The aim of this study was to translate and adapt the Portuguese version of the Questionnaire about Interpersonal Difficulties for Adolescents (QIDA) using a sample of 369 students aged 12 to 18 years (51 % girls; M = 14.50; SD = 1.60). The exploratory factor analysis of main components and oblimin rotation, identified four related factors (Assertiveness, Relations with family and friends, Heterosexual relationships and Public speaking) that explained 43.45 % of the variance. In the confirmatory factor analysis (CFA), three predefined models were analyzed: the null model (M0), the one-factor model (M1) and the four-factor model (M4). CFA showed that M4 fit the data best, and internal consistency and test-retest reliability for the total score of the QIDA were appropriate. The correlation of the QIDA with the total score of the School Anxiety Questionnaire (SAI) was positive and with a moderate effect size. This result was similar for the three school anxiety response systems measured by the SAI and slightly more variable for the five situational subscales of the SAI. In addition, the correlation of the QIDA with the Social Anxiety Scale for Adolescents (SASA) was positive and low with the Apprehension and Fear of Negative Evaluation subscale (AFNE) and moderate with the Tension and Inhibition in Social Situations subscale (TISC). Finally, the correlation of the QIDA with the Internet Personality Inventory (IPI) was positive and moderate for the Neuroticism scale, and negative and low with the Extraversion scale

Targeting Alzheimer’s disease with multimodal polypeptide-based nanoconjugates

    Get PDF
Alzheimer’s disease (AD), the most prevalent form of dementia, remains incurable mainly due to our failings in the search for effective pharmacological strategies. Here, we describe the development of targeted multimodal polypeptide-based nanoconjugates as potential AD treatments. Treatment with polypeptide nanoconjugates bearing propargylamine moieties and bisdemethoxycurcumin or genistein afforded neuroprotection and displayed neurotrophic effects, as evidenced by an increase in dendritic density of pyramidal neurons in organotypic hippocampal culture. The additional conjugation of the Angiopep-2 targeting moiety enhanced nanoconjugate passage through the blood-brain barrier and modulated brain distribution with nanoconjugate accumulation in neurogenic areas, including the olfactory bulb. Nanoconjugate treatment effectively reduced neurotoxic amyloid aggregate levels and rescued impairments to olfactory memory and object recognition in APP/PS1 transgenic AD model mice. Overall, this study provides a description of a targeted multimodal polyglutamate-based nanoconjugate with neuroprotective and neurotrophic potential for AD treatment. Copyrigh

Tobacco and alcohol consumption: academic goals, school self-concept and academic performance in portuguese adolescents

    Get PDF
Introduction. Adolescence is a phase of the human life cycle including the use of drugs such as alcohol and tobacco. Objectives. The main objective of this study was to analyze whether alcohol and tobacco use is related to psychoeducational variables (e.g., school goals, academic self-concept, and school performance). Method. A random sampling of 1 197 students selected from nine schools in Portugal was used. Results. The results of the present study found that tobacco and alcohol users had significantly lower scores on psychoeducational variables compared to non-users. It should be noted that in verbal self-concept no differences were found between users and non-users in relation to both alcohol and tobacco. Conclusions. For all these reasons, it can be established that there is a relationship between tobacco and alcohol consumption and the psychoeducational variables examined in this study of Portuguese adolescents

Non-conservation of folding rates in the thioredoxin family reveals degradation of ancestral unassisted-folding

Evolution involves not only adaptation, but also the degradation of superfluous features. Many examples of degradation at the morphological level are known (vestigial organs, for instance). However, the impact of degradation on molecular evolution has been rarely addressed. Thioredoxins serve as general oxidoreductases in all cells. Here, we report extensive mutational analyses on the folding of modern and resurrected ancestral bacterial thioredoxins. Contrary to claims from recent literature, in vitro folding rates in the thioredoxin family are not evolutionarily conserved, but span at least a ∼100-fold range. Furthermore, modern thioredoxin folding is often substantially slower than ancestral thioredoxin folding. Unassisted folding, as probed in vitro, thus emerges as an ancestral vestigial feature that underwent degradation, plausibly upon the evolutionary emergence of efficient cellular folding assistance. More generally, our results provide evidence that degradation of ancestral features shapes, not only morphological evolution, but also the evolution of individual proteins. This research was supported by FEDER Funds, grant BIO2015-66426-R from the Spanish Ministry of Economy and Competitiveness ( J.M.S.-R.), grant RGP0041/2017 from the Human Frontier Science Program ( J.M.S.-R. and E.A.G.) and National Institutes of Health 1R01AR069137 (E.A.G.), Department of Defence MURI W911NF-16-1-0372 (E.A.G.)

Generation, establishment and characterization of a pluripotent stem cell line (CVTTHi001-A) from primary fibroblasts isolated from a patient with activated PI3 kinase delta syndrome (APDS2)

APDS2 is caused by mutations in PIK3R1 gene resulting in constitutive PI3Kδ activation. PI3Kδ is predominantly expressed in leukocytes and plays critical roles in regulating immune responses. Here we first derived fibroblast primary cells from a skin biopsy of a patient carrying a heterozygous single T deletion in intron 11 of the PIK3R1 gene. We next present the derivation of an induced pluripotent stem cell (iPS) line using a non-integrative reprogramming technology. Pluripotent-related hallmarks are further shown, including: iPSCs self-renewal and expression of pluripotent and differentiation markers after in vitro differentiation towards embryonic germ layers, assessed by RT-PCR and immunofluorescence

On the basis of the heterogeneous casuistry that characterizes the students who refuse going to school, it is useful to have a classification of this population in homogeneous groups. For this, the aim of this study was, first, to identify by cluster analysis the profiles of school refusal behavior based on the functional model evaluated through the School Refusal Assessment Scale-Revised (SRAS-S). Secondly, it is intended to analyze if there are differences in social functioning scores according to the school refusal profiles identified. This study involved 1212 Spanish children between 8 and 11 years old (M=9.12, SD=1.05) who completed the SRAS-R to evaluate the school refusal behavior and the Child and Adolescent Social Adaptive Functioning Scale (CASAFS) to assess social functioning. Four profiles were identified: Non-school refusers, School refusers by mixed reinforcements, School refusers by tangible reinforcements and School refusers by negative reinforcements. The profile of Non-school refusers achieved the highest average scores in social functioning, while School refusers by mixed reinforcements group obtained the lowest average scores in social functioning. In general, the profiles found support the clusters identified in previous studies. The implications of social functioning on school refusal behavior are discussed.
