    Characterisation of polymorphic microsatellite loci in the freshwater bryozoan Fredericella sultana

    Eight polymorphic microsatellite loci were isolated from massively parallel next-generation sequencing data and tested in three populations (74 individuals) of the colonial freshwater bryozoan Fredericella sultana. Up to 13 alleles per locus were found and all loci were polymorphic in all populations. Minimum of three loci were sufficient to distinguish all unique multilocus genotypes. These highly variable markers are suitable for clonal identity assignment based on unique multilocus genotypes and provide tools for resolving fine scale population structure in a species characterised by clonal, vegetative growth and asexual reproductio

    Spin and Statistics and First Principles

    It was shown in the early Seventies that, in Local Quantum Theory (that is the most general formulation of Quantum Field Theory, if we leave out only the unknown scenario of Quantum Gravity) the notion of Statistics can be grounded solely on the local observable quantities (without assuming neither the commutation relations nor even the existence of unobservable charged field operators); one finds that only the well known (para)statistics of Bose/Fermi type are allowed by the key principle of local commutativity of observables. In this frame it was possible to formulate and prove the Spin and Statistics Theorem purely on the basis of First Principles. In a subsequent stage it has been possible to prove the existence of a unique, canonical algebra of local field operators obeying ordinary Bose/Fermi commutation relations at spacelike separations. In this general guise the Spin - Statistics Theorem applies to Theories (on the four dimensional Minkowski space) where only massive particles with finite mass degeneracy can occur.     BACKGROUND: Personalized therapy considering clinical and genetic patient characteristics will further improve breast cancer survival. Two widely used treatments, chemotherapy and radiotherapy, can induce oxidative DNA damage and, if not repaired, cell death. Since base excision repair (BER) activity is specific for oxidative DNA damage, we hypothesized that germline genetic variation in this pathway will affect breast cancer-specific survival depending on treatment. METHODS: We assessed in 1,408 postmenopausal breast cancer patients from the German MARIE study whether cancer specific survival after adjuvant chemotherapy, anthracycline chemotherapy, and radiotherapy is modulated by 127 Single Nucleotide Polymorphisms (SNPs) in 21 BER genes. For SNPs with interaction terms showing p<0.1 (likelihood ratio test) using multivariable Cox proportional hazard analyses, replication in 6,392 patients from nine studies of the Breast Cancer Association Consortium (BCAC) was performed. RESULTS: rs878156 in PARP2 showed a differential effect by chemotherapy (p=0.093) and was replicated in BCAC studies (p=0.009; combined analysis p=0.002). Compared to non-carriers, carriers of the variant G allele (minor allele frequency=0.07) showed better survival after chemotherapy (combined allelic hazard ratio (HR)=0.75, 95% 0.53-1.07) and poorer survival when not treated with chemotherapy (HR=1.42, 95% 1.08-1.85). A similar effect modification by rs878156 was observed for anthracycline-based chemotherapy in both MARIE and BCAC, with improved survival in carriers (combined allelic HR=0.73, 95% CI 0.40-1.32). None of the SNPs showed significant differential effects by radiotherapy. CONCLUSIONS: Our data suggest for the first time that a SNP in PARP2, rs878156, may together with other genetic variants modulate cancer specific survival in breast cancer patients depending on chemotherapy. These germline SNPs could contribute towards the design of predictive tests for breast cancer patients

    A Numerical Study on Temperature Distribution of Line Heated Anisotropic Carbon Fiber Composites

    Earlier we have described the various uses of infrared line scanner based thermal nondestructive testing equipment [1]. Time constants of measurements made with these kind of equipment are very suitable for testing carbon fiber composites. Scanning a line heat source over a sample surface causes a nonuniform temperature distribution in the sample. In addition to the heat flow normal to the surface, lateral heat flow exists in the surface plane. In the case of carbon fiber composites with a specific oriented structure, the surface temperature distributions depend on the direction where the line source moves. Generally, this is true of any sample having anisotropic thermal conductivity. In oriented carbon fiber composites the bulk thermal conductivity can be considered anisotropic, because the heat transfer in the composite is different in the direction of the fibers compared to perpendicular directions [2,3]. Varis et al. have discussed these phenomenon briefly with the testing of carbon fiber tubes using numerical methods [4].     In recent years a variety of NDT techniques utilizing an IR camera have been developed (for example [1–4]). In the synchronous heating method [3] an IR camera is used for detecting the surface temperature rise caused by a scanning laser beam. The movement of the heating beam is synchronized with the deflection mirror of the IR camera so the distance between the object point of the camera and the heating point remains constant. This measurement set-up allows rapid inspection of thin coatings that are otherwise problematic from the NDT point of view.     Riverbed sediments buffer phosphorus concentrations downstream of sewage treatment works across the River Wensum catchment, UK

    Purpose: Wastewater effluent discharged into rivers from sewage treatment works (STWs) represents one of the most important point sources of soluble reactive phosphorus (SRP) pollution and is a major driver of freshwater eutrophication. In this study, we assess the ability of riverbed sediments to act as a self-regulating buffering system to reduce SRP dissolved in the water column downstream of STW outflows. Materials and methods: River water and riverbed sediment samples were collected from 10 tributary outlets across the River Wensum catchment, Norfolk, UK, at monthly intervals between July and October 2016, such that 40 sediment and 40 water samples were collected in total. Of these locations, five were located downstream of STWs and five were on tributaries without STWs. Dissolved SRP concentrations were analysed and the Equilibrium Phosphorus Concentration (EPC0) of each sediment sample was measured to determine whether riverbed sediments were acting as net sources or sinks of SRP. Results and discussion: The mean SRP concentration downstream of STWs (382 µg P L-1) was double that of sites without a STW (185 µg P L-1), whilst the mean EPC0 for effluent impacted sites (105 µg P L-1) was 70% higher than that recorded at unaffected sites (62 µg P L-1). Regardless of STW influence, riverbed sediments across all 10 sites almost always acted as net sinks for SRP from the overlying water column. This was particularly true at sites downstream of STWs which displayed enhanced potential to buffer the river against increases in SRP released in sewage effluent. Conclusions: Despite EPC0 values revealing riverbed sediments were consistently acting as sinks for SRP, elevated SRP concentrations downstream of STWs clearly demonstrate the sediments have insufficient SRP sorption capacity to completely buffer the river against effluent discharge. Consequently, SRP concentrations across the catchment continue to exceed recommended standards for good chemical status, thus emphasising the need for enhanced mitigation efforts at STWs to minimise riverine phosphorus loading
