    The polarized expression of Na+,K+-ATPase in epithelia depends on the association between beta-subunits located in neighboring cells

    The polarized distribution of Na+,K+-ATPase plays a paramount physiological role, because either directly or through coupling with co- and countertransporters, it is responsible for the net movement of, for example, glucose, amino acids, Ca2+, K+, Cl-, and CO3H- across the whole epithelium. We report here that the beta-subunit is a key factor in the polarized distribution of this enzyme. 1) Madin-Darby canine kidney (MDCK) cells (epithelial from dog kidney) express the Na+,K+-ATPase over the lateral side, but not on the basal and apical domains, as if the contact with a neighboring cell were crucial for the specific membrane location of this enzyme. 2) MDCK cells cocultured with other epithelial types (derived from human, cat, dog, pig, monkey, rabbit, mouse, hamster, and rat) express the enzyme in all (100%) homotypic MDCK/MDCK borders but rarely in heterotypic ones. 3) Although MDCK cells never express Na+,K+-ATPase at contacts with Chinese hamster ovary (CHO) cells, they do when CHO cells are transfected with beta(1)-subunit from the dog kidney (CHO-beta). 4) This may be attributed to the adhesive property of the beta(1)-subunit, because an aggregation assay using CHO (mock-transfected) and CHO-beta cells shows that the expression of dog beta(1)-subunit in the plasma membrane does increase adhesiveness. 5) This adhesiveness does not involve adherens or tight junctions. 6) Transfection of beta(1)-subunit forces CHO-beta cells to coexpress endogenous a-subunit. Together, our results indicate that MDCK cells express Na+,K+-ATPase at a given border provided the contacting cell expresses the dog P,-subunit. The cell-cell interaction thus established would suffice to account for the polarized expression and positioning of Na+,K+-ATPase in epithelial cells

    Should a family firm communicate their family identity and country of origin? A cross-cultural study from Chile and Spain

    Purpose – This paper investigates the influence of firms’ communication in terms of family firm identity and country-of-origin on consumer response. Design/methodology/approach – A self-supplied online experiment in Chile and Spain is employed using as dependent variables brand trust and intention to buy. The experiment includes the following factors: family firm identity (family vs non-family), country of origin (national vs foreign) and as a manipulation check (type of product: hedonic vs utilitarian). Findings – The results indicate that communicating the family firm identity increases brand trust and purchase intention. Consumers show higher scores on trust and purchase intention when exposed to national country of origin products. The effect of the variability on the dependent variables is greater when the family firm identity is communicated. Trust and purchase intention are different in Chilean and Spanish consumers when the family firm identity is combined with a national country of origin cue. Originality/value – This article contributes to family business theory by exploring how to capitalize on the family firm identity component in brand communication. It also contributes to the theory of corporate brand identity by proposing a communication model oriented toward consumer behavior.     Ara militaris en el centro-occidente de Veracruz: registros recientes

    Based on interviews and field observations, we present information about individuals of Military Macaw (Ara militaris) in the common of Caballo Blanco, in central western of Veracruz. We obtained observations in 2014 and 2015 on a site with cliffs covered by deciduous forest relicts. However, local people said they had seen Military Macaws in this area at least since 2012. It is important to continue working with the people of Caballo Blanco in order to ensure the preservation of this population of Military Macaw. Also, it is necessary to deepen aspects of their ecology on this site, which would be the only one with recent and constant presence of this species in Veracruz.RESUMEN. Con base en entrevistas y observaciones de campo, presentamos información sobre individuos de la guacamaya verde (Ara militaris) en la comunidad de Caballo Blanco, en el centro occidente de Veracruz. Se obtuvieron observaciones en 2014 y 2015 en un sitio con acantilados cubiertos por relictos de bosques caducifolios. Sin embargo, la población local dijo que había visto guacamayas verdes en esta área por lo menos desde 2012. Es importante seguir trabajando con la gente de Caballo Blanco para asegurar la preservación de esta población de guacamayas verdes. Además, es necesario profundizar aspectos de su ecología en este sitio, que sería el único con presencia reciente y constante de esta especie en Veracruz.

    La técnica de análisis espectrométrico de aceites de motores de automoción es altamente efectiva para detectar su desgaste. Los resultados de la aplicación de esta técnica son las concentraciones metálicas de los principales metales de desgaste presentes en muestras de aceite usado; en el caso de los motores de vehículos industriales seconsideran aluminio, cromo, cobre, estaño, hierro, níquel, plomo y silicio. El objetivo de la presente ponencia es llevar a cabo una investigación estadística con el objetivo de relacionar el contenido de silicio con los contenidos de metales de desgaste, a partir de los resultados obtenidos del análisis espectrométrico de un elevado número de muestras de aceite usado, tomadas en motores de diferentes vehículos almacenados en una base de datos. Se considera además, la influencia del tipo de servicio, examinando por separado el caso de los motores de vehículos de transporte y el de los de obras públicas

    Unraveling the selective antibacterial activity and chemical composition of citrus essential oils

    ABSTRACT Post-weaning diarrhea (PWD) is an often disease affecting piglets. It is caused mainly by enterotoxigenic Escherichia coli (ETEC) colonization in pig gut. Antibiotics has been used to prevent, combat and control PWD and its negative impact on the productivity of pig breeding sector. Nonetheless, antibiotics due to their wide antibacterial spectrum also can reach beneficial gut bacteria, such as Lactobacillus. Lately, essential oils (EOs) have emerged as a potential alternative to using antibiotics in animal breeding because of their effect on bacterial growth. Commonly, citrus EOs are by-products of food industry and the availability of these EOs in the worldwide market is huge. Thus, six commercials citrus EOs were evaluated on ETEC strains, as model of pathogenic bacteria, and on Lactobacillus species, as models of beneficial bacteria. In overall, citrus EOs exhibited a selective antibacterial activity with higher effect on pathogenic bacteria (ETECs) than beneficial bacteria (Lactobacillus). Brazilian orange terpenes (BOT) oil presented the highest selective performance and caused higher disturbances on the normal growth kinetic of ETEC than on Lactobacillus rhamnosus. The action was dose-dependent on the maximal culture density (A) and the lag phase duration (λ) of the ETEC. The highest sub-inhibitory concentration (0.925 mg/mL) extended the λ duration to ETEC eight times (14.6 h) and reduced A in 55.9%. For L. rhamnosus, the λ duration was only extended 1.6 times. Despite the fact that limonene was detected as the major compound, the selective antibacterial activity of the citrus EOs could not be exclusively attributed to limonene since the presence of minor compounds could be implicated in conferring this feature

    New young planetary nebulae in IPHAS

We search for very small-diameter galactic planetary nebulae (PNe) representing the earliest phases of PN evolution. The IPHAS catalogue of H-emitting stars provides a useful basis for this study since all sources present in this catalogue must be of small angular diameter. Methods. The PN candidates are selected based on their location in two colour-colour diagrams: IPHAS (r' - H) vs. (r' - i'), and 2MASS (J - H) vs. (H - ). Spectroscopic follow-up was carried out on a sample of candidates to confirm their nature. Results. We present a total of 83 PN candidates. We were able to obtain spectra or find the classification from the literature for 35 candidates. Five of these objects are likely to be new PNe, including one large bipolar PN discovered serendipitously close to an emission-line star. PN distances deduced from extinction-distance relations based on IPHAS field-star photometry are presented for the first time. These yield distance estimates for our objects in the range 2 kpc and 6 kpc. From the data in hand, we conclude that four of the discovered objects are probably young PNe.Peer reviewe

    Efficient inhibition of iron superoxide dismutase and of Trypanosoma cruzi growth by benzo[g]phthalazine derivatives functionalized with one or two imidazole rings

    The synthesis and trypanosomatic behavior of a new series of 1,4-bis(alkylamino)benzo[g]phthalazines 1−4 containing the biologically significant imidazole ring are reported. In vitro antiparasitic activity against Trypanosoma cruzi epimastigotes is remarkable, especially for compound 2, whereas toxicity against Vero cells is very low. Conversion of epimastigotes to metacyclic forms in the presence of the tested compounds causes significant decreases in the amastigote and trypomastigote numbers. Fe-SOD inhibition is noteworthy, whereas effect on human Cu/Zn-SOD is negligible.The authors thank the Spanish CICYT for the financial support

    Climatic control on the Holocene hydrology of a playa-lake system in the western Mediterranean

    Evaporitic lakes such as playa-lakes are characteristic of many arid regions and are unique environments with respect to fauna and flora, while being very vulnerable to climate and environmental fluctuations and threatened by the current global change scenario. Water balance oscillations in these systems can trigger the precipitation or dissolution of different evaporitic minerals, negatively impacting local biodiversity and economic activities. Here, we study the sedimentary record of a small saline pond from a playa-lake complex in southwestern Iberia in order to reconstruct the paleohydrological evolution of this area and assess potential anthropogenic disturbances. The different proxies studied in the-11.9 ky old sedimentary record of the Laguna de la Ballestera suggest that the greatest lake extension and the highest water levels occurred during the Early Holocene, pointing to the wettest period of the record. Climate transitioned towards more arid conditions during the Middle Holocene, and even more dramatically during the Late Holocene. In this last stage the wetland surface and the water level largely diminished and gypsum precipitation gradually increased pointing towards a negative precipitation/ evapotranspiration balance and lowest water levels. Summer desiccation likely occurred under this scenario, especially after-1.0-0.9 cal ky BP coeval with the Medieval Climate Anomaly, when gypsum content started to rise abruptly. However, this significant gypsum precipitation was only associated with a massive drop in the siliciclastic content and scarce carbonates (dolomite and calcite) during the last-400 years. This evidence suggests a shift from a (semi) permanent to a temporal/seasonal hydrological regime. The environmental evolution of this wetland responded to the general climatic evolution of the western Mediterranean during the Holocene, being mostly controlled by changes in insolation. Our data also show that the environmental response of the studied wetland to natural climate variations was only significantly disturbed by human activities since the 20th century, especially in the second half of the century, deduced by abrupt fluctuations in the siliciclastic, gypsum and organic content in the sediments, as well as by the enhanced sedimentary accumulation rates, probably as a response to changes in the hydroperiod of the lake and in the catchment land use.Peer reviewe

    1,4-Bis(alkylamino)benzo[g]phthalazines able to form dinuclear complexes of Cu(II) which as free ligands behave as SOD inhibitors and show efficient in vitro activity against Trypanosoma cruzi

    The synthesis of a new series of 1,4-bis(alkylamino)benzo[g]phthalazines 1–3 is reported, and their ability to form dinuclear complexes with Cu(II) assayed. The geometry of the complexes is dependent on the nature of the electron-donor sites at the sidechains. Compounds 1 and 2, that contain sp3 or sp2 nitrogens at the end of the alkylamino groups, originate monopodal dinuclear complexes which seem to include endogenous OH bridges, and the sidechains seem to actively participate in complexation. However, the substitution of nitrogen by oxygen in 3 leads to a tripodal dinuclear complex in which the sidechains are not involved. The in vitro antiparasitic activity on Trypanosoma cruzi epimastigotes and amastigotes and the SOD activity inhibition have been evaluated for compounds 1–3, and, as expected, 1 and 2 show in all cases relevant results, whereas 3 is always the less active among the three substrates tested.The authors thank the Spanish Comision Interministerial de Ciencia y Tecnologia for the economical support given to this work (SAF99-0066)

    Theory of Transport Properties in the p-wave Superconducting State of Sr2RuO4 - A Microscopic Determination of the Gap Structure -

    We provide a detailed quantitative analysis of transport properties in the p-wave superconducting state of Sr2RuO4. Specifically, we calculate ultrasound attenuation rate and electronic thermal conductivity within the mean field approximation. The impurity scattering of the quasi-particles are treated within the self-consistent T-matrix approximation, and assumed to be in the unitarity limit. The momentum dependence of the gap function is determined by solving the Eliashberg equation for a three-band Hubbard model with the realistic electronic structure of Sr2RuO4. On the basis of the microscopic theory, we can naturally expect nodal structures along the c-axis on the cylindrical Fermi surfaces, even if we assume the chiral pairing state (i.e., \Delta(k) \sim k_x \pm {\rm i} k_y). Consequently, we obtain the temperature dependence of the transport coefficients in agreement with the experimental results. We can clarify that actually the thermal excitations on the passively superconducting bands contribute significantly to the thermal conductivity in a wide temperature range, in contrast to the case of other physical quantities.Comment: 12 pages, 7 figures, submitted to J.
