Dependence of ocean wave return levels on water depth and sampling length: a focus on the South Yellow Sea
A deterministic Extreme Value Analysis method is of particular importance in an engineering-oriented context. In this paper, three extreme value analysis (EVA) methods, in which annual maxima, monthly maxima or the Peaks-Over-Threshold are fitted into Generalized Extreme Value distribution (GEV) function or Generalized Pareto distribution (GP) function, are used to estimate return values of significant wave height. Sensitivity of return levels on water depth and sampling length is vigorously investigated, based on a 40-year long and high-resolution wave hindcast for the South Yellow Sea (SYS). A spatially-and-temporally varied POT sampling method combined with GP function produces most conservative and confident estimates of return levels for a large return period, but produces wider confidence level than the other two EVA approaches based on GEV function for short return period. In the SYS, the return level estimates are significantly reduced with a longer sample length. However, we find that the reduction is closely related to the long-term trend in extreme wave heights, rather than due to the sampling effect. From deep to shallow waters, spatial inhomogeneity of return levels increases, which should be considered in the engineering practice
Optimization of Flash Extraction Process and Antioxidant Activity of American Ginseng Flower Polysaccharides
Objective: To study the optimal process conditions of flash extraction and antioxidant activity of American ginseng flower polysaccharides (AGFPs). Methods: American ginseng flower (AGF) as the raw material, the effects of extraction voltage, liquid-material ratio, and extraction time on the yield of AGFPs were explored. The flash extraction process of AGFPs was improved by response surface methodology. The scavenging effect on DPPH and hydroxy free radicals and total reduction capacity of AGFPs were determined to assess the antioxidant activity of AGFPs. Results: The optimal extraction conditions were determined as 130 V of extraction voltage, 30:1 mL/g of liquid-material ratio, and 100 s of extraction time, resulting in an AGFPs yield of 11.12%±0.23%, which agreed with the model prediction. The AGFPs exhibited significant scavenging effects on DPPH and hydroxyl radicals, with IC50 values of 1.34 mg/mL and 1.42 mg/mL, respectively, and had a certain reducing power. Conclusion: These results suggested that flash extraction was an efficient and rapid method for obtaining AGFPs from AGF, and that AGFPs had promising antioxidant potential for further applications. This study can provide a theoretical basis for the development and application of AGF
ÎČ-Sitosterol Ameliorates Endometrium Receptivity in PCOS-Like Mice: The Mediation of Gut Microbiota
Background: Polycystic ovary syndrome (PCOS), one of the most common endocrine diseases in women of childbearing age, has been found to be accompanied by changes in the gut microbiota. The Bu Shen Yang Xue formula (BSYXF) is a traditional Chinese medicine widely used for the treatment of PCOS. This study aimed to investigate whether the protective effects of ÎČ-sitosterol, the main active ingredient of BSYXF, on PCOS was mediated by regulating gut microbiota.Methods: The presence of ÎČ-sitosterol in BSYXF was detected by liquid chromatography-mass spectrometry. The PCOS-like mouse model was induced by dehydroepiandrosterone. The fecal supernatant of ÎČ-sitosterol-treated mice was prepared for fecal microbiota transplantation (FMT). Body weight and wet weight of the uterus and ovary of the mice were recorded for organ index calculation. Hematoxylin and eosin stain was used to assess the endometrial morphology and microenvironment changes. Expression of endometrial receptivity markers cyclooxygenase-2 (COX-2), Integrin αΜÎČ3, leukemia inhibitory factor (LIF), and homeobox A10 (HOXA10) in the endometrium were determined by immunohistochemistry and western blot analysis. Enzyme-linked immunosorbent assay was employed to detect the expression of follicle stimulating hormone (FSH), luteinizing hormone (LH), progesterone (P), and testosterone (T) in the serum. The diversity of gut microbiota was examined by 16S rDNA gene sequencing.Results: With the treatment of ÎČ-sitosterol and ÎČ-sitosterol-FMT, the uterine index of PCOS-like mice increased, the ovarian index decreased, levels of COX-2, LH and T decreased, and levels of Integrin αΜÎČ3, LIF, HOXA10, FSH, and P increased. Under ÎČ-sitosterol treatment, the structure of the gut microbiota in PCOS-like mice was also changed.Conclusion: ÎČ-sitosterol regulates the endometrial receptivity of PCOS and harmonizes the sex hormone balance, which may be related to the changes in the structure and composition of gut microbiota, thus affecting the pathological process of PCOS
The role of school organizational conditions in teacher psychological resilience and stress during COVID-19 pandemic: A moderated mediation model
Educational revisions facilitate the relief of teacher stress by means of enhancing school organizational conditions. However, limited research has explored the effects of school organizational conditions on teacher stress in China. Using a sample of 734 primary and secondary school teachers from 30 provinces or municipalities of China, this study examined the effects of school organizational conditions on teacher stress in China, with a particular focus on the mediating role of psychological resilience and moderating role of perceived COVID-19 crisis strength. The results demonstrated that school organizational conditions were negatively associated with teacher stress. Furthermore, psychological resilience partially mediated the relation between school organizational conditions and teacher stress. In addition, perceived COVID-19 crisis strength significantly moderated the direct and indirect relations between school organizational conditions and teacher stress. The relations between school organizational conditions and teacher stress and between school organizational conditions and psychological resilience were stronger for teachers who perceived low levels of COVID-19 crisis strength. However, the indirect relation between psychological resilience and stress was stronger for teachers who perceived high levels of COVID-19 crisis strength. Implications have been provided accordingly
Aging- And Alcohol-Associated Spatial Transcriptomic Signature in Mouse Acute Pancreatitis Reveals Heterogeneity of Inflammation and Potential Pathogenic Factors
The rapidly aging population is consuming more alcohol, leading to increased alcohol-associated acute pancreatitis (AAP) with high mortality. However, the mechanisms remain undefined, and currently there are no effective therapies available. This study aims to elucidate aging- and alcohol-associated spatial transcriptomic signature by establishing an aging AAP mouse model and applying Visium spatial transcriptomics for understanding of the mechanisms in the context of the pancreatic tissue. Upon alcohol diet feeding and caerulein treatment, aging mice (18 months) developed significantly more severe AAP with 5.0-fold increase of injury score and 2.4-fold increase of amylase compared to young mice (3 months). Via Visium spatial transcriptomics, eight distinct tissue clusters were revealed from aggregated transcriptomes of aging and young AAP mice: five acinar, two stromal, and one islet, which were then merged into three clusters: acinar, stromal, and islet for the comparative analysis. Compared to young AAP mice,â\u3eâ1300 differentially expressed genes (DEGs) and approximately 3000 differentially regulated pathways were identified in aging AAP mice. The top five DEGs upregulated in aging AAP mice include Mmp8, Ppbp, Serpina3m, Cxcl13, and Hamp with heterogeneous distributions among the clusters. Taken together, this study demonstrates spatial heterogeneity of inflammatory processes in aging AAP mice, offering novel insights into the mechanisms and potential drivers for AAP development
Search for new particles in events with energetic jets and large missing transverse momentum in proton-proton collisions at root s=13 TeV
A search is presented for new particles produced at the LHC in proton-proton collisions at root s = 13 TeV, using events with energetic jets and large missing transverse momentum. The analysis is based on a data sample corresponding to an integrated luminosity of 101 fb(-1), collected in 2017-2018 with the CMS detector. Machine learning techniques are used to define separate categories for events with narrow jets from initial-state radiation and events with large-radius jets consistent with a hadronic decay of a W or Z boson. A statistical combination is made with an earlier search based on a data sample of 36 fb(-1), collected in 2016. No significant excess of events is observed with respect to the standard model background expectation determined from control samples in data. The results are interpreted in terms of limits on the branching fraction of an invisible decay of the Higgs boson, as well as constraints on simplified models of dark matter, on first-generation scalar leptoquarks decaying to quarks and neutrinos, and on models with large extra dimensions. Several of the new limits, specifically for spin-1 dark matter mediators, pseudoscalar mediators, colored mediators, and leptoquarks, are the most restrictive to date.Peer reviewe
MUSiC : a model-unspecific search for new physics in proton-proton collisions at root s=13TeV
Results of the Model Unspecific Search in CMS (MUSiC), using proton-proton collision data recorded at the LHC at a centre-of-mass energy of 13 TeV, corresponding to an integrated luminosity of 35.9 fb(-1), are presented. The MUSiC analysis searches for anomalies that could be signatures of physics beyond the standard model. The analysis is based on the comparison of observed data with the standard model prediction, as determined from simulation, in several hundred final states and multiple kinematic distributions. Events containing at least one electron or muon are classified based on their final state topology, and an automated search algorithm surveys the observed data for deviations from the prediction. The sensitivity of the search is validated using multiple methods. No significant deviations from the predictions have been observed. For a wide range of final state topologies, agreement is found between the data and the standard model simulation. This analysis complements dedicated search analyses by significantly expanding the range of final states covered using a model independent approach with the largest data set to date to probe phase space regions beyond the reach of previous general searches.Peer reviewe
