume method. The effects of Reynolds number and thermal conductivity ratio between the deposit layer and the fluid on local Nusselt number as well as length-averaged Nusselt number are investigated. It is found that heat transfer performance, represented by the local and length-averaged Nusselt number reduces significantly in a fouled channel compared with that in a clean channel. Heat transfer performance decreases with the growth of the deposit layer. Increases in Reynolds, Prandtl numbers both enhance heat transfer. Besides, heat transfer is enhanced when the thermal conductivity ratio between the deposit layer and the fluid is lower than 20 but it decreases when the thermal conductivity ratio is larger than 2

    Granular dynamics of cohesive powders in a rotating drum as revealed by speckle visibility spectroscopy and synchronous measurement of forces due to avalanching

    We have used speckle visibility spectroscopy (SVS) and synchronized force measurements to compare the granular dynamics of two cohesive lactose powders, with Sauter mean diameters of ~29 and ~151 μm, in a rotating drum. A load cell (LC) was used to measure forces on the drum mounting frame and enable monitoring of bulk powder motion; SVS is a dynamic light scattering technique particularly suited for studying dynamics in dense, non-ergodic granular systems. Our results reveal that surface slumping and intermittent collisional dynamics in the bulk of the bed are correlated, especially for the fine more cohesive particles (Geldart group C/A boundary), but not as much for the less cohesive larger particles (Geldart group A/B boundary). The specific dissipation energy of the particles in the drum is similar for both powders, and increases linearly with increasing drum speed. However, the dependencies of the load cell and SVS signals on rotation speed have opposing trends for these two powders, indicating different dissipation mechanisms for the different Geldart Groups; collisional dissipation is more important for the Geldart C/A powder, while for the Geldart A/B powder avalanche dissipation is dominant

    A comparison between yellow-green and green cultivars of four vegetable species in pigments, ascorbate, photosynthesis, energy dissipation, and photoinhibition

    Yellow-green foliage cultivars of four vegetables grown outdoors, i.e., Chinese mustard (Brassica rapa), Chinese kale (Brassica oleracea var. alboglabra), sweet potato (Ipomoea batatas) and Chinese amaranth (Amaranthus tricolor), had lower chlorophyll (Chl) (a+b) (29-36% of green cultivars of the same species), total carotenoids (46-62%) and ascorbate (72-90%) contents per leaf area. Furthermore, yellow-green cultivars had smaller photosystem II (PSII) antenna size (65-70%) and lower photosynthetic capacity (52-63%), but higher Chl a/b (107-156%) and from low (60%) to high (129%) ratios of de-epoxidized xanthophyll cycle pigments per Chl a content. Potential quantum efficiency of PSII (F(v)/F(m)) of all overnight dark-adapted leaves was ca. 0.8, with no significant difference between yellow-green and green cultivars of the same species. However, yellow-green cultivars displayed a higher degree of photoinhibition (lower F(v)/F(m) after illumination) when they were exposed to high irradiance. Although vegetables used in this study are of either temperate or tropical origin and include both C(3) and C(4) plants, data from all cultivars combined revealed that F(v)/F(m) after illumination still showed a significant positive linear regression with xanthophyll cycle-dependent energy quenching (q(E)) and a negative linear regression with photoinhibitory quenching (q(I)). F(v)/F(m) was, however, not correlated with nonphotochemical quenching (NPQ). Yet, a higher degree of photoinhibition in yellow-green cultivars could recover during the night darkness period, suggesting that the repair of PSII in yellow-green cultivars would allow them to grow normally in the field

    Effect of dried tangerine peel extract supplementation on the growth performance and antioxidant status of broiler chicks

    The aim of this study was to evaluate the effects of dried tangerine peel extract (DTPE) supplementation on the growth performance and immune and antioxidant status of broiler chicks. A total of 525day-old, male, Arbour Acres (AA) broiler chicks were fed a basal diet (CTR) either with or without 80, 160, 240 and 480mg DTPE/kg diet. The chicks were randomly assigned into five dietary groups for a 42-day experiment. There were seven replicates per group and 15 chicks per replicate. At 42 days of age, two birds from each cage were selected for blood sampling. The dietary DTPE supplementation linearly increased the body weight and the average daily gain (p=.03; p=.03) and quadratically increased the average daily feed intake (p=.02) during the starter period. In addition, the plasma lysozyme level in the 80 or 160mg/kg DTPE group increased compared to the CTR group (p=.02 and .07, respectively). Chicks fed 80-mg/kg DTPE reduced the malondialdehyde concentration (p=.02) compared with birds fed the CTR diets. Moreover, the plasma glutathione peroxidase activities in the low DTPE dietary groups (80 and 160mg/kg) were higher than in the CTR (p<.01) and 240mg/kg DTPE group (p<.01 and p=.02, respectively). The results of the present study indicate that dietary DTPE with low supplemental dosage (<= 160mg/kg) might positively modulate the systemic antioxidative defence property of chicks

    Phenotypic and Molecular Analysis of the Effect of 20-hydroxyecdysone on the Human Filarial Parasite Brugia malayi

    A homologue of the ecdysone receptor has been identified and shown to be responsive to 20- hydroxyecdysone in Brugia malayi. However, the role of this master regulator of insect development has not been delineated in filarial nematodes. Gravid adult female B. malayi cultured in the presence of 20-hydroxyecdysone produced significantly more microfilariae and abortive immature progeny than control worms, implicating the ecdysone receptor in regulation of embryogenesis and microfilarial development. Transcriptome analyses identified 30 genes whose expression was significantly up-regulated in 20-hydroxyecdysone-treated parasites compared with untreated controls. Of these, 18% were identified to be regulating transcription. A comparative proteomic analysis revealed 932 proteins to be present in greater amounts in extracts of 20- hydroxyecdysone-treated adult females than in extracts prepared from worms cultured in the absence of the hormone. Of the proteins exhibiting a greater than two-fold difference in the 20- hydroxyecdysone-treated versus untreated parasite extracts, 16% were involved in transcriptional regulation. RNA interference (RNAi) phenotype analysis of Caenorhabditis elegans orthologs revealed that phenotypes involved in developmental processes associated with embryogenesis were significantly over-represented in the transcripts and proteins that were up-regulated by exposure to 20-hydroxyecdysone. Taken together, the transcriptomic, proteomic and phenotypic data suggest that the filarial ecdysone receptor may play a role analogous to that in insects, where it serves as a regulator of egg development
