    Independent responses to ultraviolet radiation and herbivore attack in broccoli

    The plant responses to ultraviolet-B radiation (UV-B) and to insect herbivory are believed to be partially similar. In this study, responses to these factors were investigated in the crop species broccoli (Brassica oleracea L. convar. botrytis, Brassicaceae). Plants were first grown under three UV-B regimes (80%, 23%, and 4% transmittance of ambient UV-B) in greenhouses covered with either innovative materials (high and medium transmittance) or conventional glass (low transmittance). Half of the plants then remained under these conditions, but the other half were transferred to the field with ambient light and herbivore access for up to 3 d. The plant responses to distinct environmental conditions were examined by analysing the morphological and chemical parameters of plants kept inside and plants exposed in the field. Furthermore, suitability of field-exposed plants to naturally occurring insects was investigated in relation to UV-B pretreatment. High levels of UV-B radiation led to increased flavonoid concentrations, but to a lower biomass accumulation in broccoli. These patterns remained after outdoor exposure. However, UV-induced changes of plant traits did not alter attractiveness to herbivorous insects: thrips, whiteflies, and aphids attacked plants independently of UV-B pretreatment. A 3-fold increase of indolyl glucosinolate concentrations occurred in above-ground tissue of all the plants, most likely due to massive herbivore attack after 3 d of field exposure. The results show that plants respond with high specificity to different abiotic and biotic impacts, demonstrating the separate perception and processing of stress factors

    Generación de una Línea Transgénica de Pez Cebra para Estudiar la Remodelación de Uniones Adherentes

    El remodelado dinámico de los contactos adhesivos intercelulares o uniones adherentes (UAs) mediado por la proteína cadherina epitelial E (cadh-E) es clave para coordinar los movimientos migratorios celulares durante la embriogénesis animal. Debido a la simplicidad estructural y a la accesibilidad experimental, el embrión de pez cebra (Danio rerio) se ha establecido como un modelo para estudiar migración celular y para entender la dinámica del tráfico vesicular y distribución de cadh-E en UAs. En el presente proyecto se propuso generar una línea transgénica estable de pez cebra (cadh-E-KikGR) para la expresión de cadh-E fusionada a la proteína fotoconvertible KikGR para visualizar in vivo la remodelación de UAs. En una primera etapa se analizó la expresión endógena nativa de cadh-E y se comparó con la expresión transgénica de cadh-E fusionada a la proteína reportera fluorescente GFP (Green Fluorescent Protein) como una primera estrategia de monitoreo. Se observó que cadh-E no se localiza en membranas como la cadh-E endógena por lo que la fusión no sería funcional una vez expresada en el embrión. El clonado de KikGR, de menor tamaño, en reemplazo de GFP en el vector final de expresión permitirá la obtención del embrión transgénico de interés

    Purpose. To evaluate the dose effect of vitamin K3 on wound healing mechanisms. Methods. Conjunctival fibroblasts were incubated for 24 hours. An artificial wound was made and the cells were incubated with fresh medium plus doses of vitamin K3 to be tested. Wound repair was monitored at 0, 18, 24, and 48 hours. Proliferation was measured in actively dividing cells by [3H]thymidine uptake. Six different groups were tested: group 1/no drugs added, group 2/ethanol 0.1%, group 3/vitamin K3 1 mg/L, group 4/vitamin K3 2 mg/L, group 5/vitamin K3 4 mg/L, and group 6/vitamin K3 6 mg/L. Each experiment was carried out in triplicate and 4 times. Results. There were no differences among groups at the initial time. In vitro wound repair was slower in groups 4, 5, and 6. There were no differences between control and ethanol groups and between control and vitamin K3 1 mg/L groups. Fibroblast mitogenic activity was statistically decreased in all vitamin K groups; statistical differences were found among vitamin K3 1 mg/mL and higher doses too. In groups 5 and 6, cellular toxicity was presented. Conclusions. Vitamin K3 is able to inhibit fibroblast proliferation. Vitamin K3 2 mg/L or higher doses inhibit wound healing repair, exhibiting cellular toxicity at 4 and 6 mg/L

    Estudios Moleculares y Celulares del Sistema de Endotelinas en Cáncer Colorectal

    En el presente proyecto se estudió el rol que cumple el sistema de endotelinas en el tracto digestivo murino. Previa verificación de la localización del sistema en condiciones fisiológicas normales, se estudiaron los cambios en el patrón de expresión de los péptidos y sus receptores durante el desarrollo de un modelo de cáncer colo-rectal murino inducido por azoximetano.La caracterización de las diferentes etapas del desarrollo de cáncer colo-rectal se realizó utilizando técnicas de anatomía patológica clásicas, de inmunohistoquímica y de microscopia óptica no lineal. Estos datos morfológicos fueron correlacionados con los niveles de expresión de los ARNms de los péptidos y sus receptores usando técnicas de PCR semi-cuantitativa. Asimismo, las evidencias morfológicas obtenidas en los animales normales y los inducidos al desarrollo tumoral, se correlacionaron con índices de auto-fluorescencia obtenidos por microscopia de desconvolución digital; el cual fue determinado registrando la fluorescencia endógena proveniente de la superficie epitelial del colon. Además de dilucidar los posibles roles del sistema Endotelinas en un modelo murino de cáncer colo-rectal, con el presente proyecto se obtuvieron parámetros cuantitativos e indicadores, que podrían utilizarse para evaluar el proceso cancerígeno y como un posible método de diagnóstico temprano

    Dark matter search in a Beam-Dump eXperiment (BDX) at Jefferson Lab

    MeV-GeV dark matter (DM) is theoretically well motivated but remarkably unexplored. This Letter of Intent presents the MeV-GeV DM discovery potential for a 1 m3^3 segmented plastic scintillator detector placed downstream of the beam-dump at one of the high intensity JLab experimental Halls, receiving up to 1022^{22} electrons-on-target (EOT) in a one-year period. This experiment (Beam-Dump eXperiment or BDX) is sensitive to DM-nucleon elastic scattering at the level of a thousand counts per year, with very low threshold recoil energies (\sim1 MeV), and limited only by reducible cosmogenic backgrounds. Sensitivity to DM-electron elastic scattering and/or inelastic DM would be below 10 counts per year after requiring all electromagnetic showers in the detector to exceed a few-hundred MeV, which dramatically reduces or altogether eliminates all backgrounds. Detailed Monte Carlo simulations are in progress to finalize the detector design and experimental set up. An existing 0.036 m3^3 prototype based on the same technology will be used to validate simulations with background rate estimates, driving the necessary R&\&D towards an optimized detector. The final detector design and experimental set up will be presented in a full proposal to be submitted to the next JLab PAC. A fully realized experiment would be sensitive to large regions of DM parameter space, exceeding the discovery potential of existing and planned experiments by two orders of magnitude in the MeV-GeV DM mass range.Comment: 28 pages, 17 figures, submitted to JLab PAC 4

    The fine-scale structure of the trade wind cumuli over Barbados – An introduction to the CARRIBA project

    The CARRIBA (Cloud, Aerosol, Radiation and tuRbulence in the trade wInd regime over BArbados) project, focused on high resolution and collocated measurements of thermodynamic, turbulent, microphysical, and radiative properties of trade wind cumuli over Barbados, is introduced. The project is based on two one-month field campaigns in November 2010 (climatic wet season) and April 2011 (climatic dry season). Observations are based on helicopter-borne and ground-based measurements in an area of 100 km2 off the coast of Barbados. CARRIBA is accompanied by long-term observations at the Barbados Cloud Observatory located at the East coast of Barbados since early in 2010 and which provides a longer-term context for the CARRIBA measurements. The deployed instrumentation and sampling strategy are presented together with a classification of the meteorological conditions. The two campaigns were influenced by different air masses advected from the Caribbean area, the Atlantic Ocean, and the African continent which led to distinct aerosol conditions. Pristine conditions with low aerosol particle number concentrations of ∼100 cm3 were alternating with periods influenced by Saharan dust or aerosol from biomass burning resulting in comparably high number concentrations of ∼ 500 cm3. The biomass burning aerosol was originating from both the Caribbean area and Africa. The shallow cumulus clouds responded to the different aerosol conditions with a wide range of mean droplet sizes and number concentrations. Two days with different aerosol and cloud microphysical properties but almost identical meteorological conditions have been analyzed in detail. The differences in the droplet number concentration and droplet sizes appear not to show any significant change for turbulent cloud mixing, but the relative roles of droplet inertia and sedimentation in initiating coalescence, as well as the cloud reflectivity, do change substantially. © Author(s) 2013

    An approach to assess flooding and erosion risk for open beaches in a changing climate

    This paper examines the vulnerability to flooding and erosion of four open beach study sites in Europe. A framework for the quantitative estimation of present and future coastal flood and erosion risks is established using methods, data and tools from across a range of disciplines, including topographic and bathymetric data, climate data from observation, hindcast and model projections, statistical modelling of current and future climates and integrated risk analysis tools. Uncertainties in the estimation of future coastal system dynamics are considered, as are the consequences for the inland systems. Different implementations of the framework are applied to the study sites which have different wave, tidal and surge climate conditions. These sites are: Santander, Spain—the Atlantic Ocean; Bellocchio, Italy—the Adriatic Sea; Varna, Bulgaria—the Black Sea; and the Teign Estuary, UK—the northern Atlantic Ocean. The complexity of each system is first simplified by sub-division into coastal "impact units" defined by homogeneity in the local key forcing parameters: wave, wind, tide, river discharge, run-off, etc. This reduces the simulation to that of a number of simpler linear problems which are treated by applying the first two components of the Source–Pathway–Receptor–Consequence (S–P–R–C) approach. The case studies reveal the flexibility of this approach, which is found useful for the rapid assessment of the risks of flooding and erosion for a range of scenarios and the likely effectiveness of flood defences

    Interobserver agreement on definition of the target volume in stereotactic radiotherapy for pancreatic adenocarcinoma using different imaging modalities

    PURPOSE The aim of this study was to evaluate interobserver agreement (IOA) on target volume definition for pancreatic cancer (PACA) within the Radiosurgery and Stereotactic Radiotherapy Working Group of the German Society of Radiation Oncology (DEGRO) and to identify the influence of imaging modalities on the definition of the target volumes. METHODS Two cases of locally advanced PACA and one local recurrence were selected from a large SBRT database. Delineation was based on either a planning 4D CT with or without (w/wo) IV contrast, w/wo PET/CT, and w/wo diagnostic MRI. Novel compared to other studies, a combination of four metrics was used to integrate several aspects of target volume segmentation: the Dice coefficient (DSC), the Hausdorff distance (HD), the probabilistic distance (PBD), and the volumetric similarity (VS). RESULTS For all three GTVs, the median DSC was 0.75 (range 0.17-0.95), the median HD 15 (range 3.22-67.11) mm, the median PBD 0.33 (range 0.06-4.86), and the median VS was 0.88 (range 0.31-1). For ITVs and PTVs the results were similar. When comparing the imaging modalities for delineation, the best agreement for the GTV was achieved using PET/CT, and for the ITV and PTV using 4D PET/CT, in treatment position with abdominal compression. CONCLUSION Overall, there was good GTV agreement (DSC). Combined metrics appeared to allow a more valid detection of interobserver variation. For SBRT, either 4D PET/CT or 3D PET/CT in treatment position with abdominal compression leads to better agreement and should be considered as a very useful imaging modality for the definition of treatment volumes in pancreatic SBRT. Contouring does not appear to be the weakest link in the treatment planning chain of SBRT for PACA
