    Aging and CaMKII alter intracellular Ca2+ transients and heart rhythm in Drosophila melanogaster

    Aging is associated to disrupted contractility and rhythmicity, among other cardiovascular alterations. Drosophila melanogaster shows a pattern of aging similar to human beings and recapitulates the arrhythmogenic conditions found in the human heart. Moreover, the kinase CaMKII has been characterized as an important regulator of heart function and an arrhythmogenic molecule that participate in Ca2+ handling. Using a genetically engineered expressed Ca2+ indicator, we report changes in cardiac Ca 2+ handling at two different ages. Aging prolonged relaxation, reduced spontaneous heart rate (HR) and increased the occurrence of arrhythmias, ectopic beats and asystoles. Alignment between Drosophila melanogaster and human CaMKII showed a high degree of conservation and indicates that relevant phosphorylation sites in humans are also present in the fruit fly. Inhibition of CaMKII by KN-93 (CaMKII-specific inhibitor), reduced HR without significant changes in other parameters. By contrast, overexpression of CaMKII increased HR and reduced arrhythmias. Moreover, it increased fluorescence amplitude, maximal rate of rise of fluorescence and reduced time to peak fluorescence. These results suggest that CaMKII in Drosophila melanogaster acts directly on heart function and that increasing CaMKII expression levels could be beneficial to improve contractility.Centro de Investigaciones CardiovascularesCentro de Investigaciones Inmunológicas Básicas y Aplicada

    Compuestos orgánicos volátiles de especies aromáticas como promotores de crecimiento del tomate saladette (Solanum lycopersicum Mill.)

    Objetive: The present study analysed the effects of the Volatile Organic Compounds (VOCs) of the foliage and flowers of Ocimum basilicum, Tagetes erecta and Origanum majorana on the growth of the Saladette tomato. Design/Method/Approach: The aromatic species in their phase of development and flowering were transplanted between grooves of tomato plants. The treatments (Association of aromatic species 1, 2 and 3 in development + tomato plant, association of aromatic species 1, 2 and 3 in flowering + tomato plant and tomato plant without association) were randomized under a completely randomized experimental design, with arrangements in blocks. The indicators of the phenology and growth of tomato plants were measured. The information was analyzed according to the ANOVA (0.05) and based on the calculated F. Next, the Tukey method (0.5) was applied. Results: The results show that the association of aromatic species offers a positive effect on the growth of tomato plants and the flowering stage showed clear results. The Tagetes erecta species produced a highly significant difference with respect to the control and Ocimum basilicum (Tukey 0.05), this showed that Marigold improves the height, diameter of the stem and the biomass of the vegetable. In addition, the Volatile Organic Compounds of T. erecta caused early flowering in the tomato (5 days). Research Limitations/ Implications:  The plantation interspersed between aromatic species and vegetables generated costs, this can be a limiting factor in the transfer of technology. Findings/Conclusions: The Volatile Organic Compounds of the aromatic species participate as plant breeders and controlling tomato stress. Thus, the primary crop or the vegetable associated with Tagetes erecta and Origanum majorana favors the height, the diameter of the stem, the biomass and presents an early flowering (5 days) obtaining an early harvest.  Objetivo: Analizar los efectos de los Compuesto Orgánicos Volátiles (COVs) del follaje y de las flores de Ocimum basilicum, Tagetes erecta y Origanum majorana, en el crecimiento del tomate Saladette. Diseño/metodología/aproximación: Las especies aromáticas en su fase de desarrollo y floración se trasplantaron entre los surcos de las plantas de tomate; los tratamientos (Asociación de especies aromáticas  1, 2 y 3 en desarrollo + planta de tomate; asociación de especies aromáticas 1, 2 y 3 en floración + planta de tomate y planta tomate sin asociación) fueron aleatorizadas bajo un diseño experimental completamente al azar, con arreglos en bloques. Los indicadores de la fenología y crecimiento de las plantas de tomate fueron medidos; la información se analizó de acuerdo al ANOVA (0.05) y con base a la F calcula, se procedió con la prueba posteriori por el método de Tukey (0.05). Resultados: Los resultados demuestran que la asociación de especies aromáticas ofrece un efecto positivo en el crecimiento de las plantas de tomate; expresándose más notorios en etapa de floración de las especies aromáticas. La especie Tagetes erecta produjo una diferencia altamente significativa con respecto al testigo y Ocimum basilicum (Tukey 0.05), esto demostró que el Cempasúchil mejora la altura, diámetro del tallo y la biomasa de la hortaliza. Además, los Compuestos Orgánicos Volátiles de T. erecta causaron floración temprana en el tomate (5 días). Limitaciones del estudio/implicaciones: La plantación intercalada entre especies aromáticas y hortalizas generó costos, ésto puede ser un factor limitante en la transferencia de tecnología. Hallazgos/conclusiones: Los compuestos orgánicos volátiles de las especies aromáticas participan como fitomejoradores y controlan el estrés del tomate. Así, el cultivo primario o la hortaliza asociado con Tagetes erecta y Origanum majorana favorece la altura, el diámetro del tallo, la biomasa y presenta una floración temprana (5 días) obteniendo una cosecha anticipada. &nbsp

    Indexación: Scopus.Traditionally, it has been suggested that rest is the ideal tool to allow natural recovery of tissues induced by overload. However, the times necessary for this to occur are very long. It has been estimated that electrotherapy is useful when it comes to recover some overused tissues, however, these benefits in vocal function are unknown. The purpose of this study was to describe the effects of acupuncture-like transcutaneous electrical nerve stimulation on the laryngeal and cordai tissues that were previously subjected to vocal overload. In this research. 21 women without pathology or vocal training participated, whose age was 24 ± 1.28 years. The participants were subjected to vocal overload for 40 minutes and subsequently to 15 minutes of acupuncture-like transcutaneous electrical nerve stimulation. Through continuous laryngoscopy, acoustic-perceptual analysis and symptomatic study, the changes caused by electrotherapy were examined. The main results include perilaryngeal and intrinsic laryngeal muscles relaxation, and elimination of the symptoms suffered.     Paradigmatic de novo GRIN1 variants recapitulate pathophysiological mechanisms underlying GRIN1-related disorder clinical spectrum

    Background: GRIN-related disorders (GRD), the so-called grinpathies, is a group of rare encephalopathies caused by mutations affecting GRIN genes (mostly GRIN1, GRIN2A and GRIN2B genes), which encode for the GluN subunit of the N-methyl D-aspartate (NMDA) type ionotropic glutamate receptors. A growing number of functional studies indicate that GRIN-encoded GluN1 subunit disturbances can be dichotomically classified into gain- and loss-of-function, although intermediate complex scenarios are often present. Methods: In this study, we aimed to delineate the structural and functional alterations of GRIN1 disease-associated variants, and their correlations with clinical symptoms in a Spanish cohort of 15 paediatric encephalopathy patients harbouring these variants. Results: Patients harbouring GRIN1 disease-associated variants have been clinically deeplyphenotyped. Further, using computational and in vitro approaches, we identified different critical checkpoints affecting GluN1 biogenesis (protein stability, subunit assembly and surface trafficking) and/or NMDAR biophysical properties, and their association with GRD clinical symptoms. Conclusions: Our findings show a strong correlation between GRIN1 variants-associated structural and functional outcomes.     El informe está dividido en cuatro apartados: “Derecho a la información y transparencia”, “La televisión mexicana”, “Empresas y prácticas periodísticas” y “Los que se fueron”. En el primero de ellos se presenta un texto que ayuda a entender cuál es el momento en el que se encuentran las propuestas legislativas para regular a los medios y las telecomunicaciones en México, y una evaluación de los primeros cinco años del Instituto de Transparencia e Información Pública de Jalisco. El segundo apartado del informe es ecléctico, pues se compone de artículos que trabajan distintas temáticas de la televisión:la estructura y oferta de la televisión en nuestro país (en particular en la ciudad de Guadalajara), la televisión por cable (enfatizando el caso de Megacable), un recuento de cómo se gestó el Canal 44 y de sus prospectivas en 2011, y los mundiales de futbol. La tercera parte del informe documenta algunas de las situaciones más importantes que se viven en el periodismo local: estos trabajos presentan sistemas en crisis (alta vulnerabilidad de los periodistas mexicanos ante un clima de violencia que lejos de disminuir va en aumento, y la participación, por acción u omisión, del Estado mexicano en la sistemática violación de los derechos de quienes dedican su vida al trabajo periodístico. Los siguientes artículos tratan sobre las transformaciones de las empresas periodísticas, particularmente las del sector de la prensa escrita: la rápida e inexorable desaparición de los suplementos culturales, y una radiografía sobre las formas de producción de algunas secciones internacionales de los periódicos tapatíos.
