Slight respiratory irritation but not inflammation in mice exposed to (1-->3)-beta-D-glucan aerosols.
Airway irritation effects after single and repeated inhalation exposures to aerosols of beta-glucan (grifolan) were investigated in mice. In addition, the effects on serum total immunoglobulin E (IgE) production and histopathological inflammation in the respiratory tract were studied. The beta-glucan aerosols provoked slight sensory irritation in the airways, but the response was not concentration dependent at the levels studied. Slight pulmonary irritation was observed after repeated exposures. No effect was found on the serum total IgE levels, and no signs of inflammation were seen in the airways 6 h after the final exposure. The results suggest that, irrespective of previous fungal sensitization of the animals, inhaled beta-glucan may cause symptoms of respiratory tract irritation but without apparent inflammation. Respiratory tract irritation reported after inhalation of fungi may not be entirely attributed to beta-glucan
Filamentous Fungi and Yeasts Associated with Mites Phoretic on Ips typographus in Eastern Finland
The European spruce bark beetle (Ips typographus) has become a major forest pest in Finland in recent years. The beetle is a well-known vector of mainly ophiostomatoid fungi causing blue-stain of timber and pathogens that have the ability to amplify the insect damage. It also vectors other associated organisms, such as phoretic mites. The ecology of these mites remains poorly understood, including their associations with fungi. In this study, we considered filamentous fungi and yeasts associated with mites phoretic on I. typographus. Fungal identifications were based on DNA sequences and phylogenetic analyses of the ITS and/or partial β-tubulin gene regions. Fifteen fungal species were detected, including eight yeasts and seven filamentous fungi. Eleven percent of the beetles carried mites and of these 74% carried at least one fungal species. An average of two fungal species were carried per mite. The most commonly found filamentous fungi were Grosmannia penicillata (25%), Ophiostoma bicolor (19%), O. ainoae (12%) and O. brunneolum (12%). Of the yeast species, the most commonly found was Wickerhamomyces bisporus (47%). This study is the first to report yeasts associated with I. typographus and its phoretic mites in Finland. Majority of the filamentous fungal species found are those previously reported in association with I. typographus. The results also confirmed that many of the fungal species commonly found on I. typographus are also associated with its phoretic mites. However, the nature of the symbiosis between the mites, beetles and fungal associates remains to be understood

Is Decreased Xylem Sap Surface Tension Associated With Embolism and Loss of Xylem Hydraulic Conductivity in Pathogen-Infected Norway Spruce Saplings?
Increased abiotic stress along with increasing temperatures, dry periods and forest disturbances may favor biotic stressors such as simultaneous invasion of bark beetle and ophiostomatoid fungi. It is not fully understood how tree desiccation is associated with colonization of sapwood by fungi. A decrease in xylem sap surface tension (sigma(xylem)) as a result of infection has been hypothesized to cause xylem embolism by lowering the threshold for air-seeding at the pits between conduits and disruptions in tree water transport. However, this hypothesis has not yet been tested. We investigated tree water relations by measuring the stem xylem hydraulic conductivity (K-stem), sigma(xylem), stem relative water content (RWCstem), and water potential (psi(stem)), and canopy conductance (g(canopy)), as well as the compound composition in xylem sap in Norway spruce (Picea abies) saplings. We conducted our measurements at the later stage ofEndoconidiophora polonicainfection when visible symptoms had occurred in xylem. Saplings of two clones (44 trees altogether) were allocated to treatments of inoculated, wounded control and intact control trees in a greenhouse. The saplings were destructively sampled every second week during summer 2016. sigma(xylem), K(stem)and RWC(stem)decreased following the inoculation, which may indicate that decreased sigma(xylem)resulted in increased embolism. g(canopy)did not differ between treatments indicating that stomata responded to psi(stem)rather than to embolism formation. Concentrations of quinic acid, myo-inositol, sucrose and alkylphenol increased in the xylem sap of inoculated trees. Myo-inositol concentrations also correlated negatively with sigma(xylem)and K-stem. Our study is a preliminary investigation of the role of sigma(xylem)inE. polonicainfected trees based on previous hypotheses. The results suggest thatE. polonicainfection can lead to a simultaneous decrease in xylem sap surface tension and a decline in tree hydraulic conductivity, thus hampering tree water transport.Peer reviewe

Method for automated requirement checking in social housing projects
One of the possible strategies for improving the quality of construction projects is the use of Building Information Modelling (BIM), which involves innovations in information technology and changes in critical processes. In the social housing sector in Brazil, the need to evaluate a large number of projects proposed for funding in a very short time frame is a major challenge. Automated design checking has been pointed out as one of the main opportunities for the use of BIM in the building industry. The evaluation of social housing projects by governmental bodies is often based mainly on the experience of construction professionals, which may result in lack of uniformity in the criteria adopted, and in a time consuming and costly process. The aim of this research study is to propose a method for automated checking of requirements for the design of social housing projects by using BIM. This method was tested in two housing projects funded by the Minha Casa, Minha Vida programme, and compared to the manual process. This investigation also makes contributions related to the understanding of the nature of the requirements involved in this type of project, and discusses the process changes needed to introduce automated design checking.

Effect of a shelf-furnished screen on space utilisation and social behaviour of indoor group-housed cats (Felis silvestris catus)
The environment of the laboratory cat can be restrictive and may impact their welfare. Enrichment is often provided to alleviate welfare impacts but is seldom assessed or validated for efficacy. This study investigated the effect of novel room furniture (a screen) on the expression of agonistic and affiliative behaviours and space utilisation amongst colony-housed laboratory cats. Video footage of cats (N = 29) housed in social rooms (N = 4) was collected for 2 days before (baseline phase), 4 days during (test phase) and 2 days following (removal phase) introduction of the novel furniture. Space utilisation data were collected using scan sampling every 10 min and analysed using a generalised linear mixed effects model and Tukey’s HSD test. Behavioural data were collected using continuous sampling for 3 h a day in 6 × 30 min episodes and analysed using a Poisson generalised mixed effects model.
Significantly more agonistic events occurred before the morning feed compared to after feeding within all phases (pre-feed mean = 0.227; post-feed mean = 0.026; P < 0.0001). However no significant differences were observed before the morning feed compared to after feeding between phases indicating that the screen had no effect on reducing pre-feed aggression at the morning feed. Agonistic behaviours occurred significantly less following the morning feed during the test phase when compared to the baseline phase (test post-feed mean = 0.011; baseline post-feed mean = 0.029; P = 0.0342). Significant differences were also observed on removal of the screen with agonistic behaviour increasing above baseline at the afternoon pre-feed time point, possibly indicative of aggression due to frustration or a rebound effect (removal pre-feed mean = 0.151; baseline pre-feed mean 0.048; P < 0.0001). Affiliative interactions between phases were not significantly affected by screen presence. Given the ratio of the screen to existing shelving (0.58:0.42) a statistical significant proportion of cats were found to be on the screen in the test phase of the study (P < 0.0001). This study suggests that exploiting the unused vertical space by the addition of stand-alone shelving should be considered a valuable resource for the cat by increasing useable space and reducing agonistic interactions
Improved synthesis of [18F] fallypride and characterization of a Huntington’s disease mouse model, zQ175DN KI, using longitudinal PET imaging of D2/D3 receptors
Dopamine receptors are involved in pathophysiology of neuropsychiatric diseases, including Huntington’s disease (HD). PET imaging of dopamine D2 receptors (D2R) in HD patients has demonstrated 40% decrease in D2R binding in striatum, and D2R could be a reliable quantitative target to monitor disease progression. A D2/3R antagonist, [18F] fallypride, is a high-affinity radioligand that has been clinically used to study receptor density and occupancy in neuropsychiatric disorders. Here we report an improved synthesis method for [18F]fallypride. In addition, high molar activity of the ligand has allowed us to apply PET imaging to characterize D2/D3 receptor density in striatum of the recently developed zQ175DN knock-in (KI) mouse model of HD.Peer reviewe
