    Comparative Genomic Analysis of Two Vibrio toranzoniae Strains with Different Virulence Capacity Reveals Clues on Its Pathogenicity for Fish

    Vibrio toranzoniae is a Gram-negative bacterium of the Splendidus clade within the Vibrio genus. V. toranzoniae was first isolated from healthy clams in Galicia (Spain) but recently was also identified associated to disease outbreaks of red conger eel in Chile. Experimental challenges showed that the Chilean isolates were able to produce fish mortalities but not the strains isolated from clams. The aim of the present study was to determine the differences at the genomic level between the type strain of the species (CECT 7225T) and the strain R17, isolated from red conger eel in Chile, which could explain their different virulent capacity. The genome-based comparison showed high homology between both strains but differences were observed in certain gene clusters that include some virulence factors. Among these, we found that iron acquisition systems and capsule synthesis genes were the main differential features between both genomes that could explain the differences in the pathogenicity of the strains. Besides, the studied genomes presented genomic islands and toxins, and the R17 strain presented CRISPR sequences that are absent on the type strain. As it is known, since the 13th Century it's a common practice of the cultural productions of determinated sectors of the European aristocracy.     The Drama Sancho García (1842), well received at the time by the public and by the critics, occupies a strategic position in the trajectories of Spanish Romanticism and of Zorrilla’s oeuvre. However, it has not received enough attention, overshadowed as it has been by more popular works of this author. The article underlines some keys of this particularity, in its historical and cultural context. These keys outline certain features of the author’s formula or art of drama, arisen from a conjunction of good choices, at the different levels and stages of his playwriting, these choices seeking to satisfy the requirements of the popular and conservative public they were meant for. Sustaining theatricality, the principal virtue of Zorrilla’s dramaturgy, he chose, among other options, 1) an adequate dramatic model (tragic composition), closer to romantic drama than to classic tragedy; 2) a narrative frame —historical-legendary, medieval and Spanish—, with high doses of suspense and excitement, 3) his skilful recreation by means of proceedings and materials that were very well known —particularly in the legendary tradition of narration and in the theatre of that time and of Zorrilla himself—, attaining great effect, and 4) the skilful performances of the actors, etc.El drama Sancho García (1842), bien acogido por el público de su tiempo y por la crítica, ocupa una posición estratégica en las trayectorias del romanticismo español y de la obra de Zorrilla. Sin embargo, no ha recibido suficiente atención, eclipsado por títulos más populares de su autor. El artículo destaca algunas claves de esa notoriedad, en su contexto histórico-cultural. Perfilan determinados rasgos del arte o fórmula dramática / de la fórmula o arte dramático del autor, surgida de la conjunción de una serie de aciertos, en los distintos niveles y etapas de la producción teatral, encaminados a satisfacer las demandas del público, popular y conservador, al que iba dirigido. Sustentando la teatralidad, principal virtud de la dramaturgia de Zorrilla, se trata, entre otros, de la elección de un modelo dramático adecuado (composición trágica), más cerca del drama romántico que de la tragedia clásica; de un armazón narrativo —histórico-legendario, medieval y español—, con fuertes dosis de suspense y emoción; de su hábil recreación mediante procedimientos y materiales muy conocidos —en especial en la tradición legendaria del relato y en el teatro de la época y de Zorrilla—, de gran efectismo; del buen oficio de los actores, etc

    Functioning in patients with schizophrenia: A systematic review of the literature using the International Classification of Functioning, Disability and Health (ICF) as a reference

    Purpose: To identify and quantify the main concepts included in published studies focusing on individuals with schizophrenia using the International Classification of Functioning, Disability and Health (ICF). Methods: Searches (limited to those published from 2008 to 2012) were performed in MEDLINE, PsycINFO and CINAHL. Included studies described participants with schizophrenia, were original articles and included only subjects who were at least 18 years of age at study entry. All concepts underlying the measures and the text of the articles were extracted, and they were linked to ICF categories using standardized rules. Results: From the 3584 abstracts retrieved, 348 were randomly selected, and of these, 206 studies met the inclusion criteria. A total of 17,141 concepts were extracted, 84.8% of which could be linked to 491 ICF categories: 222 (45.21%) of the categories referred to Body Functions, 29 (5.91%) to Body Structures, 186 (37.88%) to Activities and Participation and 54 (11%) to Environmental Factors. Seventy second-level categories were reported in at least 5% of all studies: 30 of these categories referred to Body Functions, 2 to Body Structures, 34 to Activities and Participation and 4 to Environmental Factors. Conclusion: The study has allowed us to identify and quantify the main concepts included in studies focusing on people with schizophrenia using the ICF. The majority of the concepts refer to Body Functions and Activities and Participation, rather than to Body Structures and Environmental Factors

    Management in patient with rheumatoid arthritis and tuberculous arthritis: a case report

    Rheumatoid arthritis (RA) is mainly treated with immunosuppressive drugs, which affects the immunological system. Therefore, the risk of tuberculosis was increased two to ten times in RA patients. Moreover, immunosuppressant is contraindicated in patients with tuberculosis arthritis. A 51-year-old male was presented with pain in his left elbow after he slipped on the floor. He was diagnosed with RA for 16 years and only took prednisone for six years. Six months before, he came to a rheumatologist and was given corticosteroid for six months for RA. The left elbow radiograph and joint aspiration revealed a tuberculosis infection. The patient treated with chloroquine and oral antituberculosis for one year and showed good clinical outcomes. Other diseases should be suspected in RA with uncommon symptoms. Chloroquine is the drug of choice in RA patients with tuberculosis who are contraindicated in immunosuppressant therapy because chloroquine has no immunosuppressant effect

    Dos nuevas versiones breves del Cognitive Emotion Regulation Questionnaire y su relación con la depresión y ansiedad

    The Cognitive Emotion Regulation Questionnaire (CERQ) (Garnefski, et al., 2001) is a 36-item instrument for measuring cognitive strategies of emotional regulation. There is a brief, 18-item version that measures the same nine strategies as the full version (Garnefski and Kraaij, 2006a). The aim of this study was to develop a brief form of the CERQ, taking into account two different proposals: a 27-item and an 18-item instrument, the latter focusing solely on the assessment of the two general factors obtained in the second-order structure of the original CERQ model and identified in previous studies as adaptive strategies and less adaptive strategies. Participants in the study were 872 individuals aged 18-58 (mean 33.86, SD=8.43). The confirmatory factor analyses yield adequate overall indices in both versions, together with satisfactory validity. In the discussion, it is argued that the 27-item version is more appropriate for the specific rating of the nine regulation strategies people employ, and we propose the 18-item version as a suitable instrument in clinical context for an overall rating of an individual’s cognitive emotion regulation profile, furthermore, the criterion validity with depression and anxiety keeps similar to the larger versions. El Cognitive Emotion Regulation Questionnaire (CERQ) (Garnefski, et al., 2001) es un instrumento de 36 ítems que mide las estrategias cognitivas de la regulación emocional. Hay una versión breve de 18 elementos que mide las mismas nueve estrategias que la versión completa (Garnefski y Kraaij, 2006a). El objetivo de este estudio fue desarrollar una versión breve, teniendo en cuenta dos propuestas: un instrumento de 27 ítems y otro de 18, el cual se centra exclusivamente en la evaluación de los dos factores generales obtenidos en la estructura de segundo orden del CERQ original e identificado en estudios previos como estrategias adaptativas y estrategias menos adaptativas. Los participantes en el estudio fueron 872 personas de 18 a 58 años (M = 33.86; DT = 8.43). El análisis factorial confirmatorio, proporciona índices globales adecuados en ambas versiones, junto con una validez satisfactoria. En la discusión, se argumenta que la versión de 27 ítems es más apropiada para la evaluación específica de las nueve estrategias de regulación que emplean las personas, y proponemos la versión de 18 ítems como un instrumento adecuado en el contexto clínico para una calificación global del perfil de regulación emocional cognitiva, además, la validez de criterio con depresión y ansiedad se mantiene similar a las versiones completas

    Recombinant monovalent llama-derived antibody fragments (VHH) to rotavirus VP6 protect neonatal gnotobiotic piglets against human rotavirus-induced diarrhea

    Group A Rotavirus (RVA) is the leading cause of severe diarrhea in children. The aims of the present study were to determine the neutralizing activity of VP6-specific llama-derived single domain nanoantibodies (VHH nanoAbs) against different RVA strains in vitro and to evaluate the ability of G6P[1] VP6-specific llama-derived single domain nanoantibodies (VHH) to protect against human rotavirus in gnotobiotic (Gn) piglets experimentally inoculated with virulent Wa G1P[8] rotavirus. Supplementation of the daily milk diet with 3B2 VHH clone produced using a baculovirus vector expression system (final ELISA antibody -Ab- titer of 4096; virus neutralization -VN- titer of 256) for 9 days conferred full protection against rotavirus associated diarrhea and significantly reduced virus shedding. The administration of comparable levels of porcine IgG Abs only protected 4 out of 6 of the animals from human RVA diarrhea but significantly reduced virus shedding. In contrast, G6P[1]-VP6 rotavirus-specific IgY Abs purified from eggs of hyperimmunized hens failed to protect piglets against human RVA-induced diarrhea or virus shedding when administering similar quantities of Abs. The oral administration of VHH nanoAb neither interfered with the host's isotype profiles of the Ab secreting cell responses to rotavirus, nor induced detectable host Ab responses to the treatment in serum or intestinal contents. This study shows that the oral administration of rotavirus VP6-VHH nanoAb is a broadly reactive and effective treatment against rotavirus-induced diarrhea in neonatal pigs. Our findings highlight the potential value of a broad neutralizing VP6-specific VHH nanoAb as a treatment that can complement or be used as an alternative to the current strain-specific RVA vaccines. Nanobodies could also be scaled-up to develop pediatric medication or functional food like infant milk formulas that might help treat RVA diarrhea.Fil: Vega, Celina Guadalupe.     Temperature dependence of underdense nanostructure formation in tungsten under helium irradiation

    Recently, tungsten has been found to form a highly underdense nanostructured morphology ("W fuzz") when bombarded by an intense flux of He ions, but only in the temperature window 900-2000 K. Using object kinetic Monte Carlo simulations (pseudo-3D simulations) parameterized from first principles, we show that this temperature dependence can be understood based on He and point defect clustering, cluster growth, and detrapping reactions. At low temperatures (2300 K), all He is detrapped from clusters, preventing the formation of the large clusters that lead to fuzz growth in the intermediate temperature range. (C) 2017 Elsevier B.V.     Missense variants in ANKRD11 cause KBG syndrome by impairment of stability or transcriptional activity of the encoded protein

We characterized clinical, molecular, and functional spectra of ANKRD11 missense variants. Methods We collected clinical information of individuals with ANKRD11 missense variants and evaluated phenotypic fit to KBG syndrome. We assessed pathogenicity of variants through in silico analyses and cell-based experiments. Results We identified 20 unique, mostly de novo, ANKRD11 missense variants in 29 individuals, presenting with syndromic neurodevelopmental disorders similar to KBG syndrome caused by ANKRD11 protein truncating variants or 16q24.3 microdeletions. Missense variants significantly clustered in repression domain 2 at the ANKRD11 C-terminus. Of the 10 functionally studied missense variants, 6 reduced ANKRD11 stability. One variant caused decreased proteasome degradation and loss of ANKRD11 transcriptional activity. Conclusion Our study indicates that pathogenic heterozygous ANKRD11 missense variants cause the clinically recognizable KBG syndrome. Disrupted transrepression capacity and reduced protein stability each independently lead to ANKRD11 loss-of-function, consistent with haploinsufficiency. This highlights the diagnostic relevance of ANKRD11 missense variants, but also poses diagnostic challenges because the KBG-associated phenotype may be mild and inherited pathogenic ANKRD11 (missense) variants are increasingly observed, warranting stringent variant classification and careful phenotyping
