    Genomic Transformation of the Picoeukaryote Ostreococcus tauri

    Common problems hindering rapid progress in Plant Sciences include cellular, tissue and whole organism complexity, and notably the high level of genomic redundancy affecting simple genetics in higher plants. The novel model organism Ostreococcus tauri is the smallest free-living eukaryote known to date, and possesses a greatly reduced genome size and cellular complexity(1,2), manifested by the presence of just one of most organelles (mitochondrion, chloroplast, golgi stack) per cell, and a genome containing only ~8000 genes. Furthermore, the combination of unicellularity and easy culture provides a platform amenable to chemical biology approaches. Recently, Ostreococcus has been successfully employed to study basic mechanisms underlying circadian timekeeping(3-6). Results from this model organism have impacted not only plant science, but also mammalian biology(7). This example highlights how rapid experimentation in a simple eukaryote from the green lineage can accelerate research in more complex organisms by generating testable hypotheses using methods technically feasible only in this background of reduced complexity. Knowledge of a genome and the possibility to modify genes are essential tools in any model species. Genomic(1), Transcriptomic(8), and Proteomic(9) information for this species is freely available, whereas the previously reported methods(6,10) to genetically transform Ostreococcus are known to few laboratories worldwide. In this article, the experimental methods to genetically transform this novel model organism with an overexpression construct by means of electroporation are outlined in detail, as well as the method of inclusion of transformed cells in low percentage agarose to allow selection of transformed lines originating from a single transformed cell. Following the successful application of Ostreococcus to circadian research, growing interest in Ostreococcus can be expected from diverse research areas within and outside plant sciences, including biotechnological areas. Researchers from a broad range of biological and medical sciences that work on conserved biochemical pathways may consider pursuing research in Ostreococcus, free from the genomic and organismal complexity of larger model species

    Continuous variable remote state preparation

    We extend exact deterministic remote state preparation (RSP) with minimal classical communication to quantum systems of continuous variables. We show that, in principle, it is possible to remotely prepare states of an ensemble that is parameterized by infinitely many real numbers, i.e., by a real function, while the classical communication cost is one real number only. We demonstrate continuous variable RSP in three examples using (i) quadrature measurement and phase space displacement operations, (ii) measurement of the optical phase and unitaries shifting the same, and (iii) photon counting and photon number shift.Comment: 7 pages, RevTeX

    Static properties and spin dynamics of the ferromagnetic spin-1 Bose gas in magnetic field

    Properties of spin-1 Bose gases with ferromagnetic interaction in the presence of a nonzero magnetic field are studied. The equation of state and thermodynamic quantities are worked out with the help of a mean-field approximation. The phase diagram besides Bose-Einstein condensation contains a first order transition where two values of the magnetization coexist. The dynamics is investigated with the help of the Random Phase Approximation.     Development of the City Public Service Model on the Basis of Integrated Transport Flow Indicators

    The problem of modeling public services based on architectural and planning decisions is considered, the role of traffic in the formation of a model of city services is analyzed. An integrated criterion for the quality of public services is proposed. A method has been developed for determining segmented public services taking into account the transport areas of the city, which will make it possible to evenly disperse public service centers. The basis is a socio-planning organization, as a material-spatial system containing anthropogenic and natural components – the territory and institutions where the functional processes that take place in the urban planning environment take place. The described model has certain versatility, and is simultaneously suitable for characterizing various categories of service institutions. Thus, the task of optimizing the quality of public services in the city is reduced to a mathematical model for which, by setting the basic design criteria, the optimal result can be obtained.On the basis of a questionnaire survey and analysis of statistical data, calculation of traffic intensity, the demand and supply of the level of public services фre studied. The structural elements of this model: population, territory, transport and service institutions, are in dialectical interaction, which is described by the mathematical model in this study. The model is based on the calculation of the minimum population in the service area, which allows to have i-th type establishments using the social potential method, as well as a graph-analytical method in determining the optimal location of service institutions in the city.As a result of the research, a conceptual model of public services for cities is built, a layout of supermarkets in the territory of Uzhhorod and distribution of service areas of these institutions is proposed. This optimization will ensure uniform domestic servicing of the territory, optimal performance indicators of service establishments and minimum average service radii of points
