BIOCHEMICAL DIFFERENTIATION AMONG S. aureus, S. Intermedius AND S. hyicus ISOLATED FROM BOVINES WITH SUBCLINICAL MASTITIS
Staphylococcus aureus é, entre as espécies de estafilococos, a mais relacionada a
infecções em animais e humanos, bem como a doenças de origem alimentar. Entretanto, S.
intermedius e S. hyicus, duas espécies com características morfológicas muito similares a S. aureus,
também podem causar doenças, bem como produzir enterotoxinas em alimentos, tornando importante
a diferenciação entre essas três espécies de estafilococos. O objetivo deste trabalho foi avaliar a
eficiência de oito técnicas bioquímicas com relação à sua capacidade para distinguir entre S. aureus,
S. intermedius e S. hyicus. Para isso, 65 cepas, previamente identificadas em nível de espécie
através de técnicas moleculares, foram submetidas aos testes de produção de pigmentos
carotenóides, atividade hemolítica em ágar sangue, produção de b- galactosidase, produção de
acetoína, atividade lipolítica em polisorbato, fermentação aeróbica da maltose, fermentação
anaeróbica do manitol e crescimento em ágar Baird-Paker e ágar P suplementados com acriflavina.
Verificou-se que os testes de sensibilidade a acriflavina e de produção de b- galactosidase
apresentaram bom poder discriminatório, demonstrando serem os melhores testes bioquímicos
para a diferenciação entre essas três espécies de estafilococos.
Abstract
Among staphylococcus species, Staphylococcus aureus is the one mostly related to
animal and human infections, as well as food diseases. However, S. intermedius and S. hyicus, two
species with very similar morphological characteristics to S. aureus, can also cause diseases as well
as produce enterotoxins in food, what makes important the differentiation of these three species.
The aim of this work was to study the efficiency of eight biochemical techniques regarding their
capacity to distinguish among S. aureus, S. intermedius and S. hyicus. For that purpose, sixty five
strains, previously identified at species level through molecular techniques, were submitted to the
tests of carotenoid pigments production, hemolytic activity in blood agar, b- galactosidase production,
acetoin production, lipolytic activity in polysorbate, maltose aerobic fermentation, manitol anaerobic
fermentation and growth in Baird-Parker and P agar supplemented with acriflavine. It was verified
that tests of sensitivity to acriflavine and b-galactosidase production display good distinguishing
properties, constituting as a whole, the best biochemical tests for the identification of these three
staphylococcus species
Live-cell FRET imaging reveals clustering of the prion protein at the cell surface induced by infectious prions
Prion diseases are associated to the conversion of the prion protein into a misfolded pathological isoform. The mechanism of propagation of protein misfolding by protein templating remains largely unknown. Neuroblastoma cells were transfected with constructs of the prion protein fused to both CFP-GPI-anchored and to YFP-GPI-anchored and directed to its cell membrane location. Live-cell FRET imaging between the prion protein fused to CFP or YFP was measured giving consistent values of 10 +/- 2%. This result was confirmed by fluorescence lifetime imaging microscopy and indicates intermolecular interactions between neighbor prion proteins. In particular, considering that a maximum FRET efficiency of 17 +/- 2% was determined from a positive control consisting of a fusion CFP-YFP-GPI-anchored. A stable cell clone expressing the two fusions containing the prion protein was also selected to minimize cell-to-cell variability. In both, stable and transiently transfected cells, the FRET efficiency consistently increased in the presence of infectious prions - from 4 +/- 1% to 7 +/- 1% in the stable clone and from 10 +/- 2% to 16 +/- 1% in transiently transfected cells. These results clearly reflect an increased clustering of the prion protein on the membrane in the presence of infectious prions, which was not observed in negative control using constructs without the prion protein and upon addition of non-infected brain. Our data corroborates the recent view that the primary site for prion conversion is the cell membrane. Since our fluorescent cell clone is not susceptible to propagate infectivity, we hypothesize that the initial event of prion infectivity might be the clustering of the GPI-anchored prion protein. The effects of a plant proteinase inhibitor from Enterolobium contortisiliquum on human tumor cell lines
Supplementary to the efficient inhibition of trypsin, chymotrypsin, plasma kallikrein, and plasmin already described by the EcTI inhibitor from Enterolobium contortisiliquum, it also blocks human neutrophil elastase (K(iapp)=4.3 nM) and prevents phorbol ester (PMA)-stimulated activation of matrix metalloproteinase (MMP)-2 probably via interference with membrane-type 1 (MT1)-MMP. Moreover, plasminogen-induced activation of proMMP-9 and processing of active MMP-2 was also inhibited. Furthermore, the effect of EcTI on the human cancer cell lines HCT116 and HT29 (colorectal), SkBr-3 and MCF-7 (breast), K562 and THP-1 (leukemia), as well as on human primary fibroblasts and human mesenchymal stem cells (hMSCs) was studied. EcTI inhibited in a concentration range of 1.0-2.5 mu M rather specifically tumor cell viability without targeting primary fibroblasts and hMSCs. Taken together, our data indicate that the polyspecific proteinase inhibitor EcTI prevents proMMP activation and is cytotoxic against tumor cells without affecting normal tissue remodeling fibroblasts or regenerative hMSCs being an important tool in the studies of tumor cell development and dissemination
Fontes orgânicas de nutrientes em sistemas de produção de batata.
Nos últimos anos, diminuiu muito o número de bataticultores na região da Quarta Colônia, no centro do Estado do Rio Grande do Sul (RS), devido ao elevado risco econômico na produção da batata. A partir de uma demanda da Associação dos Produtores de Batata da Região Central do Rio Grande do Sul (ASBAT), foram instalados experimentos visando a avaliar, técnica e economicamente, fontes orgânicas alternativas à adubação mineral tradicional dos produtores de batata, objetivando diminuir os custos de produção e o impacto do seu cultivo no ambiente. Foram estabelecidas rotações envolvendo culturas comerciais e plantas de cobertura de solo para o cultivo da batata na safra e na safrinha. As fontes de nutrientes foram camas de frango e de suínos e adubo mineral tradicionalmente utilizado pelos bataticultores. Os resultados mostraram que a utilização de fontes orgânicas de nutrientes no cultivo da batata é uma alternativa viável técnica e economicamente. Contudo, sua eficiência pode ser maximizada quando associada com a adubação mineral, especialmente para aumentar a quantidade de nitrogênio disponível desde o início do ciclo da cultura e isso é particularmente importante quando do uso da cama de suínos. A cama de frango, por conter maior quantidade de nutrientes, é uma alternativa melhor do que a cama de suínos, tanto técnica quanto economicamente

Impactos ambientais do manejo agroecológico da caatinga no Rio Grande do Norte.
O objetivo deste trabalho foi avaliar os impactos ambientais do manejo agroecológico da caatinga, em unidades de produção familiar no Rio Grande do Norte, pelo método Ambitec de produção animal - dimensão ambiental, desenvolvido pela Embrapa Meio Ambiente. Foram avaliadas sete unidades de produção familiar, em quatro projetos de assentamentos de reforma agrária do Município de Apodi, RN. Os dados para o levantamento foram obtidos por meio de questionários aplicados aos representantes das unidades produtivas familiares, que atribuíram, a cada variável estudada, um valor que representou a alteração proporcionada pela implementação da tecnologia. Após a inserção dos coeficientes de alteração de cada variável dos indicadores por unidade de produção, o coeficiente de impacto foi automaticamente calculado por meio da planilha Ambitec. O manejo agroecológico da caatinga resultou num impacto ambiental positivo, e suas maiores contribuições foram relacionadas aos efeitos positivos dos seguintes indicadores: capacidade produtiva do solo, uso de insumos materiais, qualidade do produto e diminuição da emissão de poluentes à atmosfera. Dois indicadores geraram efeitos negativos: o uso de energia e o uso de recursos naturais. Pela superioridade dos benefícios gerados, o manejo agroecológico da caatinga é uma inovação tecnológica geradora de impactos ambientais positivos
Hybrid self-assembled materials constituted by ferromagnetic nanoparticles and tannic acid : A theoretical and experimental investigation
Hybrid magnetite materials are interesting for both biomedical and catalytic applications due to their well-known biocompatibility, as well as their magnetic and electric properties. In this work we prepared Fe O nanoparticles (NPs) coated with tannic acid (TA), a natural polyphenol, through two different synthetic routes, aiming to understand the influence of TA in the synthesis step and contribute to the development of water-dispersible magnetic materials. The coating process was verified by information obtained from transmission electron microscopy (TEM), zeta-potential and Fourier transform infrared (FTIR) spectroscopy. The incorporation of TA after Fe O NPs production generated spherical NPs smaller than 10 nm, suggesting that TA plays a fundamental role in the nucleation and organization of Fe O NPs. Data from both density functional theory (DFT) and FTIR allowed us to infer that Fe O interacts mainly with the carbonyl groups of TA. Hybrid materials having improved water-dispersibility are very attractive for biomedical applications
