    Chlorpyrifos Affects Phenotypic Outcomes in a Model of Mammalian Neurodevelopment: Critical Stages Targeting Differentiation in PC12 Cells

    The organophosphate insecticide chlorpyrifos (CPF) adversely affects mammalian brain development through multiple mechanisms. To determine if CPF directly affects neuronal cell replication and phenotypic fate, and to identify the vulnerable stages of differentiation, we exposed PC12 cells, a model for mammalian neurodevelopment, to CPF concentrations spanning the threshold for cholinesterase inhibition (5–50 ÎŒM) and conducted evaluations during mitosis and in early and mid-differentiation. In undifferentiated cells, exposure to 5 ÎŒM CPF for 1–3 days reduced DNA synthesis significantly without eliciting cytotoxicity. At the same time, CPF increased the expression of tyrosine hydroxylase (TH), the enzymatic marker for the catecholamine phenotype, without affecting choline acetyltransferase (ChAT), the corresponding marker for the cholinergic phenotype. Upon exposure to nerve growth factor (NGF), PC12 cells developed neuritic projections in association with vastly increased TH and ChAT expression accompanying differentiation into the two phenotypes. CPF exposure begun at the start of differentiation significantly reduced ChAT but not TH activity. In contrast, when CPF was added in mid-differentiation (4 days of NGF pretreatment), ChAT was unaffected and TH was increased slightly. Thus, CPF exerts stage-specific effects, reducing DNA synthesis in the undifferentiated state, impairing development of the cholinergic phenotype at the start of differentiation, and promoting expression of the catecholaminergic phenotype both in undifferentiated and differentiated cells. CPF administration in vivo produces deficits in the number of neurons and cholinergic function, and because we were able to reproduce these effects in vitro, our results suggest that CPF directly influences the phenotypic fate of neuronal precursors

    Dust and Gas in the Magellanic Clouds from the HERITAGE Herschel Key Project. II. Gas-to-Dust Ratio Variations across ISM Phases

    The spatial variations of the gas-to-dust ratio (GDR) provide constraints on the chemical evolution and lifecycle of dust in galaxies. We examine the relation between dust and gas at 10-50 pc resolution in the Large and Small Magellanic Clouds (LMC and SMC) based on Herschel far-infrared (FIR), H I 21 cm, CO, and Halpha observations. In the diffuse atomic ISM, we derive the gas-to-dust ratio as the slope of the dust-gas relation and find gas-to-dust ratios of 380+250-130 in the LMC, and 1200+1600-420 in the SMC, not including helium. The atomic-to-molecular transition is located at dust surface densities of 0.05 Mo pc-2 in the LMC and 0.03 Mo pc-2 in the SMC, corresponding to AV ~ 0.4 and 0.2, respectively. We investigate the range of CO-to-H2 conversion factor to best account for all the molecular gas in the beam of the observations, and find upper limits on XCO to be 6x1020 cm-2 K-1 km-1 s in the LMC (Z=0.5Zo) at 15 pc resolution, and 4x 1021 cm-2 K-1 km-1 s in the SMC (Z=0.2Zo) at 45 pc resolution. In the LMC, the slope of the dust-gas relation in the dense ISM is lower than in the diffuse ISM by a factor ~2, even after accounting for the effects of CO-dark H2 in the translucent envelopes of molecular clouds. Coagulation of dust grains and the subsequent dust emissivity increase in molecular clouds, and/or accretion of gas-phase metals onto dust grains, and the subsequent dust abundance (dust-to-gas ratio) increase in molecular clouds could explain the observations. In the SMC, variations in the dust-gas slope caused by coagulation or accretion are degenerate with the effects of CO-dark H2. Within the expected 5--20 times Galactic XCO range, the dust-gas slope can be either constant or decrease by a factor of several across ISM phases. Further modeling and observations are required to break the degeneracy between dust grain coagulation, accretion, and CO-dark H2

    Pleasure and meaningful discourse: an overview of research issues

    The concept of pleasure has emerged as a multi-faceted social and cultural phenomenon in studies of media audiences since the 1980s. In these studies different forms of pleasure have been identified as explaining audience activity and commitment. In the diverse studies pleasure has emerged as a multi-faceted social and cultural concept that needs to be contextualized carefully. Genre and genre variations, class, gender, (sub-)cultural identity and generation all seem to be instrumental in determining the kind and variety of pleasures experienced in the act of viewing. This body of research has undoubtedly contributed to a better understanding of the complexity of audience activities, but it is exactly the diversity of the concept that is puzzling and poses a challenge to its further use. If pleasure is maintained as a key concept in audience analysis that holds much explanatory power, it needs a stronger theoretical foundation. The article maps the ways in which the concept of pleasure has been used by cultural theorists, who have paved the way for its application in reception analysis, and it goes on to explore the ways in which the concept has been used in empirical studies. Central to our discussion is the division between the ‘public knowledge’ and the ‘popular culture’ projects in reception analysis which, we argue, have major implications for the way in which pleasure has come to be understood as divorced from politics, power and ideology. Finally, we suggest ways of bridging the gap between these two projects in an effort to link pleasure to the concepts of hegemony and ideology

    Spectral cinema from a phantom state: film aesthetics and the politics of identity in Divided Heaven and Solo Sunny

    In this essay I draw on close textual analysis to consider the interface between film aesthetics and the politics of identity in Konrad Wolf’s Der geteilte Himmel / Divided Heaven (1964) and Solo Sunny (1979). Both films focus on women who have to confront painful processes of self realisa-tion in specifically East German contexts. They also show Wolf and his collaborators working in two very different modes, from a nouvelle vague-inspired mix of location shooting and self-conscious formal artifice to a more laconic style and mobile camera that borrow from documentary aesthetics. Viewed from the perspective of today, the films resist the reductive stereotyping of what Christa Wolf in 1991 called the 'phantom' East Germany, and offer a more productive haunting. As living ghosts in the post-reunification era, they are a reminder of the necessity of remembering, and so confound both a negative 'master narrative' of the GDR and a collective amnesia with no interest in this history

    Production, consumers' convenience, and cynical economies: The case of Uber in Buenos Aires

    Based on twelve months of fieldwork into Uber's conflict in Buenos Aires, Argentina, this article examines convenience's role in the emergence of what I call cynical economies: a method and logic of production expressly organized on the awareness of a distance the very rhetoric of convenience exacerbates. For the city's middle class, convenience defined a democratizing, empowering arena of private relations away from the hierarchies and exclusions proper to the private sphere. As Uber's ratings translated consumers' experiences into a political economy for the trade, drivers organized the production of the ride knowing that whatever exceeded the immediate intelligibility of the experience could not matter in that political economy. In the process, cynical economies delegitimize complex and inherently social categories like risk, responsibility, and liability, as well as the social sphere that frames them, without offering an alternative order in return

    This paper looks at Alain Resnais’ Last Year in Marienbad (1961) and Chris Marker’s La JetĂ©e (1962). It rests on a premise of film as a constructed, ordered world that answers only to itself. Both films address particular questions about time: what happens to our anticipation of the future if we move back and forth in time reinventing our past and present? (Marienbad) or, can we escape our ruined present by moving into the future? (La JetĂ©e). From Jacques Lacan, it borrows the concepts of the mirror stage by which we recognise ourselves, and of the objet petit a, the looking for which (both in terms of ‘search’ and ‘seeing’) is that from which we derive our pleasure. From Jean-Luc Nancy it adopts descriptions of how film touches us, and the careful orchestration of the pleasure that is jouissance in being within this moment, not knowing where we are going

    Dust and Gas in the Magellanic Clouds from the Heritage Herschel Key Project. I. Dust Properties and Insights into the Origin of the Submm (Submillimeter) Excess Emission

    The dust properties in the Large and Small Magellanic Clouds are studied using the HERITAGE Herschel Key Project photometric data in five bands from 100 to 500 micromillimeters. Three simple models of dust emission were fit to the observations: a single temperature blackbody modified by a powerlaw emissivity (SMBB), a single temperature blackbody modified by a broken power-law emissivity (BEMBB), and two blackbodies with different temperatures, both modified by the same power-law emissivity (TTMBB). Using these models we investigate the origin of the submillimeter excess; defined as the submillimeter (submm) emission above that expected from SMBB models fit to observations < 200 micromillimeters. We find that the BEMBB model produces the lowest fit residuals with pixel-averaged 500 micromillimeters submillimeter excesses of 27% and 43% for the Large and Small Magellanic Clouds, respectively. Adopting gas masses from previous works, the gas-to-dust ratios calculated from our fitting results show that the TTMBB fits require significantly more dust than are available even if all the metals present in the interstellar medium (ISM) were condensed into dust. This indicates that the submillimeter excess is more likely to be due to emissivity variations than a second population of colder dust. We derive integrated dust masses of (7.3 plus or minus 1.7) x 10 (sup 5) and (8.3 plus or minus 2.1) x 10 (sup 4) solar masses for the Large and Small Magellanic Clouds, respectively. We find significant correlations between the submillimeter excess and other dust properties; further work is needed to determine the relative contributions of fitting noise and ISM physics to the correlations
