    Dietary Supplementation with Soluble Plantain Non-Starch Polysaccharides Inhibits Intestinal Invasion of Salmonella Typhimurium in the Chicken

    Soluble fibres (non-starch polysaccharides, NSP) from edible plants but particularly plantain banana (Musa spp.), have been shown in vitro and ex vivo to prevent various enteric pathogens from adhering to, or translocating across, the human intestinal epithelium, a property that we have termed contrabiotic. Here we report that dietary plantain fibre prevents invasion of the chicken intestinal mucosa by Salmonella. In vivo experiments were performed with chicks fed from hatch on a pellet diet containing soluble plantain NSP (0 to 200 mg/d) and orally infected with S.Typhimurium 4/74 at 8 d of age. Birds were sacrificed 3, 6 and 10 d post-infection. Bacteria were enumerated from liver, spleen and caecal contents. In vitro studies were performed using chicken caecal crypts and porcine intestinal epithelial cells infected with Salmonella enterica serovars following pre-treatment separately with soluble plantain NSP and acidic or neutral polysaccharide fractions of plantain NSP, each compared with saline vehicle. Bacterial adherence and invasion were assessed by gentamicin protection assay. In vivo dietary supplementation with plantain NSP 50 mg/d reduced invasion by S.Typhimurium, as reflected by viable bacterial counts from splenic tissue, by 98.9% (95% CI, 98.1–99.7; P<0.0001). In vitro studies confirmed that plantain NSP (5–10 mg/ml) inhibited adhesion of S.Typhimurium 4/74 to a porcine epithelial cell-line (73% mean inhibition (95% CI, 64–81); P<0.001) and to primary chick caecal crypts (82% mean inhibition (95% CI, 75–90); P<0.001). Adherence inhibition was shown to be mediated via an effect on the epithelial cells and Ussing chamber experiments with ex-vivo human ileal mucosa showed that this effect was associated with increased short circuit current but no change in electrical resistance. The inhibitory activity of plantain NSP lay mainly within the acidic/pectic (homogalacturonan-rich) component. Supplementation of chick feed with plantain NSP was well tolerated and shows promise as a simple approach for reducing invasive salmonellosis

    The development of the würde + infinitive construction in Early Modern German (1650–1800)

    This paper presents a corpus-based analysis of the evolution of the würde + infinitive construction in German during the Early Modern period (1650– 1800), using newly available data from the GerManC-corpus. We demonstrate how this construction occupies a unique position orthogonal to both the tense and mood systems of German through an analysis of the syntax and semantics of würde + infinitive clauses, beginning with Modern Standard German and then subsequently with a historical focus on the late seventeenth and eighteenth centuries. Through both quantitative and qualitative analyses, we examine the contexts in which the construction occurs and see how, as it came to be used more frequently over the period in question, it encroached more and more into contexts which had been the preserve of the synthetic preterite subjunctive, even being used in some where the latter is still the norm in modern German. Thus, by the end of the eighteenth century it had become difficult to identify a clear difference in meaning and use between these forms, and the reasons why the würde + in- finitive construction may be preferred over the synthetic preterite subjunctive are by no means clear. We conclude our discussion with an overview of how the würde + infinitive construction was received in the prescriptive tradition during this key period in the standardization of German, seeing its stigmatization in some contexts in part as an attempt by prescriptive grammarians to establish an explicit and clearly justified role for it in the language

    Some Consequences of Thermosolutal Convection: The Grain Structure of Castings

    The essential principles of thermosolutal convection are outlined, and how convection provides a transport mechanism between the mushy region of a casting and the open bulk liquid is illustrated. The convective flow patterns which develop assist in heat exchange and macroscopic solute segregation during solidification; they also provide a mechanism for the transport of dendritic fragments from the mushy region into the bulk liquid. Surviving fragments become nuclei for equiaxed grains and so lead to blocking of the parental columnar, dendritic growth front from which they originated. The physical steps in such a sequence are considered and some experimental data are provided to support the argument

    When Correction Turns Positive:Processing Corrective Prosody in Dutch

    Current research on spoken language does not provide a consistent picture as to whether prosody, the melody and rhythm of speech, conveys a specific meaning. Perception studies show that English listeners assign meaning to prosodic patterns, and, for instance, associate some accents with contrast, whereas Dutch listeners behave more controversially. In two ERP studies we tested how Dutch listeners process words carrying two types of accents, which either provided new information (new information accents) or corrected information (corrective accents), both in single sentences (experiment 1) and after corrective and new information questions (experiment 2). In both experiments corrective accents elicited a sustained positivity as compared to new information accents, which started earlier in context than in single sentences. The positivity was not modulated by the nature of the preceding question, suggesting that the underlying neural mechanism likely reflects the construction of an interpretation to the accented word, either by identifying an alternative in context or by inferring it when no context is present.     Biomarkers of stroke recovery: consensus-based core recommendations from the Stroke Recovery and Rehabilitation Roundtable

    The most difficult clinical questions in stroke rehabilitation are ‘‘What is this patient’s potential for recovery?’’ and ‘‘What is the best rehabilitation strategy for this person, given her/his clinical profile?’’ Without answers to these questions, clinicians struggle to make decisions regarding the content and focus of therapy, and researchers design studies that inadvertently mix participants who have a high likelihood of responding with those who do not. Developing and implementing biomarkers that distinguish patient subgroups will help address these issues and unravel the factors important to the recovery process. The goal of the present paper is to provide a consensus statement regarding the current state of the evidence for stroke recovery biomarkers. Biomarkers of motor, somatosensory, cognitive and language domains across the recovery timeline post-stroke are considered; with focus on brain structure and function, and exclusion of blood markers and genetics. We provide evidence for biomarkers that are considered ready to be included in clinical trials, as well as others that are promising but not ready and so represent a developmental priority. We conclude with an example that illustrates the utility of biomarkers in recovery and rehabilitation research, demonstrating how the inclusion of a biomarker may enhance future clinical trials. In this way, we propose a way forward for when and where we can include biomarkers to advance the efficacy of the practice of, and research into, rehabilitation and recovery after stroke

    Strontium reacts with nitrogen and hydrogen or deuterium under defined conditions to form a phase in which hydride, imide and nitride ions are present side by side.Sr is evaporated at 1000 ° C. The reaction with N 2 and H 2 or D 2 (p (N 2 ) = p (H 2 / D 2 ) = 50 mbar) takes place in the gas phase. The product crystallizes as orange-yellow or black crystal needles at 800 ° C.Neither from X-ray single crystal nor from X-ray or neutron diffraction powder data could the crystal structure be clearly determined. Only a high-resolution diffraction experiment with synchrotron radiation at the powder gave decisive indications for a structural model:Fractions of reflections can only be explained with a model in R 3 m with a = 3,870 (1) Å and c = 18,958 (3) Å. Additional reflections show that, in addition to Sr 2 (H), there are N regions with SrNH as the second phase. The refinement of neutron diffraction data (measurements at 2 K, 10 K, 300 K and 505 K) with a two-phase model Sr 2 (D) N / SrND leads to a meaningful result: in a slightly distorted cubic dense arrangement of Sr atoms occupy layers in Domains Hydride and nitride ions or imide ions Octahedral gaps completely and ordered to one third each

    Fecal Microbiota Transfer Reliable Indications, Donor Screening, and Modes of Application

    Background: Fecal microbiota transfer (FMT) is increasingly being used in Germany, as in other countries, for the treatment of recurrent Clostridioides difficile infection (rCDI). FMT is now being performed both for research and in individual patients outside of clinical trials. No compulsory standards have been established to date for donor screening or for the method of fecal transfer. Given the potential dangers of FMT, this would seem to be urgently necessary. Methods: This review is based on pertinent literature retrieved by a selective search, including the reports of consensus conferences from Germany and abroad. Results: Because of its high efficacy, FMT is the treatment of choice for rCDI. It is largely free of adverse side effects, even in immune-deficient patients, as long as comprehensive and repeated donor screening has been carried out, with extensive clinical and microbiological testing and with the use of structured questionnaires. The ingestion of frozen, encapsulated microbiota is just as effective as other modes of delivery for the treatment of rCDI. Conclusion: Encapsulation of the fecal microbiome (FM) and storage at -20 degrees C is the method of choice, because it can be standardized with the necessary quality controls and it is readily available. Patients with rCDI should undergo FMT by orally ingesting the capsules. There are ongoing research efforts to identify the active components of the FM. It is not yet clear when the ultimate goal of recombinant production can be achieved
